Abstract
本发明公开了一种治疗风湿的苗药,本发明所述药物由半边荷,豆珍空,嘎炯豆丢劳独,莴强牛,豆顿、豆莴岗佳菲幼、独活制备而成。本本发明具有补肝肾,强筋骨,祛风除湿的功效,用于治疗寒湿腰痛所致腰部冷痛重着,转侧不利,逐渐加重,每遇阴雨天或腰部感寒后加剧,痛处喜温,得热则减,兼有手足不温,少气乏力,舌淡红,苔白腻而润,脉沉细紧或沉迟。适用于患有风湿的人群。其副作用比西药小。

发明领域:
本发明涉及一种治疗风湿的苗药及其制备方法,属于药品、尤其是苗药技术的领域。
背景技术
风湿病是一组侵犯关节、骨骼、肌肉、血管及有关软组织或结缔组织为主的疾病,其中多数为自身免疫性疾病。发病多较隐蔽而缓慢,病程较长,且大多具有遗传倾向。风湿性疾病是一类侵犯多种组织多系统和内脏器官的自身免疫性疾病。程度不同的免疫性炎症反应,可致成各种组织和器官损伤,严重影响其正常功能。甚至造成致命性损害。此外,大多数风湿性疾病都有关节症状;每个病人,同一种疾病,不同病程都有其特殊性。应该仔细评价,以制订出其个人的治疗计划。治疗目标应包括缓解症状,改善病情,恢复功能,提高生活质量,尽可能延续患者的生命。由于大部分风湿性疾病目前还不能根治,因此要争取病人的合作,长期坚持治疗。治疗的方法包括药物,理疗,休息及锻练,矫形及手术。要教育病人了解自己的病情,配合治疗。
治疗风湿性疾病,西医目前有使用非甾类抗炎药(NSAIDs)和肾上腺皮质激素药物治疗,非甾类抗炎药(NSAIDs)主要为解热,消炎和镇痛,而达到减轻炎症反应和目的,但对胃肠,肾,肝和血液系统副作用大。肾上腺皮质激素药物有抗炎和免疫抑制作用,有较强和快速的消除炎症及炎症反应带来的各种症状,但由于其并非根治药物,长期大量使用可诱发感染,骨质疏松,股骨头坏死,糖尿病,消化性溃疡,高血压,精神异常等;且如停药过快易产生病情反跳现象。
在风湿治疗方面苗医药具有明显突出的优势。苗医源远流长,发展至今,苗家医药已经有三、四千年的历史。苗族的医药常常与神秘、神奇这样的词汇联系在一起,自成体系,尤以其内病外治的疗法闻名中外,成为民族医药的一枝奇葩。苗族民间还有“千年苗医,万年苗药”之说。苗医认为一个人身体某部位、或心理或精神抵御不住过量的负荷,必然会导致气血、经络的运动不正常,产生疼痛难受的现象,而治病就是要采取种种手段使气血、经络疏通,恢复正常,达到疼痛消除,各个器官发挥正常功能的目的。而苗医所称的“两纲”即“冷病热治、热病冷治”,是他们治病的两大法则,具体治法则分内治法和外治法,其外治法尤为丰富,并体现了浓郁的民族特色和治疗特点。本申请采取外制药浴的方法,使用补肝肾,强筋骨,祛风除湿的药物,以治疗寒湿腰痛所致腰部冷痛重着,转侧不利,逐渐加重,每遇阴雨天或腰部感寒后加剧,痛处喜温,得热则减,兼有手足不温,少气乏力,舌淡红,苔白腻而润,脉沉细紧或沉迟。以治愈风湿性疾病,其副作用比西药小,安全性比西药大。
发明内容
本发明所要解决的技术问题在于提供一种治疗风湿的苗药及其制备方法。本发明具有补肝肾,强筋骨,祛风除湿的功效,用于治疗寒湿腰痛所致腰部冷痛重着,转侧不利,逐渐加重,每遇阴雨天或腰部感寒后加剧,痛处喜温,得热则减,兼有手足不温,少气乏力,舌淡红,苔白腻而润,脉沉细紧或沉迟。适用于患有风湿的人群。
为解决以上技术问题,本发明采用以下技术方案实现:
一种治疗风湿的苗药,其药用有效成分按照重量组分计算:由半边荷10-150份,豆珍空10-150份,嘎炯豆丢劳独10-150份,莴强牛10-150份,豆顿10-150份、豆莴岗佳菲幼10-150份、独活10-150份制备而成。
具体的说,所述的治疗风湿的苗药,其药用有效成分按照重量组分计算:半边荷30-100份,豆珍空30-100份,嘎炯豆丢劳独30-100份,莴强牛30-100份,豆顿30-100份、豆莴岗佳菲幼30-100份、独活30-100份制备而成。
更具体的说,所述的治疗风湿的苗药,其药用有效成分按照重量组分计算:半边荷50份,豆珍空50份,嘎炯豆丢劳独50份,莴强牛50份,豆顿50份、豆莴岗佳菲幼50份、独活50份制备而成。
前述治疗风湿的苗药的制备方法为:按照比例称取方中所有药材,并按照常规的制备方法进行加工,制成相应的药物制剂。
具体地说,前述治疗风湿的苗药的制备方法为:按照比例称取方中所有药材,将其粉碎后混合均匀后,加水煎煮后得煎煮液,并按照常规的制备方法进行加工,制成相应的药物制剂。
所述药物制剂包括水煎剂、洗剂。
所述水煎液这样制备:按照比例称取方中所有药材,将其粉碎后混合均匀后,打包备用,使用前取该药包加1000-2000ml水浸泡30-60分钟后,先大火煎煮20-40分钟,再文火煎煮1-2小时,水煎液倒入浴盆中加入适量冷水,泡浴30-60分钟即得。
所述洗剂这样制备:按照比例称取方中所有药材,将其粉碎后混合均匀后,加10倍量的水浸泡30-60分钟,再煎煮2-3次,每次1-2小时,合并两次水煎液,过滤后每500ml灌装装袋,密封即得。
与现有技术相比,本发明具有以下有益效果:
本发明所述治疗风湿的苗药由由半边荷、豆珍空、嘎炯豆丢劳独、莴强牛、豆顿、豆莴岗佳菲幼、独活制成制备而成。各成分性质及功效如下:半边荷又名半枫荷(学名:Semiliquidambar cathayensis Hung T.Chang)是金缕梅科;全年可采,切片晒干;味甘,性温;具有祛风湿、舒筋活血的功效;用于风湿性关节炎,类风湿关节炎,腰肌劳损,慢性腰腿痛,半身不遂,跌打损伤,扭挫伤;外用治刀上出血。豆珍空(又名透骨香)透骨香为杜鹃花科白珠树属植物滇白珠Gaultheria leucocarpa Bl.var.crenulata(Kurz)T.Z.Hsu的全株或根;植物滇白珠,分布于我国长江流域及其以南各省区;性热,味麻、辣,属热药,入冷经;具有清热解毒,活血化瘀之功效;主治风湿,胃寒疼痛,跌打损伤,咳嗽痰多。嘎炯豆丢劳独(又名八角枫)为八角枫科八角枫属植物华瓜木Alangium chinenese(Lour.)Harms[Stylidiumchinense Lour.]和瓜木Alangium Platanifolium(Sieb.Et Zucc.)Harms[Marleaplatanifolia Sieb.Et Zucc.]的根、须根或根皮;根全年可采,挖出后,除去泥沙,斩取侧根和须状根,晒干即得;夏、秋采叶及花,晒干备用或鲜用;性热,味麻、辣,属热药,入冷经,有大毒;具有祛风除湿,舒筋活络的功效。莴强牛(又称续断)为川续断科植物川续断Dipsacus asper Wall.Ex Henry的干燥根;秋季采挖,除去根头和须根,用微火烘至半干,堆置“发汗”至内部变绿色时,再烘干;性热,味甜,属热药,入冷经;具有补肝肾,强筋骨,壮腰,续折伤,止崩漏的功效;用于肝肾不足,腰膝酸软,风湿痹痛,跌扑损伤,筋伤骨折,崩漏,胎漏。豆顿(又称杜仲)为杜仲科植物杜仲Eucommia ulmoides Oliv.的干燥树皮;4~6月剥取,刮去粗皮,堆置“发汗”至内皮呈紫褐色,晒干;性热,味甜,属热药,入冷经;具有补肝肾,强筋骨的功效;用于肝肾不足,腰膝酸痛,筋骨无力,头晕目眩。豆莴岗佳菲幼(又名桑寄生)为桑寄生科植物桑寄生Taxillus chinensis(DC.)Danser的干燥带叶茎枝;冬季至次春采割,除去粗茎,切段,干燥,或蒸后干燥;性冷,味苦、微甜,属冷药,入热经;具有祛风湿,补肝肾,强筋骨,养血,安胎元的功效;用于风湿痹痛,腰膝酸软,筋骨无力,崩漏经多,妊娠漏血,胎动不安,头晕目眩。独活为伞形科植物重齿毛当归Angelica pubescensMaxim.f.biserrata Shan et Yuan的干燥根;春初苗刚发芽或秋末茎叶枯萎时釆挖,除去须根和泥沙,烘至半干,堆置2~3天,发软后再烘至全干;味辛、苦,性微温;归肾、膀胱经;具有祛风除湿,通痹止痛的功效;用于风寒湿痹,腰膝疼痛,少阴伏风头痛,风寒挟湿头痛。
方解:
母药:嘎炯豆丢劳独(八角枫),性热,味麻、辣,属热药,入冷经,有大毒,祛风除湿,舒筋活络;豆珍空(透骨香)性热,味麻、辣,属热药,入冷经,清热解毒,活血化瘀;独活,性辛、苦、微温,归肾、膀胱经,祛风除湿,通痹止痛;
子药:莴强牛(续断),性热,味甜,属热药,入冷经,补肝肾,壮筋骨,壮腰;豆顿(杜仲),性热,味甜,属热药,入冷经,补肝肾,强筋骨。豆莴岗佳菲幼(桑寄生),性冷,味苦、微甜,属冷药,入热经,补肝肾,强筋骨,养血;
本发明进行了大量的实验研究,以下为本发明实验研究的结果:一、急性毒性试验
1、实验动物:健康、皮肤完好的新西兰大白兔,体重2.0-3.0kg,由贵州医科大学实验动物中心提供,实验动物使用许可证:SYXK(黔)2018-001。环境温度:18-23℃,湿度:40%-60%,饲养条件:单笼饲养。
2、样品处理:将本发明所述药物实施例1按比例混合后,用适量温水调匀备用。
3、试验方法:试验动物检疫适应性喂养5天后,选用健康、成年、皮肤无损的新西兰大白兔5只。试验前24h,将试验动物背部脊柱两侧被毛除去,勿损伤表皮,去毛范围为每处6cm*6cm。分别取所述实施例1本发明药物1g涂抹于脊柱一侧去毛皮肤上,涂抹面积为5.5*5.5cm,另一侧作为对照,每天涂抹一次,连续涂抹14天。从第二次涂抹开始,每次涂抹前剪毛,并用温水清除残留药物,1h后观察结果,同时观察除外刺激性以外的其他毒性作用。
4、结果见表1。

二、长期毒性试验:
1、实验动物:SD大鼠,200±20g,雌雄各半,购于贵州医科大学实验动物中心,实验动物使用许可证:SYXK(黔)2018-001。饲养条件:动物分笼饲养,室温18-23℃,湿度:40%-60%,保持昼夜节律,自由进食、饮水。
2、试验方法:将本发明实施例1、实施例2、实施例3、实施例4、实施例5、实施例6制备的洗剂或水煎剂在动物体表进行3个月的长期毒性试验。将大鼠置于含上述本发明药物的容器内进行泡浴用药,每天一次,每次30min。剂量分别相当于所述用量1倍、2倍、5倍、10倍。观察项目包括一般性观察和血液学检查。结果表明,各实施例的四种给药剂量皆未出现明显不良反应,也无动物死亡,表明根据本发明实施例1、实施例2、实施例3、实施例4、实施例5、实施例6制备的洗剂或水煎剂用于临床使用是安全的。
三、对佐剂性类风湿关节炎大鼠的治疗作用
1、实验动物和材料:Wistar大鼠,SPF级,雄性,体重160~180g,由贵州医科大学实验动物中心提供,实验动物使用许可证:SYXK(黔)2018-001。大鼠适应性喂养7天,给予正常光照,自由进食进水。弗氏完全佐剂(CFA),购买于Sigma公司。托法替尼(MCE公司);ELISA试剂盒(南京建成);多功能酶标仪(美国BioTek)。
2、实验方法:
(1)建立佐剂关节炎(AIA)模型:取健康雄性Wistar大鼠60只,适应性喂养7天,随机分为空白对照组、模型组、阳性对照组(托法替尼,15mg/kg)、本发明实施例1低剂量组(75mg/kg)、本发明实施例1中剂量组(150mg/kg)、本发明实施例1高剂量组(300mg/kg)。75%酒精消毒后,于每只大鼠右后足趾皮下注射弗氏完全佐剂0.1mL致炎,空白对照组大鼠采用相同方法注射同体积的0.9%氯化钠溶液。约7天左右,大鼠右后足出血肉眼可见的红肿,活动受限,即为造模成功。
(2)分组给药:空白对照组和模型组给予蒸馏水灌胃;阳性对照组给药剂量为15mg/kg灌胃;本发明药物实施例1低、中、高剂量组分别进行泡浴,每次30min,隔一天给药1次,连续给药28天。
3、检测指标:
(1)大鼠一般状态观察:根据大鼠的生物学特点,重点观测和记录大鼠精神状态、活动情况、关节红肿情况、毛发光泽度、摄食量、体重、大小便情况等。
(2)肿胀度和肿胀抑制率测定:采用排水体积法测量致炎足(原发性)和对侧足(继发性)致炎前、后间隔一定时间的体积,计算肿胀度、肿胀抑制率。肿胀度=致炎后体积-致炎前体积;肿胀抑制率(%)=[(对照组肿胀度-给药组肿胀度)/对照组肿胀度]×100%。
(3)大鼠关节炎指数评分:全身病变按5级分法评价,大鼠致炎后观察并记录全身关节病变程度,每7天1次,根据未注射佐剂的余下肢体的病变程度累计积分,计算出关节炎指数(AI)。把各个关节的积分累计起来,即为每只大鼠的AI。

(4)大鼠炎症细胞因子水平测定:大鼠给药28天后,大鼠禁食不禁水12h,腹腔注射3.5%水合氯醛(10mL/kg)将大鼠麻醉,腹主动脉取血约5mL,室温静置2h后离心取上清液,分装保存于-20℃冰箱备用。按照ELAISA试剂盒说明书操作,测量各组大鼠血清致炎细胞因子IL-1β、IL-6、TNF-α和抗炎细胞因子IL-10的水平。
5、结果:
(1)各组大鼠一般情况:除空白对照组外,模型组和其余各组大鼠表现出精神倦怠、活动减少、食欲下降,背毛失去光泽、体重减轻等。造模侧和前爪足趾均有不同程度的红肿,受累关节畸形等,表明造模成功。上述现象在各给药组大鼠均有所改善。
(2)对大鼠肿胀度的影响:实验结果显示各组大鼠右踝关节肿胀度始终处于增高状态。与正常对照组比较,模型对照组大鼠右踝关节肿胀度7-21天均明显增高,差异具有统计学意义(P<0.05);与模型组比较,阳性对照组大鼠右踝关节肿胀度降低且差异具有统计学意义(P<0.05);与模型对照组比较,本发明实施例1中、高剂量组对肿胀出现明显的抑制作用(P<0.05);与模型对照组比较,本发明实施例1低剂量组大鼠右踝关节肿胀度降低,但无统计学意义(P>0.05)。结果见表3、表4。

(3)对佐剂性关节炎大鼠关节炎指数的影响:结果显示,本发明药物实施例1能够明显改善大鼠关节炎指数。与模型对照组比较,阳性对照组和本发明药实施例1低、中、高剂量组大鼠关节炎指数显著降低,差异具有统计学意义。结果见表5。

(4)对大鼠血清炎性因子水平的影响:如表6结果所示,与空白对照组比较,模型组大鼠血清IL-1β、IL-6、TNF-α水平显著升高(P<0.05),IL-10水平显著降低(P<0.05);与模型组比较,阳性对照组和本发明药物实施例1低、中、高剂量组IL-1β、IL-6、TNF-α水平显著下降(P<0.05),IL-10水平显著上升(P<0.05);

6、总结:本发明以Wistar大鼠佐剂性关节炎动物模型试验测试药物的治疗效果,是比较经济、经典的动物模型,具有可参照性。用本发明中药组合物对佐剂性关节炎大鼠进行干预,结果显示其能减轻足趾关节肿胀和关节炎指数,改善大鼠的一般状况,并通过降低血清IL-1β、IL-6、TNF-α的表达水平,促进抗炎因子IL-10水平,从而阻滞风湿疾病的发生与发展,改善大鼠的全身状况。综上所述,本发明药物组合可有效治疗风湿疾病,延缓关节炎的进展,提示本发明药物所含半边荷、豆珍空、嘎炯豆丢劳独、莴强牛、豆顿、豆莴岗佳菲幼、独活成分可有效治疗风湿疾病。
与现有技术相比,本发明选用由半边荷,豆珍空,嘎炯豆丢劳独,莴强牛,豆顿、豆莴岗佳菲幼、独活组方,具有补肝肾,强筋骨,祛风除湿的功效,用于治疗风痰瘀血、痹阻脉络所致的半身不遂,口舌歪斜,舌强言蹇或不语,偏身麻木,头晕目眩、舌质暗淡,舌苔薄白或白腻,脉弦滑。适应于患有风湿的人群。
具体实施方式
下面结合实施例对本发明作进一步的说明,但并不作为对本发明限制的依据。
实施例1。
处方:半边荷10g,豆珍空10g,嘎炯豆丢劳独10g,莴强牛10g,豆顿10g、豆莴岗佳菲幼10g、独活10g制成。
工艺:按照比例称取方中所有药材,将其粉碎后混合均匀后,打包备用,使用前取该药包加1000ml水浸泡30分钟后,先大火煎煮20分钟,再文火煎煮1小时,水煎液倒入浴盆中加入适量冷水,泡浴30分钟即得水煎液。
用法用量:每7天一次泡浴,一次30分钟,早期患者连续泡浴3次可见疗效。
实施例2。
处方:半边荷150g,豆珍空150g,嘎炯豆丢劳独150g,莴强牛150g,豆顿150g、豆莴岗佳菲幼150g、独活150g制成。
工艺:按照比例称取方中所有药材,将其粉碎后混合均匀后,加10倍量的水浸泡30分钟,再煎煮2次,每次1小时,合并两次水煎液,过滤后每500ml灌装装袋,密封即得洗剂。
用法用量:每次取两袋洗剂,加适量温水泡浴30分钟,每7天一次,早期患者连续泡浴3次可见疗效。
实施例3。
处方:半边荷50g,豆珍空50g,嘎炯豆丢劳独50g,莴强牛50g,豆顿50g、豆莴岗佳菲幼50g、独活50g制成。
工艺:按照比例称取方中所有药材,将其粉碎后混合均匀后,打包备用,使用前取该药包加2000ml水浸泡60分钟后,先大火煎煮40分钟,再文火煎煮2小时,水煎液倒入浴盆中加入适量冷水,泡浴60分钟即得水煎液。
用法用量:每7天一次泡浴,一次30分钟,早期患者连续泡浴3次可见疗效。
实施例4。
半边荷10-150g,豆珍空10-150g,嘎炯豆丢劳独10-150g,莴强牛10-150g,豆顿10-150g、豆莴岗佳菲幼10-150g、独活10-150g制成。
工艺:按照比例称取方中所有药材,将其粉碎后混合均匀后,加8倍量的水浸泡60分钟,再煎煮3次,每次2小时,合并两次水煎液,过滤后每500ml灌装装袋,密封即得洗剂。
用法用量:每次取两袋洗剂,加适量温水泡浴30分钟,每7天一次,早期患者连续泡浴3次可见疗效。
实施例5。
半边荷10-150g,豆珍空10-150g,嘎炯豆丢劳独10-150g,莴强牛10-150g,豆顿10-150g、豆莴岗佳菲幼10-150g、独活10-150g制成。
工艺:按照比例称取方中所有药材,将其粉碎后混合均匀后,打包备用,使用前取该药包加1500ml水浸泡45分钟后,先大火煎煮30分钟,再文火煎煮1.5小时,水煎液倒入浴盆中加入适量冷水,泡浴45分钟即得水煎液。

工艺:按照比例称取方中所有药材,将其粉碎后混合均匀后,加12倍量的水浸泡45分钟,再煎煮3次,每次1.5小时,合并两次水煎液,过滤后每500ml灌装装袋,密封即得洗剂。
用法用量:每次取两袋洗剂,加适量温水泡浴30分钟,每7天一次,早期患者连续泡浴3次可见疗效。

1.一种治疗风湿的苗药,其特征在于:其药用有效成分按照重量组分计算:由半边荷10-150份,豆珍空10-150份,嘎炯豆丢劳独10-150份,莴强牛10-150份,豆顿10-150份、豆莴岗佳菲幼10-150份、独活10-150份制成。
2.如权利要求1所述的治疗风湿的苗药,其特征在于:其药用有效成分按照重量组分计算:由半边荷30-100份,豆珍空30-100份,嘎炯豆丢劳独30-100份,莴强牛30-100份,豆顿30-100份、豆莴岗佳菲幼30-100份、独活30-100份制成。
3.如权利要求1或2所述的治疗风湿的苗药,其特征在于:其药用有效成分按照重量组分计算:由半边荷50份,豆珍空50份,嘎炯豆丢劳独50份,莴强牛50份,豆顿50份、豆莴岗佳菲幼50份、独活50份制成。
4.如权利要求1-3中所述治疗风湿的苗药的制备方法,其特征在于:按照比例称取方中所有药材,并按照常规的制备方法进行加工,制成相应的药物制剂。
5.如权利要求4所述治疗风湿的苗药的制备方法,其特征在于:按照比例称取方中所有药材,将其粉碎后混合均匀后,加水煎煮后得煎煮液,并按照常规的制备方法进行加工,制成相应的药物制剂。
6.如权利要求5所述治疗风湿的苗药的制备方法,其特征在于:所述药物制剂包括水煎液、洗剂。
7.如权利要求6所述治疗风湿的苗药的制备方法,其特征在于:所述水煎液这样制备:按照比例称取方中所有药材,将其粉碎后混合均匀后,打包备用,使用前取该药包加1000-2000ml水浸泡30-60分钟后,先大火煎煮20-40分钟,再文火煎煮1-2小时,水煎液倒入浴盆中加入适量冷水,泡浴30-60分钟即得。
8.如权利要求6所述治疗风湿的苗药的制备方法,其特征在于:所述洗剂这样制备:按照比例称取方中所有药材,将其粉碎后混合均匀后,加10倍量的水浸泡30-60分钟,再煎煮2-3次,每次1-2小时,合并两次水煎液,过滤后每500ml灌装装袋,密封即得。
