Abstract

发明构思的另一个实施方案是大麻素受体激动剂在针对病毒的改善测定中的用途。此类病毒测定法可以基于病毒检测、病毒遗传物质检测(例如通过PCR、RT-PCR和其他依赖特定核酸复制的方法)、病毒抗原检测(例如,通过免疫测定、Western印迹、质谱法等),和/或观察对培养细胞的细胞病变效应。在此类方法中,可在存在导致培养细胞中脂质积累的无毒性量的大麻素受体激动剂(例如(R)-methanandamide)的情况下将获得用于分析的样品(或此类样品的含有病毒的级分)引入细胞培养物中。增强的病毒复制导致可检测的病毒标志物(遗传和/或抗原性)的产生增加,从而与不存在大麻素受体激动剂的情况相比,允许更早和/或更灵敏的检测,并且还导致可直接观察到的细胞病变效应增加。
此类细胞培养步骤不需要进行到细胞裂解或可观察到的细胞病变效应的点,而是可以进行足以导致细胞内病毒或可鉴定病毒组分复制的时间段。在采用此类缩短的温育时段的方法中,可以将培养中的细胞裂解(例如,通过研磨、超声处理、添加表面活性剂、渗透性休克等)以释放病毒和/或病毒产物用于随后的检测。在一些实施方案中,快速诊断方法(例如,实时定量PCR、流式免疫测定、即时检验等)可用于此类基于细胞的测定或方法,以提供具有改善的灵敏度和有利地具有改善的特异性(根据病毒与培养物之间的特异性相互作用而定)的快速诊断方法。

Claims (39)
1.用于改善对疫苗接种的免疫反应的方法,其包括:
在疫苗接种时或紧接疫苗接种之前向受试者提供包含米替福新(miltefosine)的佐剂;和
在疫苗接种部位处引入疫苗,所述疫苗包含疫苗接种物质,其中选择所述佐剂以提供足以增强所述免疫反应的量的米替福新。
2.权利要求1的方法,其中所述免疫反应选自下组:IgG滴度、IgM滴度、血凝抑制滴度和微量中和滴度。
3.权利要求1或2的方法,其中所述佐剂作为局部配制剂局部应用于所述疫苗接种部位上,并且进一步包括将屏障应用于所述疫苗接种部位上的步骤。
4.权利要求3的方法,其中所述局部配制剂包含应用装置的一部分。
5.权利要求1至4中任一项的方法,其中所述佐剂提供1μg至100mg的米替福新。
6.权利要求1至5中任一项的方法,其中所述疫苗包含三价流感疫苗、多价流感疫苗或冠状病毒疫苗。
7.米替福新在制备提供针对疫苗的增强反应的佐剂中的用途。
8.权利要求7的用途,其中所述佐剂配制为提供1μg和100mg之间的米替福新。
9.权利要求7或8的用途,其中所述佐剂作为局部配制剂或器具的一部分供应。
10.权利要求7至9中任一项的用途,其中所述增强的反应选自下组:相对于在尚未用所述佐剂治疗的经疫苗接种的受试者中实现的疫苗接种反应,增加的IgG滴度、增加的IgM滴度、增加的血凝抑制滴度和增加的微量中和滴度。
11.权利要求7至10中任一项的用途,其中所述疫苗包含三价流感疫苗、多价流感疫苗或冠状病毒疫苗。
12.用于增强针对皮内注射的疫苗的免疫反应的佐剂,其包含米替福新。
13.权利要求12的佐剂,其中所述免疫反应选自下组:IgG滴度、IgM滴度、血凝抑制滴度和微量中和滴度。
14.权利要求12或13的佐剂,其中所述佐剂提供1μg至100mg的米替福新。
15.权利要求12至14中任一项的佐剂,其中所述佐剂配制为局部配制剂。
16.权利要求15的佐剂,其中所述局部配制剂包含应用装置的一部分。
17.权利要求12至16中任一项的佐剂,其中所述疫苗包含三价流感疫苗、多价流感疫苗或冠状病毒疫苗。
18.用于提供治疗性或保护性免疫力的疫苗配制剂,其包含免疫原性疫苗接种物质和米替福新。
19.权利要求18的疫苗配制剂,其包含1μg至100mg的米替福新。
20.权利要求18或19的疫苗配制剂,其中所述免疫原性疫苗接种物质包含三价流感疫苗、多价流感疫苗或冠状病毒疫苗。
21.用于增加来自细胞培养物的病毒的产率的方法,其包括:
使培养中的细胞与来自所述病毒的遗传物质接触;和
使所述细胞与无毒浓度下的大麻素受体激动剂接触,其中选择所述大麻素受体激动剂以增加所述细胞的脂质含量。
22.权利要求21的方法,其中所述大麻素受体激动剂是花生四烯酸乙醇胺或花生四烯酸乙醇胺类似物。
23.权利要求22的方法,其中所述花生四烯酸乙醇胺类似物是(R)-methanandamide。
24.权利要求21至23的方法,其中所述病毒或其一部分适合用作免疫原。
25.权利要求21至24中任一项的方法,其中所述病毒是冠状病毒或流感病毒。
25.生产疫苗配制剂的方法,其包括:
使培养中的细胞与来自病毒的遗传物质接触;
使所述细胞与无毒浓度下的大麻素受体激动剂接触,其中选择所述大麻素受体激动剂以增加所述细胞的脂质含量;
在足以使所述细胞中的所述病毒复制的时间段后,从培养物中收集所述病毒;和
在药学上可接受的载体中提供所述病毒或所述病毒的一部分。
26.权利要求25的方法,其中所述大麻素受体激动剂是花生四烯酸乙醇胺或花生四烯酸乙醇胺类似物。
27.权利要求26的方法,其中所述花生四烯酸乙醇胺类似物是(R)-methanandamide。
28.权利要求25至27的方法,其中所述病毒或其一部分适合用作免疫原。
29.权利要求25至28中任一项的方法,其中所述病毒是冠状病毒或流感病毒。

31.用于检测病毒的方法,其包括:
获取疑似含有所述病毒的样品;
使培养中的细胞与所述样品的至少一部分或其级分接触;
使所述细胞与无毒浓度下的大麻素受体激动剂接触,其中选择所述大麻素受体激动剂以增加所述细胞的脂质含量;
在足以使所述细胞中的所述病毒复制的时间段后,从培养物中收集测试样品;和
确定所述测试样品中所述病毒特征性的分子的存在。
32.权利要求30的方法,其中确定所述病毒特征性的分子的存在的步骤包括核酸扩增步骤。
33.权利要求30或31的方法,其中确定所述病毒特征性的分子的存在的步骤包括核酸杂交步骤。
34.权利要求30至32中任一项的方法,其中确定所述病毒特征性的分子的存在的步骤包括免疫测定。
35.权利要求29至32中任一项的方法,其中所述病毒是冠状病毒或流感病毒。
36.权利要求30至34中任一项的方法,其中所述足以使所述细胞中的所述病毒复制的时间段在观察到细胞病变效应之前。
37.权利要求30至35中任一项的方法,其中使用快速或即时检验来进行确定所述病毒特征性的分子的存在。
38.权利要求30至36中任一项的方法,其中所述大麻素受体激动剂是花生四烯酸乙醇胺或花生四烯酸乙醇胺类似物。
