Abstract
本发明公开了一种过氧乙酸消毒液及其制备方法,属于消毒剂制备技术领域。其技术方案为:包括A剂和B剂,具体如下,A剂包括:过氧化氢1‑7%,稳定剂0.1‑10%,螯合剂0.1‑10%,缓蚀剂0.1‑5%,增溶剂5‑30%、溶剂10‑60%,B剂包括:酰化剂25‑40%、pH调节剂1‑10%、溶剂50‑70%;本发明制备的过氧乙酸溶液性质稳定,方便运输,制备方法简单,反应速度快,泡沫量低,稳定性好,腐蚀性低,可用于医疗器械的高水平消毒和灭菌。

技术领域
本发明涉及消毒剂制备技术领域,具体涉及一种过氧乙酸消毒液及其制备方法。
背景技术
过氧乙酸消毒液属于过氧化物类消毒剂,是目前国际公认的高水平消毒剂和灭菌剂。过氧乙酸消毒液可杀灭肠道致病菌、化脓性球菌、分枝杆菌、绿脓杆菌、致病性酵母菌及细菌芽孢,并能灭活病毒。过氧乙酸消毒液具有高效快速、绿色环保、分解后无残留等的优点,在医疗、食品等领域发挥着越来越重要的作用,应用前景非常广泛。
目前市售的过氧乙酸消毒液主要有一元过氧乙酸消毒液和二元过氧乙酸消毒液。《一种高浓度高稳定性一元过氧乙酸消毒剂及其制备方法》(CN201410542587)公开了一种高浓度高稳定性一元过氧乙酸消毒剂及其制备方法,按照该方法制备的过氧乙酸浓度高,有效成分达到25%(w/w)以上,用于饮料无菌冷灌线的消毒。该方法制备的过氧乙酸浓度较高,在运输过程中存在安全隐患,《过氧乙酸消毒液及其制备方法》(CN201310667274)公开了一种二元过氧乙酸消毒液及其制备方法,该方法改善了传统过氧乙酸不稳定易分解的问题,同时有效的降低了过氧乙酸自身对金属的腐蚀性。但是由于方法所选用的A剂包含过氧乙酸(5%-7%)、过氧化氢(7%-9%),按照《危险化学品名录》(2015版)的管理要求,A剂为危险化学品,在运输和储存上需要额外的解决措施,增加了运输成本。《过氧乙酸消毒剂及其应用》(CN201811603771)公开了一种过氧乙酸消毒剂及其应用,该发明的过氧乙酸消毒剂具有稳定性高、反应速率快等优点。但是该发明的A剂包括30%的过氧化氢,按照《危险化学品名录》(2015版)的管理要求,A剂为危险化学品,同样需要增加成本解决运输和储存问题。《一种过氧乙酸液体发生剂及其制备方法》(CN202011572711)公开了一种过氧乙酸液体发生剂及其制备方法,该方法制备的过氧乙酸溶液具有pH值接近中性、腐蚀性低、气味小、稳定性好的特点。但是该发明所选用的助溶剂为十二烷基苯磺酸钠、十二烷基磺酸钠、十二烷基硫酸钠等。这些助溶剂泡沫量大,如果制备的过氧乙酸溶液用于全自动内镜清洗消毒机,存在不易漂洗的问题,极易造成医院感染事件的发生。
发明内容
本发明要解决的技术问题是:克服现有技术的不足,提供一种过氧乙酸消毒液及其制备方法,制备的过氧乙酸溶液性质稳定,方便运输,反应速度快,泡沫量低,稳定性好,腐蚀性低,可用于医疗器械的高水平消毒和灭菌。
本发明的技术方案为:
第一方面,公开了一种过氧乙酸消毒液,包括A剂和B剂,按照重量百分含量具体如下,具体如下,A剂包括:过氧化氢1-7%,稳定剂0.1-10%,螯合剂0.1-10%,缓蚀剂0.1-5%,增溶剂5-30%,溶剂10-60%;B剂包括:酰化剂25-40%、pH调节剂1-10%、溶剂50-70%。
优选地,所述的稳定剂为有机膦酸,包括氨基三甲叉膦酸、乙二胺四甲叉膦酸、羟基乙叉二膦酸、二乙烯三胺五甲叉膦酸和2-羟基磷酸基乙酸中的一种或多种。
优选地,所述的螯合剂为氨基三乙酸、乙二胺四乙酸和酒石酸的一种或多种。
优选地,所述的缓蚀剂为葡萄糖酸钠、氨基三甲叉膦酸四钠、乙二胺四甲叉膦酸五钠、二乙烯三胺五甲叉膦酸和苯并三氮唑中的一种或多种。
优选地,所述的增溶剂为N-乙基己基-β-亚氨基二丙氨酸钠Ampholak YJH-40(阿克苏诺贝尔公司生产)和磷酸聚醚脂钾盐H-66(美国陶氏化学公司生产)中的一种或两种。
优选地,所述A剂和B剂中的中的溶剂均为乙醇、丙二醇、丙三醇、异丙醇、聚丙二醇-200和聚醚50-HB-660中的一种或多种。
优选地,所述的酰化剂为N-乙酰戊内酰胺、N-乙酰己内酰胺、N-乙酰庚内酰胺和N-乙酰辛内酰胺中的一种或多种。
优选地,所述的pH调节剂为三乙醇胺、二甲基乙醇胺、二异丙基乙胺、2-氨基-2-甲基-1-丙醇、N,N,N-三乙胺和N,N,N-二异丙基乙胺中的一种或多种。
第二方面,公开了过氧乙酸消毒液的制备方法,包括将所述的含有酰化剂的B剂加入到含过氧化氢的A剂中,得到质量浓度为0.21%-0.39%的过氧乙酸消毒液。
优选地,B剂和A剂的体积比为:1:30-1:100,常温条件下混合5-15分钟。
本发明与现有技术相比,具有以下有益效果:
1.本发明制备的过氧乙酸溶液性质稳定,方便运输,制备方法简单,反应速度快,泡沫量低,稳定性好,腐蚀性低,可用于医疗器械的高水平消毒和灭菌;2.本发明制备过氧乙酸的液体发生剂,由于增溶剂的加入,使酰化剂在水中的表面张力降低,导致其快速溶解,从而加快了过氧乙酸的制取速度;3.用低泡的增溶剂N-乙基己基-β-亚氨基二丙氨酸钠Ampholak YJH-40或磷酸聚醚脂钾盐H-66代替十二烷基苯磺酸钠、十二烷基磺酸钠、十二烷基硫酸钠等阴离子表面活性剂,增加了溶解性,降低了整个体系的泡沫量,一方面提高了过氧乙酸的制取速度,另一方面可以使该过氧乙酸用于全自动内镜清洗消毒机,解决不易漂洗的问题;4.制备的过氧乙酸稳定性强,1年有效期加速试验合格,过氧乙酸下降率≤10%。

具体实施方式
实施例1
一种过氧乙酸消毒液,包括A剂和B剂,具体如下,A剂:
过氧化氢1918.8mL,羟基乙叉二膦酸246mL,氨基三乙酸123g,乙二胺四甲叉膦酸五钠75.2g,苯并三氮唑97.0g,磷酸聚醚脂钾盐H-66 1230mL,丙二醇710mL和聚丙二醇-200520mL;B剂:N-乙酰己内酰胺25.58mL,N-乙酰辛内酰胺6.40mL,2-氨基-2-甲基-1-丙醇2.0mL,N,N,N-三乙胺2.1mL,丙二醇25.72mL、异丙醇5.1mL和聚醚50-HB-660 15.1mL,分别按以上比例配制A剂和B剂,将B剂加入到A剂中,B剂和A剂的体积比为:1:60,常温条件下混合时间5分钟,即可得质量浓度为0.3%的过氧乙酸消毒液。
实施例2
过氧化氢1918.8mL,羟基乙叉二膦酸246mL,氨基三乙酸123g,乙二胺四甲叉膦酸五钠75.2g,苯并三氮唑97.0g,N-乙基己基-β-亚氨基二丙氨酸钠1230mL,丙二醇710mL和聚丙二醇-200 520mL;N-乙酰己内酰胺25.58mL,N-乙酰辛内酰胺6.40mL,2-氨基-2-甲基-1-丙醇2.0mL,N,N,N-三乙胺2.1mL,丙二醇25.72mL、异丙醇5.1mL和聚醚50-HB-660 15.1mL,分别按以上比例配制A剂和B剂,将B剂加入到A剂中,B剂和A剂的体积比为:1:100,常温条件下混合时间5分钟,即可得质量浓度为0.3%的过氧乙酸消毒液。
对比例1
对比例1为一种过氧乙酸液体发生剂,A剂和B剂分别采用申请号为202011572711.8的专利申请中的实施例1制备的A剂和B剂。
实施例3
实施例3为实施例1和对比例1制备的过氧乙酸消毒液生成速度对比,具体实验方法如下:
将实施例1和对比例1的A剂和B剂分别按照活化时间1min、3min、5min、7min、9min、11min、13min、15min进行活化。活化方法为将B剂导入A剂中,上下翻转数次,静置一定时间。活化后的过氧乙酸溶液立即按照下述方法进行过氧乙酸含量的测定。过氧乙酸含量的测定的检测依据:《消毒技术规范》(2002年版)第2.2.1.2.3项。
试验方法:精密称取适量样品的A液置于容量瓶中用蒸馏水稀释至刻度。精密吸取适量稀释液置于碘量瓶中,加2mol/L硫酸5mL,100g/L硫酸锰3滴,摇匀并用高锰酸钾滴定液滴定至溶液呈粉红色。随即加100g/L碘化钾溶液10mL与30g/L钼酸铵3滴,摇匀并用硫代硫酸钠滴定液滴定至淡黄色。加入5g/L淀粉溶液3滴(溶液立即变为蓝色),继续用硫代硫酸钠滴定液滴定至蓝色消失,记录硫代硫酸钠滴定液的总用量,每份重复测定2次。
实施例1和对比例1的过氧乙酸生产速度对比图如图1所示,从图中可以看出,实施例1过氧乙酸A剂和B剂活化5min,过氧乙酸浓度达到0.3%。而对比例1活化11min时,过氧乙酸浓度达到0.3%左右。实施例1与对比例1相比,反应速度更快,活化时间短。
实施例4
实施例4为实施例1和对比例1制备的过氧乙酸消毒液的泡沫量对比,具体操作如下:
分别将实施例1和对比例1的A剂和B剂按照实施例3的活化方法活化,用SITA泡沫量测试仪分别测试活化好的实施例1和对比例1过氧乙酸消毒液的泡沫量,具体结果如图2所示,
从图2中看出,实施例1的最大泡沫量体积为82mL,而对比例1的最大泡沫量体积为267mL。实施例1的过氧乙酸泡沫量远低于对比例1,在用于全自动内镜清洗消毒机时,实施例1更便于漂洗。
实施例5
实施例5为实施例1和对比例1制备的过氧乙酸消毒液的稳定性对比,具体操作如下:
1)保存条件:将完全活化好的实施例1和对比例1过氧乙酸消毒液分别置于54℃隔水式恒温培养箱中保存14d。按照下述实验方法分别测试初始浓度、第2天、4天、6天、8天、10天、12天、14天的过氧乙酸含量。
2)试验方法:精密称取保存后的过氧乙酸样品适量置于容量瓶中用蒸馏水稀释至刻度。精密吸取适量稀释液置于碘量瓶中,加2mol/L硫酸5mL,100g/L硫酸锰3滴,摇匀并用高锰酸钾滴定液滴定至溶液呈粉红色。随即加100g/L碘化钾溶液10mL与30g/L钼酸铵3滴,摇匀并用硫代硫酸钠滴定液滴定至淡黄色。加入5g/L淀粉溶液3滴(溶液立即变为蓝色),继续用硫代硫酸钠滴定至蓝色消失,记录硫代硫酸钠滴定液的总用量,每份重复测定2次。
实施例1和对比例1过氧乙酸的稳定性对比结果见图3,从图3中可以看出,54℃14天加速实验中,实施例1的过氧乙酸含量下降率为9.6%,对比例1的过氧乙酸含量下降率为16.3%,实施例1的过氧乙酸稳定性优于对比例1。
实施例6
实施例6为实施例1和对比例1制备的过氧乙酸消毒液的腐蚀性对比,具体操作如下:
1)检测依据:《消毒技术规范》(2002年版)第2.2.4项。
2)试验方法:将金属片在有表面活性作用的清洁剂中浸泡10min充分去油,洗净。用砂纸磨去金属片表面的氧化层、自来水冲净。用无水乙醇脱脂、置50℃电热恒温干燥箱中干燥1h、称重。将金属片用塑料线系以标签、编号并注明日期,分别悬挂浸泡于盛有实施例1和对比例1样品的广口试剂瓶中,浸泡72h。浸泡至规定时间后,取出金属片,自来水冲洗后,用毛刷去除腐蚀产物并清洗。粗滤纸吸干水分后,置50℃电热恒温干燥箱中干燥1h,称重。试验同时设蒸馏水浸泡不锈钢的空白对照。每600mL样品的试验用液浸泡同一种金属片3片,每片相隔1cm以上。每一张砂纸只磨一种金属材料,同时观察金属样片的外观变化。
实施例1和对比例1过氧乙酸的腐蚀性对比见表1,从表1中实施例1的过氧乙酸对于不锈钢为基本无腐蚀,对于碳钢为基本无腐蚀,对于铜为轻度腐蚀,对于铝为轻度腐蚀。对比例1的过氧乙酸对于不锈钢为基本无腐蚀,对于碳钢为重度腐蚀,对于铜为重度腐蚀,对于铝为中度腐蚀。对于金属的腐蚀性,实施例1均明显优于对比例1。

实施例7
实施例7为实施例1制备的过氧乙酸消毒液的杀菌效果内镜机洗消毒模拟现场试验应用例,具体操作如下:
1)检测依据:GB/T 38497-2020《内镜消毒效果评价方法》6.2。
2)染菌载体制备:取枯草杆菌黑色变种芽孢悬液滴染于聚四氟乙烯管载体内壁,干燥,制成染菌载体备用。将染菌载体分别连接在模拟内镜的50mm、1000mm和1950mm处。
3)试验方法:试验前将实施例1过氧乙酸消毒液活化完全,加入内镜清洗消毒机内并与之连接。将制备好的染菌模拟内窥镜装放于清洗消毒机规定位置。启动机器清洗消毒程序[清洗(酶洗)5min-漂洗30s-漂洗30s-消毒5.0min-末漂洗30s-末漂洗30s-吹干30s],结束后无菌操作取下染菌载体,分别移入含10mL中和剂的试管内,作为试验组。另取2个染菌载体,放置室温环境中,不做消毒处理,待试验组作用时间结束后,按试验组同法处理,作为阳性对照组。同时设置中和剂、稀释液以及未用过的同批次培养基为阴性对照组。将各组接种培养基,置37℃培养箱培养72h,菌落计数。试验重复3次。
经3次重复试验,应用实施例1过氧乙酸消毒液,对模拟内镜进行一次清洗消毒程序[清洗(酶洗)5min-漂洗30s-漂洗30s-消毒5.0min-末漂洗30s-末漂洗30s-吹干30s],对载体上的枯草杆菌黑色变种芽孢的杀灭对数值均>3.00(见表2)。符合GB/T 38497-2020《内镜消毒效果评价方法》的规定。

实施例8
实施例8为实施例1制备的过氧乙酸消毒液的杀菌效果内镜机洗灭菌模拟现场试验应用例,具体操作如下:
1)检测依据:《消毒技术规范》(2002年版)第2.1.1.7.5(2)项和GB/T 38497-2020《内镜消毒效果评价方法》6.2。
2)染菌载体制备:取枯草杆菌黑色变种芽孢悬液滴染于聚四氟乙烯管载体内壁,干燥,制成染菌载体备用。将染菌载体分别连接在模拟内镜的50mm、1000mm和1950mm处。
3)试验方法:试验前将实施例1过氧乙酸消毒液活化完全,加入内镜清洗消毒机内并与之连接。将制备好的染菌模拟内窥镜装放于清洗消毒机规定位置。启动机器清洗消毒程序[清洗(酶洗)5min-漂洗30s-漂洗30s-消毒5.0min-末漂洗30s-末漂洗30s-吹干30s],结束后无菌操作取下染菌载体,分别移入含10mL中和剂的TSB试管内,做为试验组样本。将3个染菌载体移入含10mL中和剂的TSB试管内,作为阳性对照。同时设置中和剂、稀释液以及未用过的同批次培养基为阴性对照。将各组接种培养基,置37℃培养箱培养7d。另取2个染菌载体进行活菌培养计数,作为菌数对照。试验重复5次。
经5次重复试验,应用实施例1过氧乙酸消毒液,对模拟内镜进行一次清洗消毒程序[清洗(酶洗)5min-漂洗30s-漂洗30s-消毒5.0min-末漂洗30s-末漂洗30s-吹干30s],各次试验组3个不同位置上的载体均无菌生长(见表3)。符合《消毒技术规范》(2002年版)灭菌合格的规定。

本发明制备的过氧乙酸溶液性质稳定,方便运输,制备方法简单,反应速度快,泡沫量低,稳定性好,腐蚀性低,可用于医疗器械的高水平消毒和灭菌;本发明制备过氧乙酸的液体发生剂,由于增溶剂的加入,使酰化剂在水中的表面张力降低,导致其快速溶解,从而加快了过氧乙酸的制取速度;用低泡的增溶剂N-乙基己基-β-亚氨基二丙氨酸钠Ampholak YJH-40或磷酸聚醚脂钾盐H-66代替十二烷基苯磺酸钠、十二烷基磺酸钠、十二烷基硫酸钠等阴离子表面活性剂,增加了溶解性,降低了整个体系的泡沫量,一方面提高了过氧乙酸的制取速度,另一方面可以使该过氧乙酸用于全自动内镜清洗消毒机,解决不易漂洗的问题;制备的过氧乙酸稳定性强,1年有效期加速试验合格,过氧乙酸下降率≤10%。
尽管通过参考附图并结合优选实施例的方式对本发明进行了详细描述,但本发明并不限于此。在不脱离本发明的精神和实质的前提下,本领域普通技术人员可以对本发明的实施例进行各种等效的修改或替换,而这些修改或替换都应在本发明的涵盖范围内/任何熟悉本技术领域的技术人员在本发明揭露的技术范围内,可轻易想到变化或替换,都应涵盖在本发明的保护范围之内。因此,本发明的保护范围应所述以权利要求的保护范围为准。
Claims (6)
1.一种过氧乙酸消毒液,其特征在于,包括A剂和B剂,按照重量百分含量具体如下,
A剂包括:过氧化氢1-7%,稳定剂0.1-10%,螯合剂0.1-10%,缓蚀剂0.1-5%,增溶剂5-30%,溶剂10-60%;
B剂包括:酰化剂25-40%、pH调节剂1-10%、溶剂50-70%;
所述的稳定剂为有机膦酸,包括氨基三甲叉膦酸、乙二胺四甲叉膦酸、羟基乙叉二膦酸、二乙烯三胺五甲叉膦酸和2-羟基磷酸基乙酸中的一种或多种;
所述的螯合剂为氨基三乙酸、乙二胺四乙酸和酒石酸的一种或多种;
所述的缓蚀剂为葡萄糖酸钠、氨基三甲叉膦酸四钠、乙二胺四甲叉膦酸五钠、二乙烯三胺五甲叉膦酸和苯并三氮唑中的一种或多种;
所述的增溶剂为N-乙基己基-β-亚氨基二丙氨酸钠Ampholak YJH-40和磷酸聚醚脂钾盐H-66中的一种或两种。
2.如权利要求1所述的过氧乙酸消毒液,其特征在于:所述A剂和B剂中的溶剂均为乙醇、丙二醇、丙三醇、异丙醇、聚丙二醇-200和聚醚50-HB-660中的一种或多种。
3.如权利要求1所述的过氧乙酸消毒液,其特征在于:所述的酰化剂为N-乙酰戊内酰胺、N-乙酰己内酰胺、N-乙酰庚内酰胺和N-乙酰辛内酰胺中的一种或多种。
4.如权利要求1所述的过氧乙酸消毒液,其特征在于:所述的pH调节剂为三乙醇胺、二甲基乙醇胺、二异丙基乙胺、2-氨基-2-甲基-1-丙醇、N,N,N-三乙胺和N,N,N-二异丙基乙胺中的一种或多种。
5.如权利要求1-4任意一项所述的过氧乙酸消毒液的制备方法,其特征在于:包括将含有酰化剂的B剂加入到含过氧化氢的A剂中,得到质量浓度为0.21%-0.39%的过氧乙酸消毒液。
6.如权利要求5所述的过氧乙酸消毒液的制备方法,其特征在于:B剂和A剂的体积比为:1:30-1:100,常温条件下混合5-15分钟。
