本发明公开了一种用于辅助改善鱼类体外细菌感染的菌液复合物,按质量体积比计,包括如下组分:聚羟基脂肪酸酯PHA 178.5~258.0g/L;氮源1.5~2.5g/L;其他微生物生存的必需元素余量。本发明采用特定种类及特定复配质量比的聚羟基脂肪酸酯PHA,与特定质量体积比的氮源、其他微生物生存的必需元素进行复配,通过各组分的协同配合,制备得到菌液复合物,将其应用至对鱼类体外细菌感染过程中,采用不同种类的微生物之间的拮抗作用,对鱼类体外感染的有害菌具有较强的拮抗作用,能够有效杀灭鱼类体外感染的有害菌,从而有效延长菌液复合物的保质期;此外,由于没有抗生素进入鱼类机体,所以没有药物残留的风险,有效保证鱼类健康。

本发明涉及水产养殖技术领域,特别涉及一种用于辅助改善鱼类体外细菌感染的菌液复合物及其应用。
背景技术
拮抗作用是指一类微生物抑制或杀死它类微生物的作用。拮抗作用是微生物界的普遍现象。微生物之间的拮抗作用分为非特异性拮抗和特异性拮抗。可产生抗生物的微生物能够抑制甚至杀死其他类微生物,例如青霉菌产生的青霉素能抑制革兰氏阳性菌,链霉菌产生的制霉菌素能够抑制酵母菌和霉菌等,这些都属于特异性的拮抗作用。
目前治疗鱼类体外细菌感染的主要方法是使用抗生素拌料饲喂或抗生素类药液进行浸泡机体,但这两种方法会在鱼类机体残留大量抗生素。同时抗生素在机体的代谢周期远长于水产鱼类的养殖周期,不良商家会将带有抗生素残留的水产品上市,因此容易造成食品安全问题。

本发明的另一目的是提供上述菌液复合物在对鱼类体外细菌感染具有辅助改善作用中的应用。

一种用于辅助改善鱼类体外细菌感染的菌液复合物,按质量体积比计,包括如下组分:
聚羟基脂肪酸酯PHA 178.5~258.0g/L;
氮源 1.5~2.5g/L;
其他微生物生存的必需元素 余量。
进一步作为本发明技术方案的改进,所述的用于辅助改善鱼类体外细菌感染的菌液复合物,按质量体积比计,包括如下组分:
聚羟基脂肪酸酯PHA 238.0g/L;
氮源 2.0g/L;
其他微生物生存的必需元素 余量。
进一步作为本发明技术方案的改进,所述聚羟基脂肪酸酯PHA选自聚-β-羟丁酸PHB、聚(3-羟基丁酸酯-co-4-羟基丁酸酯)P34HB、3-羟基丁酸酯和3-羟基戊酸酯共聚物PHBV、3-羟基丁酸与3-羟基己酸共聚酯PHBHHx中的一种或几种;
进一步作为本发明技术方案的改进,所述聚羟基脂肪酸酯PHA是很多细菌可合成的胞内聚酯,是微生物自身储藏的能源物质,具有优良的生物降解性,可被生物吸收的优良碳源,选自选自PHB、P34HB、PHBV、PHBHHx四种组分的复配,所述复配质量比为(4~8):(1~3):1:1,优选为6:2:1:1。

进一步作为本发明技术方案的改进,所述其他微生物生存必需元素中的微生物为可产生抗病菌次级代谢产物的放线菌类微生物种类,同时在一定浓度范围内对鱼类无明显毒理作用;所述必需元素选自铜离子、锌离子、铁离子、钾离子、镁离子或钙离子;具体选自硫酸锌、硫酸铜、磷酸二氢钾、硫酸镁、氯化钙中的一种或几种。

以下将结合实施例对本发明的构思及产生的技术效果进行清楚、完整地描述,以充分地理解本发明的目的、特征和效果。显然,所描述的实施例只是本发明的一部分实施例,而不是全部实施例,基于本发明的实施例,本领域的技术人员在不付出创造性劳动的前提下所获得的其他实施例,均属于本发明保护的范围。
以下具体实施方式中,如无特殊说明,均为常规方法;以下具体实施方式所用的原料、试剂材料等,如无特殊说明,均为市售购买产品。
聚羟基脂肪酸酯PHA:PHB,珠海麦得发生物科技股份有限公司;
P34HB,珠海麦得发生物科技股份有限公司;
PHBV,珠海麦得发生物科技股份有限公司;
PHBHHx,珠海麦得发生物科技股份有限公司;
氮源:硫酸铵,国药、分析纯;
氯化铵,国药、分析纯;
其他微生物生存的必需元素:硫酸锌,国药、分析纯;
硫酸铜,国药、分析纯。
各性能指标的测试方法:
菌活性的测试方法:平计数术法;
保质期的测试方法:以菌液活性(有效活菌数≥5×108CFU/ml)和产品性状(无明显臭味,无沉淀或少量沉淀)作为产品的保存期限。
实施例A1-A5及对比例B1-B5:菌液复合物的制备
按照表1的配比将聚羟基脂肪酸酯PHA、氮源、其他微生物生存的必需元素进行混合得到菌液复合物,制备得到的菌液复合物的活性、保质期等性能指标如表1所示。

应用例:
采用25L的实施例A1-A5及对比例B1-B5制备得到的菌液复合物分别加入至如图1和图2所示的水箱2中,所述水箱2满足鱼类的生存空间的需要,所述增氧装置3安装在所述水箱2底部,增加水体氧气含量,保证鱼类的呼吸需要,所述pH在线监测装置4安装在所述水箱侧壁,所述pH在线监测装置4完全浸没于水体中,在pH值高于或低于设置范围时发出警报,实时监测水体酸碱度;将体外受伤感染的石斑鱼分别投入至上述含有25L的实施例A1-A5及对比例B1-B5制备得到的菌液复合物1的水箱2中,观察实施例A1-A5及对比例B1-B5制备得到的菌液复合物对体外受伤感染的石斑鱼的辅助改善作用,具体测试结果如表2所示。

Claims (10)
1.一种用于辅助改善鱼类体外细菌感染的菌液复合物,按质量体积比计,包括如下组分:
聚羟基脂肪酸酯PHA 178.5~258.0g/L;
氮源 1.5~2.5g/L;
其他微生物生存的必需元素 余量。
2.根据权利要求1所述的用于辅助改善鱼类体外细菌感染的菌液复合物,按质量体积比计,包括如下组分:
聚羟基脂肪酸酯PHA 238.0g/L;
氮源 2.0g/L;
其他微生物生存的必需元素 余量。
3.根据权利要求1或2所述的用于辅助改善鱼类体外细菌感染的菌液复合物,其特征在于,所述聚羟基脂肪酸酯PHA选自聚-β-羟丁酸PHB、聚(3-羟基丁酸酯-co-4-羟基丁酸酯)P34HB、3-羟基丁酸酯和3-羟基戊酸酯共聚物PHBV、3-羟基丁酸与3-羟基己酸共聚酯PHBHHx中的一种或几种。
4.根据权利要求3所述的用于辅助改善鱼类体外细菌感染的菌液复合物,其特征在于,所述聚羟基脂肪酸酯PHA选自PHB、P34HB、PHBV、PHBHHx四种组分的复配,所述复配质量比为(4~8):(1~3):1:1,优选为6:2:1:1。
5.根据权利要求1或2所述的用于辅助改善鱼类体外细菌感染的菌液复合物,其特征在于,所述氮源选自硫酸铵、氯化铵、尿素中的一种或几种。
6.根据权利要求1或2所述的用于辅助改善鱼类体外细菌感染的菌液复合物,其特征在于,所述其他微生物生存必需元素中的微生物为可产生抗病菌次级代谢产物的放线菌类微生物种类;所述必需元素选自硫酸锌、硫酸铜、磷酸二氢钾、硫酸镁、氯化钙中的一种或几种。
7.如权利要求1-6任一项所述的菌液复合物在对鱼类体外细菌感染具有辅助改善作用中的应用。
8.根据权利要求7所述的应用,其特征在于,包括如权利要求1-6任一项所述的菌液复合物和水箱(2)。
9.根据权利要求8所述的应用,其特征在于,所述水箱(2)底部装有增氧装置(3)。
10.根据权利要求8所述的应用,其特征在于,所述水箱(2)侧壁装有pH在线监测装置(4)。
