背景技术

实施例
实施例1.二价BRD4抑制剂AZD5153与维奈托克组合在AML临床前模型中的功效
DFAM-68555离体筛选:PDX细胞分离自携带DFAM-68555异种移植物的雌性NSG小鼠的脾脏。将PDX细胞重悬于HS-5细胞调整培养基和IMDM+10%FBS的50/50混合物中。将细胞接种于96孔板中并用维奈托克、AZD5153+维奈托克和5-氮杂胞苷+维奈托克处理。处理72h之后,通过流式细胞术评估存活AML细胞的数目。结果示于图1。
DFAM-68555PDX疗效:通过尾静脉注射将106DFAM-68555PDX细胞植入雌性NSG小鼠。当循环疾病(通过huCD45+huCD33+细胞%测量)达到约4%时,将小鼠随机分组并用媒介物、维奈托克(100mpk qd po)+5-氮杂胞苷(1mpk bid 3on/4off ip)或维奈托克(100mpkqd po)+AZD5153(0.075mpk qd po)处理。将维奈托克在10%乙醇、30%PEG-400和60%Phosal 50PG中配制。将AZD5153在0.5%HPMC+0.1%Tween-80中配制。将氮杂胞苷在0.9%盐水中配制。处理动物14天。通过流式细胞术每周监测循环疾病。在研究结束时,收集血液、脾脏和骨髓(胸骨)用于流式细胞术和组织病理学分析。结果示于图2。
结果:如图1和图2所示,维奈托克+5-氮杂胞苷在DFAM-68555PDX模型离体(图1)和体内(图2)几乎没有功效。然而,维奈托克+AZD5153显著降低血液、脾脏和骨骼中的离体和体内肿瘤负荷。
Claims (14)
1.一种治疗恶性血液病的方法,其包括向有需要的受试者施用有效量的(3R)-4-[2-[4-[1-(3-甲氧基-[1,2,4]三唑并[4,3-b]哒嗪-6-基)-4-哌啶基]苯氧基]乙基]-1,3-二甲基-哌嗪-2-酮或其药学上可接受的盐和有效量的维奈托克或其药学上可接受的盐。
2.如权利要求1所述的方法,其中所述方法包括与维奈托克或其药学上可接受的盐依序、分别或同时施用(3R)-4-[2-[4-[1-(3-甲氧基-[1,2,4]三唑并[4,3-b]哒嗪-6-基)-4-哌啶基]苯氧基]乙基]-1,3-二甲基-哌嗪-2-酮或其药学上可接受的盐。
3.如权利要求1或2所述的方法,其中所述恶性血液病选自急性髓性白血病(AML)、骨髓增生异常综合征(MDS)、慢性髓性白血病(CML)、慢性淋巴细胞性白血病(CLL)和慢性骨髓单核细胞性白血病(CMML)。
4.(3R)-4-[2-[4-[1-(3-甲氧基-[1,2,4]三唑并[4,3-b]哒嗪-6-基)-4-哌啶基]苯氧基]乙基]-1,3-二甲基-哌嗪-2-酮或其药学上可接受的盐,其用于治疗恶性血液病,其中所述治疗包括分别、依序或同时施用维奈托克或其药学上可接受的盐。
5.维奈托克或其药学上可接受的盐,其用于治疗恶性血液病,其中所述治疗包括分别、依序或同时施用(3R)-4-[2-[4-[1-(3-甲氧基-[1,2,4]三唑并[4,3-b]哒嗪-6-基)-4-哌啶基]苯氧基]乙基]-1,3-二甲基-哌嗪-2-酮或其药学上可接受的盐。
6.如权利要求4或5所述的用途,其中所述恶性血液病选自急性髓性白血病(AML)、骨髓增生异常综合征(MDS)、慢性髓性白血病(CML)、慢性淋巴细胞性白血病(CLL),以及慢性骨髓单核细胞性白血病(CMML)。
7.如权利要求1至6中任一项所述的方法或用途,其还包括施用有效量的5-阿扎胞苷。

第一药物组合物,其包含(3R)-4-[2-[4-[1-(3-甲氧基-[1,2,4]三唑并[4,3-b]哒嗪-6-基)-4-哌啶基]苯氧基]乙基]-1,3-二甲基-哌嗪-2-酮或其药学上可接受的盐和药学上可接受的载剂;以及
第二药物组合物,其包含维奈托克或其药学上可接受的盐;以及使用说明书。
11.如权利要求10所述的药盒,其还包括包含5-阿扎胞苷的第三药物组合物。
12.一种药物产品,其包含i)(3R)-4-[2-[4-[1-(3-甲氧基-[1,2,4]三唑并[4,3-b]哒嗪-6-基)-4-哌啶基]苯氧基]乙基]-1,3-二甲基-哌嗪-2-酮或其药学上可接受的盐,以及ii)维奈托克或其药学上可接受的盐。
