背景技术
目前的抗癌药物疗法效果有限。常用的抗癌药物可能会使肿瘤暂时缓解,并有助于延长患者的寿命,但在大多数情况下不能治愈。它们通常不能完全消除癌症,且肿瘤随后可能会复发。小分子化疗是目前应用最广泛的癌症治疗方法,但它们的作用在很大程度上是非特异性的。化疗试剂具有严重的脱靶毒性作用,对正常、健康的细胞和组织造成附带损害。因此,患者因毒性而遭受严重的副作用。基于免疫的疗法有望降低毒性并提高生存率,但尽管它们有希望,但它们只是逐渐改善了癌症治疗的前景和有利的长期患者预后。尽管靶向疗法和免疫疗法越来越多地被用作传统化疗的补充或替代品,但这类疗法通常缺乏长期疗效,因为癌症通常会适应并迅速产生耐药性,从而逃避靶向疗法效果。由于上述原因,化疗、免疫疗法和靶向疗法很少是治愈性的。

小干扰RNA(siRNA)具有特异性沉默细胞中的靶基因的潜力。成功实施siRNA疗法的主要障碍之一是难以将有效量的siRNA递送到身体的相关作用部位。siRNA是小分子,大约只有21-23个核苷酸,容易被肾脏从体内清除,被诸如核酸酶和溶菌酶的内源酶降解,并被免疫系统攻击。此外,裸siRNA的治疗活性很难利用,因为它们的体内半衰期非常短,且体内清除速度很快。因此,成功将取决于匹配适当的siRNA以抑制某些有问题的基因的能力,同时提供一个使新的siRNA递送到靶位点如癌细胞的可能性最大化的递送系统。
鉴于上述情况,需要用于调节和抑制癌症相关基因的表达和进一步提高基于siRNA的治疗的能力和有效性的分子组合物和方法。本发明解决了这些和其他需求。
发明内容
本发明包括包含靶向在癌症中表达的基因的siRNA分子的药物组合物,以及使用此类组合物沉默此类基因的表达从而抑制癌细胞产生的方法。本发明的实施方案包含新的化学实体,其包含装载有siRNA的靶向药物递送系统,该siRNA被设计用于敲低PRDM14基因的表达。PRDM14是主调控基因,其编码PRDM14蛋白。PRDM14的调节异常与多种癌症类型密切相关,包括乳腺癌和肺癌以及其他癌症。
根据本发明,设计用于调节PRDM14基因的表达的新药物(包括相关药物递送系统)的开发预期满足对某些癌症类型的有效长期治疗且甚至治愈的迫切需求,现有的化疗和免疫疗法通常无法满足这一需求。根据本发明,这些新药物将提供一种有效的解决方案,其可通过使用针对特定癌细胞的补充药物递送系统来增强,从而将毒性副作用和对健康细胞和组织的损害降至最低。

具体实施方式
本发明的实施方案包含用于治疗人类和动物的各种癌症的新化学实体。具体地,本发明包含装载有siRNA的靶向药物递送系统,该siRNA被设计用于沉默PRDM14基因的表达,从而抑制癌性生长。
在第一方面中,本发明包含长度为16-30个核苷酸的分离的寡核苷酸(siRNA),其具有与人PRDM14基因序列(在本文中识别为SEQ ID NO:1且识别为RefSeq登录号:NM_024504)的连续部分互补的序列。
下表1列出了根据本发明主题设计用于下调PRDM14基因的表达并抑制癌症生长的siRNA序列。在某些实施方案中,可将位点特异性碱基和骨架修饰设计到siRNA中,以保护siRNA不被降解和消除,防止免疫原性,并将脱靶效应的可能性降至最低。

下表1中列出的siRNA在递送给患者时,可通过药物递送系统例如脂质体或由脂质、环糊精、壳聚糖、碳水化合物聚合物、类弹性蛋白聚合物(ELP)、磷酸钙聚合物或其组合组成的纳米颗粒增强。
siRNA以及任何伴侣有效载荷优先通过癌症特异性靶向纳米颗粒或其他类型的癌症靶向药物递送系统(如适体)递送至癌细胞,以避免损伤正常健康细胞并保护裸siRNA不被降解,稳定循环系统中的整个有效载荷,并介导siRNA有效内化到癌细胞中。
与siRNA递送相关的纳米颗粒可由载体材料的组合组装而成,载体材料可包括但不限于脂质、脂质体、糖、葡聚糖、磷酸钙、壳聚糖、肽和塑料聚合物。
在某些实施方案中,药物组合物进一步包括癌细胞靶向配体,以增强针对癌细胞的siRNA有效载荷相对于正常健康细胞的选择性。选择靶向配体以特异性结合在癌细胞表面上过度表达的受体或其他部分,并将癌细胞与正常细胞区分。在某些实施方案中,靶向配体可以是但不限于,多肽、适体或其他类别的能够以高特异性和亲和力结合到靶向的癌细胞的分子。
在某些实施方案中,靶向配体可以是但不限于,肽(例如,线性或环状形式的肽DMPGTVLP,以高特异性和亲和力与乳腺癌细胞结合)。
在某些实施方案中,通过siRNA与磷硅酸钙载体材料络合形成纳米药物颗粒。在其他实施方案中,纳米颗粒可由类弹性蛋白多肽(ELP)组装而成。仍在其他实施方案中,药物递送系统可以是脂质体。在一些实施方案中,药物递送系统可涂覆有保护层(例如,脂质体的聚乙二醇化),以延长药物在血液中的寿命,并保护药物递送系统免受免疫系统的破坏。
本发明主题主要针对治疗由PRDM14基因调节异常驱动的癌症,然而其他癌症可能受到影响。在某些实施方案中,药物组合物可被配制用于但不限于,治疗患有乳腺癌、肺癌、食管癌、胰腺癌、卵巢癌、肾癌、膀胱癌、肾癌、生殖细胞癌、白血病、头颈癌或宫颈癌的患者。
在优选的实施方案中,将具有位点特异性碱基和骨架修饰的抗PRDM14 siRNA与化疗伴侣有效载荷复合到基于ELP的纳米颗粒中,并使用DMPGTVLP靶向配体靶向乳腺癌细胞。
下面提供的表1是根据本发明主题针对PRDM14表达的那些siRNA双链体的列表,其中4=2’OMe-U;粗体=硫代磷酸酯;P=磷酸酯。

以下实施方案描述了根据本发明主题的药物组合物的预期用途和功效。
在根据本发明主题的第一实施方案中,描述了说明根据本发明的siRNA用于杀死人乳腺癌细胞(细胞系MCF-7)的用途和功效的组合物。如图3的图表所示的结果反映体外实验。
将MCF-7癌细胞在96孔板(1,000个细胞/孔)中铺板,并且在24小时后,将MCF-7癌细胞用20nM siRNA(包括ARIZ-040(SEQ ID NO:22和23)、ARIZ-044(SEQ ID NO:24和25),以及ARIZ-026(SEQ ID NO:10和11),使用0.3μl的Lipofectamine(Invitrogen,Inc.)转染。在两天后更换培养基,并且在四天后,测定相对于未处理的细胞的细胞活力百分比。作为对照,用同一组的siRNA处理正常结肠成纤维细胞(细胞系CC112)。
现在参考图3和图4的图表,我们示出了MCF-7乳腺癌细胞与正常结肠成纤维细胞(细胞系CCD112)相比在暴露于包括ARIZ-040(SEQ ID NO:22和23)、ARIZ-0044(SEQ ID NO:24和25),以及ARIZ-026(SEQ ID NO:10和11)的各种siRNA后产生的细胞活力。
因此,如图所示,抗PRDM14 siRNA(ARIZ-040(SEQ ID NO:22和23)、ARIZ-0044(SEQID NO:24和25),以及ARIZ-026(SEQ ID NO:10和11)相对于加扰siRNA阴性对照在杀死MCF-7乳腺癌细胞方面有效。结果进一步证明,根据本发明主题和本第一实施方案的抗PRDM14siRNA特异性攻击癌细胞,因为对正常结肠成纤维细胞的杀伤最小。
现在参考图5,在根据本发明主题的第二实施方案中,描述了说明siRNA用于杀死人肺癌细胞(细胞A549)的用途和功效的组合物。如图4的图表所示的结果反映体外结果。

(i)治疗有效量的siRNA,其中,所述siRNA包含具有序列5'-GUGAAGACC4ACGGAGACAdTdT-3'的有义链和具有序列3'-dTdTCACUUCUGGAUGCC4C4G4-P-5'的反义链,其中4=2'OMe-U;粗体=硫代磷酸酯;并且P=磷酸酯;
(ii)药物递送系统,所述药物递送系统包含类弹性蛋白多肽纳米颗粒,所述纳米颗粒包含所述siRNA;和
(iii)细胞靶向配体DMPGTVLP。
38.根据权利要求28-37中任一项所述的药物组合物,其中所述有义链或所述反义链包含2’-OMe。
39.根据权利要求28-37中任一项所述的药物组合物,其中所述药物组合物被配制用于瘤内注射、皮下注射或静脉注射或用于吸入。
40.根据权利要求28-37中任一项所述的药物组合物在制备用于治疗的癌症的药物中的用途,其中所述癌症是乳腺癌、肺癌、食管癌、胰腺癌、卵巢癌、肾癌、膀胱癌、白血病、头颈癌或宫颈癌。
