Abstract
The invention discloses a sword fungus adhesion, which is classified and named as sword fungus adhesion (A)Ensiferadhaerens) The strain number is CN-02, and the strain number is CGMCC 21357, which is preserved in the China general microbiological culture Collection center; further discloses the application of the Zygomyces invicta in enhancing the salt stress resistance of crops. The invention discloses a microbial inoculum which takes sword-shaped bacteria as active microorganisms; further discloses application of the microbial inoculum in enhancing salt stress resistance of crops. The Zygomyces adhesins can be adhered to the root system of the plant and can also enter the plant to be symbiotic with the plant, so that the absorption of the plant to external nutrient elements is improved, the metabolic system of the plant is improved, the water potential in the plant is maintained, and the like, thereby promoting the growth and development of the plant, improving the yield, and simultaneously enhancing the salt stress resistance of the plant. To give full play to the ecological fertility of soil and maintainThe balance of the agricultural ecological environment and the like have extremely important significance and application value.
Description
Technical Field
The invention relates to the field of microbial agents, and particularly relates to sword-shaped bacterium adhesion and a microbial agent and application thereof.
Background
Soil salinization and soil secondary salinization caused by irrigation are main environmental risks caused by natural or human activities and generally occur in regions with dry climate, high soil evaporation strength, high underground water level and more soluble salts. Statistically, 20% of cultivated land and nearly half of irrigated land in the world are threatened by salt damage to different degrees, and the proportion is rising. The total area of the salinized land in China is about 1 hundred million hectares, and the salinized land is mainly distributed in 17 provinces including northwest, northeast, north China and coastal areas. When the soil salt concentration reaches a certain value, the normal life activities of plant cells can be damaged, the growth of plants is inhibited, the land output rate is low, the comprehensive agricultural productivity is seriously insufficient, and the ecological safety and the economic sustainable development of the regions are directly influenced.
At present, how to improve saline-alkali resistance and saline-alkali resistance of crops becomes a major challenge for researchers. The key to the problem of promoting the growth of plants in the halophytic environment is to improve the soil and improve the saline-alkali resistance of crops. Plant growth-promoting rhizobacteria (PGPR) is a type of microorganism in Plant rhizosphere soil which can promote Plant growth, prevent and control diseases and increase crop yield. The use of PGPR will help to reduce the adverse effects of salinity on plants.
Disclosure of Invention
The purpose of the invention is as follows: in order to solve the problem of salt resistance of plants, the invention provides a sword-shaped bacterium adhesion, and further provides a microbial inoculum taking the sword-shaped bacterium adhesion as an active microorganism and application of the microbial inoculum in enhancing the salt stress resistance of crops.
The technical scheme is as follows: the sword-like fungus adhesion is classified and named as sword-like fungus adhesion (Ensiferadhaerens), the strain number is CN-02, the sword-like fungus adhesion has been preserved in the China general microbiological culture Collection center, the preservation time is 12 months and 11 days in 2020, the preservation number is CGMCC NO.21357, the preservation address is No. 3 of Xilu No.1 of Xingyang district in Beijing, the preservation center of the institute of microbiology in China, and the postal code is 100101. The strain is a strain bred by the inventor from the soil of the paddy field of the Putouu region of Nanjing in 2020 and 5 months.
Ensifer adhesins CN-02 has the following properties:
1. morphological characteristics of colonies
After culturing for 16-24h at 30 ℃ on LB solid culture medium, the cells are observed to be single cells and rod-shaped by a microscope, the length of the bacteria is 1.0-1.5 μm, and the width is 0.4-0.6 μm. Belongs to gram-negative bacteria and no spores. The thalli can grow in a large amount after being cultured in the culture medium for 12 hours at the temperature of 30 ℃, and bacterial colonies are milky, opaque, smooth, sticky, convex and regular in edge.
2. 16SrDNA sequence analysis
The length of the 16SrDNA sequence is 1388bp, and the nucleotide sequence is shown in a sequence table SEQ. NO. 1. and dissolving the collected soil sample in sterile water, shaking and uniformly mixing. The sample solution is diluted in gradient, 10 is taken-4、10-5、10-6The dilutions of the three concentration gradients were spread evenly on solid medium (medium composition: sucrose 20g/L, peptone 5g/L, beef extract 1g/L, NaCl 5g/L, agar 20 g/L). Culturing in 30 deg.C constant temperature incubator for 48h, viewing the plate every day during the period, the colony characteristics were observed, and the sticky colonies were used as the screening target. Selecting a single colony which can produce high-viscosity extracellular polysaccharide from the culture medium, and purifying the single colony on a solid culture difficult plate by a scribing method.
And respectively inoculating the single colonies obtained in the previous step into a liquid culture medium (the components of the culture medium are 20g/L of cane sugar, 5g/L of peptone, 1g/L of beef extract and 5g/L of NaCl), performing shake fermentation culture at 30 ℃ on a shaking table, and comparing the viscosity of fermentation liquids of different strains after 48 hours, wherein the strain with the highest viscosity of the fermentation liquid is the Ensifer adhesivelis CN-02.
Ensifer adhesins CN-02 has the following properties:
