本发明属于医药材料技术领域,公开了一种纳米材料及其制备方法和应用。该纳米材料,包括聚合物改性的聚多巴胺,和负载于聚合物改性的聚多巴胺上的染料和造影剂;聚合物选自双氨基聚乙二醇、聚乙烯亚胺、带巯基的聚乙二醇或透明质酸中的一种。本发明提供的纳米材料能够与淋巴特异性结合,能够作为淋巴示踪剂在手术中获得实时的肉眼/荧光/热信号的淋巴结成像,实现了术前/术中淋巴结示踪,实现淋巴结精准、高效切除。本发明能够制备出不同粒径大小的纳米材料,能够提供灵活多变的术前、术中注射时间点,以满足复杂和变化的临床需求。

技术领域
本发明属于医药材料技术领域,具体涉及一种纳米材料及其制备方法和应用。
背景技术
淋巴结是呈椭圆形或蚕豆形的淋巴组织小体,穿插于淋巴管的行程中,并与淋巴管相连。淋巴结一侧凸隆,与此侧相连的淋巴管称为输入淋巴管;另一侧凹陷形成淋巴结门,与此侧(门)相连的淋巴管称输出淋巴管。淋巴结的主要功能是过滤淋巴,清除细菌和异物,产生淋巴细胞和抗体。
淋巴结系统性清扫是将区域淋巴结及周围软组织切除,是提高实体肿瘤患者生存率的有效手段。精准的术前/术中淋巴结示踪手段能提高淋巴结清扫的准确性。然而,目前临床上使用的淋巴示踪剂无法实现术前示踪成像,也无法实现多模态精准成像;此外部分临床应用的淋巴结示踪剂无淋巴特异性,可进入血管对手术造成干扰;最后,目前临床应用的淋巴结示踪剂的注射时间相对固定,无法满足复杂多变的临床需求。
因此,亟需提供一种具有淋巴特异性的纳米淋巴示踪剂,能够实现多模态精准成像。
发明内容
本发明旨在至少解决上述现有技术中存在的技术问题之一。为此,本发明提出一种纳米材料,能够与淋巴特异性结合,作为淋巴示踪剂能够实现多模态精准成像。
一种纳米材料,包括聚合物改性的聚多巴胺和负载于所述聚合物改性的聚多巴胺上的染料和造影剂;所述聚合物选自双氨基聚乙二醇、聚乙烯亚胺、带巯基的聚乙二醇或透明质酸中的一种。
所述带巯基的聚乙二醇如巯基聚乙二醇、二巯基聚乙二醇等。
聚多巴胺分子中含有酚羟基、氨基等多种活性基团,在盐溶液中性质不稳定,易沉淀,经过聚合物改性的聚多巴胺,其稳定性强,负载染料和造影剂后,结构稳定,形成的所述纳米材料光热性能稳定,成像稳定性强;且能够与淋巴特异性结合,作为淋巴示踪剂能够实现多模态精准成像。
优选地,所述聚合物为双氨基聚乙二醇。
优选地,所述造影剂的负载量为1-3%;进一步优选地,所述造影剂的负载量为1-2%。
优选地,所述染料的负载量为60-80%;进一步优选地,所述染料的负载量为65-75%。
优选地,所述造影剂为顺磁性金属盐;进一步优选地,所述造影剂选自钆盐、铁盐或锰盐中的一种。
优选地,所述染料为近红外荧光染料;进一步优选地,所述近红外染料为吲哚菁绿(ICG)、新吲哚菁绿(IR820)、IR-780碘化物或近红外二区荧光探针中的一种。
优选地,所述纳米材料的粒径为20-300nm;进一步优选地,所述纳米材料的粒径为50-200nm。
本发明还提供了一种纳米材料的制备方法,包括以下步骤:
(1)将聚多巴胺溶解,调节pH值至碱性,加入聚合物,搅拌,过滤,去滤液,制得聚合物改性的聚多巴胺;
(2)向步骤(1)制得的聚合物改性的聚多巴胺中加入染料和造影剂,搅拌,过滤,去滤液,制得所述纳米材料。
优选地,步骤(1)中所述聚多巴胺的制备方法,包括以下步骤:将多巴胺或盐酸多巴胺溶解,加入碱液,搅拌,过滤,去滤液制得。
优选地,所述碱液为氢氧化钠、氢氧化钾、碳酸氢钾或氨水中的至少一种;进一步优选地,所述碱液为氢氧化钠。
优选地,所述氢氧化钠的浓度为0.5-2mol/L;进一步优选地,所述氢氧化钠的浓度为0.5-1.5mol/L。
优选地,所述多巴胺或盐酸多巴胺与所述氢氧化钠的物质的量的比为1:(0.1-1.2);进一步优选地,所述多巴胺或盐酸多巴胺与所述氢氧化钠的物质的量的比为1:(0.2-1);更优选地,所述多巴胺或盐酸多巴胺与所述氢氧化钠的物质的量的比为1:(0.8-1)。
通过控制所述碱液与所述多巴胺或盐酸多巴胺的物质的量的比,可以得到不同粒径的聚多巴胺,进而经改性后得到不同粒径的纳米材料,根据不同粒径的纳米材料在淋巴结中滞留的时间不同(粒径为50-100nm的纳米材料在淋巴结中滞留的时间约为4h、100-150nm的纳米材料在淋巴结中滞留的时间约为12h、150-200nm的纳米材料在淋巴结中滞留的时间约为24h),能够提供灵活多变的术前、术中注射时间点,以满足复杂和变化的临床需求。
优选地,步骤(1)中所述聚多巴胺和所述聚合物的质量比为(1-2):(1-2);进一步优选地,步骤(1)中所述聚多巴胺和所述聚合物的质量比为1:(1-1.5)。
优选地,步骤(1)中所述pH值为8-10;进一步优选地,步骤(1)中所述pH值为8.5-9.5。
优选地,步骤(2)中所述聚合物改性的聚多巴胺、染料和造影剂的质量比为1:(0.01-0.08):(0.01-0.08)。进一步优选地,步骤(2)中所述聚合物改性的聚多巴胺、染料和造影剂的质量比为1:(0.02-0.06):(0.02-0.06)。
具体的,一种纳米材料的制备方法,包括以下步骤:
(1)将多巴胺或盐酸多巴胺溶解于去离子水中,加入氢氧化钠,搅拌反应6-12h,超滤,制得聚多巴胺;
(2)将步骤(1)制得的聚多巴胺溶解,调节pH值至8-10,加入聚合物,搅拌6-15h,超滤,去滤液即为聚合物改性的聚多巴胺;
(3)向步骤(2)制得的聚合物改性的聚多巴胺中加入染料和造影剂,搅拌2-8h,超滤,去滤液即为所述纳米材料。
优选地,步骤(1)中所述过滤使用的是超滤管,所述超滤管的截留分子量为10-100kD;进一步优选地,所述超滤管的截留分子量为30-50kD;更优选地,所述超滤管的截留分子量为30kD。
本发明也提供了上述纳米材料的应用。
所述纳米材料在制备医药材料中的应用。
一种示踪剂,包含所述纳米材料。
一种淋巴示踪剂,包含所述纳米材料。
相对于现有技术,本发明的有益效果如下:
(1)本发明提供的纳米材料由经过聚合物改性的聚多巴胺和负载于所述聚合物改性的聚多巴胺上的染料和造影剂组成,其结构稳定,光热性能优异,成像稳定性强;且具有嗜淋巴性,能特异性进入淋巴系统,作为淋巴示踪剂,能够在手术前获得淋巴结精准的MR成像,同时能够在手术中获得实时的肉眼/荧光/热信号(多模态)的淋巴结精准成像,实现淋巴结精准、高效切除。
(2)本发明通过控制碱液的量制备出不同粒径的聚多巴胺,进而经改性后得到不同粒径的纳米材料,根据不同粒径的纳米材料在淋巴结中滞留的时间不同,本发明能够实现0-24小时不同时长的淋巴结示踪,提供了灵活多变的术前、术中注射时间点,能够满足复杂和变化的临床需求。
(3)本发明提供的纳米材料的制备方法简单,易操作。

具体实施方式
为了让本领域技术人员更加清楚明白本发明所述技术方案,现列举以下实施例进行说明。需要指出的是,以下实施例对本发明要求的保护范围不构成限制作用。
以下实施例中所用的原料、试剂或装置如无特殊说明,均可从常规商业途径得到,或者可以通过现有已知方法得到。
现以造影剂选用钆双胺,染料选用IR820,聚合物选用双氨基聚乙二醇,碱液选用NaOH溶液为例,制备不同粒径的纳米材料。以上原料的选择只是选取代表性物质,其他聚合物、染料、造影剂和碱液也能达到类似的技术效果,以下就不一一列举。
实施例1
一种纳米材料的原料如表1。

一种纳米材料的制备方法,包括以下步骤:
(1)制备聚多巴胺纳米粒子:将432mg(2.28mmol)盐酸多巴胺加入到216mL去离子水中溶解,调节至50度后加入2280μL 1mol/L的NaOH溶液,充分搅拌反应10h,使用超滤管(30kd)过滤获得聚多巴胺纳米粒子(PDA)。
(2)对聚多巴胺粒子进行聚乙二醇改性(PEG化):取100mg溶于10mL去离子水中,加入适量NaOH溶液,将pH值调节至9,加入双氨基聚乙二醇(NH2-PEG5000-NH2)120mg,常温下充分搅拌12h。最后使用超滤管(30KD)获得PEG改性的聚多巴胺纳米粒子(PEG-PDA)。
(3)制备纳米材料:以投料比1:0.04,分别向经PEG改性的聚多巴胺纳米粒子(PEG-PDA)中加入钆双胺及IR820,充分搅拌4h后过滤(30KD),获得负载了钆双胺及IR820的PEG聚多巴胺纳米粒子(PPIG),记为纳米材料PPIG-1。
纳米材料的制备过程的示意图及试验示意图见图1,由图1可知,将制备的纳米材料进行动物实验和离体人标本试验,在动物实验中实现进行一体化多模态淋巴结示踪(核磁共振、肉眼成像、荧光成像、光热成像),在人离体标本中实现一体化双模态淋巴结示踪(肉眼成像、荧光成像)。
实施例2
一种纳米材料的原料如表2。

一种纳米材料的制备方法,包括以下步骤:
(1)制备聚多巴胺纳米粒子:将432mg(2.28mmol)盐酸多巴胺加入到216mL去离子水中溶解,调节至50度后加入1368μL 1mol/L的NaOH溶液,充分搅拌反应10h,使用超滤管(30kd)过滤获得聚多巴胺纳米粒子(PDA)。
(2)对聚多巴胺粒子进行聚乙二醇改性(PEG化):取100mg溶于10mL去离子水中,加入适量NaOH溶液,将pH值调节至9,加入双氨基聚乙二醇(NH2-PEG5000-NH2)120mg,常温下充分搅拌12h。最后使用超滤管(30KD)获得PEG改性的聚多巴胺纳米粒子(PEG-PDA)。
(3)制备纳米材料:以投料比1:0.04,分别向经PEG改性的聚多巴胺纳米粒子(PEG-PDA)中加入钆双胺及IR820,充分搅拌4h后过滤(30KD),获得负载了钆双胺及IR820的PEG聚多巴胺纳米粒子(PPIG),记为纳米材料PPIG-2。
实施例3
一种纳米材料的原料如表3。

一种纳米材料的制备方法,包括以下步骤:
(1)制备聚多巴胺纳米粒子:将432mg(2.28mmol)盐酸多巴胺加入到216mL去离子水中溶解,调节至50度后加入456μL 1mol/L的NaOH溶液,充分搅拌反应10h,使用超滤管(30kd)过滤获得聚多巴胺纳米粒子(PDA)。
(2)对聚多巴胺粒子进行聚乙二醇改性(PEG化):取100mg溶于10mL去离子水中,加入适量NaOH溶液,将pH值调节至9,加入双氨基聚乙二醇(NH2-PEG5000-NH2)120mg,常温下充分搅拌12h。最后使用超滤管(30KD)获得PEG改性的聚多巴胺纳米粒子(PEG-PDA)。
(3)制备纳米材料:以投料比1:0.04,分别向经PEG改性的聚多巴胺纳米粒子(PEG-PDA)中加入钆双胺及IR820,充分搅拌4h后过滤(30KD),获得负载了钆双胺及IR820的PEG聚多巴胺纳米粒子(PPIG),记为纳米材料PPIG-3。
由实施例1-3可知,本发明通过调节反应的pH值,就能合成不同粒径大小的聚多巴胺,合成方法简单,容易控制,具有巨大的临床转化前景。
产品效果测试
(1)对实施例1-3制得的纳米材料的粒径、紫外吸收波长、光热性能等进行测试。
图2为纳米材料的扫描电镜图,图2中a为纳米材料PPIG-1的扫描电镜图,其粒径为50-100nm;图2中b为纳米材料PPIG-2的扫描电镜图,其粒径为100-150nm;图2中c为纳米材料PPIG-3的扫描电镜图,其粒径为150-200nm。对实施例1-3制得的纳米材料的水合粒径进行分析,图3为纳米材料PPIG-1的水合粒径图,由图3可知,纳米材料PPIG-1的水合粒径约为87nm;图4为纳米材料PPIG-2的水合粒径图,由图4可知,纳米材料PPIG-2的水合粒径约为134nm;图5为纳米材料PPIG-3的水合粒径图,由图5可知,纳米材料PPIG-3的水合粒径约为170nm。
图6为实施例1-3制得的纳米材料的zeta电位图,其中,横坐标分别为粒径为50-100nm、100-150nm和150-200nm的纳米材料,纵坐标为zeta电位,由图6可知,实施例1-3制得的纳米材料的zeta电位值在-44至-38mV之间。
图7为实施例1-3制得的纳米材料的紫外吸收波长图;图7中,a指纳米材料PPIG-1,b指纳米材料PPIG-2,c指纳米材料PPIG-3。三种纳米材料均表现出特有的IR820紫外吸收峰(波长=820nm)。
图8为实施例1-3制得的纳米材料的MR强度曲线,图8中,横坐标为Gd(NH2)2的浓度,纵坐标为相对MR信号强度,图8中曲线分别为粒径为50-100nm、100-150nm、150-200nm的纳米材料以及Gd(NH2)2。由图8可知,三种纳米材料均能实现MRI下成像,且与MRI造影剂钆双胺具有相类似的信号-浓度曲线。
图9为实施例1-3制得的纳米材料的光热性能图,其中横坐标为时间,纵坐标为温度增加值,图9中三条曲线分别表示,在808nm激光照射(1W/cm2)下持续5分钟,PBS缓冲液中不同粒径大小的纳米材料(浓度为200μg/mL)的温度变化情况。图9中上方插图为在恒定的808nm激光下照射5分钟后,浓度为200μg/mL的三种纳米材料溶液与PBS缓冲液的热图像。
图10为实施例3制得的纳米材料的不同浓度的光热性能图,其中横坐标为时间,纵坐标为温度增加值,图10中曲线分别表示,在808nm激光辐射(1W/cm2)下持续5分钟,PBS缓冲液中实施例3制得的纳米材料(粒径为150-200nm)在不同浓度(0、25、50、100、200、400μg/mL)下的温度变化。图10中上方插图为在恒定的808nm激光下照射5分钟后,在不同浓度下(从左往右依次为浓度0、25、50、100、200、400μg/mL)纳米材料溶液的热图像。
图11为实施例3制得的纳米材料的在不同功率下的光热性能图;其中横坐标为时间,纵坐标为温度增加值,图11中曲线分别表示,在808nm激光辐照下,在不同功率密度(0.125、0.25、0.5、0.75、1W/cm2)下实施例3制得的纳米材料(浓度为200μg/mL)的温度变化情况。图11中上方插图为在恒定的808nm激光下照射5分钟后,在不同功率下(从左往右依次为功率密度0.125、0.25、0.5、0.75、1W/cm2)实施例3制得的纳米材料溶液(浓度200μg/mL)的热图像。
由图9-11可知,在各浓度和各光照功率下,实施例1-3制得的纳米材料的温度曲线平稳光滑,约在5min时达到稳定温度,其光热性能优异,有助于成像的稳定性。
图12为实施例3制得的纳米材料在五个NIR激光辐照周期内的温度变化图;其中横坐标为时间,纵坐标为温度增加值,图12中曲线分别表示,在五个NIR激光辐照(808nm,1W/cm2)周期内,浓度为400μg/mL的纳米材料的温度变化图。每个周期中温度变化情况一致,可见该纳米材料光热性能稳定。
(2)将实施例1-3制得的纳米材料作为示踪剂应用于淋巴结清扫手术中。
图13为小鼠术前/术中一体化多模态淋巴结示踪导航下的淋巴结切除手术示意图。从图13上部分表示,通过局部注射纳米材料后,可实现小鼠腘窝淋巴结的MR成像;从图13下部分表示,基于小鼠腘窝淋巴结MRI定位,随后进行肉眼/荧光/光热三模态成像,在三模态成像定位下进行淋巴结术中精准定位并行淋巴结切除。由13可知,该纳米材料作为示踪剂,具有多模态成像功能,能够有效提升手术淋巴结清扫的准确性和清扫效率。
图14为实施例1制得的纳米材料在人离体胃标本淋巴结光学/荧光双模态示踪示踪导航下淋巴结切除术图。图14中左侧为光学示踪导航下淋巴结切除术的淋巴结光学/荧光图;右侧为光学/荧光双模态示踪导航下淋巴结切除术的光学/荧光图,淋巴组织均被完整切除,未见残留淋巴组织。通过本实验可知,本发明提供的纳米材料作为示踪剂,能够实现淋巴结光学/荧光双模态成像,淋巴组织能够被完整切除,不残留淋巴组织。但对比单纯的淋巴结光学成像,光学/荧光双模态成像能更好地提升淋巴清扫的完整性。
图15为小鼠注射不同粒径纳米材料后的淋巴结MR成像图,在图15中,纵向依次为粒径为50-100nm、100-150nm、150-200nm的纳米材料和Gd(NH2)2,横向代表不同的时间。
图16为小鼠注射不同纳米材料后的淋巴结肉眼成像图,在图16,纵向依次为粒径为50-100nm、100-150nm、150-200nm的纳米材料,横向代表不同的时间。本发明提供的纳米材料能够在淋巴结中滞留不同的时间,可为临床提供灵活的淋巴示踪剂注射时间点。图17为小鼠注射不同粒径纳米材料及IR820染料后的淋巴结荧光成像图,图17中小鼠分别注射50-100nm,100-150nm,150-200nm的纳米材料及IR820染料,并跟踪1,2,4,12,24小时显示的荧光图。由图17可知,本发明提供的三种粒径大小的纳米材料,作为示踪剂,均能专一性地进入淋巴组织,其中注射粒径为50-100nm的纳米材料后,能实现小鼠腘窝淋巴结0-2h的淋巴结示踪;注射粒径为100-150nm纳米材料后,实现2-12h的淋巴结示踪,注射粒径为150-200nm纳米材料后,实现2h-24h的淋巴结示踪。而注射IR820染料后,无法实现淋巴结特异性成像,2h开始出现明显的晕染,IR820染料进入周围组织,无法特异性示踪淋巴结。
图18为小鼠注射不同粒径纳米材料后,利用808nm激光照射局部淋巴结实现淋巴结光热成像图,横向代表不同的激发时间。本发明提供的纳米材料能够在实现不同照射时间下淋巴结的光热成像。
Claims (10)
1.一种纳米材料,其特征在于,包括聚合物改性的聚多巴胺和负载于所述聚合物改性的聚多巴胺上的染料和造影剂;所述聚合物选自双氨基聚乙二醇、聚乙烯亚胺、带巯基的聚乙二醇或透明质酸中的一种。
2.根据权利要求1所述的纳米材料,其特征在于,所述造影剂的负载量为1-3%;所述染料的负载量为60-80%。
3.根据权利要求1或2所述的纳米材料,其特征在于,所述染料为近红外荧光染料;所述造影剂为顺磁性金属盐。
4.根据权利要求1所述的纳米材料,其特征在于,所述纳米材料的粒径为20-300nm。
5.权利要求1-4中任一项所述的纳米材料的制备方法,其特征在于,包括以下步骤:
(1)将聚多巴胺溶解,调节pH值至碱性,加入聚合物,搅拌,过滤,去滤液,制得所述聚合物改性的聚多巴胺;
(2)向步骤(1)制得的所述聚合物改性的聚多巴胺中加入染料和造影剂,搅拌,过滤,去滤液,制得所述纳米材料。
6.根据权利要求5所述的制备方法,其特征在于,步骤(1)中所述聚多巴胺的制备方法,包括以下步骤:将多巴胺或盐酸多巴胺溶解,加入碱液,搅拌,过滤,去滤液制得;所述碱液为氢氧化钠、氢氧化钾、碳酸氢钾或氨水中的至少一种。
7.根据权利要求6所述的制备方法,其特征在于,所述氢氧化钠的浓度为0.5-2mol/L;步骤(1)中所述多巴胺或盐酸多巴胺与所述氢氧化钠的物质的量的比为1:(0.1-1.2)。
8.根据权利要求5所述的制备方法,其特征在于,步骤(3)中,所述聚合物改性的聚多巴胺、染料和造影剂的质量比为1:(0.01-0.08):(0.01-0.08)。
9.权利要求1-4中任一项所述的纳米材料在制备医药材料中的应用。
10.一种淋巴示踪剂,其特征在于,包含权利要求1-4中任一项所述的纳米材料。
