Abstract
本发明公开了一种抗炎抑菌中药复方漱口水及其制备方法,所述中药复方漱口水,由漱口水膏体基质结合中药金银花、地丁、水牛角、赤苓、连翘、丹皮、川连、夏枯草黄芪的提取物制备而成;本发明所述漱口水的通过两次水煮、一次醇沉获得中药组合提取物,接着添加基质进一步制备而得,所述制备方法简单,工艺稳定可规模化生产,能够清新口气、清洁牙齿以及具有抑菌抗炎、免疫调节效力的作用,便于随身携带使用。

技术领域
本发明涉及一种抗炎抑菌中药复方漱口水及其制备方法,还涉及其制备方法。
背景技术
口腔疾病是全世界最普遍的健康问题之一,由口腔疾病造成的负担也在日益加剧。常规的生物或化学制剂用于日常的抑菌或杀菌会造成耐药性的潜在危险,且炎症引起的红肿热痛,会造成患者极度不适。中药成分温和,在一定程度上可以减小耐药性和患者的不适度,在口腔防护具有一定的优势,如中国专利CN201811078894.0公开了一种包含由苍术和桂枝挥发油制备的中漱口水,具有抑制口腔微生物滋生的作用。中国专利CN200810301495.6是一种加入大蓟提取物的用于消炎止血作用的植物漱口水。
目前漱口水在国内市场尚有待开发,大部分仍为国外进口漱口水。市场中的中药类漱口水,一部分漱口水的中药种类较单一,功能也较单一,未发挥出传统中药材相互配伍产生的协同作用;一部分是这些漱口水所使用的抗菌有效成分针对性不强,具有广谱的抗菌性,且进一步导致口腔免疫力下降和降低各组织对损伤的自愈能力。
发明内容
发明目的:本发明针对现有技术中存在的广谱抗菌性漱口水易破坏口腔菌群的问题,提供一种抗炎抑菌中药复方漱口水及其制备方法;还提供了上述中药复方漱口水的制备方法。
技术方案:本发明的所述的一种抗炎抑菌中药复方漱口水及其制备方法,由金银花、地丁、水牛角、赤苓、连翘、丹皮、川连、夏枯草和黄芪的提取物和漱口水膏体基质制备而成;所述的中药提取物各原料的重量份是:金银花30-40份,地丁25-30份,水牛角10-15份,赤苓20份,连翘15-30份,丹皮15份,川连3份,夏枯草15份,黄芪15-20份。
优选的,重量配比是金银花30份,地丁30份,水牛角15份,赤苓20份,连翘15份,丹皮15份,川连10份,夏枯草15份,黄芪15份。
优选的,所述提取物的重量百分比为0.1-1.5%,余量为漱口水基质。
优选的,所述漱口水基质组成的重量百分比为:12-35%保湿剂、0.8-2%表面活性剂、0.1-0.5%甜味剂、0.5-1%香味剂、0.01-0.15%防腐剂、0.1-0.2%焦磷酸钠、0.5-0.15%氟化钠,余量为水。
优选的,所述保湿剂为甘油、聚乙二醇或山梨醇中的一种,表面活性剂为十二烷基硫酸钠或月桂酰肌氨酸钠,甜味剂为木糖醇或甜菊糖,香味剂为薄荷醇、西柚香精、茉莉花香精或西瓜香精中的一种,防腐剂为苯甲酸钠或对羟基苯甲酸乙酯,美白剂为焦磷酸钠和氟化钠。
本发明所述中药复方漱口水的制备方法,包括以下步骤:
(1)称取水牛角,将水牛角加入其质量10-15倍量的水先煎制得混合水溶液A;称取金银花、地丁、赤苓、连翘、丹皮、川连、夏枯草和黄芪混合得中药混合物,将中药混合物与其质量的6-8倍水加入混合水溶液A煮沸提取,过滤得药液;
(2)药渣再加入其总质量的6-8倍水煮沸提取,过滤得药液,合并提取的过滤液,减压浓缩得浓缩液,向浓缩液中加乙醇,搅拌混匀后静置;
(3)过滤除去沉淀得醇沉滤液即为中药组合提取物;
(4)称取中药组合提取物与漱口水基质混合均匀,制得所述中药复方漱口水。
优选的,步骤(2)中向浓缩液中加乙醇,达到浓缩液中乙醇含量为40-60%。
优选的,步骤(4)中中药组合提取物与漱口水基质的比例为1:49。
本发明采用MTT比色法进行了黄芪提取物与中药组合提取物对淋巴细胞增殖影响的实验,发现黄芪中有效成分能够促进淋巴细胞增殖从而达到提高自身免疫力作用,做到正气存内而外邪不可干,增强口腔对病原微生物的抵抗力。护理产品添加中药成分作用于口腔,大多数还停留在利用中药发挥抗菌消炎的作用,从而缓解牙龈出血、促进口腔溃疡恢复。而口腔周围淋巴组织分布较多,黄芪与金银花结合,协同增效,使得在抗菌消炎的同时能够提升自身免疫力,达到有益健康的效果。
有益效果:与现有技术相比,本发明具有如下显著优点:
(1)本发明的一种抑菌抗炎、具有免疫调节效力的中药复方漱口水,可以起到清新口气、保持口腔卫生、清洁和美白牙齿、、防龋齿以及抑菌抗炎,而且具有免疫调节效力的作用;
(2)中药混合物采用两次水煮、一次醇沉获得中药组合提取物,制备方法简单,工艺稳定可规模化生产。

实施例1
中药复方漱口水,包括以下重量份的原料:
1%中药组合提取物、20%甘油与山梨醇、1.5%十二烷基硫酸钠、0.2%木糖醇、0.8%薄荷醇、0.1%苯甲酸钠、0.2%焦磷酸钠、0.1%氟化钠、及余量水,其中,中药提取物的原料药包括:金银花30份,地丁30份,水牛角15份,赤苓20份,连翘15份,丹皮15份,川连10份,夏枯草15份,黄芪15份。
所述的中药复方漱口水的制备方法,包括以下步骤:
按比例先称取水牛角,再另称取金银花,地丁,赤苓,连翘,丹皮,川连,夏枯草及黄芪混合得中药混合物,将水牛角加入其质量15倍量的水先煎15分钟,后加入中药混合物与其质量的7倍水煮沸提取45分钟。过滤得药液,药渣再加入其总质量的7倍水煮沸提取30分钟,过滤得药液,合并2次提取的过滤液,减压浓缩得浓缩液,浓缩至生药量1g/mL,向浓缩液中加乙醇使醇含量为40-60%,搅拌混匀后静置24小时。过滤除去沉淀得醇沉滤液即为中药组合提取物。

1%中药组合提取物、20%聚乙二醇与山梨醇、1.5%月桂酰肌氨酸钠、0.2%木糖醇、0.8%茉莉花香精、0.1%苯甲酸钠、0.2%焦磷酸钠、0.1%氟化钠、及余量水,其中,中药提取物的原料药包括:金银花30份,地丁30份,水牛角15份,赤苓20份,连翘15份,丹皮15份,川连10份,夏枯草15份,黄芪15份。
所述的中药复方漱口水的制备方法,包括以下步骤:

抗菌测试(采用的细菌均为国际标准株)
测试(扩散法-滤纸片法)
对照组:庆大霉素、克林霉素、特比萘芬、市售同类型漱口水
取4组(组A、组B、组C、组D)培养皿,每组10个,将固体培养基趁热与上述4种菌液混匀,均匀铺在培养皿后冷却(组A(接种金黄色葡萄球菌)与克林霉素对照、组B(接种大肠杆菌)与庆大霉素对照、、组C(接种枯草芽孢杆菌)与庆大霉素对照、组D(接种白色念珠菌)与特比萘芬对照),得培育培养皿;
称取实施例1-5制备的中药组合提取物、对照组抗生素、市售同类型漱口水及空白对照各5mL,分别将滤纸片浸泡其中2小时,再将小圆滤纸片按照每个培育培养皿内均匀对称贴附4个;
将贴附有滤纸片的组A、组B、组C培育培养皿置于37℃的恒温培养箱中培养24小时,以及将贴附有滤纸片的组D培育培养皿置于28℃的恒温培养箱中培养24小时后,测量每个培养皿中的四个类圆形抗菌区域的直径并取平均值。
取4组(组A、组B、组C、组D)培养皿,每组10个,将固体培养基趁热与上述4种菌液混匀,均匀铺在培养皿后冷却(组A(接种金黄色葡萄球菌)与克林霉素对照、组B(接种大肠杆菌)与庆大霉素对照、组C(接种枯草芽孢杆菌)与庆大霉素对照、组D(接种白色念珠菌)与特比萘芬对照),得培育培养皿;

将贴附有滤纸片的组A、组B、组C培育培养皿倒置于37℃的恒温培养箱中培养24小时,以及将贴附有滤纸片的组D培育培养皿倒置于28℃的恒温培养箱中培养24小时后,测量每个培养皿中的四个类圆形抗菌区域的直径并取平均值。

采用扩散法进行抗菌测试,如果对应样品具有抗菌性,则会在培养基上显示抗菌区域,通过抗菌区域的直径大小来评估样品抗菌性,直径越大表示抗菌性越强。其中,金黄色葡萄球菌、白色念珠菌、枯草芽孢杆菌、大肠杆菌、均为口腔常见菌,金黄色葡萄球菌是一种革兰氏阳性球菌,常堆积成葡萄串状,易引发口腔炎症,枯草芽孢杆菌和大肠杆菌为常见的革兰氏阴性菌,白色念珠菌是一种真菌,通常存在于正常人口腔,上呼吸道,当机体免疫功能下降时,易侵入细胞引起疾病。
淋巴细胞增殖检测
无菌条件下取出小鼠脾脏,用淋巴细胞分离液分离出淋巴细胞,再用不完全1640培养液调细胞浓度至1×105/mL备用;
在96孔培养板加小鼠脾淋巴细胞悬液(1×105/mL)100μL/孔,对照组加生理盐水100μL/孔。实验组依次加入实施例1-5制备的中药组合提取物100μL;轻轻混匀后置于5%CO2、37℃孵箱中培养24小时,再在无菌条件下向每孔加MTT溶液40mL,轻轻混匀,继续培养4小时,后放入离心机1500r/min离心10分钟,用滤纸吸弃上清液,向每孔加二甲基亚砜150μL,室温下放置15分钟后,测定各孔吸光度A值;
数据处理
促细胞增殖率按下式计算:
促细胞增殖率=(A实验-A对照)/A实验×100%
如图2所示为中药单方对PBMC淋巴细胞增值活性的促进作用:针对加入的黄芪各类化学成分的总提取物进行淋巴细胞增殖试验检测,与空白对照组相比,0~25mg/ml黄芪总皂苷提取物可以增强细胞活性,0~50mg/ml黄芪总黄酮提取物可以增强细胞活性,0~200mg/ml黄芪总多糖提取物可以增强细胞活性,黄芪的皂苷、黄酮、多糖类成分均对淋巴细胞增殖有促进作用。其次是方剂中经典药对和君药金银花和连翘水提物淋巴细胞增殖试验检测,结果显示与空白对照组相比,25~200mg/ml连翘水提物可以增强细胞活性,0~12.5mg/ml连翘水提物可以增强细胞活性,实验证明在一定浓度范围内,金银花和连翘水提物均对淋巴细胞增殖有促进作用。图1为本发明中药组合提取物,结果显示,与空白对照组相比,0.25~1mg/ml中药组合提取物可以增强细胞活性,且活性增强呈现上升趋势,因此中药组合提取物浓度进行淋巴细胞增殖试验检测对淋巴细胞增殖有显著的促进作用。
最小抑菌浓度和最小杀菌浓度的测定
细菌种类:金黄色葡萄球菌、大肠杆菌、枯草芽孢杆菌、白色念珠菌;
测试(二倍稀释法)
准备11个1.5mL的无菌离心管,标号,在第1号离心管中加入100μL中药提取物(实施例1-5制备的中药组合提取物),用移液枪混合均匀,吸取100μL加入2号离心管,以此类推,使各个中药提取物的浓度分别为500.00、250.00、125.00、62.50、31.25、15.62、7.81、3.90、1.95、0.97μg/L。第11号离心管中不添加药物作为阳性对照;
再准备11个大号离心管,标号,分别加入40μL对应序号的中药提取物和4ml培养基,混匀后趁热倒平板,冷却后在每个培养皿三个区域分别接入10μL的菌液,(组A(接种金黄色葡萄球菌)、组B(接种大肠杆菌)、组C(接种枯草芽孢杆菌));
将培养皿放入37℃恒温生化培养箱中培养24小时,记录最小抑菌浓度和最小杀菌浓度。

如表1所示,※代表具有抑菌效应,※越多,抑菌圈平均值越大;中药复方组合物浓度经预实验暂定为0.5g/mL进行试验。每组中药复方组合物加入其余药物包括地丁30份,赤苓20份,丹皮15份,夏枯草15份。由表1可得知:中药复方组合物组成为:金银花30-40份,地丁30份,水牛角15份,赤苓20份,连翘15-30份,丹皮15份,川连3份,夏枯草15份,黄芪15份的抑菌效果最为明显,对金黄色葡萄球菌、大肠杆菌、枯草芽孢杆菌都具有抑制作用,对白色念珠菌没有抑制作用。

1.一种抗炎抑菌中药复方漱口水,其特征在于,所述漱口水由金银花、地丁、水牛角、赤苓、连翘、丹皮、川连、夏枯草和黄芪的提取物和漱口水膏体基质制备而成;所述的中药提取物各原料的重量份是:金银花30-40份,地丁25-30份,水牛角10-15份,赤苓20份,连翘15-30份,丹皮15份,川连3份,夏枯草15份,黄芪15-20份。
2.根据权利要求1所述的中药复方漱口水,其特征在于,所述提取物的重量百分比为0.1-1.5%,余量为漱口水基质。
3.根据权利要求1所述的中药复方漱口水,其特征在于,所述漱口水基质组成的重量百分比为:12-35%保湿剂、0.8-2%表面活性剂、0.1-0.5%甜味剂、0.5-1%香味剂、0.01-0.15%防腐剂、0.1-0.2%焦磷酸钠、0.5-0.15%氟化钠,余量为水。
4.根据权利要求1所述的中药复方漱口水,其特征在于,所述保湿剂为甘油、聚乙二醇或山梨醇中的一种,表面活性剂为十二烷基硫酸钠或月桂酰肌氨酸钠,甜味剂为木糖醇或甜菊糖,香味剂为薄荷醇、西柚香精、茉莉花香精或西瓜香精中的一种,防腐剂为苯甲酸钠或对羟基苯甲酸乙酯,美白剂为焦磷酸钠和氟化钠。
5.权利要求1所述的抑菌抗炎中药复方漱口水的制备方法,其特征在于,包括以下步骤:
(1)称取水牛角,将水牛角加入其质量10-15倍量的水先煎制得混合水溶液A;称取金银花、地丁、赤苓、连翘、丹皮、川连、夏枯草和黄芪混合得中药混合物,将中药混合物与其质量的6-8倍水加入混合水溶液A煮沸提取,过滤得药液;
(2)药渣再加入其总质量的6-8倍水煮沸提取,过滤得药液,合并提取的过滤液,减压浓缩得浓缩液,向浓缩液中加乙醇,搅拌混匀后静置;
(3)过滤除去沉淀得醇沉滤液即为中药组合提取物;
(4)称取中药组合提取物与漱口水基质混合均匀,制得所述中药复方漱口水。
6.根据权利要求5所述的中药复方漱口水,其特征在于,步骤(2)中,向浓缩液中加乙醇,达到浓缩液中乙醇含量为40-60%。
7.根据权利要求5所述的中药复方漱口水,其特征在于,步骤(4)中,所述漱口水基质组成为保湿剂、表面活性剂、甜味剂、香味剂、防腐剂、焦磷酸钠和氟化钠,余量为水。
