Abstract
本发明提出一种用于抗犬细小病毒的单链抗体、制备方法及其应用,属于基因工程领域,能够解决常规抗体对犬细小病治疗效果不理想的技术问题。其中,用于抗犬细小病毒的单链抗体为耦联信号肽的单链抗体,其氨基酸序列如SEQ ID NO.1所示,核苷酸序列如SEQ ID NO.3所示,具体通过以下步骤制备得到:(1)基因合成与表达载体构建;(2)质粒转化与双酶切鉴定;(3)目的蛋白诱导表达;(4)目的蛋白纯化;(5)目的蛋白抗原结合活性、细胞增殖、细胞毒性以及对犬细小病毒抑制效果检测。本发明能够应用于抗犬细小病以及制备抗细小病毒药物制品方面。
Description
技术领域
本发明属于基因工程领域,尤其涉及一种用于抗犬细小病毒的单链抗体、制备方法及其应用。
背景技术
犬细小病毒(Canineparvovirus,CPV),属细小病毒科,细小病毒属,猫细小病毒亚群的单链DNA病毒,其结构蛋白VP2具有中和抗体的抗原表位。犬细小病主要有两种临床表现型,一种是出血性肠炎型,表现为剧烈呕吐、血便、白细胞大量减少;另一种是非化脓性心肌炎型,病犬会突然死亡。该病传播速度快,发病率高,幼犬死亡率高达70%,成年犬无明显临床症状,但是隐性带毒的现象普遍存在。
目前,常用的犬细小病治疗策略以注射疫苗和注射特异性抗体为主。其中,有研究者对CPV疫苗的免疫效果进行检测发现,仅有不到20%的疫苗能产生具有保护力的抗体。而且临床上治疗犬细小病的方法主要针对其急性脱水症状,给病犬输液进行补液,此种治疗效果有限;市场上常用的抗体是鼠源性IgG,病犬反复或者长期使用,lgG中Fc片段的鼠源成分会使犬对其免疫应答后产生特异性抗体,从而不利于犬细小病的治疗。另外,大分子抗体不能有效中和细胞内病毒,影响治疗效果。可见,这两种方式的治疗效果均不理想。
鉴于上述两种治疗方式的局限性,具有可溶性好、稳定性强、亲和力高等特点的单链抗体为治疗犬细小病提供了新的可能,但是仅采用常规的单链抗体进行犬细小病治疗的效果仍未达到理想状态,如何采用基因工程手段增强单链抗体的治疗效果,从而研发出一种体内组织渗透性好、治疗效果显著的新型单链抗体是解决上述问题的关键。
发明内容
本发明针对常规单链抗体对犬细小病治疗效果不显著等的技术问题,提出一种体内组织渗透性好、治疗效果显著的用于抗犬细小病毒的单链抗体、制备方法及其应用。
为了达到上述目的,本发明采用的技术方案为:
用于抗犬细小病毒的单链抗体,所述用于抗犬细小病毒的单链抗体为耦联信号肽的单链抗体,其氨基酸序列如SEQ ID NO.1所示。
在一实施方式中,所述耦联信号肽的单链抗体中信号肽的核苷酸序列如SEQ IDNO.2所示,耦联信号肽的单链抗体的核苷酸序列如SEQ ID NO.3所示。
本发明还提供了一种用于抗犬细小病毒的单链抗体的制备方法,包括以下步骤:
基因合成与表达载体构建:人工合成耦联信号肽的单链抗体基因全长序列,并构建含有耦联信号肽的单链抗体基因全长序列的原核表达载体pET-32a(+)-TAT-VR质粒;
质粒转化与双酶切鉴定:将所述原核表达载体pET-32a(+)-TAT-VR质粒转化至宿主菌中,依次经过平板涂布与培养、双酶切鉴定以及测序后,确定耦联信号肽的单链抗体基因是否插入成功;
目的蛋白诱导表达:将插入成功的宿主菌加入含IPTG的液体培养基中进行蛋白诱导表达;
目的蛋白纯化:蛋白诱导表达结束后,收集宿主菌菌体进行蛋白纯化,获得纯化后的TAT-VR蛋白。
在一实施方式中,在所述目的蛋白诱导表达步骤中,诱导表达条件为37℃,8-10h。
在一实施方式中,在所述目的蛋白纯化步骤中,采用不同浓度咪唑的平衡缓冲液进行目的蛋白洗脱。
在一实施方式中,所述不同浓度咪唑的平衡缓冲液中咪唑浓度分别为50mM、100mM、300mM。
在一实施方式中,所述制备方法还包括对纯化后的TAT-VR蛋白进行抗原结合活性检测、细胞增殖试验、细胞毒性试验以及对犬细小病毒抑制检测。
本发明又提供了一种用于抗犬细小病毒的单链抗体在抗犬细小病中的应用。
本发明又提供了一种用于抗犬细小病毒的单链抗体在制备抗犬细小病毒药物制品中的应用。
与现有技术相比,本发明的优点和积极效果在于:
1、本发明提供的用于抗犬细小病毒的单链抗体是一种在原核表达系统中制备得到的耦联信号肽的单链抗体,其中,信号肽可通过静电相互作用,引起磷脂肽丝氨酸从质膜内转移到质膜外,进而介导信号肽共价结合的单链抗体被转移至细胞内部,中和胞内病毒,达到快速治愈的目的,同时该单链抗体也能够弥补常规抗体的不足,其分子质量小,体内组织渗透性好,极易通过血管或组织到达靶部位;
2、本发明提供的用于抗犬细小病毒的单链抗体在抗犬细小病以及制备抗犬细小病药物制品中具有广阔的应用前景。

具体实施方式
下面将对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
本发明实施例提供了一种用于抗犬细小病毒的单链抗体,所述用于抗犬细小病毒的单链抗体为耦联信号肽的单链抗体,其氨基酸序列如SEQ ID NO.1所示。
在上述技术方案中,本发明通过在原核表达系统中制备得到一种耦联信号肽的单链抗体,该抗体中的信号肽可通过静电相互作用,引起磷脂肽丝氨酸从质膜内转移到质膜外,进而介导信号肽共价结合的单链抗体被转移至细胞内部,中和胞内病毒,达到快速治愈的目的,同时该单链抗体也能够弥补常规抗体的不足,其分子质量小,体内组织渗透性好,极易通过血管或组织到达靶部位。
在一具体实施方式中,所述耦联信号肽的单链抗体中信号肽的核苷酸序列如SEQID NO.2所示,耦联信号肽的单链抗体的核苷酸序列如SEQ ID NO.3所示。
本发明实施例还提供了一种用于抗犬细小病毒的单链抗体的制备方法,包括以下步骤:
S1、基因合成与表达载体构建:人工合成耦联信号肽的单链抗体基因全长序列,并构建含有耦联信号肽的单链抗体基因全长序列的原核表达载体pET-32a(+)-TAT-VR质粒;
S2、质粒转化与双酶切鉴定:将所述原核表达载体pET-32a(+)-TAT-VR质粒转化至宿主菌中,依次经过平板涂布与培养、双酶切鉴定以及测序后,确定耦联信号肽的单链抗体基因是否插入成功;
S3、目的蛋白诱导表达:将插入成功的宿主菌加入含IPTG的液体培养基中进行蛋白诱导表达;
S4、目的蛋白纯化:蛋白诱导表达结束后,收集宿主菌菌体进行蛋白纯化,获得纯化后的TAT-VR蛋白。
在一具体实施方式中,在所述目的蛋白诱导表达步骤中,诱导表达条件为37℃,8-10h。
在一具体实施方式中,在所述目的蛋白纯化步骤中,采用不同浓度咪唑的平衡缓冲液进行目的蛋白洗脱。
在一具体实施方式中,所述不同浓度咪唑的平衡缓冲液中咪唑浓度分别为50mM、100mM、300mM。
在一具体实施方式中,所述制备方法还包括对纯化后的TAT-VR蛋白进行抗原结合活性检测、细胞增殖试验、细胞毒性试验以及对犬细小病毒抑制检测。
本发明实施例又提供了一种用于抗犬细小病毒的单链抗体在抗犬细小病中的应用。
本发明实施例又提供了一种用于抗犬细小病毒的单链抗体在制备抗犬细小病毒药物制品中的应用。
为了更清楚详细地介绍本发明实施例所提供的用于抗犬细小病毒的单链抗体、制备方法及其应用,下面将结合具体实施例进行描述。
实施例1
本实施例提供了用于抗犬细小病毒的单链抗体的制备方法,具体为:
(1)人工合成TAT-VR基因片段,约为800bp,序列信息如下,其中,单下划线所标注序列为信号肽序列,两条双下划线所标注序列为Linker序列:
将酶切后的TAT-VR与载体pET-32a(+)进行连接,获得含有TAT-VR基因的原核表达载体pET-32a(+)-TAT-VR质粒;
(2)质粒转化与双酶切鉴定:将pET-32a(+)-TAT-VR质粒转化至宿主菌感受态细胞中,涂布氨苄抗性平板,过夜培养后,挑取单菌落,提取质粒,进行EcoRI和HindIII双酶切鉴定,酶切体系见表1,按照该体系双酶切结束后,将酶切产物进行琼脂糖凝胶电泳,紫外检测并分析结果(见图1);

从图1可知,TAT-VR基因片段约为800bp,pET-32a约为5900bp,与预期相符,将酶切鉴定正确的质粒送往测序公司测序,结果正确,确定TAT-VR基因插入成功。
(3)目的蛋白诱导表达:
将插入成功的宿主菌接种于含有IPTG的LB液体培养基中,37℃条件下进行蛋白诱导表达8h,其中,IPTG浓度为1mmol/L;将诱导后的菌体8000rpm/min离心10min,使用预冷的PBS溶液洗涤1次,使用包涵体破碎Buffer重悬沉淀后,用超高压低温破碎机破碎菌体,至溶液为半透明;收集破碎产物分离上清和沉淀,处理后进行Western Blot检测确定目的蛋白的表达;
实验结果如图2所示,通过Western Blot检测发现,目的蛋白TAT-VR以包涵体形式表达。
(4)目的蛋白纯化:将目的蛋白样品通过平衡好的Ni-IDA亲和层析柱,收集流穿液,然后使用咪唑浓度分别为50mM、100mM、300mM的平衡缓冲液洗脱目的蛋白,并收集洗脱液进行检测,纯化结果见图3,该图的泳道信息如下:
泳道M:SDS-PAGE Protein Marker,泳道1:包涵体溶解离心后上清,泳道2:上清同Ni-IDA孵育后流出液,泳道3-4:50mM咪唑的洗脱组分;泳道5-8:100mM咪唑的洗脱组分;泳道9-11:300mM咪唑的洗脱组分。
从图中可知,当咪唑浓度为100mM时,洗脱效率最高。将收集的纯化蛋白加入到处理后的透析袋中,4℃环境下,透析到透析缓冲液Buffer中复性,复性后透析于储存液Buffer中,离心收集上清蛋白,用0.22μm滤器过滤除菌后分装,冻存至-80℃储存备用。
实施例2
本实施例提供了ELISA测定TAT-VR蛋白结合抗原活性方法,该方法利用双抗夹心ELISA检测TAT-VR蛋白结合VP2抗原的活性,具体为:
分别将浓度为0μM、5μM、11μM、16μM的TAT-VR蛋白包被ELISA板,孵育CPV-VP2蛋白,以PBS作为阴性对照,然后继续孵育VP2一抗、二抗,TMB显色后使用Epoch微孔分光光度计测定每孔的OD450值,实验结果见图4。
从图4可知,在体外,本发明所提供的TAT-VR蛋白能够有效地结合CPV结构蛋白VP2,且呈现剂量依赖性。
本发明实施例的实验数据采用SPSS(20.5)软件的two-wayANOVA进行双因素方差分析,数据以(P<0.05)为显著水平,图中*表示差异显著(P<0.05);**表示差异极显著(P<0.01)。
实施例3
本实施例提供了TAT-VR蛋白对F81细胞增殖的影响试验及对F81细胞毒性试验,具体为:
(1)TAT-VR蛋白对F81细胞增殖的影响试验
首先将F81细胞(接种密度为5000个cell/孔)接种于96孔板中,每孔100μL,置于培养箱内培养,然后细胞贴壁后每孔加入10μL的CCK8试剂,继续在培养箱孵育(0.5h、1h、1.5h、2h、2.5h、3h、3.5h、4h)后,测定在450nm处的吸光度,测定结果如图5所示。
实验结果:从图5可知,当加入CCK8试剂后孵育3h,细胞孔吸光度达到平台期,因此,CCK8最佳孵育时间为3h。图中*表示差异显著(P<0.05);**表示差异极显著(P<0.01)。
(2)TAT-VR蛋白对F81细胞毒性试验
首先将F81细胞(接种密度为5000个cell/孔)接种于96孔板中,每孔100μl,置于培养箱内培养,然后分别加入10μL不同浓度(4μM、6μM、8μM、10μM、12μM)的TAT-VR蛋白到孔内培养基,在培养箱内培养24h后,每孔内加入10μLCCK8试剂,孵育3h后测定吸光值,按照计算公式计算细胞存活率。结果如图6所示。
实验结果:从图6可知,当TAT-VR蛋白的浓度低于8μM时,F81细胞活性接近于未处理的对照组,但是当蛋白浓度达大于或等于10μM时,细胞活性降低到对照细胞90%之下,这表明TAT-VR浓度在小于或等于8μM时对F81细胞没有毒性。
实验数据采用SPSS(20.5)软件的one-wayANOVA进行单因素方差分析,数据以(P<0.05)为显著水平,图中*表示差异显著(P<0.05);**表示差异极显著(P<0.01)。
实施例4
本实施例提供了F81细胞对TAT-VR蛋白的摄取实验,具体为:
将F81细胞接种于48孔板,待细胞长到80%,加入8μM的TAT-VR蛋白,孵育不同时间(0h、6h、12h、24h)后进行间接免疫荧光试验。F81细胞摄取TAT-VR蛋白的结果如图7-8所示。
实验结果:从图7所示结果可知,TAT-VR蛋白能够进入细胞内部,随着时间延长,细胞内TAT-VR含量增加;将细胞图片按照10张为一组,统计3组,计算细胞对TAT-VR蛋白摄取率,结果如图8所示,TAT-VR蛋白能够进入细胞内部,并在24h后在细胞中分布达到80%上下。
实施例5
本实施例提供了TAT-VR蛋白对犬细小病毒的抑制实验,具体为:
将F81细胞接种于24孔板,待细胞达到80%左右时,接种0.1TCID50的CPV(犬细小病毒),孵育6h后更换培养基为含不同浓度(1μM、2μM、3μM、4μM)TAT-VR蛋白的细胞维持液,同时设置未加蛋白的空白对照组,培养24h后观察细胞病变,实验结果如图9所示。
实验结果:与空白对照组相比,随着TAT-VR蛋白浓度(1μM、2μM、3μM、4μM)的不断增加,当使用TAT-VR蛋白的浓度为3μM时,F81细胞边界越来越清晰,细胞状态也越来越好。结果表明,TAT-VR蛋白对于犬细小病毒有一定程度的抑制作用。
实施例6
本实施例提供了TAT-VR蛋白对胞内犬细小病毒的抑制实验,具体为:
将F81细胞接种于48孔板,孵育0.1个TCID50的CPV(犬细小病毒),培养6h后更换培养基为含不同浓度(0μM、1μM、2μM、3μM、4μM)TAT-VR蛋白的细胞维持液,不同时间点(24h、36h、48h)收集样品,分为细胞悬液总样、上清、细胞沉淀三组,分别测定其中病毒滴度,实验结果见图10。
实验结果:从图10可知,孵育4μM的TAT-VR蛋白后,无论是胞内还是胞外的CPV均被显著抑制。并且随着孵育时间延长,TAT-VR蛋白对病毒抑制效果更明显。注:实验数据采用SPSS(20.5)软件的one-wayANOVA进行单因素方差分析,数据以(P<0.05)为显著水平,图中*表示差异显著(P<0.05);**表示差异极显著(P<0.01);***表示差异及其显著(P<0.001)。
实施例7
本实施例提供了TAT-VR蛋白对犬细小病毒vDNA的抑制实验,具体为:
将F81细胞接种于6孔板,待细胞长到80%左右时,接种0.1个TCID50的CPV(犬细小病毒),孵育6h后更换培养基为含不同浓度(0μM、1μM、2μM、3μM、4μM)的TAT-VR蛋白的维持液,孵育48h后收集样品(结果如图11所示)。同时将TAT-VR蛋白(4μM)组在不同时间点(12h、24h、36h、48h)收集样品。提取细胞内Hirt DNA,设计CPV vp基因特异性引物,进行qPCR分析vDNA的含量,实验结果如图12所示。

实验结果:如图12所示,和VR组相比,TAT-VR蛋白对vDNA抑制效果更佳,并存在剂量依赖和时间依赖性。注:实验数据采用SPSS(20.5)软件的one-wayANOVA进行单因素方差分析,数据以(P<0.05)为显著水平,图中*表示差异显著(P<0.05);**表示差异极显著(P<0.01);***表示差异及其显著(P<0.001)。

Claims (7)
1.用于抗犬细小病毒的单链抗体,其特征在于,所述用于抗犬细小病毒的单链抗体为耦联信号肽的单链抗体,其核苷酸序列如SEQ ID NO.1所示。
2.如权利要求1所述的用于抗犬细小病毒的单链抗体的制备方法,其特征在于,包括以下步骤:
基因合成与表达载体构建:人工合成耦联信号肽的单链抗体基因全长序列,其序列如SEQ ID NO.1所示,并构建含有耦联信号肽的单链抗体基因全长序列的原核表达载体pET-32a(+)-TAT-VR质粒;
质粒转化与双酶切鉴定:将所述原核表达载体pET-32a(+)-TAT-VR质粒转化至宿主菌中,依次经过平板涂布与培养、双酶切鉴定以及测序后,确定耦联信号肽的单链抗体基因是否插入成功;
目的蛋白诱导表达:将插入成功的宿主菌加入含IPTG的液体培养基中进行蛋白诱导表达;
目的蛋白纯化:蛋白诱导表达结束后,收集宿主菌菌体进行蛋白纯化,获得纯化后的TAT-VR蛋白。
3.根据权利要求2所述的用于抗犬细小病毒的单链抗体的制备方法,其特征在于,在所述目的蛋白诱导表达步骤中,诱导表达条件为37℃,8-10h。
4.根据权利要求2所述的用于抗犬细小病毒的单链抗体的制备方法,其特征在于,在所述目的蛋白纯化步骤中,采用不同浓度咪唑的平衡缓冲液进行目的蛋白洗脱。
5.根据权利要求4所述的用于抗犬细小病毒的单链抗体的制备方法,其特征在于,所述不同浓度咪唑的平衡缓冲液中咪唑浓度分别为50mM、100mM、300mM。
6.根据权利要求2所述的用于抗犬细小病毒的单链抗体的制备方法,其特征在于,所述制备方法还包括对纯化后的TAT-VR蛋白进行抗原结合活性检测、细胞增殖试验、细胞毒性试验以及对犬细小病毒抑制检测。
7.如权利要求1所述的用于抗犬细小病毒的单链抗体在制备抗犬细小病毒药物制品中的应用。
