Wherein the total amount of the Chinese medicinal materials is 44, the total weight is 2.145kg, the addition amount of the original stock mature vinegar is 6.435kg, and the transdermal agent is added according to 2.5% of the weight of the upper layer mixed liquid medicine after solid-liquid separation.
2. The preparation process is shown in figure 1:
step 1: soaking Chinese medicinal materials
Crushing the traditional Chinese medicines, then putting the crushed traditional Chinese medicines into a low-temperature reaction kettle, adding raw stock mature vinegar to soak the traditional Chinese medicines to obtain a solid-liquid mixture, wherein the soaking time is 22 days;
step 2: adding a solvent, wherein the solvent is methanol and isopropanol, and the adding amount of the solvent is 6% of the mass of the solid-liquid mixture;
and step 3: cooling the solid-liquid mixture soaked in the low-temperature reaction kettle to-10 ℃, increasing the gas pressure to 0.5MPa, and reacting for 30 minutes;
and 4, step 4: continuously cooling the solid-liquid mixture soaked in the low-temperature reaction kettle to-15 ℃, increasing the pressure to 1.5MPa, converting gas into liquid, increasing the volume of the material, and reacting for 40 minutes;
and 5: heating the solid-liquid mixture soaked in the low-temperature reaction kettle to 5 ℃ of critical temperature, reducing the pressure to the critical pressure, and reacting for 40 minutes to obtain an extraction mixture;
step 6: carrying out solid-liquid separation on the extraction mixture and precipitating for 6 hours to obtain upper-layer mixed liquid medicine;
and 7: transdermal treatment of mixed medicinal liquid
Adding a transdermal agent into the mixed liquid medicine obtained in the step 6, stirring for 20 minutes, and uniformly mixing to obtain the tumor cancer traditional Chinese medicine, wherein the transdermal agent is artemisia seed penetrating fluid, and the addition amount of the transdermal agent is 2.5% of the mixed liquid medicine;

Wherein the total weight of the Chinese medicinal materials is 1.88kg, the addition amount is 5.64kg, and the transdermal agent is added in an amount of 2.5% of the weight of the upper layer mixed medicinal liquid after solid-liquid separation.

Wherein the total weight of the Chinese medicinal materials is 2.41kg, the addition amount is 7.23kg, and the transdermal agent is added in an amount of 2.5% of the weight of the upper layer liquid medicine after solid-liquid separation.

Secondly, the invention is used for the inhibition effect experiment of breast cancer cells
Experimental methods
(1) Collecting breast cancer cells MCF-7 of logarithmic growth phase, and digesting with trypsin to obtain cell suspension with cell sap concentration of about 1 × 105Per liter;
(2) inoculating the cell sap into a 96-well plate, and adding 100ul of the cell sap into each well;
(3) the culture medium containing the Chinese medicine of example 1, the Chinese medicine of example 2, the Chinese medicine of example 3, the cyclophosphamide as a control drug and a solvent control was added to the well plate, and the concentration of the culture medium was controlled to 5g/L, and 100ul was added per well.
(4) After culturing the culture medium at 37 ℃ for 3 days, removing the old culture medium, adding 100ul of MTT culture solution into each well, wherein the concentration of the MTT culture solution is 0.5g/L, continuously culturing for 4 hours at 37 ℃, removing the old culture medium again, adding 100ul of DMSO into each well, and uniformly shaking.
(5) The optical density value was measured with a microplate reader at 570 nm.
(6) Based on the measured values, the survival rate of cancer cells was calculated according to the following formula:

As can be seen from the above Table 1, the drug of the present invention has a very good inhibitory effect on breast cancer cells in vitro.
Thirdly, the treatment effect of the medicine of the invention on the pain symptoms of patients
60 breast cancer patients are divided into two groups, wherein 30 breast cancer patients are divided into a control group and 30 breast cancer patients are divided into an experimental group, wherein 60 breast cancer patients are female and are classified between 45-60 years old according to the pain degree of the chief complaints, the scoring standard is shown in the following table 2, in order to reduce the pain of the patients, the control group of the experiment adopts tramadol sustained-release tablets for treatment, the experimental group adopts tramadol sustained-release tablets and the external traditional Chinese medicine prepared in the embodiment 1 of the invention to be externally applied to the related acupuncture points and pain parts of the pain parts, the dosage is 3-4 times per day, and the external traditional Chinese medicine is repeatedly applied for 2-3 times after each air drying.

The collected data were analyzed using statistical software SPSS 22.0, and the differences were statistically significant (P < 0.05) as shown in table 3 below.

From the results, it can be seen that the effect of the pain treatment only by adopting western medicines is inferior to that of the pain treatment by adding the western medicines into the traditional Chinese medicine of the invention, particularly, after the medicine is stopped, the western medicine treatment method cannot cure the root cause, and the external traditional Chinese medicine of the invention can play a role in fundamentally treating the pain, which has a very important significance for cancer patients.
Fourth, the typical clinical experiment of the medicament of the invention
60 breast cancer patients were divided into two groups, 30 in the control group and 30 in the experimental group, 60 of which were female and aged between 45-60 years, and breast cancer tumor sizes were recorded.
The application of the medicine of the invention is then carried out on the experimental group, and the medicine is applied to the cancer area or the corresponding acupuncture points, generally 3-4 times per day.
Statistics shows that the effective rate reaches 98.5 percent after 20-30 times of treatment by using the medicament of the embodiment 1 of the invention.
The clinic for other cancers is as follows:
patient 1: lower limb edema, chest distress and extremely difficult pharynx and food of patients with squamous carcinoma, and gastroscopy result; squamous cancer cells and esophagus are about 28-32 cm from incisor teeth, a nodular lump is seen on the right front wall and protrudes to the cavity, the surface is eroded, becomes brittle and is easy to bleed, the external liquid is applied between the dove tail point and the lower epigastric point, and the nodular lump is obviously reduced after the external liquid for tumor is used for 20 days.
Patient 2: the liquid for external use is difficult to swallow, is fond of drinking water, is stuffy in chest and stomachache, is diagnosed as esophagus cancer in a hospital, has a lump at the 28 cm position of the lower section of the esophagus, and has reduced symptoms and greatly increased appetite after being used for 3 weeks.
Therefore, the external traditional Chinese medicine has the following characteristics: the external liquid for treating the tumor is simple and convenient to use, quick in effect, free of injection and medication, free of wound and pain, exact in curative effect and suitable for various tumors.
The externally applied traditional Chinese medicine for treating the tumor is scientifically matched with the datura flower which is used for treating pain of anesthesia in ancient Huatuo, and the raw myrrh and the raw frankincense, and has strong analgesic effect in cooperation, particularly has the effect of relieving pain of long-term severe pain; can relieve pain caused by various tumors. The product has strong medicine property, safe intervention of medicine effect and no toxic or side effect.
The acupoint pain point administration technology of the externally applied traditional Chinese medicine for treating the tumor directly acts on a focus, continuously acts on cancer cells along meridians and collaterals and blood vessels, and initiates attack on the cancer cells; reducing tumor, controlling tumor growth, reducing compression of tumor on peripheral nerve, promoting penetration of effective components of topical liquid medicine for treating tumor to focus part, and reducing cancer cell diffusion.

step 1: soaking Chinese medicinal materials
Soaking the traditional Chinese medicinal materials of any one of claims 1-5 in a low-temperature reaction kettle by using original stock mature vinegar to obtain a solid-liquid mixture;
step 2: adding a solvent, wherein the solvent is methanol and isopropanol;
and step 3: cooling the solid-liquid mixture soaked in the low-temperature reaction kettle to-8 to-12 ℃, increasing the gas pressure to 0.3 to 0.6MPa, and reacting for at least 20 minutes;
and 4, step 4: continuously cooling the solid-liquid mixture soaked in the low-temperature reaction kettle to-13 to-16 ℃, increasing the pressure to 1.3 to 1.6MPa, converting gas into liquid, increasing the volume of the material, and reacting for at least 30 minutes;
and 5: heating the solid-liquid mixture soaked in the low-temperature reaction kettle to 5 ℃ of critical temperature, reducing the pressure to the critical pressure, and reacting for 30-60 minutes to obtain an extraction mixture;
step 6: carrying out solid-liquid separation on the extraction mixture to obtain upper-layer mixed liquid medicine;
and 7: transdermal treatment of mixed medicinal liquid
And (4) adding a transdermal agent into the mixed liquid medicine obtained in the step (6) and mixing to obtain the tumor cancer traditional Chinese medicine.
