Description
Technical Field
The invention belongs to the technical field of poultry breeding, and particularly relates to a sperm diluent suitable for a large-scale poultry farm and a preparation method thereof.
Background
At present, artificial insemination is the most main breeding mode in chicken farms with intensive cultivation. The artificial insemination is realized by collecting semen of the cock, adding the semen diluent according to a certain dilution ratio to achieve the purpose of increasing the semen volume, and a technician injects the diluted semen into the reproductive tract of the hen by a means of turning the anus of the hen by using a rubber-tipped dropper to complete the insemination process. Sperm are highly sensitive to reactive oxygen species because their plasma membranes are rich in polyunsaturated fatty acids, which are the main targets of oxidation, while their cytoplasm is largely confined to the mid-segment, with few antioxidant mechanisms to provide adequate protection against oxidative damage. Active oxygen in semen may lead to lipid peroxidation, DNA fragmentation, cellular communication and alterations in the enzymatic pathways, which in turn are associated with loss of viability, low fertilization rates or embryo developmental disorders. In the process of collecting the semen of the cock to artificially inseminate the hen, the sperms of the poultry are easily damaged by active oxygen, which causes damage to plasma membranes, thereby reducing the vitality of the sperms. For large-scale poultry farms, there is a long time lag from harvest to hen insemination and the sperm are more vulnerable to reactive oxygen species. Under the conditions of large-scale intensive breeding and artificial insemination of poultry, salpingitis of hen groups occurs occasionally, and a certain negative influence is generated on the artificial insemination process, for example, the insemination is difficult due to the pathological changes of the oviduct, and the sperm is adversely affected by inflammatory factors in the oviduct, so that the egg laying and death and culling of the breeding hens can be directly reduced. In order to deal with the situation, a sperm diluent needs to be developed, which not only needs to provide sperm energy, keep acid-base balance and maintain the balance of internal and external osmotic pressures of the sperm, but also needs to provide additional antioxidant protection, further can reduce the incidence rate of poultry salpingitis, is beneficial to improving the fertilization rate of poultry eggs, and thus increases the economic benefit of a farm.
