Abstract
本发明公开了能提高肉鸡饲料转化效率和促生长的中草药添加剂,根据中医药理及其配方中各组分所含的主要生物活性物质种类等因素,用连翘、当归、黄芪、山楂等十味中草药组成添加剂,对广西东兰乌鸡进行饲养和代谢试验,研究其对乌鸡生长性能及饲料养分利用效率的影响,探索其作为抗生素替代物的可能性,为“禁抗”后寻找抗生素替代物提供科学依据。

饲料作为鸡每天的营养来源,与鸡的身体机能、免疫能力和生长性能息息相关。在现代的规模化与集约化养殖过程中,疾病的防控、家禽的饲养量、成活率、饲料报酬和生长性能是养殖效益的主要来源。目前,畜禽养殖现代化、规模化、集约化程度越来越高,养殖者采取了提高饲养密度、饲喂高营养饲料、长期在饲料中添加抗生素和化学药物来预防疫病、频繁使用消毒剂、改进家禽的养殖环境等措施。而这些措施的实行极容易造成应激反应,导致家禽机体内环境紊乱,免疫力降低;饲喂高营养饲料、也易造成较大的消化和代谢负担;大剂量、长期使用抗生素及化学药物添加入饲料,则常常会损伤家禽的消化器官与机能,引发采食量下降,而且导致病原耐药性和药物残留的产生,甚至引起肝脏药物代谢性中毒,这样会间接对经常食用鸡肉和鸡蛋的人体健康造成危害;使家禽处于长期高强度的抗生素和化学药物压力以及高负荷、高应激的生理临界状态。在这种状态下家禽极易发生疫病,反而降低生产性能,同时造成肉、蛋等产品的药物残留,由于高密度的养殖,鸡机体的相应机能、免疫能力也会下降,作为被人类食用的家禽,也会相应影响人的身体健康。
饲用抗生素具有良好的抗病与促生长性能,曾长期在我国畜禽生产中应用,并为我国畜禽养殖的发展起过重要作用。但大量使用后所产生的副作用及风险使我国不得不采取了“禁抗”。然而,在“禁抗”后的当今,却没有适当的替代物,因此,寻找抗生素相应的替代物已成了当务之急。诸多研究表明:传统中草药含有多种生物活性物质,具有良好的抗菌、抗病和促生长作用,且复方中草药添加剂因含有更多种类的生物活性物质,故作用效果更好。目前已有相关的研究,例如中国专利申请号201410225931.1一种绿色无药残肉鸡养殖中使用的中药制剂、制备方法及使用中药制剂养殖肉鸡的方法,所述中药制剂包括:(1)用于肉鸡幼禽阶段的增效免疫中药制剂,含有以下原料组分:黄芪95-105份、党参95-105份、当归55-65份、柴胡35-45份、贯众35-45份和防风35-45份;(2)用于肉鸡新城疫防疫的增效免疫中药制剂,含有以下原料组分:黄芪95-105份、白术95-105份、连翘75-85份、菟丝子75-85份、杜仲75-85份和甘草35-45份;(3)用于肉鸡呼吸道疾病防疫的增效免疫中药制剂,含有以下原料组分:石膏95-105份、桑叶25-35份、麦冬25-35份和甘草10-20份。
虽然这些研究已初步显示了中草药作为抗生素替代物的可行性,但还有诸多问题尚待解决:如添加剂的质量、其中生物活性物质的作用效率、作用效果的稳定性及其机理等等。因此,仍需进一步深入研究。
【发明内容】
针对现有技术中缺乏对东兰乌鸡生长性能及日粮养分代谢的研究,本发明提供了能提高肉鸡饲料转化效率和促生长的中草药添加剂,根据中医药理及其配方中各组分所含的主要生物活性物质种类等因素,用连翘、当归、黄芪、山楂等十味中草药组成添加剂,对广西东兰乌鸡进行饲养和代谢试验,研究其对乌鸡生长性能及饲料养分利用效率的影响,探索其作为抗生素替代物的可能性,为“禁抗”后寻找抗生素替代物提供科学依据。

本发明中,各组分作用如下:
连翘:属性苦,微寒,归肺、心、胆经,具有清热解毒,消肿散结,疏散风热,有广谱抗菌作用;主要含有苯乙醇苷类、木脂素及其苷类、C6-C2天然醇及其苷类、黄酮类、萜类、挥发油等有效成分。
当归:温、甘、辛、归肝、心、脾经,补血、活血、止痛、润肠;主要有效成分为挥发油、多糖、黄酮、有机酸和氨基酸等。
黄芪:微温、甘、归脾、肺经。补气升阳、益卫固表,利水清肿,托疮生肌;主要化学成分为多糖、皂苷类、黄酮类及氨基酸、微量元素、甾醇类物质等。
穿心莲:寒、苦,归肺、胃、大肠经,清热解毒、凉血清肿;穿心莲含有二萜内酯类、黄酮类、苯丙素类、环烯醚萜类、生物碱等活性成分。
玄参:性微寒,味甘、苦、成,归肺、胃、肾经。具有清热解毒、消肿止痛、凉血滋阴,泻火解毒的功效;同时也具有较好的抗炎和广谱抗菌作用;用于治疗热病伤阴、舌绛烦渴、温毒发斑、津伤便秘、咽痛、瘰疠、白喉、痈肿疮毒等。其主要化学活性成分包括环烯醚萜类、倍半萜类、三萜类、简单苯丙素类、黄酮类、生物碱、有机酸及多糖类等。
板蓝根:具有清热解毒,凉血利咽的功效,可用于瘟疫时毒、发热咽痛、温毒发斑和丹毒等病症。板蓝根主要含有吲哚类生物碱、含硫类化合物、有机酸类、甾醇类化合物等。
山楂:味酸、甘,性微温。归脾、胃、肝经。具有消食健胃、行气散瘀、化浊调脂;用于治疗肉食积滞、胃脘胀满、泻痢腹痛、心腹刺痛、高脂血症;主要含有有机酸类、黄酮类和三萜类等活性成分。
陈皮:行气健脾、和胃降逆、燥湿化痰、止咳平喘。陈皮主要含有黄酮类、挥发油类、生物碱类等活性物质。
甘草:性平,味甘,归心、肺、脾、胃经,具有补脾益气、清热解毒、祛痰止咳、缓急止痛、调和诸药等功效。甘草主要含有三萜类、黄酮类、多糖类、香豆素类、挥发油类以及氨基酸等活性成分,其中三萜类和黄酮类是主要成分。
神曲:(由面粉、麦麸、赤小豆、苦杏仁、青篙、苍耳草、辣寥按一定比例发酵制成);具有健脾和胃,消食调中,兼有发表的功效;临床主要用于因饮食失节,食积停滞所引起的脱腹满胀,呕吐泻痢等消化不良症状;神曲主要活性成分为挥发油、苷类、生物碱、维生素B族、麦角固醇、淀粉酶和蛋白酶等。
上述能提高肉鸡饲料转化效率和促生长的中草药添加剂的制备方法,包括如下步骤:
1)将连翘、当归、黄芪、穿心莲、玄参、板蓝根、山楂、陈皮、甘草、神曲等按确定的份数比例混合,经100目筛粉碎后均匀成粉剂,装袋备用;
2)饲喂前将配合饲料和步骤1)得到的物料放入搅拌机搅拌均匀,再行饲用。
步骤2)所述的饲喂前将配合饲料和步骤1)得到的物料,按照99.75:0.25的重量比放入搅拌机搅拌均匀。
上述拌好的饲料在南方的春夏季节可保存8-10天;秋冬季节可用12-14天。
和现有技术相比,本发明具有如下优点:
本发明所述的能提高肉鸡饲料转化效率和促生长的中草药添加剂,组方中的连翘、当归、黄芪、穿心莲、玄参、甘草等所具有的清热解毒、活血消肿、补气升阳、益卫固表等药理功能及其所含有的黄酮、三萜、多糖、挥发油和生物碱等生物活性物质所具有的抗菌、抗炎、提高免疫力和抗氧化能力,由此可提高鸡的抗病力,使鸡群能在高度规模化和集约化的高密度养殖条件下消除因此而产生的不良应激,保持鸡群有健康的体况和良好的生长状态;然后在此基础上再利用山楂、陈皮、神曲、当归、黄芪和甘草等所具有的健胃消食、补脾益气、润肠活血、和胃降逆等药理作用及其所含有的有机酸、多糖、挥发油、生物碱、氨基酸、消化酶类等活性物质提高鸡的食欲,促进鸡的肠道发育;改善肠道的微生态系统,保持肠道健康;同时还能提高部分消化酶的活性和肠道的消化机能,从而提高营养物质的吸收利用和饲料的转化效率,促进鸡的生长。

实验例:
1 材料与方法
1.1 中草药添加剂
本试验用实施例1的连翘、当归、黄芪、陈皮、山楂、甘草等十味中草药构成的添加剂(以下称为“连黄”中草药添加剂),由广西南宁市一心医药有限公司提供,用前经100目筛粉碎后装袋备用。
1.2试验日粮及营养水平
试验用饲料由南宁市漓源粮油饲料有限公司提供(配方保密;原料组成有:玉米、小麦、高粱等谷物及豆粕、鱼粉、磷酸氢钙、L-赖氨酸硫酸盐、蛋氨酸羟基类似物、维生素A、维生素D3、维生素E、维生素K3、硫酸铜、硫酸亚铁、硫酸锌、丙酸钙等),不同阶段日粮的主要养分水平见表1。

注:鸡花料喂至15d,16-35d喂小鸡料,35d后喂中鸡料。
1.3试验动物、设计及饲养管理
1.3.1饲养试验
试验用1d龄东兰乌鸡480羽(均为♀鸡),由广西立藤农牧发展有限公司提供;用单因子完全随机设计,把试验鸡分4组,每组设6个重复,每重复20羽;其中A组喂基础日粮为对照组;B、C、D组分别在其基础日粮中添加0.25%、0.50%和1.00%的“连黄”中草药添加剂;试验于2020年11月9日在广西畜牧研究所养禽研究室鸡场进行,试期70d。21d前养在育雏室,人工控温,初始室温控制在33℃,1周后每2天降1℃,最终(21d前)维持在30℃;试验期间试鸡自由采食和饮水,饲养管理及免疫程序按生产常规进行。
1.3.2代谢试验
在70d的饲养试验结束时,即从每个试验组中选取24羽体重相似的试验鸡,共96羽,用原饲养试验相同设计,分4组,每组设6个重复,每重复4羽;继续饲喂先前试验各组所用的日粮;进行6天代谢试验;其中前3天为预试期,收粪采样在后3天进行;采样期间于每天早上8:00时始采集前一天试验鸡的所有鸡粪,去除羽毛、皮屑等杂物,均匀后采样,用10%的硫酸喷洒并均匀后装袋密封,于-20℃下保存;然后由广西畜牧研究所动物营养研究室进行各种养分及AIA含量的测定。代谢试验方法:用4N盐酸不溶性灰分(4NHCl·AIA)法。
1.4测定指标与方法
1.4.1体重、采食量及饲料报酬的测定
分别于试验初始、21、35、49和70d时早上(8:00)饲喂前称各组试验鸡空腹体重;以此计算相应的日均增重(ADG);同时测定期间的试验鸡饲料采食量并计算相应的日均饲料采食量(ADFI);最后根据各阶段的ADG和AAFI计算试鸡的料重比(F/G)(即饲料报酬),计算公式:(F/G)=ADFI(g)/ADG(g)。
1.4.2日粮养分代谢率测定
代谢试验期间收集的样品,在正式开始测定前统一解冻、60℃烘干、粉碎后装袋备用;最后测定样品中各养分和AIA含量。并根据4NHCl·AIA法所用公式计算日粮各种养分的代谢率;计算公式:DC(%)(养分代谢率)=100-100[(A1/A2)*(F2/F1)]

1.5试验数据处理
试验数据用Excel作初步处理,用SPSS16.0统计软件进行单因素方差分析,组间均数比较用Duncan氏法,结果表示为:平均数±标准差。

2.1“连黄”中草药添加剂对试验鸡生长性能的影响
2.1.1试验鸡的ADG结果见表2:

同行各组的均数值肩标相同小写字母或无字母时表示差异不显著(P>0.05);肩标不同相邻小写字母表示差异显著(P<0.05);不同相隔小写字母表示差异极显著(P<0.01),下同。
表2数据显示:在整个试验期间,除50-70d之外,B组在其他各阶段的ADG和70d平均个体重均显著高于A组(P<0.05);C组在36-49、1-49、1-70d的ADG和70d时平均个体重比A组均有提高趋势,但未达到统计差异标准(P>0.05),此外,两组之间的其它阶段均无明显差异(P>0.05);D组在36-49d和1-70d的ADG及70d的平均个体重较A组也有提高趋势(P>0.05),但两组之间的其它阶段也无明显差异(P>0.05)。由此可见,日粮添加0.25%的“连黄”中草药添加剂能明显提高试鸡的增重性能;且在三个试验组中B组的增重效果最好。
2.1.2对饲料采食量的影响

从表3数据看来:虽然在36-49d时A组的ADFI极显著高于B组(P<0.01),也显著高于C、D组(P<0.05),且A组在1-49d时也还明显高于C组(P<0.05),但在除此之外的其他阶段,尤其在1-70d时,4组试验鸡的ADFI均无显著差异(P>0.05)。可见“连黄”中草药添加剂的三种添加量均不能提高试验鸡的日粮采食量,但也没有产生不良影响。
2.1.3对饲料报酬的影响

表4数据表明:在1-21和50-70d时,A、B组间的F/G并无明显差异(P>0.05),但B组在22-35和1-70d时明显低于A组(P<0.05),且在36-49和1-49d时更是极显著低于A组(P<0.01);在36-49d时C组的F/G显著低于A组(P<0.05),且在1-49d时还极显著低于A组(P<0.01),但在此外的其他阶段,两组之间并无显著差异(P>0.05);虽然在36-49d时D组的F/G明显低于A组(P<0.05),但A组在1-21d时却显著低于D组(P<0.05),此外,两组之间并无明显差异(P>0.05)。可见在三个试验组之中,添加量为0.25%的“连黄”中草药添加剂的B组试验鸡能显著降低料重比,换言之,即是显著提高了饲料转化效率(或饲料报酬)。
2.2对日粮养分代谢效率的影响
试验鸡对日粮养分代谢率的结果见表5:

养分代谢率:B、C两组的DM和CF代谢率均极显著高于A组(P<0.01),D组也显著高于A组(P<0.05);B、C、D三试验组的CP和TE代谢率均显著高于A组(P<0.05);但4组之间的EE代谢率并无明显差异(P>0.05);B组的NFE代谢率极显著高于A组(P<0.01),C、D组亦显著高于A组(P<0.05);C组的CA代谢率极显著高于A组(P<0.01),B、D两组也明显高于A组(P<0.05);B、C两组的Ca、P代谢率均极显著高于A组(P<0.01),D组也明显高于A组(P<0.05)。可见“连黄”中草药添加剂的3种不同添加量,对日粮主要养分代谢效率均有不同程度的提高,其中以0.25%和0.50%添加量的“连黄”中草药添加剂的提高程度较为显著。
3讨论
东兰乌鸡是广西优良地方品种鸡,主要分布在广西东兰县的东兰、隘洞、长江等14个乡镇及周边县乡,年出栏100万羽以上;东兰乌鸡全身黑色,皮、肉、骨及内脏呈浅黑色(韦善等,2017)。该鸡体形为中小型,成年(150d)公鸡1.3-2.5kg,母鸡1.1-2.0kg;120d时公鸡平均体重1336.6g,日增重11.1g,母鸡913.9g,日增重8.4g(罗城,2011);东兰乌鸡具有耐粗饲、抗病力强、肉质细嫩、肉味鲜美及药用价值高等优点而广受人们青睐(吴亮等,2018)。
本研究在4组东兰乌鸡的B、C、D三个试验组日粮中分别添加0.25%、0.50%和1.00%的“连黄”中草药添加剂,饲养至70d时,B组的ADG比对照A组显著提高,其中1-49或1-70d时分别提高了6.76%和5.00%,且差异显著;虽然C组较A组在一定程度上有提高的趋势,但未达到统计差异水准。可见日粮添加0.25%的“连黄”中草药添加剂对试验鸡的生长具有良好促进作用,同时也表明日粮的这一添加水平是适宜的。
适宜的中草药添加剂能促进鸡的生长,其原因可能与添加剂中各种中草药所含的生物活性物质有关,本试验所用的黄芪、当归、陈皮、山楂、甘草等分别不同程度地含有多糖、黄酮、挥发油、生物碱等之类的活性物质(姜辉等,2020;陈琳等,2021;郭强等,2009;邓桃妹等,2021;董嘉琪等,2021;李思琦等,2020),研究显示:多糖能在促进消化道有益菌增殖的同时可抑制有害菌繁殖,因此能改善肠道的微生态系统,保持肠道健康;同时还能促进十二指肠、空肠微肠毛的生长,增加肠道对养分吸收的表面积;此外多糖还能上调碳水化合物活性酶(CAZymes)基因的表达,提高CAZymes的活性(唐圆等,2020;金俊奇等,2021);因此能提高饲料养分的消化吸收,从而促进动物生长。方磊涵等(2018)和石念进等(2013)分别在鸡日粮中添加复方中草药多糖进行研究时,结果均能显著提高鸡的生长性能。而黄酮类化合物具有抗氧化、调节机体免疫及促生长功能,江阳等(2021)在AA公鸡日粮中添加750mg/kg的艾蒿黄酮时显著提高了22-42d的ADG并明显降低了F/G;杨季等(2021)在AA公雏鸡日粮中添加橙皮苷(也叫生物黄酮)饲喂时,也明显提高了22-42d或1-42d的ADG,并显著降低了F/G。研究认为:黄酮类化合物能够提高小肠绒毛高度和隐窝深度,并通过增加细胞内碱性磷酸酶的活性以及肠道上皮细胞DNA的合成而促进肠道发育;同时黄酮类化合物也能促进胃肠的蠕动,加快食糜的消化,提高饲料的利用效率。挥发油又称为精油,研究表明:植物精油(PEO)可以增加饲料的适口性,刺激胃肠道消化液和黏液的分泌;并且能抑制消化道内有害菌群的繁殖,调整并稳定消化道微生物区系;PEO还能提升肠道的绒毛高度和隐窝深度,增加小肠绒毛的表面积,改善肠道的消化吸收(赵琛等,2020;杨敏等,2017);在肉鸡饲粮中添加PEO的研究还表明;PEO可显著提高胰蛋白酶、α-淀粉酶和肠麦芽糖酶活性(JANG,2007),故能提高饲料营养物质的消化率,改善养分的吸收与利用,因此在日粮中添加适量的PEO可显著提高鸡的生长性能及饲料利用效率(马伟斌等,2021;张文静等,2016),研究还表明:添加复合PEO比单一PEO效果更好(汪天龙等,2020);此外,上述生物活性物质还具有抗菌、抗病毒和抗氧化等作用(朱永刚等,2016;陈志刚等,2015;刘旺景等,2016),通过这些作用能消除某些负面因素对鸡生长的影响,进一步促进鸡的生长发育。研究显示:由黄芪、当归、陈皮、山楂、甘草等作为主要组分构成的中草药添加剂对鸡的生长性能均有较好促进效果(于佳楠等,2020;李姣清等,2021;贾玉刚等,2018;张琼娣等,2011)。本研究结果也与之极为相似,由此再次证明了此类添加剂对鸡的促生长作用。
试验期间,因B组的ADG显著高于A组,而两组间的ADFI无明显差异,因此B组的F/G就低于A组,说明日粮添加0.25%的“连黄”中草药添加剂能明显降低鸡的F/G,换言之,即提高了饲料转化效率,由此说明:B组试验鸡生长性能的提高是通过饲料转化效率的提高来实现的,这对生产实践很有意义,因为不用增加饲料成本,也可提高生产效益。
从代谢试验的结果看来,“连黄”中草药添加剂的两种不同添加量均能不同程度提高了日粮主要养分的代谢率,其中尤以0.25%添加量的提高效果最好,由此再次说明日粮添加0.25%的B组试验鸡生长性能的提高是通过饲料转化效率的提高而实现的。
小结
1、日粮中0.25%“连黄”中草药添加剂可以明显提高东兰乌鸡的生长性能,并且显著降低了乌鸡的料/重比;
2、“连黄”中草药添加剂的0.25%、0.50%的添加量均不能提高东兰乌鸡的日粮采食量,但也没有对试验鸡的采食产生负面影响;
3、“连黄”中草药添加剂的0.25%、0.50%的添加量均能不同程度地提高日粮主要养分的代谢率,其中以“连黄”中草药添加剂0.25%和0.50%添加量的提高程度显著。
以上所述仅为本发明的优选实施方式,应当指出,对于本领域的普通技术人员来说,在不脱离本发明创造构思的前提下,做出若干改进和变化,这些都属于本发明的保护范围。
Claims (5)
4.权利要求1-3任一所述的能提高肉鸡饲料转化效率和促生长的中草药添加剂的制备方法,其特征在于:包括如下步骤:
1)将连翘、当归、黄芪、穿心莲、玄参、板蓝根、山楂、陈皮、甘草、神曲等按确定的份数比例混合,经100目筛粉碎后均匀成粉剂,装袋备用;
2)饲喂前将配合饲料和步骤1)得到的物料放入搅拌机搅拌均匀,再行饲用。
5.根据权利要求4所述的能提高肉鸡饲料转化效率和促生长的中草药添加剂的制备方法,其特征在于:步骤2)所述的饲喂前将配合饲料和步骤1)得到的物料,按照99.75:0.25的重量比放入搅拌机搅拌均匀。
