Background
Fatigue is a subjective feeling of discomfort, but objectively loses its ability to perform the normal activities or work originally undertaken under the same conditions. With the progress of modern society and the increase of living pressure, more and more people feel that the physical condition is not as good as before, and many people bear great psychological pressure for a long time or are in overtime overload working state, thereby causing a series of fatigue complications such as lassitude, dizziness, headache, limb weakness, poor spirit, hypomnesis and the like. According to the investigation of the world health organization, more than 35% of people worldwide are in fatigue state, especially 60% -75% of middle-aged male people in fatigue state. Medical experts point out that sustained over-fatigue can lead to a series of adverse symptoms, resulting in "fatigue syndrome". Fatigue is one of the main factors endangering human health in the 21 st century.
At present, most of the medicines for relieving fatigue in the market are chemical medicines, and the traditional Chinese medicines occupy relatively less market compared with western medicines. However, the chemical products mainly containing adenosine disodium triphosphate, vitamins and stimulants are mainly used for symptomatic treatment to relieve discomfort, and the chemical products can generate side effects after being taken for a long time, and can generate different degrees of damage to organs of a body or form drug dependence, so that the chemical products are not suitable for long-term use. Therefore, the development of an anti-fatigue traditional Chinese medicine which is efficient, safe, mild in medicine property, wide in applicable population and suitable for long-term administration is a technical problem which needs to be solved by technical personnel in the field urgently.

Detailed Description
In order to more clearly illustrate the present invention, the present invention will be described in further detail below with reference to examples and the accompanying drawings. It is to be understood by persons skilled in the art that the following detailed description is illustrative and not restrictive, and is not to be taken as limiting the scope of the invention.
In the following examples, unless otherwise specified, materials, reagents and the like used are commercially available, and the relevant experimental means may be realized by conventional techniques.
EXAMPLE 1 preparation of Green Tangerine Peel essential oil by steam distillation
Weighing 500g of dried green tangerine peel, grinding into coarse powder, putting into an extraction container, and adding 6 times (6-12 times) of distilled water. Heating and distilling at 100 deg.C for 2 hr (2-10 hr).
And after the extraction is finished, standing until oil and water are completely layered, taking out the essential oil, and removing residual water in the essential oil by using anhydrous sodium sulfate to finally obtain the pericarpium citri reticulatae viride essential oil.
The overall flow of the embodiment of the present invention is shown in fig. 1.
Example 2 mouse fatigue model establishment and administration of Green Tangerine Peel essential oil
The experimental animals were male, 7-week-old C57 mice. All mice were free to ingest drinking water, using a day-night cycle of 12h (8: 00-20: 00 for the night, 20: 00-8: 00 for the day).
Green tangerine peel essential oil is dissolved in Tween 80 according to the volume to prepare green tangerine peel essential oil with the volume concentration of 30% (experiments prove that green tangerine peel essential oil solutions with the volume concentration of 0.1% -50% have curative effect on anxiety neurosis, and the concentration is taken as an example and is not limited). Mice were randomized into 4 groups: model group, blank group, administration group and negative vehicle group, corresponding drugs were administered by gavage, and administered at a dose of 0.1ml/10 g. (the negative solvent group was Tween 80 solvent.)
Except for the blank group of mice, the other groups of mice need to establish a fatigue model.
The model is established by weight swimming of the mouse. The specific implementation steps are as follows:
1. first, weight swimming was started in an amount of 5% of the body weight of each group of mice, and the swimming time was recorded.
2. And (4) rejecting the mice with larger swimming time difference to enable the average swimming time of each group to be similar.
3. The swimming was carried out under a weight of 5% of the body weight until the mice were completely submerged and did not float on the water within 5 seconds.
The period of model building was 7 days.
After the model is built, each model group needs to be dosed for 1 time each day. The administration is carried out separately according to the scheduled groups, and in order to prevent fatigue during the administration and free recovery of anxiety symptoms accompanying fatigue, a mode of maintaining a fatigue model while administering the drug during the administration is adopted. And the administration period is seven days.
Example 3 exhaustive swimming test of mice
The weight-bearing swimming method comprises the following steps: the weight of the mice was weighed and recorded before the test was started, and then the lead was cut out in an amount of 5% of the weight of the mice. And winding the lead wire at a position 1-2cm away from the root of the mouse tail. And controlling the water temperature to be 25-30 ℃, putting the mice into the water, starting timing until the mice are completely immersed into the water and can not float out of the water surface within 5 seconds, and recording the time.
The experimental results are expressed as the standard error of the mean soil and statistically processed by the t test.
Exhaustive swimming tests are routine fatigue tests, and effective anti-fatigue drugs increase the weight bearing swimming time of mice. As shown in fig. 2, gavage of pericarpium citri reticulatae viride essential oil significantly increased the swimming time of mice compared to the model group (P < 0.05). The pericarpium citri reticulatae viride essential oil can increase the swimming time of the mouse in the exhaustive swimming experiment by intragastric administration.
Example 4 mouse fatigue rod rotation experiment
Rod turning experiment: the mice were placed on a roller-type fatigue tester with their heads rotating in the opposite direction to the roller, and given 1min of acclimation time. Setting the rotating speed of the bar rotating instrument to be 30r/min, and setting the total experimental time to be 5 min; after starting, the instrument accelerates the mixture to a set rotating speed. The mice first fall time and the number of falls were recorded. Each mouse was assayed 3 times, and the average value was taken as the measurement result. The motor coordination ability of the mice is evaluated through the first falling time and falling times of the mice.
The experimental results are expressed as the standard error of the mean soil and statistically processed by the t test.
The fatigue rolling bar experiment is a classic anti-fatigue screening model, and the effective anti-fatigue medicine can prolong the time of the mice falling for the first time and reduce the falling times. As shown in FIGS. 3 and 4, the drop time of the administration group was significantly prolonged, and the number of drops was significantly reduced (P < 0.05). The green tangerine peel essential oil has obvious anti-fatigue effect.
Example 5 mouse open field experiment
The behavioral detection method comprises the following steps: the open field device is a black wooden open box with the length of 42cm, the width of 42cm and the height of 40 cm. The center of the bottom surface is marked with a 20cm × 20cm square by a white line. The mice are placed in the central square of the open field, and the residence time of the mice in the central square within 6min and the running track of the mice in the open field are recorded. Video analysis was performed using Smart3.0 software. Each mouse required a floor scrub with alcohol after the test was completed.
The experimental results are expressed as the standard error of the mean soil and statistically processed by the t test.
Open field experiments are a classic anxiolytic screening model, and effective anxiolytic drugs can increase the movement distance of mice in the center. As shown in fig. 5, 6 and 7, gavage of pericarpium citri reticulatae viride essential oil significantly increased the distance of mouse movement in the center (P <0.05) compared to the model group. The result shows that the lavage of the pericarpium citri reticulatae viride essential oil can increase the movement distance of the mouse in the center in an open field experiment.
Example 6 mouse Tail suspension experiment
The behavioral detection method comprises the following steps: a mouse tail suspension experiment model is constructed according to an experiment method recorded in European Journal of Pharmaco logy, 2001, 415:197, the mouse is hung upside down on a cross bar 60cm away from the ground for 6min, the fixing point of the mouse is a position 1cm away from the tail, after 1min of adaptation, the mouse does not move for 5min after observation, and the motionless state is defined as the state that the mouse stops struggling and the body is in a relaxed state.
The experimental results are expressed as the standard error of the mean soil and statistically processed by the t test.
The mouse tail suspension experiment is a classical experimental model for screening the anxiolytic drugs, and the mouse tail suspension presents an despair state after a period of time and stops struggling. Effective anxiolytic drugs can shorten the immobility time of mice during tail suspension. As shown in figure 8, the study of the experiment shows that the lavage of the pericarpium citri reticulatae viride essential oil can obviously shorten the immobility time (P <0.05) in the tail suspension process of the mice. The result shows that the total immobility time of the mice in the tail suspension process can be shortened by the lavage of the pericarpium citri reticulatae viride essential oil.
Example 7 mouse elevated plus maze experiment
Elevated Plus Maze (EPM) experiments: mice were placed one at a time from the central square of the maze, after which the maze curtains were closed. And observing the activity condition of the mouse in the maze within 5min by using high-definition video software, and recording the behavioural data. After each experiment, the mouse excrement in the maze is removed, the maze is wiped by alcohol cotton to eliminate the smell of the mouse, and then the next experiment is carried out.
The experimental results are expressed as the standard error of the mean soil and statistically processed by the t test.
The elevated cross experiment is a classical experimental model for screening anxiolytic drugs, a mouse is placed in the center of a cross maze, and the residence time of the mouse in an opening and closing arm in the cross maze is recorded and observed. As shown in fig. 9 and fig. 10, the study of the experiment shows that the pericarpium citri reticulatae viride essential oil can significantly improve the standing time of the mouse in the open-close arm in the elevated plus maze (P < 0.05).
The experimental model of the animal above verifies, including mouse exhaustion swimming experiment, mouse fatigue commentaries on classics stick experiment, mouse open field experiment (OFT), mouse hangs tail experiment (TST), mouse elevated cross maze Experiment (EPM), it can increase mouse exhaustion swimming time to have proved that green tangerine peel essential oil gavage can increase, the time of first falling is prolonged, the number of times of falling of reduction, the mouse is at the moving distance of central authorities in the open field experiment, shorten the motionless total time of mouse tail process of hanging, and the dwell time of the arm that opens and shuts in the elevated cross maze of mouse has been improved. In conclusion, the pericarpium citri reticulatae viride essential oil has a definite curative effect on relieving fatigue and anxiety neurosis caused by fatigue, and meanwhile, the pericarpium citri reticulatae viride belongs to a pure natural extract, is small in side effect, safe and convenient to use, and has a great significance for preventing and treating depression and anxiety neurosis.
