Abstract
本发明属于护肤保健品技术领域,尤其是一种包裹中药提取物的缓释微囊及其制备方法和应用,解决了现有技术中直接将中药提取物直接添加于护肤品中混合乳化使用,由于中药活性成分的特殊化学结构,在皮肤表面极易丧失生物活性,而不能抵达皮肤深层,进而不能起到修复皮肤屏障,以及不能起到显著的护肤效果的问题,所述包裹中药提取物的缓释微囊,包括囊心物和包裹所述囊心物的囊材;所述囊心物由中药提取物、抗坏血酸棕榈酸酯和木糖醇脂肪酸酯混合复配而得;所述囊材由脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯混合复配而得。本发明能够有效的保证中药活性成分能够抵到皮肤深层,缓慢释放,进而起到修复皮肤屏障的作用。

技术领域
本发明涉及护肤保健品技术领域,尤其涉及一种包裹中药提取物的缓释微囊及其制备方法和应用。
背景技术
皮肤屏障狭义主要指物理性屏障,即由皮脂膜、角质层角蛋白、脂质、“三明治”结构、砖墙结构、真皮粘多糖类、粘多糖类等共同构成。健康的皮肤屏障能够抵御外界有害物、刺激物和日光进入,同时具有保湿及调节作用。然而,由于环境、饮食和错误的皮肤护理等原因,皮肤屏障受损,使得皮肤自身防御能力不足,皮肤极易敏感受损。皮肤屏障受损后,保湿功能就会变差,出现外油内干、皮肤干燥、脱屑、干痒等现象。其次,皮肤屏障受损,外界刺激物(紫外线、微生物等)很容易刺激皮肤,进而导致皮肤出现炎症,产生红斑、丘疹、痤疮、脂溢性皮炎等问题。而且,皮肤保湿能力严重下降,加上炎症,就会导致皮肤整体的防御能力下降,对外界刺激非常敏感,容易产生瘙痒、灼烧等问题。
功效活性物是护肤产品配方的“灵魂”,护肤品配方中使用的活性物来源很广。而近几年,随着消费者安全意识的不断提高,市场上对于天然、绿色、安全等原料的呼声越来越大,中药提取物也成为了关注的焦点。中药提取物是采用先进的工艺技术对中药材或中药复方进行提取分离加工而得到的单一活性物质或混合物,天然生物活性物质具有抗菌、消炎、抗癌、抗氧化等生理活性,被广泛应用于食品、饮料、医药、护肤品、保健品等行业。然后,现有技术多是直接将中药提取物直接添加于护肤品中混合乳化使用,由于中药活性成分的特殊化学结构,在皮肤表面极易丧失生物活性,而不能抵达皮肤深层,进而不能起到修复皮肤屏障,以及不能起到显著的护肤效果。基于上述陈述,本发明提出了一种包裹中药提取物的缓释微囊及其制备方法和应用。
发明内容
本发明的目的是为了解决现有技术中直接将中药提取物直接添加于护肤品中混合乳化使用,由于中药活性成分的特殊化学结构,在皮肤表面极易丧失生物活性,而不能抵达皮肤深层,进而不能起到修复皮肤屏障,以及不能起到显著的护肤效果的问题,而提出的一种包裹中药提取物的缓释微囊及其制备方法和应用。
一种包裹中药提取物的缓释微囊,所述缓释胶囊包括囊心物和包裹所述囊心物的囊材;所述囊心物由中药提取物、抗坏血酸棕榈酸酯和木糖醇脂肪酸酯混合复配而得;所述囊材由脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯混合复配而得。
优选的,所述中药提取物、抗坏血酸棕榈酸酯和木糖醇脂肪酸酯的质量比为20-24:1-2:1-3。
优选的,所述中药提取物由以下方法制得:按固液比1:12-18,将干燥中药混合粉碎物加浓度为68%的乙醇溶液搅拌分散,分散混合均匀后,于52-72℃的温度下,超声微波提取7-10h后抽滤,滤液浓缩至自身体积比的28-35%得稠膏,将稠膏冷冻干燥后研磨得粒径为1-100nm的中药提取物。
优选的,所述中药混合粉碎物为质量比3-5:8-12:1-2的刺五加、芍药和天竺葵的混合粉碎物。
优选的,所述超声微波提取条件为:超声波功率为900-1200W,微波功率为300-500W。
优选的,所述脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯的质量比为20-30:12-18:1-3:1-2。
本发明还提出了一种包裹中药提取物的缓释微囊的制备方法,包括以下步骤:
S1、制备囊心物:按质量比20-24:1-2:1-3,将中药提取物、抗坏血酸棕榈酸酯和木糖醇脂肪酸酯研磨混合均匀后,得囊心物;
S2、囊材的制备:按质量比20-30:12-18:1-3:1-2,将脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯研磨混合均匀后,得囊材;
S3、按质量比2-3:2-3:1,将囊材加水研磨制成混悬液后加入囊心物,于48-58℃的温度下,以500-800r/min的转速,搅拌30-50min,然后缓慢滴加醋酸溶液,调节pH值至4-5,继续搅拌20-30min,加入总体积量1.5-2倍的温度为48-58℃的蒸馏水,边搅拌边停止加热,搅拌至温度降至室温后,放入冰水浴搅拌,加甘油搅拌20-30min,静置至微囊沉降完全,倾去上清液,烘干即得所需的包裹中药提取物的缓释微囊。
优选的,所述包裹中药提取物的缓释微囊的制备方法制备得到的缓释微囊在制备护肤保健品或护肤药品中的应用。
本发明提出的一种包裹中药提取物的缓释微囊,具有以下有益效果:
1、本发明采用刺五加、芍药和天竺葵为中药原料提取活性成分,将其与抗坏血酸棕榈酸酯和木糖醇脂肪酸酯混合复配制成囊心物;本发明制得的囊心物具有较强的抗氧化功效,能够促进新陈代谢,抑制氧自由基的产生,增强内源性超氧化歧化酶活性,其能够新生关键蛋白,强化细胞间连接,补充皮脂膜,重塑强韧皮肤屏障,进而有效抵御肌肤外界刺激能力,延缓衰老。
2、本发明采用脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯为原料混合制得囊材,所得囊材具有良好的稳定性、溶解性和乳化性,能够有效的包裹囊心物,使其生物活性稳定的同时达到缓释效果,其还能在皮肤表面形成一层多孔类脂膜,能够滋润皮肤,有效防止皮肤干燥,同时能够促进皮肤新陈代谢、缓解氧化,从而减缓皮肤氧化和黑色素沉积,能减少面部皮肤的细微皱纹,且具有抗氧化及美白的作用。
3、本发明将囊材包裹囊心物制得包裹中药提取物的缓释微囊,采用微囊包裹缓释技术,能够有效的保证中药活性成分能够抵到皮肤深层,缓慢释放,进而起到新生关键蛋白,强化细胞间连接,补充皮脂膜,修复皮肤屏障等作用,其原料选用科学合理,制备方法简单,制备条件温和,所得缓释微囊无毒、无味、无嗅,对皮肤无刺激性,安全性高,其用于制备护肤保健品或护肤药品,适用于各种肌肤状况,能平衡皮脂分泌、促进血液循环,进而使肌肤变得饱满、红润、有活力,添加有本发明缓释微囊的护肤品具有改善肌肤干燥、粗糙、晦暗、发黄,淡化肌肤色斑、美白、保湿、延缓肌肤衰老等功效,特别适用于脸部、颈部及眼部护肤。

下面结合具体实施例对本发明作进一步解说。
实施例一
本发明提出的一种包裹中药提取物的缓释微囊,所述缓释胶囊包括囊心物和包裹所述囊心物的囊材;所述囊心物由质量比为20:1:1的中药提取物、抗坏血酸棕榈酸酯和木糖醇脂肪酸酯混合复配而得;所述囊材由质量比为20:12:1:1的脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯混合复配而得;
所述中药提取物由以下方法制得:按固液比1:12,将干燥中药混合粉碎物加浓度为68%的乙醇溶液搅拌分散,分散混合均匀后,于温度为52℃,超声波功率为900W,微波功率为300W的条件下,超声微波提取7h后抽滤,滤液浓缩至自身体积比的28%得稠膏,将稠膏冷冻干燥后研磨得粒径为1nm的中药提取物,其中,中药混合粉碎物为质量比3:8:1的刺五加、芍药和天竺葵的混合粉碎物。

S3、按质量比2:2:1,将囊材加水研磨制成混悬液后加入囊心物,于48℃的温度下,以500r/min的转速,搅拌30min,然后缓慢滴加醋酸溶液,调节pH值至4,继续搅拌20min,加入总体积量1.5倍的温度为48℃的蒸馏水,边搅拌边停止加热,搅拌至温度降至室温后,放入冰水浴搅拌,加甘油搅拌20min,静置至微囊沉降完全,倾去上清液,烘干即得所需的包裹中药提取物的缓释微囊。
实施例二
本发明提出的一种包裹中药提取物的缓释微囊,所述缓释胶囊包括囊心物和包裹所述囊心物的囊材;所述囊心物由质量比为22:1.5:2的中药提取物、抗坏血酸棕榈酸酯和木糖醇脂肪酸酯混合复配而得;所述囊材由质量比为25:15:2:1.5的脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯混合复配而得;
所述中药提取物由以下方法制得:按固液比1:15,将干燥中药混合粉碎物加浓度为68%的乙醇溶液搅拌分散,分散混合均匀后,于温度为62℃,超声波功率为1050W,微波功率为400W的条件下,超声微波提取8.5h后抽滤,滤液浓缩至自身体积比的32%得稠膏,将稠膏冷冻干燥后研磨得粒径为50nm的中药提取物,其中,中药混合粉碎物为质量比4:10:1.5的刺五加、芍药和天竺葵的混合粉碎物。
本发明还提出了一种包裹中药提取物的缓释微囊的制备方法,包括以下步骤:
S1、制备囊心物:按质量比22:1.5:2,将中药提取物、抗坏血酸棕榈酸酯和木糖醇脂肪酸酯研磨混合均匀后,得囊心物;
S2、囊材的制备:按质量比25:15:2:1.5,将脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯研磨混合均匀后,得囊材;
S3、按质量比2.5:2.5:1,将囊材加水研磨制成混悬液后加入囊心物,于53℃的温度下,以650r/min的转速,搅拌40min,然后缓慢滴加醋酸溶液,调节pH值至4.5,继续搅拌25min,加入总体积量1.8倍的温度为53℃的蒸馏水,边搅拌边停止加热,搅拌至温度降至室温后,放入冰水浴搅拌,加甘油搅拌25min,静置至微囊沉降完全,倾去上清液,烘干即得所需的包裹中药提取物的缓释微囊。
实施例三
本发明提出的一种包裹中药提取物的缓释微囊,所述缓释胶囊包括囊心物和包裹所述囊心物的囊材;所述囊心物由质量比为24:2:3的中药提取物、抗坏血酸棕榈酸酯和木糖醇脂肪酸酯混合复配而得;所述囊材由质量比为30:18:3:2的脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯混合复配而得;
所述中药提取物由以下方法制得:按固液比1:18,将干燥中药混合粉碎物加浓度为68%的乙醇溶液搅拌分散,分散混合均匀后,于温度为72℃,超声波功率为1200W,微波功率为500W的条件下,超声微波提取10h后抽滤,滤液浓缩至自身体积比的35%得稠膏,将稠膏冷冻干燥后研磨得粒径为100nm的中药提取物,其中,中药混合粉碎物为质量比5:12:2的刺五加、芍药和天竺葵的混合粉碎物。
本发明还提出了一种包裹中药提取物的缓释微囊的制备方法,包括以下步骤:
S1、制备囊心物:按质量比24:2:3,将中药提取物、抗坏血酸棕榈酸酯和木糖醇脂肪酸酯研磨混合均匀后,得囊心物;
S2、囊材的制备:按质量比30:18:3:2,将脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯研磨混合均匀后,得囊材;
S3、按质量比3:3:1,将囊材加水研磨制成混悬液后加入囊心物,于58℃的温度下,以800r/min的转速,搅拌50min,然后缓慢滴加醋酸溶液,调节pH值至5,继续搅拌30min,加入总体积量2倍的温度为58℃的蒸馏水,边搅拌边停止加热,搅拌至温度降至室温后,放入冰水浴搅拌,加甘油搅拌30min,静置至微囊沉降完全,倾去上清液,烘干即得所需的包裹中药提取物的缓释微囊。
对比例一
本发明提出的一种包裹中药提取物的缓释微囊,所述缓释胶囊包括囊心物和包裹所述囊心物的囊材;所述囊心物为中药提取物;所述囊材由质量比为20:12:1:1的脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯混合复配而得;
所述中药提取物由以下方法制得:按固液比1:12,将干燥中药混合粉碎物加浓度为68%的乙醇溶液搅拌分散,分散混合均匀后,于温度为52℃,超声波功率为900W,微波功率为300W的条件下,超声微波提取7h后抽滤,滤液浓缩至自身体积比的28%得稠膏,将稠膏冷冻干燥后研磨得粒径为1nm的中药提取物,其中,中药混合粉碎物为质量比3:8:1的刺五加、芍药和天竺葵的混合粉碎物。

S1、制备囊心物:按质量比1:1,将抗坏血酸棕榈酸酯和木糖醇脂肪酸酯研磨混合均匀后,得囊心物;
S2、囊材的制备:按质量比20:12:1:1,将脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯研磨混合均匀后,得囊材;

S3、按质量比2:2:1,将囊材加水研磨制成混悬液后加入囊心物,于48℃的温度下,以500r/min的转速,搅拌30min,然后缓慢滴加醋酸溶液,调节pH值至4,继续搅拌20min,加入总体积量1.5倍的温度为48℃的蒸馏水,边搅拌边停止加热,搅拌至温度降至室温后,放入冰水浴搅拌,加甘油搅拌20min,静置至微囊沉降完全,倾去上清液,烘干即得所需的包裹中药提取物的缓释微囊。
按以下配方方法,分别将本发明实施例一-三、对比例一-三中制备的包裹中药提取物的缓释微囊添加制得护肤品(即实施例一-三、对比例一-三),或直接添加本发明制备的中药提取物制得护肤品(即对比例四)
取包裹中药提取物的缓释微囊(对比例四替换为中药提取物)80份、甘油50份、丁二醇10份、双丙甘醇10份,加去离子水150份,加热溶解至搅拌均匀,冷却即得所需护肤品。
选取选取年龄、健康状况和面部皮肤情况相似(存在角质化皮损、色斑、细纹等皮肤问题)的女性志愿70名,每10名一组,随机分成7组,分别于每日早晚涂抹添加有本发明实施例一-三、对比例一-三中的包裹中药提取物的缓释微囊的护肤品,以及直接添加本发明中药提取物的护肤品(即对比例四),连续使用1个月后,评价使用效果,得出如下结果:

2.根据权利要求1所述的一种包裹中药提取物的缓释微囊,其特征在于,所述中药提取物、抗坏血酸棕榈酸酯和木糖醇脂肪酸酯的质量比为20-24:1-2:1-3。
3.根据权利要求1所述的一种包裹中药提取物的缓释微囊,其特征在于,所述中药提取物由以下方法制得:按固液比1:12-18,将干燥中药混合粉碎物加浓度为68%的乙醇溶液搅拌分散,分散混合均匀后,于52-72℃的温度下,超声微波提取7-10h后抽滤,滤液浓缩至自身体积比的28-35%得稠膏,将稠膏冷冻干燥后研磨得粒径为1-100nm的中药提取物。

S1、制备囊心物:按质量比20-24:1-2:1-3,将中药提取物、抗坏血酸棕榈酸酯和木糖醇脂肪酸酯研磨混合均匀后,得囊心物;
S2、囊材的制备:按质量比20-30:12-18:1-3:1-2,将脂肪酸蔗糖聚酯、乳酸铵、羟丙基二淀粉磷酸酯和对羟基苯甲酸乙酯研磨混合均匀后,得囊材;
