Abstract
本发明提供一种添加4株乳酸菌的哺乳仔猪代乳料添加剂、代乳料及其制备方法。本发明的方法采用原料预处理工艺和超微粉碎工艺,提高了大米粉等常规原料的消化利用率,增加了大豆浓缩蛋白,减少了高成本乳源原料的用量,降低了产品成本;同时添加了含复合乳酸菌添加剂,维持哺乳仔猪的肠道健康,有效的减少抗生素的使用。试验表明本发明的哺乳仔猪代乳料产品具有产品成本低、适口性好、腹泻率低、生长速度快、抗病力强、无抗生素等特点。
Description
技术领域
本发明涉及一种哺乳仔猪代乳料添加剂,具体涉及一种适用于哺乳仔猪代乳料的添加剂、相应哺乳仔猪代乳料及其配制方法。
背景技术
在现代集约化养猪生产中,为了提高母猪的利用效率,有效控制疾病的垂直传播,普遍采用仔猪3-4周龄早期断奶的养殖方式(Duncan等,1997),甚至是10-14日龄的超早期隔离断奶(Segregated Early Weaning,SEW)。由于日龄较小的断奶仔猪仍未适应固体饲料,采食量低(Metz,1990);消化系统不成熟,内源酶不适于消化植物原料为主的日粮;饲料中的免疫原性成分(大豆蛋白等)容易引起肠道过敏反应和损伤(Dureau等,1994;WANG T等,2014);免疫系统不完善,对病原菌的抗病力弱(Vega等,2001);再叠加离开母猪和混群的心理应激和转群等环境应激(Kelley等,1980;Prerremans等,2001),断奶仔猪往往存在生长受到抑制、死淘率高等一系列问题,成为养猪生产中面临的重大挑战之一。
在哺乳仔猪代乳料产品的设计中,需要同时解决消化率和仔猪健康问题,才能满足实际生产的需要。目前使用的哺乳仔猪代乳料产品通常会选择全脂奶粉、浓缩乳清蛋白粉、乳糖等适口性好且易于消化的乳制品原料,并在使用的过程中添加大量的广谱抗生素控制仔猪的腹泻。而我国的乳制品原料非常稀缺,主要依靠进口,价格也非常昂贵,从而增加了养殖成本。抗生素的大量使用也会带来一系列的负面影响:(1)病原菌的耐药性问题凸显,猪场的疾病越来越复杂,治疗用药的效果也越来越差,为控制疾病而不得不增加用药的剂量,这使耐药性问题进一步恶化,并威胁到人类的健康;(2)大剂量抗生素的长期使用,破坏了仔猪肠道的微生态平衡,抑制了仔猪的生长速度,甚至会造成僵猪,给养猪场带来巨大损失;(3)造成严重的土壤和水体环境污染,威胁着人类的健康;(4)抗生素等添加剂的不合理使用,导致猪肉产品药物残留和品质下降,既影响食品安全,又阻碍肉产品进入国际市场,降低了经济效益。近年来替抗产品已成为饲料添加剂领域的研究热点,其中作用效果和机理研究较深入的有酸化剂、植物提取物(精油)、酶制剂、益生菌、寡糖、抗菌肽(苏丁丁等,2011;胡睿智等,2018;崔海燕等,2018;刘军等,2018)等。在替代抗生素的众多产品中,研究和应用实践都证明乳酸菌在哺乳仔猪阶段具有显著的效果:乳酸菌耐受胃肠道酸性环境,抑制有害菌的生长恢复失调的微生态平衡;可以竞争性地抑制病原菌在肠道内的定植,抑制病原体附到肠细胞上,形成良好的屏障作用;乳酸菌可刺激动物肠道,促进免疫器官的发育成熟,产生局部型免疫反应,提高机体抗体水平或巨噬细胞的活性,增强机体免疫功能(提高疫苗效价);乳酸菌在肠道内可产生乳酸、乙酸、丙酸和丁酸等挥发性脂肪酸,产生蛋白酶、脂肪酶、淀粉酶,合成维生素、氨基酸等营养物质,提高动物的生长速度,提高养殖效益。
发明内容
针对现有技术现存的上述问题和发展趋势,本发明首先提供一种专门适用于仔猪代乳料的乳酸菌组合物,其由以下四种菌株组成:嗜酸乳杆菌(Lactobacillusacidophilus),菌株保藏号为CGMCC No.7.423;长双岐杆菌长亚种(Bifidobacteriumlongum subsp.longum),菌株保藏号为CGMCC No.7.427;植物乳杆菌(Lactobacillusplantarum),菌株保藏号为CGMCC No.7.426;以及,屎肠球菌(Enterococcus faecium),菌株保藏号为CGMCC No.7.428。上述四株菌株于2021年07月01日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏地址为:北京市朝阳区北辰西路1号院3号中国科学院微生物研究所。其中,乳酸菌组合物中,各菌株的添加质量比为1:1:1:1。
具体地,所述嗜酸乳杆菌的具体获得方式及相关施用参数为:

所述植物乳杆菌的具体获得方式及相关施用参数为:
菌株编号:SY-R4;采集地:北京;具体采集位置:北京市通州区永乐店渠头牛场;分离基物:青贮玉米;分离方法:平板划线法;培养基(成分g/L):蛋白胨10.0g、牛肉膏10.0g、酵母提取物5.0g、葡萄糖20.0g、吐温801.0g、柠檬酸氢二铵2.0g、醋酸钠5.0g、硫酸镁0.1g、硫酸锰0.05g、磷酸氢二钾2.0g;特征特性:厌氧或兼性厌氧,菌种为直或弯的杆状,单个、有时成对或成链状,最适pH6.5左右,属于同型发酵乳酸菌;细胞形状:杆菌;培养温度:37℃;培养时间:16~18h;是否扩增:是;混合前菌落数要求:≥106;测序序列如SEQ ID NO:2所示。主要作用:抗逆性强,耐酸碱、可发酵多种碳源;分泌有机酸、消化酶及抗菌成分,平衡肠道菌群,提高饲料消化率。
所述屎肠球菌的具体获得方式及相关施用参数为:
菌株编号:SY-R6;采集地:北京;具体采集位置:北京养猪育种中心种猪资源场;分离基物:猪肠道;分离方法:平板划线法;培养基(成分g/L):蛋白胨10.0g、牛肉膏10.0g、酵母提取物5.0g、葡萄糖20.0g、吐温801.0g、柠檬酸氢二铵2.0g、醋酸钠5.0g、硫酸镁0.1g、硫酸锰0.05g、磷酸氢二钾2.0g;特征特性:圆形或椭圆形,呈单个、成对或短链状排列的革兰氏阳性球菌,无芽孢、无鞭毛,为需氧或兼性厌氧菌,油镜下观察屎肠球菌菌体多单生,对生和链状较少;细胞形状:球菌;培养温度:37℃;培养时间:16~18h;是否扩增:是;混合前菌落数要求:≥106;测序序列如SEQ ID NO:4所示。主要作用:增强动物免疫力,合成抗菌物质;分泌有机酸,维持肠道菌群平衡,预防腹泻。
除上述生理特征外,本申请的乳酸菌组合物在耐高温、适应仔猪的强酸性胃液环境以及耐胆碱的特性:
韩庆功等(2018)试验表明,植物乳杆菌在温度70℃条件下处理2min,存活率可以达到88.9%,但当温度升高到80℃时处理2min,存活率则下降至34.95%。而本发明的上述乳酸菌组合物的复合菌在65℃理2.5min、5min及7.5min以及85℃处理2.5min存活率均在96%以上,对存活率几乎没有影响,适于例如沸水冲泡等高温施用条件;
仔猪出生时胃内pH为5~6,数小时后pH降至4左右,2月龄前胃内pH始终保持在3左右。本发明的上述复合乳酸菌pH为3时处理90min,存活率高80.54%,pH为4、5、6时处理90min,存活率均在90%以上;
仔猪消化道中的胆盐含量为0.03%~0.3%,本发明的上述复合乳酸菌在0.3%猪胆盐处理1h,存活率为73.52%;
同时,本发明的上述复合乳酸菌对大肠埃希氏菌有抑制作用,抑制圈直径为10.2mm;对杆菌肽锌、亚甲基水杨酸杆菌肽的耐受性最强,最小抑菌浓度(MIC)大于1280mg/kg,其次是黄霉素,以下依次是土霉素钙、莫能菌素、盐霉素、金霉素均有较强的耐受性。
综上,由上述4种菌株组成的复合乳酸菌组合物组成的添加剂可以作为仔猪饲料预混料(或饲料/代乳料)的一重要组成部分来应用。添加该乳酸菌组合物的预混料(或饲料/代乳料)可以无需加入任何形式的抗生素,且同样能够实现仔猪腹泻率低、生长速度快、抗病力强、无抗生素等方面的需求。
进一步的,本发明还涉及一种添加上述乳酸菌组合物的哺乳仔猪代乳料及其制备方法。

具体地,本发明的仔猪代乳料的制备方法具体包括如下步骤:
1.按照原料配方配制0.2%吡啶甲酸铬预混剂;
2.按照各自的原料配方比例配制乳酸菌组合物、哺乳仔猪多矿、哺乳仔猪有机多矿和哺乳仔猪多维;
3.按照原料配方配制哺乳仔猪预混料;
4.按照原料配方将复合预混料与其他组分混合均匀,制得哺乳仔猪代乳料成品。
其中,为保证产品的效果,需选用优质大米(或碎米)和鱼粉(大米经湿法膨化处理)经过超微粉碎,粉碎粒度达到95%过60目。
本发明上述原料均为市售可得原料。
生产工艺多级预混保证混合均匀度。仔猪代乳料以及复合预混料配方中涉及的所有原料,经过品控部门的质量检验合格后才能接收入库,各组分按照配方比例使用电子称准确计量,投入混合机中混合均匀后即可称量、打包、入库。整个生产过程严格按照ISO9001质量管理体系和HACCP食品安全管理体系的要求操作。
本发明的代乳料涉及4株复合乳酸菌的添加,采用原料预处理工艺和超微粉碎工艺,提高了原料的消化利用率,减少了代乳料产品中高成本乳源原料的用量,降低了产品成本;同时添加的4株乳酸菌组合物,维持哺乳仔猪的肠道健康,有效的减少抗生素的使用。
具体地,本发明的代乳料适用于7日龄至28日龄的仔猪的喂养或补饲。施用方法为:可直接饲喂;与四倍温水(1:4质量比)混匀后饲喂效果更好。试验表明本发明的哺乳仔猪代乳料产品具有产品成本低、适口性好、腹泻率低、生长速度快、抗病力强、无抗生素等特点。

试验例:仔猪复合代乳料效果试验
1.材料与方法
1.1试验材料
上述制备实施例制备的哺乳仔猪代乳料(上述实施例1-3)为试验组日粮;申请人公司此前出品的一款哺乳仔猪代乳料,按产品使用说明饲喂,为对照组。
该对照代乳料的配方为(按重量份计):膨化玉米301;全脂牛乳粉300;乳清粉120;白糖50;浓缩乳清蛋白116;鸡肝脏粉40;酵母水解物25;酸度调节剂8;石粉5;磷酸二氢钙10;和复合预混合饲料25。
其中,复合预混合料饲料有效成分见下表:
1.2试验动物及分组
试验选取14日龄健康的断奶仔猪160头,随机分为4个处理组,每个处理组4个重复,每个重复10头仔猪,公母各半,进行为期14天的饲养试验;其中,试验猪在7日龄左右开始补饲对应的代乳料,随后,在15-28日龄期间进行对应代乳料的断奶饲养,具体的试验分组见下表:

对照组和各试验组猪只均在断奶当天有个别猪只腹泻,无药物治疗。
2.3不同代乳料对仔猪肤色毛色的影响
对照组和各试验组的肤色毛色眼观无明显差异。
2.4试验结果分析
本试验结果表明,15日龄断奶的哺乳仔猪使用按照实施例1-3配制的哺乳仔猪代乳料,试验期间仔猪健康状况良好,毛色正常;平均日采食量和日增重表现良好,且都显著高于对照组(市售现有技术产品)。
与上述对比例比较,本发明产品主要是用膨化米粉代替了膨化玉米、增加了大豆浓缩蛋白和鱼粉、降低了浓缩乳清蛋白和全脂奶粉用量,每吨成本约降低1000元;另外对比例产品含常用饲料抗生素添加剂:金霉素3000PPM,吉他霉素2000PPM,而本发明的实施例代乳料产品中不含任何抗生素添加剂,试验结果表明腹泻率和抗病力可达到添加抗生素的水平。

7.根据权利要求1-6任一所述的复合预混添加剂,其特征在于,所述微量元素有机预混料按质量分数计包括以下原料:蛋白铜9~10%;蛋白铁31.5~34%;蛋白锌18~22%;酵母硒18~22%;稀释剂9~11%;和沸石粉14.5~1%。
8.根据权利要求7所述的复合预混添加剂,其特征在于,所述稀释剂按质量分数计包括以下原料:吡啶甲酸铬16.6%;沸石粉33.4%;和米糠粕50%。
9.一种仔猪代乳料,其特征在于,所述仔猪代乳料包括权利要求1-8所述的复合预混添加剂。
10.根据权利要求9所述的仔猪代乳料,其特征在于,所述仔猪代乳料按重量份计由以下原料组成:膨化大米478~365;全脂牛乳粉216~264;乳清粉90~110;白糖45~55;浓缩乳清蛋白36~44;鸡肝脏粉36~44;酵母水解物22.5~27.5;鱼粉18~22;大豆浓缩蛋白18~22;酸度调节剂5.4~8.6;石粉3.4~4.7;磷酸二氢钙6.7~8.2;和所述复合预混合饲料25。
