The invention aims to provide a freeze-dried milk block for protecting teeth, which achieves the purposes of balanced nutrition proportion, easy absorption, good effect of preventing decayed teeth and popular flavor by adjusting the components and the proportion of raw materials.
In order to achieve the purpose, the invention adopts the technical scheme that:
a freeze-dried milk block for protecting teeth is prepared from the following raw materials: 60-80 parts of raw milk, 1.5-4 parts of xylitol, 2-5 parts of whey protein powder, 20-30 parts of pulp, 5-8 parts of bean powder, 2-5 parts of nut powder, 1-3 parts of zymophyte powder and 3-6 parts of composite bacterial powder;
the pulp is prepared from the following components in percentage by weight of 4: 4: 1-3 parts of apples, pears and raisins;
the apples and the pears in the pulp are fresh apples and pears which are peeled and denucleated.
The bean flour is prepared from (by weight ratio) 1: 1-2 of hyacinth bean powder and pea powder;
the nut powder comprises the following components in percentage by weight: 1: 1-2 of hazelnut powder, black sesame powder and sunflower seed powder;
the zymophyte powder is prepared from the following components in parts by weight of 1: 1-1.5 of lactobacillus plantarum N3117 bacterial powder and lactobacillus paracasei bacterial powder;
the composite bacterial powder is prepared from the following components in parts by weight of 1: 1-1.5 of lactobacillus rhamnosus x253 bacteria powder and animal bifidobacterium bacteria powder.
The viable bacteria of the lactobacillus plantarum N3117, the lactobacillus paracasei, the lactobacillus rhamnosus x253 and the bifidobacterium animalis in the zymocyte powder and the composite powder are all 1 x 107cfu/g。
The Lactobacillus plantarum N3117 is selected from fermented dairy products of inner Mongolia, and is preserved in China general microbiological culture Collection center (CGMCC) at 12/05 in 2014, the preservation number is CGMCC NO.10133, the Latin name is Lactobacillus plantarum, and the Lactobacillus plantarum is disclosed for the first time in a patent application document with the application number of 201510936243.0 and can be obtained by the public;
the Lactobacillus rhamnosus X253 is separated and screened from traditional fermented dairy products in Xinjiang in China, the strain is preserved in the China general microbiological culture Collection center in 8-20 th month in 2019 with the preservation number of CGMCC No.18404, the Latin article name is Lactobacillus rhamnosus, and the Lactobacillus rhamnosus is disclosed for the first time in a patent application document with the application number of 202010258772.0 and can be obtained by the public;
the lactobacillus paracasei and the bifidobacterium animalis are common strains or published strains on the market and can be obtained by the public.
The invention also provides a preparation method of the tooth-protecting freeze-dried milk block, which is prepared by a fermentation freeze-drying process and comprises the following steps of:
s1, material preparation:
heating raw milk, adding xylitol, whey protein powder and bean powder, homogenizing, and sterilizing to obtain solution A;
s2, fermentation:
taking the solution A, cooling, adding zymophyte powder, dissolving, fermenting and demulsifying to obtain a solution B;
s3, precooling and drying:
and adding the pulp, the nut powder and the compound bacteria powder into the solution B, uniformly mixing, cooling, adding into a mold, and pre-freezing and vacuum freeze-drying to obtain the tooth-protecting freeze-dried milk block.
As a limitation, the temperature is raised to 50-65 ℃; the homogenizing pressure is 16-20 MPa; the sterilization is performed at 93-100 ℃ for 300-310 s.
As another limitation, in step S2, the temperature is reduced to 36-40 ℃.
As a further limitation, the fermentation time is 5-8 h.
As a third limitation, in step S3, the temperature is decreased to 10 to 30 ℃.
As a fourth limitation, the pre-freezing is freezing at-25 to-45 ℃ for 1 to 2 hours.
And as a fifth limitation, the vacuumizing is carried out until the pressure is 0-10 Pa.
Due to the adoption of the technical scheme, compared with the prior art, the invention has the technical progress that:
(1) the freeze-dried tooth-protecting milk block comprises a plurality of nutrient components for protecting teeth, wherein,
pectin contained in fructus Mali Pumilae and fructus Pyri in the pulp composition can inhibit growth of pathogenic bacteria of dental caries; the oleanolic acid, oleanal and betulin contained in the raisin can inhibit the growth of pathogenic bacteria of dental caries, is beneficial to the health of teeth and gum, and can effectively prevent dental caries, gingivitis and periodontitis;
the hyacinth bean powder and the pea powder in the bean powder composition contain a small amount of anti-nutritional factors such as tannin, phytic acid, oligosaccharide and the like, but contain high phosphorus, so that a phosphate buffer system is formed, and dental caries caused by over acidification of the oral cavity is prevented;
the hazelnut powder, the black sesame powder and the sunflower seed powder in the nut powder all contain rich arginine, and are utilized by the composite bacteria powder and an arginine deiminase system in probiotics planted in the oral cavity, so that the metabolic alkali is generated, the balance between tooth demineralization and remineralization is regulated, the acid-producing bacteria and the acid-resisting bacteria are prevented from forming dominant flora, and the formation of cariogenic dental plaque is prevented; meanwhile, vegetable oil contained in the hazelnut powder, the black sesame powder and the sunflower seed powder in the nut powder forms a hydrophobic layer on the surface of teeth, so that dentin is prevented from being dissolved, and the teeth are protected;
the probiotic combination of the lactobacillus rhamnosus x253 bacteria powder and the bifidobacterium animalis bacteria powder in the composite bacteria powder can inhibit the growth of oral pathogenic bacteria through competition, maintain the ecological balance of the oral cavity, and simultaneously utilize arginine in the nut powder to produce alkali, thereby protecting teeth;
(2) according to the preparation method of the tooth-protecting freeze-dried milk block, apples and pears are subjected to freeze-drying process, so that the tooth-protecting freeze-dried milk block has a mechanical scrubbing effect in the eating process, and dirt adhered to the surfaces of teeth can be wiped off; raw milk and bean flour are sterilized to inhibit mixed bacteria and then added with zymophyte powder, so that the bean flour is fermented by the zymophyte powder, anti-nutritional factors are reduced, and gastrointestinal adverse reactions are prevented; the raw milk is easier to be absorbed by human body through fermentation.
The preparation method is economical, simple and quick, and can be applied to industrial production.

Detailed Description
The present invention will be described in further detail with reference to specific examples. It should be understood that the described embodiments are only for illustrating the present invention and do not limit the present invention.
The experimental conditions in the following examples, if not indicated, are generally according to conventional conditions or according to the conditions recommended by the reagents company; reagents, consumables and the like used in the following examples can be obtained commercially without special instructions, analytically pure reagents meeting national standards are used during analysis, experimental water is pure water, and the used methods are conventional process methods in the field without special instructions.
EXAMPLE 1A teeth-protecting Freeze-dried milk Block
The embodiment comprises the following steps which are carried out in sequence:
s1, material preparation:
heating 60kg of raw milk to 65 ℃, adding 1.5kg of xylitol, 2.6kg of whey protein powder and 6kg of bean powder (3kg of hyacinth bean powder and 3kg of pea powder), stirring until the xylitol, the whey protein powder and the bean powder are fully dissolved, homogenizing under the pressure of 20MPa, and sterilizing at 98 ℃ for 300s to obtain a solution A;
s2, fermentation:
taking the solution A, cooling to 37 ℃, and adding a mixture of a 1: 1.5 Lactobacillus plantarum N3117(1kg, viable count 1X 10)7cfu/g) powder and Lactobacillus paracasei N1115 powder (1.5kg, viable bacteria amount is 1 × 10)7cfu/g) of 2.5kg of zymocyte powder, stirring to fully dissolve the zymocyte powder, keeping the temperature unchanged, fermenting for 6 hours, and demulsifying to obtain a solution B;
wherein, Lactobacillus paracasei N1115(Lactobacillus paracasei N1115) has been preserved in 17.3.2011 in China general microbiological culture Collection center (China Committee for culture Collection of microorganisms) No. 3 of West Lu No.1 Hospital, Beijing, Tokyo, Chaoyang, with the preservation number: CGMCC No.4691, which is disclosed in Chinese invention patent with the patent number of 201110357058.8 for the first time;
s3, precooling and drying:
adding the solution B into a mixture prepared by mixing the components in a weight ratio of (B) to (B) of 2: 2: 1 of 25kg of pulp consisting of 10kg of apples, 10kg of pears and 5kg of raisins, the weight ratio c being 2: 1: 2.5kg of nut powder consisting of 1kg of hazelnut powder, 0.5kg of black sesame powder and 1kg of sunflower seed powder, and 1 × 10 of active bacteria with equal weight (weight ratio d is 1: 1)7cfu/2 kg of composite bacterial powder consisting of 1kg of lactobacillus rhamnosus x253 bacterial powder and 1kg of bifidobacterium animalis subsp lactis i797 bacterial powder, and fully stirring and mixingMixing, cooling to 15 deg.C, adding into a mold, pre-freezing at-45 deg.C for 2 hr, vacuumizing to pressure of 10Pa, and vacuum freeze-drying for 24 hr to obtain freeze-dried block labeled as K1.
The Bifidobacterium animalis subsp 797 is separated and screened from intestinal flora of infants or children, and the strain is preserved in the China general microbiological culture collection center of the China Committee for culture Collection of microorganisms (CGMCC NO. 18403) in 8 months and 20 months in 2019, has the name of Bifidobacterium animalis subsp.lactis, and is disclosed for the first time in the Chinese patent application with the application number of 202010259210.8.
Example 2-6 tooth-protecting Freeze-dried milk block
Examples 2 to 6 are tooth-protecting freeze-dried milk blocks, and the preparation methods thereof are basically the same as those in example 1, except for the differences in the raw material ratios and the process parameters, which are specifically shown in table 1:

The contents of the other portions of examples 2 to 6 are the same as those of example 1.
Example 7 Effect test of tooth-protecting Freeze-dried milk Block
The embodiment is an effect experiment of the tooth-protecting freeze-dried milk block, and the detection of the bacterial membrane inhibition rate proves the tooth-protecting effect of the invention.
The streptococcus mutans is common cariogenic bacteria, has strong acid production and acid resistance, can generate insoluble extracellular polysaccharide by utilizing sucrose, further promotes bacterial colonies to gather on the surface of teeth, forms dental plaque, namely bacterial membrane, and finally causes caries.
The freeze-dried milk block CK1 is prepared by a method basically the same as that of the example 1, and the preparation method is different only in that compound bacterial powder is not added in raw materials;
a freeze-dried milk block CK2 was prepared in substantially the same manner as in example 1, except that nut flour was not added to the raw materials;
the detection method comprises the following steps: the inhibition rate of the bacterial membrane of the streptococcus mutans is detected for eight kinds of milk blocks, namely the tooth-protecting freeze-dried milk block K1-K6, the freeze-dried milk block CK1 and the freeze-dried milk block CK2, and the specific method comprises the following steps:
the bacterial liquid concentration of the activated streptococcus mutans is adjusted to 1.0 x 10 by using BHI liquid culture medium8CFU/mL for standby; respectively dissolving 10g of the above eight milk block samples in 100g of distilled water, centrifuging at 7000r/min for 10min, and filtering with a filter membrane to obtain supernatant;
uniformly mixing 100 mu L of streptococcus mutans bacterial liquid and 100 mu L of supernatant, adding the mixture into a 96-hole plate, taking 200 mu L of distilled water as negative control, culturing at 37 ℃ for 24h, discarding the culture solution in the hole of the 96-hole plate, and washing each hole for 3 times by PBS (phosphate buffer solution); removing PBS, adding 95% methanol into each well, fixing for 15min, removing methanol, and naturally drying at room temperature;
adding 2% of crystal violet into each hole, performing gram staining for 5min, washing with deionized water, removing redundant crystal violet dye, naturally drying at room temperature, adding 160 mu L of glacial acetic acid with volume fraction of 33% into each hole, and dissolving the crystal violet dye adhered to bacteria to obtain a solution;
transferring 125 mu L of the solution from each well to a new 96-well plate, measuring the OD value at the wavelength of 630nm, and calculating the bacterial membrane inhibition rate of each milk block sample;
the calculation formula is as follows: the biofilm inhibition (%) (1-OD milk block sample/OD negative control) × 100.
The results are shown in fig. 1, and the results show that the bacterial membrane inhibition rates of the tooth-protecting freeze-dried milk blocks K1-K6 are more than 70%, the bacterial membrane inhibition rate of the freeze-dried milk block CK1 is 14.6%, the bacterial membrane inhibition rate of the freeze-dried milk block CK2 is 41.3%, and the bacterial membrane inhibition rates of the tooth-protecting freeze-dried milk blocks K1-K6 are all significantly higher than the sum of the bacterial membrane inhibition rates of the freeze-dried milk blocks CK1, CK2 and the sum of the bacterial membrane inhibition rates of the freeze-dried milk blocks CK 3583 and the CK 2; the freeze-dried tooth-protecting milk block has good tooth-protecting effect, can effectively inhibit the generation of mycoderm causing dental caries, simultaneously, the composite bacteria powder and the nut powder interact with each other to realize synergistic interaction, and probiotics in the composite bacteria powder utilizes arginine in the nut powder to generate alkali, so that the tooth-protecting effect is better.
It should be noted that the above-mentioned embodiments are only preferred embodiments of the present invention, and are not intended to limit the present invention in other forms, and any person skilled in the art may use the above-mentioned technical content as a teaching to make changes or modifications to the equivalent embodiments with equivalent changes, but all those simple changes, equivalent changes and modifications made to the above-mentioned embodiments without departing from the technical spirit of the present invention, and still all those embodiments are within the scope of the present invention as claimed in the claims.
Claims (9)
1. The freeze-dried tooth-protecting milk block is characterized in that the raw materials for preparing the active ingredients of the freeze-dried tooth-protecting milk block comprise the following components in parts by weight: 60-80 parts of raw milk, 1.5-4 parts of xylitol, 2-5 parts of whey protein powder, 20-30 parts of pulp, 5-8 parts of bean powder, 2-5 parts of nut powder, 1-3 parts of zymophyte powder and 3-6 parts of composite bacterial powder;
the pulp is prepared from the following components in percentage by weight of 4: 4: 1-3 parts of apples, pears and raisins;
the soybean powder is prepared from the following raw materials in parts by weight of 1: 1-2 of hyacinth bean powder and pea powder;
the nut powder comprises the following components in percentage by weight: 1: 1-2 of hazelnut powder, black sesame powder and sunflower seed powder;
the zymophyte powder is prepared from the following components in parts by weight of 1: 1-1.5 of lactobacillus plantarum N3117 bacterial powder and lactobacillus paracasei bacterial powder;
the composite bacterial powder is prepared from the following components in parts by weight of 1: 1-1.5 of lactobacillus rhamnosus x253 bacteria powder and animal bifidobacterium bacteria powder.
2. The freeze-dried tooth-protecting milk block as claimed in claim 1, wherein the viable bacteria amount of lactobacillus plantarum N3117, lactobacillus paracasei, lactobacillus rhamnosus x253 and bifidobacterium animalis in the fermented bacteria powder and the composite bacteria powder is 1 x 107cfu/g。
3. A process for the preparation of a freeze-dried tooth-protecting milk block as claimed in claim 1 or 2, characterized in that it comprises the following steps carried out in succession:
s1, material preparation:
heating raw milk, adding xylitol, whey protein powder and bean powder, homogenizing, and sterilizing to obtain solution A;
s2, fermentation:
taking the solution A, cooling, adding zymophyte powder, dissolving, fermenting and demulsifying to obtain a solution B;
s3, precooling and drying:
and adding the pulp, the nut powder and the compound bacteria powder into the solution B, uniformly mixing, cooling, adding into a mold, and pre-freezing and vacuum freeze-drying to obtain the tooth-protecting freeze-dried milk block.
4. The method for preparing the freeze-dried tooth-protecting milk block according to claim 3, wherein the temperature rise is to 50-65 ℃; the homogenizing pressure is 16-20 MPa; the sterilization is performed at 93-100 ℃ for 300-310 s.
5. The method for preparing a freeze-dried tooth-protecting milk block according to claim 3 or 4, wherein in step S2, the temperature is reduced to 36-40 ℃.
6. The method for preparing the freeze-dried tooth-protecting milk block according to claim 5, wherein the fermentation time is 5-8 h.
7. The method for preparing a freeze-dried tooth-protecting milk block according to claim 3, 4 or 6, wherein in step S3, the temperature is reduced to 10-30 ℃.
8. The method for preparing the freeze-dried tooth-protecting milk block according to claim 3, 4 or 6, wherein the pre-freezing is carried out at-25 to-45 ℃ for 1 to 2 hours.
9. The method for preparing a freeze-dried tooth-protecting milk block according to claim 3, 4 or 6, wherein the vacuum is applied until the pressure is 0-10 Pa.
