本发明提供一种含有奥西替尼的荧光探针,所述荧光探针包括吲哚菁绿和奥西替尼,重量比为1:0.1‑5,所述荧光探针为自组装结构,在电子显微镜下能够形成一种稳定的结构,能够有效识别EGFR受体表达的肿瘤细胞,并持久示踪,且具有肿瘤细胞杀伤功能,成为一种多功能示踪探针,并能和某些疗法联用,具有更好的肿瘤杀伤效果,具有医药生产的应用前景。

技术领域
本发明涉及一种含有奥西替尼的荧光探针、制备方法及其应用,属于药品和示踪领域。
背景技术
癌症是威胁人类生命健康的重大疾病之一,其中肺癌的死亡率居全球第一,而早期诊断与有效的治疗可显著提高患者的生存率。
然而,如何找到肿瘤病灶已经成为示踪领域的一个难题,特别是针对隐匿性早期肺癌病灶,找到相关的病灶部位才能进行后期的治疗与观察,临床目前主要用于识别肺癌的方法仍为胸部CT扫描,尽管随着CT分辨率升高其敏感性大大增高,但其特异性差,易出现误诊及漏诊。此外,对于晚期肺癌,尤其是小分子靶向药物耐药者,缺乏有效治疗方法。
因此,本领域存在一种技术难题,即癌症检测中,早期对微小或隐匿性病灶显影困难,易出现漏诊;而晚期肺癌治疗效果差,口服抗癌药物来进行治疗,透过多层屏障,药效差且不良反应较多。
上述问题对于检测领域提出了产品研发需求,是否可以开发一种探针,能够特异性的结合相关病灶部位,且稳定结合时间长,进而伴随有治疗功能,实时示踪肿瘤细胞的变化,为检测病人体征提供可靠的参考证据,直观地反映治疗进程。
但此时带来一种技术问题,需要采用何种给药方式、采用何种探针才能达到这一目的,成为了本领域开发时的难题。
发明内容
为解决上述的技术问题,本发明提供了一种含有奥西替尼的荧光探针,其中所述荧光探针由吲哚菁绿和奥西替尼组成,所述吲哚菁绿和奥西替尼的结合为自组装,重量比为1:0.1-5。
这是一种全新的探针组合,首先发明人进行了前期的筛选,采用的荧光探针为染料与药物结合的方式进行筛选,而染料需要具有光学激发和吸收功能,药物应当与染料稳定的结合,不容易发生解离,所构成的荧光探针不应具有明显的毒性。
基于上述考虑,发明人对于肺癌肿瘤进行文献检索,发现非小细胞肺癌占肺癌的85%,且普遍存在EGFR受体高表达,因此发明人阐释开发一种针对于EGFR+肺癌细胞的荧光探针,初步确定其靶向性。
目前以EGFR为靶点的抗肿瘤药物主要分为两大类:一是作用于受体胞外区的单克隆抗体,临床常用的是西妥昔单抗;二是作用于受体胞内区的小分子酪氨酸激酶抑制剂,临床常用的是第一代吉非替尼、厄洛替尼,第二代阿法替尼和第三代奥西替尼。
而关于染料的选择,发明人选取了吲哚菁绿(ICG),这是一种具有近红外特征吸收峰的三碳花菁染料,是唯一一种被美国食品药品监督管理局(FDA)批准的可用于临床诊断的近红外荧光染料,但ICG具有稳定性差,代谢快,非靶向性等缺点,因此,检测领域开发,很少单独使用ICG进行探针的开发,ICG具有其作为探针的稳定性缺点。
目前的荧光探针主要通过发光基团和某些物质的外部修饰,使其具有某种结构,从而特异地结合到细胞或器官的某些位点上,这种探针存在的缺点:生产复杂,质控要求高,存在某些安全性风险,体内实验可能特异性不高。
因此发明人力求开发一种简单的荧光探针,最好是自然状态下,染料和药物能够发生自然结合,且结合稳定。
根据临床试验对于EGFR+肺癌的治疗效果、缓解率及副作用等因素考虑目前优先选择吉非替尼与奥西替尼。
申请人前期分别把吉非替尼与奥西替尼(Osi)和吲哚菁绿(ICG)荧光染料结合,结果发现吉非替尼与吲哚菁绿的结合率较低、溶解性较差、黏连不分散,有趣的是,奥西替尼能有效地与吲哚菁绿结合并且分散性及溶解性良好,初步符合了荧光探针的要求。并且意外发现,奥西替尼和ICG的结合能够在体内实验中稳定存在较长时间,稳定结合靶细胞,并发挥持久杀伤功能,这成为了本发明开发得到的亮点,因为并不是所有药物都可以与ICG进行有效结合的。
进一步地进行浓度筛选,所述吲哚菁绿和奥西替尼的重量比为1:0.5-2。优选1:0.8-1.2,最优选1:0.8。
根据荧光探针本身的特点,推荐将该探针使用药学常规冻干技术制备成冻干注射粉剂,前期实验,实验人员发现,将其进行冻干后,室温避光保存,存在1年内,水复溶后,荧光探针依旧可以正常使用,说明该类产品复溶稳定性。
申请人进一步地提供一种上述荧光探针的制备方法,包括以下步骤:
1)称取奥西替尼,溶解于有机溶剂,配成奥西替尼溶液;
2)称取吲哚菁绿,溶于水中,配成吲哚菁绿溶液;
3)将奥西替尼溶液滴入吲哚菁绿溶液,避光搅拌1-12h,离心;
4)弃上清,水重悬,得到所述荧光探针。
所述有机溶剂为甲醇、乙醇或DMSO,优选DMSO。
奥西替尼是难溶性物质,不溶于水,在二甲基亚砜中的溶解性最高,还能溶解在甲醇和乙醇中,但甲醇和乙醇具有较强的挥发性及较低的溶解性,所以优先考虑二甲基亚砜。
所述方法中,步骤4)还包括离心洗涤的步骤,即,弃上清,水重悬,离心,弃上清,水重悬,得到所述荧光探针溶液。多次洗涤(1-3次),有助于洗涤探针中游离的吲哚菁绿和奥西替尼。
本发明进一步提供一种上述荧光探针在制备肿瘤示踪试剂盒中的应用,其中,所述肿瘤为EGFR表达的肿瘤。
研究发现,本发明所提供的荧光探针具有以下两个优点:
1)能够识别EGFR表达的肺癌细胞,包括野生型EGFR表达和突变型EGFR表达,具有更广泛的应用特性。
2)能够识别Osi抵抗的肺癌细胞,为后期的治疗提供辅助功能,由于光热治疗需要确定相关的病灶部位,而收到激光的刺激,ICG能够得到激发,辅助相关的治疗,增加了所述荧光探针的意义,这也是目前其他荧光探针所不具有的特性,即,绕开了原来的Osi口服治疗途径,为精准光热治疗奠定了基础。
本发明进一步提供一种荧光探针在制备治疗肿瘤的药物中的应用,其特征在于,所述肿瘤为EGFR表达的肺癌肿瘤。
本发明进一步提供一种荧光探针在制备光热治疗剂中的应用,所述光热治疗剂应用于肺癌示踪和治疗。
在上述应用中,所述EGFR包括野生型EGFR和突变型EGFR。即具有更加广泛的应用范围,不受EGFR突变的影响。
进一步地,所述肺癌肿瘤细胞包括奥西替尼不敏感(口服Osi治疗抵抗)的细胞。
有益效果:
申请人经过筛选,开发出一种自组装的的荧光探针,具有组装简单、示踪时间长(96h,常规的ICG荧光探针的示踪时间通常为48h),低毒作用(极高剂量下,1.5mg/kg(以吲哚菁绿计算)未产生相关的细胞伤害),并进一步提供一种试剂盒,包含了光源激发设备,该探针在吲哚菁绿近红外区具有较强的荧光及光学吸收性能,而且可以杀死肿瘤细胞,在细胞和动物水平上实现联合治疗与成像。可以实现实时活体成像,无创,具有准确性高,副作用小等特点。
1)应用范围广,不仅适用于肺癌,也适用于EFGR表达的其他肿瘤细胞的示踪活动。
2)识别能力强,不仅能够识别EGFR野生型肿瘤细胞,也能EGFR突变型肿瘤细胞。
3)功能更强,不仅能够提供肿瘤细胞的示踪作用,更具有肿瘤细胞杀伤作用。
4)进一步结合光热治疗,提高了Osi抗性细胞的杀伤作用,解决了Osi抵抗性肺癌的治疗难题,成为重要的光热治疗剂。

具体实施方式
以下优选实施例仅用以说明本发明的技术方案而非限制,尽管通过下述优选实施例已对本发明进行了详细的描述,但本领域技术人员应当理解,可以在形式上和细节上对其作出各种各样的改变,而不偏离本发明权利要求书所限定的范围。
本发明中未特殊规定溶剂的情况下,均指水溶液,在未规定温度时,均指室温。
实施例1
(一)吲哚菁绿和奥西替尼荧光探针的制备
(1)取0.8mg奥西替尼(Selleckchem,规格:100mg,S7297)粉末溶解于100ul二甲基亚砜(DMSO)中,配成浓度为8mg/ml的溶液;
(2)取1mg吲哚菁绿(丹东医创药业有限责任公司,规格:25mg,H20055881)溶于1ml去离子水中,配成浓度为1mg/ml的溶液;
(3)将浓度为1mg/ml的吲哚菁绿溶液置于磁力搅拌器上,用100ul移液枪取8mg/ml奥西替尼溶液100ul,一滴一滴缓慢滴加入吲哚菁绿溶液中;
(4)在磁力搅拌器上室温避光过夜搅拌;
(5)搅拌后的溶液以20000g,30min室温离心;
(6)离心后,弃上清,并用1ml去离子水重悬,20000g,30min室温离心;(离心洗涤)
(7)弃上清,用1ml去离子水重悬,得到吲哚菁绿与奥西替尼的复合物(ICG-Osi)如图1所示。
(二)ICG-Osi荧光探针的表征
(1)通过透射电子显微镜(TEM)观察到ICG-Osi探针呈现大小均一、圆形形态,粒径约200-300nm之间,如图1所示。
(2)通过Malvern粒径仪测得ICG-Osi粒径约为260nm左右。
(3)通过多功能酶标仪测得ICG与ICG-Osi的紫外吸收光谱,与ICG相比,ICG-Osi的紫外吸收峰出现明显的红移,证明了两者之间是通过π-π堆积共轭作用自组装形成的复合物,而并未改变其本身的化学结构与性能,如图3所示。
(4)通过荧光分光光度计测得ICG与ICG-Osi的荧光吸收光谱,结果证明了两者之间的结合并未影响ICG本身的荧光性能,如图4所示。
结论:通过对ICG-Osi相关性能的检测,证明了通过自组装的方式成功合成具有一定功能的荧光探针。
(三)细胞水平验证ICG-Osi对于EGFR+肺癌细胞的特异靶向性
(1)首先通过WesternBlot实验检测不同肺癌细胞系的EGFR的表达水平,如图5所示,EGFR+的肺癌细胞系为PC9、A549、H1975,EGFR-肺癌细胞系为H520,并分别作为阳性实验组与阴性对照组进行以下实验。
(2)把ICG-Osi荧光探针以2μg/ml的浓度分别与PC9、A549、H1975、H520细胞系孵育6h,并用DAPI染料染细胞核20min,置于荧光显微镜下检测ICG-Osi对于不同肺癌细胞系的靶向性,结果发现,与EGFR-肺癌细胞系H520相比,ICG-Osi荧光探针对于EGFR+肺癌具有特异的靶向性,并且定位于细胞膜,与EGFR表达的部位相同。其中A549是wtEGFR,H1975和PC9是mutEGFR,说明本探针可以识别和靶向EGFR的肺癌细胞,不论野生型还是突变型,具有更广泛的应用前景。
(3)进一步通过流式细胞术定量检测ICG-Osi荧光探针对于EGFR+肺癌细胞的靶向性,通过检测平均荧光强度反应其细胞水平的靶向性,结果如图6所示,与EGFR-肺癌细胞系H520相比,ICG-Osi荧光探针对于EGFR+肺癌具有特异的靶向性,且结果具有统计学差异。
(4)通过CCK8细胞增殖试验检测ICG-Osi在细胞水平上的生物安全性,如图7所示,ICG-Osi荧光探针以不同浓度与正常支气管细胞(BEAS-2B)共孵育24h后测其细胞活性,结果表明随着ICG-Osi荧光探针浓度的增加,其细胞活性未受明显影响,证明ICG-Osi荧光探针在细胞水平上具有一定的生物安全性,不影响正常细胞的生长与增殖。
结论:通过以上实验数据表明该探针能特异靶向EGFR+肺癌细胞,并且对于正常支气管上皮细胞无明显的生物毒性。
(四)动物水平验证ICG-Osi对于EGFR+肺癌细胞的特异靶向性
(1)首先通过小动物荧光成像仪器检测ICG-Osi的荧光性能,随着ICG-Osi浓度的增加,其荧光强度逐渐增强,表明其具有荧光成像的性能,能进一步应用于小动物活体成像。
(2)皮下接种EGFR+肺癌细胞系A549于裸鼠大腿皮下,约3-4周后成瘤,肿瘤体积约为100mm3时,以1.5mg/kg的ICG浓度通过尾静脉注射ICG与ICG-Osi荧光探针,检测其不同时间点体内荧光强度的分布情况,结果如图8所示,与ICG组相比,ICG-Osi组在第3小时时肿瘤部位就有明显的荧光富集,并且在第48小时时,其他部位的荧光基本代谢完了,只有肿瘤部位有显著的荧光富集,并且在48h后,取出心、肝、脾、肺、肾、肿瘤检测离体组织的荧光强度,如图9所示,48小时后,心、肝、脾、肺、肾的荧光基本代谢完,只有ICG-Osi组肿瘤部位有明显的荧光富集,后期发现ICG-Osi能够持续96个小时(黑白图像显示不太清晰),并且与ICG组相比具有明显的统计学差异。
(3)通过取对照组(PBS)及实验组(ICG-Osi)小鼠的主要脏器,如心、肝、脾、肺、肾,做石蜡切片HE染色,检测该探针在动物水平上的生物安全性,结果如图10所示,与对照组相比,实验组(ICG-Osi)的器官病理未见明显损伤,证明该探针在动物水平上也具有一定的生物安全性。(1.5mg/Kg,奥西替尼临床口服给药是每天每人80mg的剂量(无需考虑年龄、体重等相关因素);
本发明静脉给药剂量范围是1-5mg/kg(这是ICG的人体安全浓度)的剂量范围,按照ICG:Osi1:0.8计算,裸鼠体重为20g左右,根据这样的范围计算平均一只小鼠奥西替尼的用量大概是16ug-80ug之间,Osi静脉给药剂量相当于奥西替尼的人体口服剂量的千分之一,这足以保证其高安全性。)
结论:通过以上实验数据表明该探针能在体特异靶向EGFR+肺癌,并且其具有高效靶向性、持续时间长、特异性好等优点,且对于主要脏器无明显损伤,具有良好的生物安全性。
(五)ICG-Osi联合光热治疗抑制EGFR+肺癌细胞
(1)通过CCK8细胞增殖试验检测ICG-Osi对于肺癌细胞系H1975的抑制能力,结果如图11所示,与ICG相比,随着ICG-Osi浓度的增加,H1975细胞活性逐渐降低,表明该探针本身在一定程度上具有抑制EGFR+肺癌细胞的能力。
进一步检测ICG-Osi联合光热治疗对于肺癌细胞系H1975的杀伤作用,结果如图12所示,ICG-Osi联合光热治疗后对H1975肺癌细胞的抑制作用进一步增强(与图11相比),表明H1975细胞的死亡是由于奥西替尼联合ICG的光热作用而产生的较强的杀伤作用,这说明ICG-Osi对于EGFR+肺癌不仅有靶向治疗的性能,还具有良好体外光热治疗的效果,且两者的联合治疗效果比单一治疗效果更显著,这也是荧光探针普遍不具有的性质。
(2)进一步地,由于晚期肺癌细胞系,常常出现具有抗药性的肺癌细胞,而这类细胞对于小分子靶向药物(例如Osi抗性肺癌细胞)是具有抗性的,例如口服Osi无法治疗的肺癌细胞,研究人员专门选取H1975(对奥西替尼敏感的肺癌细胞)和H1975-OR(对于奥西替尼具有抗性的肺癌细胞)进行研究,发现H1975-OR对于实施例1的探针具有抵抗性,研究人员进一步对于H1975-OR肺癌细胞进行ICG-Osi联合光热治疗方式,发现这种方式能够杀灭这类细胞,这也为开发治疗Osi抗性肺癌的试剂盒提供了新的工具。图13的流式定量方法确认(本发明荧光探针对于H1975和H1975-OR均有识别结合的功能,而后起到示踪作用,为光热治疗奠定基础)
注:光热治疗是一种新颖的肿瘤细胞热消融方法,其主要的原理是在激光的照射下,利用光热转换产生热效应来直接杀死肿瘤细胞。近红外是一种非侵入性的用于肿瘤光热治疗的新工具,能够有效穿透正常组织达到肿瘤部位,可以减少对正常组织的伤害。
本发明中涉及的相关实验步骤,未详述部分为本领域公知技术和本领域普通技术人员能够理解并实现的部分。
一、细胞光热治疗实验步骤
(1)细胞铺板,96孔板,每孔5000个细胞,100ul培养基,分为四组,ICG、ICG-Osi、ICG-laser、ICG-Osi-laser,每组3个副孔,置于细胞培养箱中培养1-2天左右
(2)分别加不同浓度的ICG和ICG-Osi共孵育12h(浓度设置0、2、4、6、8、10ug/mL)
(3)弃原培养基,用PBS洗3次,换新的培养基
(4)ICG-laser、ICG-Osi-laser于红外热成像仪808nm激光器下照射5min(1W/cm2)
(5)四组同时置于细胞培养箱中继续培养4h
(6)加CCK8试剂,置于培养基中2-3h,测波长450nm处的OD值,计算细胞活性=(实验组OD值/对照组OD值)*100%。
二、动物光热治疗实验步骤
(1)通过在裸鼠(免疫缺陷小鼠)大腿皮下接种H1975肺癌细胞构建动物肺癌模型,待肿瘤体积约为100mm3时,以1.5mg/kg的ICG浓度通过尾静脉注射PBS(空白对照)、ICG、ICG-Osi;
(2)注射后48h时(因为前面实验发现48h时体内的其他部位的荧光基本代谢完,只有肿瘤部位有明显的荧光富集),用红外热成像仪808nm激光器下照射5min(1W/cm2);
(3)两天监测一次小鼠的体重及肿瘤体积的变化,持续监测2周;
(4)2周后取小鼠的肿瘤组织比较其肿瘤体积大小,取小鼠主要脏器(心、肝、脾、肺、肾)进行HE染色病理检测。
三、动物水平上ICG-Osi的示踪能力检测
(1)通过在裸鼠(免疫缺陷小鼠)大腿皮下接种A549肺癌细胞构建动物肺癌模型,待肿瘤体积约为100mm3时;
(2)以1.5mg/kg的ICG浓度通过尾静脉注射ICG与ICG-Osi荧光探针,用小动物活体成像系统检测其不同时间点体内荧光强度的分布情况;
(3)48h时取小鼠的肿瘤组织、心、肝、脾、肺、肾检测离体器官的荧光水平;
四、ICG-Osi对奥西替尼敏感的肺癌细胞(H1975)及耐药的肺癌细胞(H1975-OR)都具有靶向性,见图13。(6h内探针与靶细胞的结合)
通过流式细胞术检测发现,ICG-Osi荧光探针不仅可以靶向对奥西替尼敏感的EGFR+肺癌细胞,还能靶向对奥西替尼耐药的EGFR+肺癌细胞,并且有望实现对奥西替尼耐药肺癌细胞进行光热治疗来抑制肿瘤的增殖。
结论:ICG-Osi不仅本身对EGFR+肺癌细胞具有靶向示踪和治疗的作用,还能协同光热治疗,起到对EGFR+奥西替尼抗性的肺癌细胞较强的杀伤作用。
实施例2
为了优化吲哚菁绿与奥西替尼结合的最佳比例,分别以吲哚菁绿与奥西替尼配比为1:1、5:4、4:5制备荧光探针,结果发现当吲哚菁绿与奥西替尼两者以5:4的比例结合时,其复合物的载药量更高、分散性更好、大小更均一,而研究发现荧光探针配置为溶液,浓度为吲哚菁绿0.95-1mg/ml,奥西替尼0.75-0.8mg/ml,效果均有效。

实施例3
一种肿瘤示踪的荧光探针试剂盒,所述试剂盒包括吲哚菁绿和奥西替尼通过实施例1制备得到的荧光探针。肿瘤针对EGFR表达的肿瘤细胞,例如肺癌细胞。
实施例4
一种治疗肿瘤的荧光探针试剂盒,所述试剂盒包括吲哚菁绿和奥西替尼通过实施例1制备得到的荧光探针。肿瘤针对EGFR表达的肿瘤细胞,例如肺癌细胞。
实施例5
一种荧光探针的冻干注射粉剂,通过实施例1制备得到的荧光探针,进行药学上的冻干制剂工艺,制备得到该荧光探针注射粉剂。使用方法,结合光热治疗仪进行相关肿瘤的质量,例如肺癌肿瘤,具体为非小细胞癌等。
实施例6
一种光热治疗剂,包括了实施例1所述的荧光探针。使用方法,结合光热治疗仪进行相关肿瘤的质量,例如肺癌肿瘤,具体为非小细胞癌等。

1.一种含有奥西替尼的荧光探针,其特征在于,所述荧光探针由吲哚菁绿和奥西替尼组成,所述吲哚菁绿和奥西替尼的结合为自组装,重量比为1:0.1-5。
2.根据权利要求1所述的荧光探针,其特征在于,所述吲哚菁绿和奥西替尼的重量比为1:0.5-2。
3.根据权利要求1所述的荧光探针,其特征在于,所述吲哚菁绿和奥西替尼的重量比为1:0.8-1.2。
4.根据权利要求1所述的荧光探针,其特征在于,所述吲哚菁绿和奥西替尼的重量比为1:0.8。
5.根据权利要求1所述的荧光探针,其特征在于,所述荧光探针为冻干注射粉剂。
6.如权利要求1所述的荧光探针的制备方法,其特征在于,包括以下步骤:
1)称取奥西替尼,溶解于有机溶剂,配成奥西替尼溶液;
2)称取吲哚菁绿,溶于水中,配成吲哚菁绿溶液;
3)将奥西替尼溶液滴入吲哚菁绿溶液,避光搅拌1-12h,离心;
4)弃上清,水重悬,得到所述荧光探针。
7.根据权利要求6所述制备方法,其特征在于,所述有机溶剂为甲醇、乙醇或DMSO。
8.根据权利要求7所述制备方法,其特征在于,所述有机溶剂为DMSO。
9.权利要求1所述的荧光探针在制备肿瘤示踪试剂盒中的应用,其特征在于,所述肿瘤为EGFR表达的肺癌肿瘤。
10.权利要求1所述的荧光探针在制备治疗肿瘤的药物中的应用,其特征在于,所述肿瘤为EGFR表达的肺癌肿瘤。
11.根据权利要求9所述的荧光探针在制备肿瘤示踪试剂盒中的应用,其特征在于,所述EGFR包括野生型EGFR和突变型EGFR。
