Abstract
本发明属于生物技术领域,公开了一种肿瘤相关性血管周细胞亚群及制备方法与应用,所述肿瘤相关性血管周细胞亚群表达GJB2。本发明利用高维单细胞测序技术对浸润性肺腺癌患者的原发灶、转移阳性淋巴结和非转移淋巴结进行单细胞转录组分析并下游细胞功能学验证,首次揭示浸润性肺腺癌肿瘤微环境中存在独立的肿瘤相关性血管周细胞亚群,特异性表达PDGFRb+GJB2+CSPG4‑为“身份标签”,后经特征抗体流式纯化及免疫荧光鉴定,成功分选并扩增、保种该亚群,进一步细胞功能学验证其可显著促进肺癌细胞的侵袭能力,并具有幼稚化、畸形的血管形成能力,是领域内研究肺癌肿瘤血管异常机制和靶向血管正常化药物的理想细胞工具。

技术领域
本发明属于生物技术领域,具体涉及一种肿瘤相关性血管周细胞亚群及制备方法与应用。
背景技术
近年来肿瘤领域的研究热点是肿瘤微环境内各类细胞与肿瘤细胞间的互作关系,探讨其调控肿瘤发生发展和转移的功能机制,随着单细胞组学的深入应用,研究人员得以从更高维度探索肿瘤微环境(Tumor Microenvironment,TME)异质性对肿瘤进展的影响,其中,基质细胞形成的纤维骨架和异质血管结构为肿瘤的生存侵袭和免疫逃逸提供了实质基础。
血管周细胞(Pericyte,PC)在肿瘤微环境中普遍存在,最新的研究发现,周细胞可通过高表达黏着斑激酶FAK介导Gas6-Axl信号轴主动调控肿瘤血管新生及肿瘤生长;另有研究指出周细胞的覆盖缺失可抑制肿瘤生长,但同时会增加肿瘤的侵袭能力,其中机制与缺氧、肿瘤上皮间质化(Epithelial to Mesenchymal Transition,EMT)和Met受体激活相关。另一项有趣的研究发现,经黑色素瘤驯化的周细胞可通过RGS5-IL-6信号通路诱导CD4+T细胞失能并降低T细胞的增殖,从而协助肿瘤细胞逃避免疫监视,据此提示周细胞或在肿瘤转移中扮演重要角色。此外,肿瘤血管具有高度增殖、异常扩张和广泛渗漏的特点,是肿瘤塑造乏氧高间质压等免疫抑制微环境的结构基础。其中血管周细胞位于内皮细胞、免疫细胞和肿瘤细胞三者交界,在转移微环境中发挥重要的门户作用,目前肿瘤抗血管治疗的核心是抑制血管新生,旨在剥夺肿瘤获氧及营养物质的途径,但越来越多的证据显示抗血管治疗在抑制肿瘤生长的同时,会增加肿瘤转移的风险,而这一经典悖论的机制与肿瘤血管正常化理念密切相关,其中血管正常化的核心是重塑肿瘤血管结构,改善临床药物的血管输送途径,理论上可显著增敏小分子靶向药和免疫治疗。因此针对血管正常化治疗联合靶向或免疫治疗的转化探索正成为领域内新的研究热点。

本发明的第二个方面,提供本发明第一方面的肿瘤相关性血管周细胞亚群的制备方法,包括以下步骤:使血管周细胞群与结合剂接触,所述结合剂与GJB2基因或其表达产物相结合;分选与所述结合剂结合的周细胞,得到富集的肿瘤相关性血管周细胞亚群。
优选地,所述结合剂选自核酸、配体、酶、底物和/或抗体。

本发明提供了一种肿瘤相关性血管周细胞亚群,所述肿瘤相关性血管周细胞亚群表达GJB2。该肿瘤相关性血管周细胞亚群可以显著促进肺癌细胞系的侵袭,显著抑制PC-9、A549、H1299的生长,且随着共培养的时间的增加,其抑制效果增加,符合实际肿瘤转移的生物学特征,为肿瘤基质血管领域研究提供了特异性的周细胞亚群,为未来肿瘤治疗的精准化研究提供了针对性的靶细胞,作为基础科研和肿瘤血管类新药转化的特征细胞,可用于筛选治疗肿瘤的候选药物。
本发明提供了第一方面的肿瘤相关性血管周细胞亚群的制备方法,包括以下步骤:将肿瘤组织与消化液混合,消化,重悬,加入结合剂,所述结合剂与GJB2基因或其表达产物相结合,分选与所述结合剂结合的周细胞,得到富集的肿瘤相关性血管周细胞亚群,该方法简单,可快速获得肿瘤相关性血管周细胞亚群。
本发明还提供了一种试剂盒,包含能与基因GJB2或其表达的蛋白相结合的结合剂,可用于鉴定、筛选或分选本发明第一方面的肿瘤相关性血管周细胞亚群,诊断肿瘤。

实施例1浸润性肺腺癌肿瘤微环境单细胞转录分析以及CAP特征表型蛋白鉴定
利用10X Genomics公司单细胞转录组测序技术对四例突变背景浸润性肺腺癌患者的原发灶、转移阳性淋巴结和阴性淋巴结共十二个样本(近七万细胞)进行单细胞转录组测序(图1);并通过质控、标准化校正、缺失整合和可视化预处理,利用SingleR内嵌参考数据集对所得到的序列进行全细胞簇注释(图2、图3、图4),全景展示出浸润性肺腺癌肿瘤微环境的基因表达谱,其中针对肿瘤微环境中的基质细胞和血管周细胞(Pericyte,PC)进行特征选择(即使用降维无监督算法的同时,利用CD31-PDPN-PDGFRb+周细胞经典分子标志物聚类)下的重聚类分析(图5、图6、图7),首次揭示出浸润性肺腺癌肿瘤转移微环境中存在两个独立的血管周细胞亚群,即PDGFRb+CSPG4+PC和PDGFRb+CSPG4-PC(图8),前者因表达经典周细胞特征标志物PDGFRb和CSPG4,将其定义为经典周细胞亚群,后者经分析发现该亚群的基因集显著富集在肿瘤EMT通路并高表达EMT核心转录因子TWIST1(图9、图10、图11、图12、图13、图14),其特征基因亦多与预后不良相关(图15),据此鉴定该亚群为肿瘤相关性血管周细胞亚群(Cancer Associated Pericyte,CAP)。
针对肿瘤相关性血管周细胞亚群的发生和分化,通过利用Monocle算法对肺腺癌转移微环境血管周细胞亚群的转分化过程进行轨迹分析(图16、图17、图18),动态展示亚群内差异基因的变化过程,其中发现缝隙连接蛋白GJB2是经典周细胞亚群向CAP亚群分化过程中变化最早且变化丰度最显著的分子;结合TCGA生存分析表示肺腺癌I期病人高表达GJB2与预后不良显著相关(图15),可见,GJB2为CAP特异蛋白标志物。
实施例2肺癌肿瘤相关性周细胞亚群分离纯化
通过自建酶消化系统对肺癌血管周细胞进行分离,具体步骤如下:临床获取手术切除后的新鲜肺癌组织块,PBS冲洗三次,置于装有50mL RPMI1640细胞培养基的离心管中,封口胶封闭,立即4℃保存运输至实验室;在生物安全柜中取出肺癌组织块于10cm培养皿中,用手术刀片配合眼科剪切碎肺癌组织至小于0.5cm的小块,并剔除肺癌组织块周围脂肪及纤维接连组织;向培养皿中加入2mL含有4mg/mL分离酶II(Dispase II)、0.6mg/mL I型胶原酶(Collagenase I)、0.6mg/mL IV型胶原酶(Collagenase IV)和0.3mg/mL DNA酶I(DNase I)的RPMI-1640培养基;将装有肺癌组织碎块的培养皿放入细胞培养箱(37℃,5%CO2),静置消化60min。
将消化好的肺癌组织碎块置于离心管中,室温下5000rpm离心5min,弃去上清,加入1mL RPMI1640细胞培养基重悬细胞,过80μm尼龙网,取滤液,2500rpm离心5min,弃去上清,继续加入1mL RPMI1640细胞培养基悬浮细胞,过45μm尼龙网,得到肺癌周细胞单细胞悬液,利用细胞计数仪对单细胞悬液进行计数,并利用台酚蓝染色法检测单细胞悬液的细胞活力(单细胞悬液的活细胞率大于90%)。
取肺癌血管周细胞单细胞悬液(5×106个细胞),加入2mL含有目标抗体(anti-CD45,anti-CD31,anti-PDPN,anti-GJB2,浓度均为0.5%)的FACS buffer(流式分选策略为混合细胞悬液去粘去死细胞后排除髓系来源抑制性细胞(CD45-)、血管内皮细胞(CD31-)和淋巴内皮细胞(PDPN-),同时富集表达特征表面抗原(GJB2+)的CAP,如图19所示),4℃避光孵育20min,使用FACS Aria III流式仪(美国BD公司)分析荧光细胞,得到肿瘤相关性血管周细胞亚群(CAP)。将CAP转至24孔板内(细胞密度为(0.5~2)×105/孔),加入肿瘤相关性血管周细胞完全培养基进行原代培养,24h后移至培养皿中扩大培养,根据细胞增殖及生长状态进行传代和细胞冻存;同时,进行免疫荧光鉴定(如图20所示);选择活力强(活细胞率大于90%)、免疫荧光鉴定正确(PDGFRb+GJB2+)的细胞建库。
肿瘤相关性血管周细胞完全培养基的制备方法为:在H-DMEM培养基中加入10%胎牛血清(Fetal Bovine Serum,FBS)和1%Penicillin/Streptomycin,即得到肿瘤相关性血管周细胞完全培养基。
效果实施例
1.Transwell侵袭实验及肺癌细胞系体外共培养实验
利用Transwell小室(含预涂基质胶Matrigel的8μm嵌套膜,3422-48EA,美国康宁Corning公司)对CAP进行侵袭实验。实验组:3个肺癌细胞组(PC-9、A549、H1299),其中上室接种肺癌细胞(1×104个/孔,3复孔),下室接种CAP(5×104/孔,3复孔);阳性对照组:3个肺癌细胞组(PC-9、A549、H1299),其中上室接种肺癌细胞(1×104个/孔,3复孔),下室接种人淋巴管内皮细胞(LEC)(5×104/孔,3复孔);空白对照组:3个肺癌细胞组(PC-9、A549、H1299),其中上室接种肺癌细胞(1×104个/孔,3复孔),下室不接种任何细胞。在各组实验的下室加入500μL含10%血清的RPMI1640细胞培养基,在细胞培养箱(37℃,5%CO2)中培养24h;24h后取出嵌套膜将其下表面浸泡在70%甲醇溶液中,加入1mL 0.1%结晶紫染色固定30min,用常规正置显微镜镜检拍照(图21中A),并采用直接计数法计算嵌套膜下表面穿透细胞膜的细胞数(参考九宫格形式,选取9个视野计数破膜的紫染细胞,取平均值),结果输出为相应的柱状图(图21中B)。
由图21可以看出,通过Transwell侵袭实验,CAP可以显著促进肺癌细胞系的侵袭,尤其是对H1299的侵袭能力极为显著;对比LEC对肺癌细胞系的侵袭能力,CAP的侵袭能力显著强于LEC。
利用康宁0.4μm嵌套膜及24孔培养板,构建CAP与肺癌细胞系体外共培养体系,结合CCK-8试剂评估CAP对肿瘤细胞的增殖情况。实验组分4组,3个肺癌细胞组(PC-9、A549、H1299)和1个正常肺上皮组(BEAS-2B),其中上室接种肺癌细胞和正常肺上皮细胞增殖(5x104个/孔,3复孔),下室接种CAP(1×104/孔,3复孔),同理,对照组分4组,上室接种肺癌细胞和正常肺上皮细胞增殖(5×104个/孔,3复孔)下室不接种CAP;在下室加入50μLCCK-8溶液,采用多时间点的培养方式(24h、48h、72h)在细胞培养箱(37℃,5%CO2)中进行培养,用常规正置显微镜镜检拍照(图22中A),同时利用Cytation多功能酶标仪(美国博腾BioTek公司)检测培养孔细胞在450nm吸光度(OD值),并输出为增殖曲线(图22中B)。
利用CAP与肺癌细胞进行共培养,结果如图22所示,CAP可以显著抑制PC-9、A549、H1299的生长,且随着共培养的时间的增加,其抑制效果增加,其中H1299抑制效果更为显著,而CAP对正常肺上皮细胞BEAS-2B无抑制作用。横向比较各肺癌细胞系的增殖、侵袭效率,侵袭转移力强的细胞(H1299)其增殖效率较低,而侵袭转移力较弱的细胞(A549)其增殖能力强,该结果符合实际肿瘤转移的生物学特征。
2.体外血管形成实验
利用肺癌细胞条件培养基与EBM-2内皮细胞培养基验证肺癌细胞对CAP成管能力的影响,实验组为H1299肿瘤细胞条件培养基(H1299-CM)+CAP、EBM-2内皮细胞培养基+CAP,对照组为H1299肿瘤细胞条件培养基(H1299-CM)+人脐静脉内皮细胞(HUVEC)、EBM2内皮细胞培养基+HUVEC。具体实验步骤如下:将Corning Matrigel matrix冻融后,用预冷的移液枪头混匀成浆状;在预冷后的96孔板中加入50μL/孔Corning Matrigel matrix基质,加入时避免产生气泡,置于37℃细胞培养箱中孵育1h固定;在CAP和HUVEC中加入0.25%(v/v)胰酶消化液,置于进行消化对培养箱(37℃,5%CO2)消化2min,10000rpm离心3.5min,计数后用EBM-2内皮细胞培养基重悬细胞,使其细胞密度为1×105个/mL;取出96孔板,每孔加入50μL细胞重悬液(即接种密度为1×104个细胞/孔),每个处理3个平行,做好标记后置于细胞培养箱中37℃孵育4h;利用常规正置显微镜,拍照采集96孔板中各实验孔的图像(图23中A),通过ImageJ插件Angiogenesis Analyzer对图像进行分析统计,并对成管长度(Tubelength)以及分支点数量(bracnch point)和长度(branch length)进行量化,结果输出为相应的树状图(图23中B、C、D),以估组间的成管差异。
H1299肿瘤细胞条件培养基(H1299-CM)的制备方法为:取H1299肺癌细胞于T75细胞培养瓶中(细胞密度1×106/瓶),加入12mL RPMI1640培养基,细胞培养箱(37℃,5%CO2)培育48h后收集培养瓶内细胞上清液,即为H1299肿瘤条件培养基。
结果如图23所示,H1299-CM较EBM-2内皮细胞培养基显著抑制CAP的血管形成能力,而HUVEC则呈现相反的效果,H1299-CM较EBM-2内皮细胞培养基显著促进HUVEC的血管形成管能力。从图23中B、C、D可以看出,H1299-CM较EBM-2内皮细胞培养基显著抑制CAP的成管长度以及分值点数量和长度,H1299-CM较EBM-2内皮细胞培养基显著促进HUVEC的的成管长度以及分值点数量和长度,表明肺癌肿瘤可通过不同机制改变血管组成细胞的功能,调控肿瘤血管的形成发展。
3.肺癌预后模型的构建
从癌症基因组图谱(TCGA)数据集(https://portal.gdc.com)获得513个肺腺癌LUAD及59个癌旁正常组织的RNA测序数据的原始计数和相应的临床信息(TCGA数据集下载时间为2021.5.17)构建肺癌预后模型。采用单因素Cox比例风险回归分析筛选出与预后相关的基因,并利用LASSO(Least Absolute Shrinkage and Selection Operator)回归进一步分析预后相关基因,得到最佳的预后相关基因,并利用该预后相关基因将样本划分为高表达组和低表达组,用于生存分析(图24中A)。利用log rank检验KM生存分析(Kaplan-Meier survival estimate)比较高表达组和低表达组之间的生存差异,并通过ROC分析以比较最佳预后基因的预测准确性和风险评分(图24中C),同时利用log rank检验和单变量Cox比例危险回归得到KM曲线(图24中B)、P值和具有95%置信区间(CI)的危险比(HR)。上述分析均使用R软件包(v4.0.3版R软件,R Foundation for Statistical Computing,2020)进行。
结果如图24所示,由ROC曲线可知,GJB2基因的ROC在第1、3、5年的预测AUC分别为0.832,0.826,0.821,表明GJB2基因具有稳定且优越的预测能力,含有GJB2基因的肿瘤相关性血管周细胞亚群(CAP)可用于肺癌患者预后的诊断。
上面结合附图对本发明实施例作了详细说明,但是本发明不限于上述实施例,在所属技术领域普通技术人员所具备的知识范围内,还可以在不脱离本发明宗旨的前提下作出各种变化。此外,在不冲突的情况下,本发明的实施例及实施例中的特征可以相互组合。
Claims (10)
1.一种血管周细胞亚群,其特征在于,所述血管周细胞表达GJB2。
2.权利要求1所述血管周细胞亚群的制备方法,包括以下步骤:使血管周细胞群与结合剂接触,所述结合剂与GJB2基因或其表达产物相结合;分选与所述结合剂结合的周细胞,得到血管周细胞亚群。
3.根据权利要求2所述的制备方法,其特征在于,包括以下步骤:将肿瘤组织与消化液混合,消化,重悬,加入结合剂,所述结合剂与GJB2基因或其表达产物相结合,分选与所述结合剂结合的周细胞,得到血管周细胞亚群。
4.根据权利要求3所述的制备方法,其特征在于,所述消化液由1640培养基与消化酶混合制成;所述消化酶包括Dispase II、Collagenase I、Collagenase IV和DNase I。
5.根据权利要求2~4中任一项所述的制备方法,其特征在于,所述结合剂选自核酸、配体、酶、底物和/或抗体;
优选地,所述分选包括通过荧光激活细胞分选、磁性细胞分选、底物辅助细胞分选、激光介导切割、荧光测定法、流式细胞术或显微技术进行分选。
6.一种试剂盒,包含能与基因GJB2或所述基因表达的蛋白相结合的结合剂。
7.权利要求1所述的血管周细胞亚群在(1)~(2)中任一项所述的应用:
(1)制备促进肿瘤细胞侵袭的产品;
(2)筛选治疗肿瘤的候选药物。
8.权利要求1所述的血管周细胞亚群的抑制剂在制备治疗肿瘤的药物中的应用;
优选地,所述抑制剂为抑制所述血管周细胞亚群中GJB2表达的试剂。
9.检测GJB2的试剂在制备诊断肿瘤的产品中的应用。
10.权利要求6所述的试剂盒在(3)~(4)中任一项所述中的应用;
(3)制备诊断肿瘤的产品;
(4)制备鉴定、筛选或分选权利要求1所述的血管周细胞亚群的产品。
