Abstract
The invention provides a special processing method and application for simultaneously improving maltopentaose and maltohexaose in dendrobium officinale. The method disclosed by the invention is used for processing the dendrobium officinale by using a special method, and in the processing process, after the fresh picked dendrobium officinale fresh strips are subjected to special treatment under the low temperature condition of-20 ℃, the dendrobium officinale fresh strips are baked for 12-36 hours under the temperature condition of 50-80 ℃, and then the dendrobium officinale processed products are obtained after cooling. The processed dendrobium officinale product is beneficial to the enrichment of maltopentaose and maltohexaose, has the content obviously higher than that of fresh strips, and can be directly eaten as functional food or used as a main raw material or an auxiliary material of functional cosmetics such as anti-aging and whitening cosmetics in the fields of cosmetics and medicine application.

The invention relates to the field of traditional Chinese medicine processing, functional food and cosmetics, and mainly relates to a special processing method for simultaneously improving maltopentaose and maltohexaose in dendrobium officinale, and application of the dendrobium officinale processed product in preparation of medicines, functional food and cosmetics.
Background
Dendrobium officinale, also known as "black clover", is a herb of Dendrobium of Orchidaceae, originally listed in Shen nong's herbal Jing and listed as the top grade, has the international title of panda in the pharmaceutical field, is the first of Jiuda Xiancao in Daozha, and attracts attention as a precious Chinese herbal medicine. The maltopentaose and maltohexaose are straight chain malto-oligosaccharide, which is formed by condensing alpha (1-4) chain D-glucose monomer and trace amount of (1-6) chain glucose, and the polymerization degrees are respectively 5 and 6. The content of maltopentaose and maltohexaose as secondary metabolites of dendrobium officinale has an important effect on the pharmacological activity of dendrobium officinale. The above object of the present invention is achieved by the following means.
The dendrobium officinale processed product is prepared by the following method:
(1) selecting fresh stems of dendrobium officinale, removing fibrous roots, leaves and impurities, keeping stems, cleaning and then airing indoors;
(2) freezing the dried fresh stems in a refrigerator at-20 ℃ for 12 h;
(3) taking out the frozen fresh strips, placing in an oven, baking at 50-80 deg.C until the leaf sheaths are opened, taking out, kneading to remove membranous leaf sheaths, and avoiding damage to stem epidermis to expose internal meat quality, thereby oxidizing compounds;
(4) and continuously placing the dendrobium officinale stems without membranous leaf sheaths in an oven to bake for 24 hours at 50-80 ℃ to obtain the dendrobium officinale processed product.

The invention also provides a processing method capable of simultaneously improving the maltopentaose and the maltohexaose in the dendrobium officinale, which comprises the following steps:

(4) continuously placing the dendrobium officinale stems without membranous leaf sheaths in an oven to bake for 24 hours at 50-80 ℃ to obtain dendrobium officinale processed products;

Detailed Description
In order to make the objects, technical solutions, effects and advantages of the present invention more clear and obvious, the present invention is described below with reference to examples, but the present invention is not limited thereto. Simple modifications of the invention in accordance with its spirit fall within the scope of the invention.
Example 1
The special method is used for preparing the dendrobium officinale processed product which can simultaneously improve the maltopentaose and the maltohexaose in the dendrobium officinale.

(3) And (3) taking out the frozen fresh strips, placing the fresh strips in an oven, baking the fresh strips at 60 ℃ until the leaf sheaths are opened, taking out the fresh strips, rubbing and removing the membranous leaf sheaths, and avoiding damaging the stem epidermis to expose the internal meat quality in the rubbing process so as to prevent the compound from being oxidized.
(4) And continuously placing the dendrobium officinale stems without membranous leaf sheaths in an oven to bake for 24 hours at the temperature of 60 ℃ to obtain the processed dendrobium officinale product.
Example 2
In this embodiment, the content changes of maltopentaose and maltohexaose in the processed dendrobium officinale product prepared in example 1 are mainly detected and analyzed, and the specific steps are as follows:
(1) the required medicines are as follows: methanol, formic acid, water, and acetonitrile were all obtained from Thermo, and L-2-chlorophenylalanine was obtained from Hengchuang Biotech, Inc. All chemicals and solvents were either analytically pure or chromatographic grade.
(2) Sample treatment: respectively weighing 500mg of fresh dendrobium officinale stems and the processed dendrobium officinale product prepared in example 1, completely grinding the samples, respectively weighing 80mg of the samples, and adding 20 mu L of internal standard (L-2-chlorophenylalanine, 0.3 mg/mL; methanol preparation) and 1mL of methanol-water (V: 7: 3); adding two small steel balls, standing at-20 deg.C for 2min for precooling, and adding into a grinder (60Hz for 2 min); ultrasonic extracting in ice water bath for 30min, standing overnight at-20 deg.C; centrifuge for 10min (13000rpm, 4 ℃), aspirate 150 μ L of supernatant with syringe, filter using a 0.22 μm organic phase pinhole filter, transfer to LC injection vial, store at-80 ℃ until LC-MS analysis. The quality control sample (QC) is prepared by mixing the extracting solutions of all samples in equal volume. All extraction reagents were pre-cooled at-20 ℃ before use.
(3) Analysis of metabolites: metabolic curves in ESI positive and ESI negative ion mode were analyzed using an ultra high performance liquid chromatograph (UHPLC, Dionex Ultimate 3000RS) equipped with a mass spectrometer (Q-active + quadrapole Orbitrap) with a heated electrospray ionization source (Thermo Fisher Scientific). Eluting with (A) water containing 0.1% (v/v) formic acid and (B) acetonitrile containing 0.1% (v/v) formic acid at a column temperature of 40 deg.C and a gradient of 0min, 5% B at 0.35 mL/min; 1min, 5% B; 2.5min, 30% B; 6min, 50% B; 7 min, 70% B; 10min, 80% B; 12 minutes, 100% B; 14min, 100% B; 14.2 min, 5% B; 16min, 5% B. During the analysis, the sample was kept at 4 ℃. The injection volume was 5. mu.L. The full MS scan used 70000 resolution and the high energy collision dissociation MS/MS scan used 17500 resolution with collision energies of 10, 20 and 40eV, respectively. The mass detected ranged between 100 and 1200 mass-to-charge ratios (m/z). The mass spectrometer works as follows: spray voltage, 3800V (+) and 3200V (-); sheath gas flow rate, 40 arbitrary units; assist gas flow rate, 15 arbitrary units; capillary temperature, 320 ℃; auxiliary gas heater temperature, 350 ℃; one QC sample was injected every fourth sample in order for the present invention to assess reproducibility.

Therefore, the dendrobium officinale processed product prepared by the embodiment of the invention can simultaneously improve the content of maltopentaose and maltohexaose, and has wide application in the fields of functional foods, medicines, cosmetics and the like.

(2) Adding the dendrobium officinale extract, the vitamin C and the citric acid in sequence according to the component proportion, supplementing the mixture to 100 percent by using deionized water, and uniformly mixing.
(3) After the dendrobium officinale extract is dissolved, the dendrobium officinale extract is sterilized by a filter membrane of 0.24um, and all operating bacteria are carried out under the aseptic condition.

The dendrobium officinale skin care essence of example 4 is prepared according to the following steps:
(1) preparing a dendrobium officinale extract: the dried dendrobium officinale processed by the method is extracted by reflux for 4 times at the temperature of 80 ℃ according to the liquid-material ratio of 20:1, and the dendrobium officinale extract is obtained by freeze drying after the filtrates are combined.
(2) Adding oil, lipid and emulsifier into dissolving pan, heating at 70-80 deg.C and stirring to completely melt.
(3) Adding water soluble substance and deionized water into dissolving pot, heating at 90-100 deg.C and stirring for 20-30min to dissolve completely, and cooling to 70-80 deg.C.
(4) Adding the above prepared oil phase and water phase into emulsifying pot, heating and stirring at 70-80 deg.C for emulsifying for 10-20min, naturally cooling to room temperature, adding thermosensitive component, and stirring.
Example 5
Examples of pharmaceutical preparations:
firstly, preparing a dendrobium officinale extract: the dried dendrobium officinale processed by the method is extracted by reflux for 4 times at the temperature of 80 ℃ according to the liquid-material ratio of 20:1, and the dendrobium officinale extract is obtained by freeze drying after the filtrates are combined.
Then:
1. dissolving herba Dendrobii extract with small amount of DMSO, adding water for injection, fine filtering, bottling, and sterilizing to obtain injection.
2. Dissolving herba Dendrobii extract in small amount of DMSO, dissolving in sterile water for injection, stirring to dissolve, filtering with sterile suction filter funnel, performing sterile fine filtration, packaging, freeze drying at low temperature, and sealing under sterile condition to obtain powder for injection.
3. Taking the dendrobium officinale extract, adding an excipient according to the weight ratio of the dendrobium officinale extract to the excipient of 9:1, and preparing into powder.
4. Adding excipient into the dendrobium officinale extract according to the weight ratio of the dendrobium officinale extract to the excipient of 5:1, and preparing tablets.
5. Taking the dendrobium officinale extract, and preparing the dendrobium officinale extract into oral liquid according to a conventional oral liquid preparation method.
6. Adding excipient into the dendrobium officinale extract according to the weight ratio of the dendrobium officinale extract to the excipient of 5:1, and preparing into capsules.
7. Adding excipient into the dendrobium officinale extract according to the weight ratio of the dendrobium officinale extract to the excipient of 3:1, and preparing into capsules.
8. Adding excipient into the dendrobium officinale extract according to the weight ratio of the dendrobium officinale extract to the excipient of 5:1, and preparing into granules.
