claudin18.2(CLDN18.2)的抗原结合域的胞外域;(b)铰链区;(c)跨膜结构域;和(d)胞内信号域。

实施例2:鼠抗CLDN18.2单抗的人源化
将鼠抗CLDN18.2单抗人源化以减轻其用于人类患者时的潜在免疫原性,如2019年3月6日提交的PCT/US19/020872所述,其通过引用整体并入本文。表7显示人源化VH和VL区序列。

人源化VH和VL区分别与人IgG1重链的恒定区与κ轻链的恒定区融合。人源化单抗命名如下:2-C3-H1L1指含重链可变区2-C3-H1和轻链可变区2-C3-L1的单抗,其余所有人源化单抗命名规则同上。
测试了几种人源化单抗结合CLDN18.2和CLDN18.1的能力。测试中也应用了嵌合单抗15-D6。FACS实验中使用分别表达人CLDN18.2和CLDN18.1的稳定细胞系(HEK293-CLDN18.2和HEK293-CLDN18.1),实验使用Alexa
488进行检测,如PCT/US19/020872所述。单抗浓度10μg/mL。结果如图1A-1B所示。“MFI”为“平均荧光强度(Mean FluorescenceIntensity)”。
其他的人源化单抗经过了CLDN18.2结合能力的测试,测试使用HEK293-CLDN18.2稳定细胞系和相同的FACS方案,使用碘化丙啶(PI)与二抗一起孵育以标记死细胞。结果如图2A-2D所示。
实施例3:人源化单抗向scFv转化
将人源化单抗转化为scFv,每个scFv由一个VH和一个VL组成,中间有一个(G4S)n连接肽(“n”代表G4S重复序列的数量)。VH或VL都位于融合蛋白N端以确定最有效的scFv设计。表8显示scFv设计序列。scFv命名如下:2-C3-H2(G4S)3L2指含有2-C3-H2重链可变区、(G4S)3连接肽和2-C3-L2轻链可变区的scFv;其他所有scFv命名规则同上。

测试了与(G
4S)连接肽和人IgG4 Fc相融合的scFv融合蛋白(从N端到C端的顺序为scFv、G
4S连接肽和Fc)结合CLDN18.2的能力。FACS实验中使用表达人CLDN18.2的稳定细胞系(HEK293-CLDN18.2),使用基于Alexa
488进行检测,如PCT/US19/020872所述。碘化丙啶与二抗一起孵育以标记死细胞。结果如图3A-3L所示。
实施例4:构建包含抗CLDN18.2抗原结合域的嵌合抗原受体构建体
为了构建CAR,使用VH、VL和(G4S)n连接肽将mAb转化为scFv,并将scFv融合到源自人类CD8α的铰链和跨膜结构域(aa 114-188,Boursier JP等,J Biol Chem.1993;268(3):2013-20)的N端。CAR细胞内信号转导域的C端通过融合CD28的细胞内共刺激结构域(aa162-202,Aruffo A和Seed B,Proc Natl Acad Sci USA.1987;84(23):8573-7)和CD3ζ链的激活域(aa 52-162,Letourneur F和Klausner RD,Proc Natl Acad Sci USA.1991;88(20):8905-9)来构建。将编码CAR的DNA序列组装并克隆到表达载体(逆转录病毒、慢病毒、染色体外或整合)中,以使用标准分子生物学克隆技术生成CAR构建体。
实施例5:用肿瘤细胞杀伤试验评估CAR T细胞活性
