Abstract

背景技术
微流控装置已在药物研发行业得到开发,并且用于改进生物学各个领域的实验,例如细胞培养。微量分析系统的优势包括减少样本量、精确的微环境控制以及在单个装置中并行操作,从而能够进行高通量分析。
由于微流控装置上的流体通道和反应室的微尺度尺寸,通常需要复杂的外围设备来操纵此类装置上的流体流动。通过利用微通道的表面化学产生的毛细管效应,毛细管微流控技术可在无需外围设备的情况下以预先编程的方式输送液体。然而,可能需要进行精密平版印刷和通道表面处理。
对于包含高密度微孔阵列且没有微通道互连的平台,装载孔阵列并保持孔中的内容物彼此隔离可能是一个挑战。尤其,如果该平台是由疏水性材料(诸如疏水性塑料)制成的,则将水溶液装载到微孔阵列中可能会受到表面张力和微孔中的滞留空气阻碍。
发明内容
在一方面,本公开提供了一种改性微制造芯片的方法,该微制造芯片具有包括多个微孔的顶部表面,每个微孔具有底部和侧壁,以及微孔之间的间隙空间,该微制造芯片由疏水性材料制成。该方法包括,按以下顺序:(a)处理微制造芯片,以使微孔的底部的表面和侧壁以及间隙空间具有亲水性;(b)选择性地处理间隙空间的表面以使其具有疏水性。
在该方法的一些实施例中,步骤(a)包括用等离子体处理微制造芯片。在该方法的一些实施例中,步骤(a)包括用电晕放电、臭氧和铜增强氧化中的一者对微制造芯片进行处理。
在该方法的一些实施例中,步骤(a)包括例如通过光化学表面改性在微孔的底部的表面和侧壁以及微制造芯片的间隙空间上形成小分子或聚合物的亲水层。

本发明具体实施方式
本发明的目的是提供一种具有限定微孔阵列的顶部表面以及微孔之间的间隙空间的微制造装置(或芯片),该微制造装置由疏水性聚合物材料制成,其中微孔的内表面是亲水的,并且微孔之间的间隙空间是疏水的。每个微孔均具有底部和侧壁。本文中所用的术语“底部”表示微孔在微制造装置的厚度方向上具有有限深度并且不是穿过微制造装置的通孔。底部和侧壁之间可以有明确的边界,但也可以平滑地接合而不存在明显的分界线。本发明的另一个目的是提供改性由疏水性材料制成的微制造芯片的方法,使得微孔的内表面变成亲水并且微孔之间的间隙空间保持疏水性。这些特性将显著简化在微孔中装载、密封、细胞保留以及下游操作,诸如从微孔中获取和转移样本。

微制造芯片可具有两个主表面:顶部表面和底面,其中微孔在顶部表面具有开口。微孔中的每个微孔可具有任何形状的开口或横截面,例如圆形、六边形、方形或其他形状。每个微孔可包括侧壁。对于其开口或横截面不是圆形的微孔,本文所述的微孔的直径是指具有等效面积的圆形的有效直径。例如,对于边长为10微米×10微米的方形微孔,具有等效面积(100平方微米)的圆的直径为11.3微米。每个微孔可包括一个或多个侧壁。侧壁可具有笔直、倾斜和/或弯曲的横截面轮廓。每个微孔包括底部,底部可以是平的、圆形的或其他形状。微制造芯片(其上具有微孔)可由聚合物(例如环烯烃聚合物)通过精密注射成型或压花等一些其他工艺制造。也可以使用其他结构材料,诸如硅和玻璃。芯片可具有基本上平坦的主表面。图1示出了微制造芯片的示意图,其边缘大体上平行于芯片上微孔的行和列的方向。
微制造芯片上的高密度微孔可用于容纳包括至少一种生物实体(例如,至少一种细胞)的样本。术语“生物实体”可包括但不限于生物体、细胞、细胞组分、细胞产物和病毒,术语“物种”可用于描述分类的单元,包括但不限于,操作分类单元(OTU)、基因型、系统发育型、表型、生态型、履历、行为或相互作用、产物、变体和进化显著单元。微制造芯片上的高密度微孔可用于进行各种实验,诸如各类物种的细菌和其他微生物体(或微生物)诸如需氧、厌氧和/或兼性需氧微生物的生长、培养或筛选。微孔可用于进行真核细胞诸如哺乳动物细胞的实验。此外,微孔可用于进行各种基因组或蛋白质组学实验,并可包含细胞产物或组分,或其他化学或生物物质或实体,诸如细胞表面(例如,细胞膜或细胞壁)、代谢物、维生素、激素、神经递质、抗体、氨基酸、酶、蛋白质、糖类、ATP、脂质、核苷、核苷酸、核酸(例如,DNA或RNA)、化学品,例如,染料、酶底物等。
在一些实施例中,高密度细胞培养平台可以是基于液滴的,例如,分散液滴群可用于保持细胞、培养基以及用于细胞培养的其他组分,从而代替在微制造芯片上用作实验单元的孔阵列。液滴生成方法(尤其是与芯片上细胞分选仪类型的仪器结合使用时)可用于使复杂的环境样本中的微生物生长并对其进行筛选。可在几百赫兹下产生液滴,这意味着可在几个小时内产生数百万滴。可使用简单的基于芯片的装置来生成液滴,并且可以将液滴设计成包含单个细胞。用于生成含有细胞悬浮液的液滴的系统可含有一个细胞或少量细胞。液滴可以是乳液、双乳液、水凝胶、气泡和复合颗粒等。例如,水性液滴可悬浮在不混溶的液体中,使它们彼此分开并且不接触或污染任何表面。液滴的体积可介于10fl至1μL之间,高度单分散的液滴可制成直径为从几纳米至500μm的范围。基于液滴的微流控系统可用于生成、操纵和/或培养小液滴。细胞存活和增殖可类似于本体溶液中的对照实验。可在芯片上以例如每秒500滴的速率进行液滴的荧光筛选。可将液滴合并以形成新液滴或添加到液滴中的试剂。可将液滴通过排成单行的微通道,并通过光谱法进行检测,例如,使用荧光检测器检测液滴发出的荧光,并且可通过分流至可汇集或收集液滴的分支通道来选择确定满足特定标准的液滴(例如,发出特定波长的荧光)。可通过阀门、泵、施加外部电场等来实现流体的分流或切换。
在各种实施例中,细胞可以是古细菌、细菌或真核生物(例如真菌)。例如,细胞可以是微生物,诸如需氧、厌氧或兼性需氧微生物。病毒可以是噬菌体。其他细胞组分/产物可包括但不限于蛋白质、氨基酸、酶、糖类、三磷酸腺苷(ATP)、脂质、核酸(例如DNA和RNA)、核苷、核苷酸、细胞膜/细胞壁、鞭毛、菌毛、细胞器、代谢物、维生素、激素、神经递质和抗体。
通常提供营养物以培养细胞。营养物可以是限定的(例如限定化学成分培养基或合成培养基)或不限定的(例如,基础培养基或复合培养基)。营养物可包括实验室配制的和/或商业制造的培养基(例如,两种或多种化合物的混合物),也可以是其组分。营养物可包括液体营养培养基(即,营养肉汤),诸如海洋肉汤、溶原肉汤(例如,Luria肉汤)等,也可以是其组分。营养物可包括与琼脂混合的液体培养基以形成固体培养基和/或市售制造的琼脂平板,诸如血琼脂,也可以是其组分。
营养物可包括选择性培养基,也可以是选择性培养基的组分。例如,选择性培养基可用于仅特定生物实体的生长或仅用于具有特定特性(例如,抗生素抗性或特定代谢物的合成)的生物实体的生长。营养物可包括差异培养基,也可以是其组分,以在存在特定指示剂(例如,中性红、酚红、曙红y或亚甲基蓝)的情况下,通过使用生化特征将一种类型的生物实体与另一种类型的生物实体或其他类型的生物实体区分开来。
营养物可包括源自自然环境的提取物或培养基,也可以是其组分。例如,营养物可源自对特定类型的生物实体而言是天然的环境、不同的环境或多个环境。环境可包括但不限于以下中的一者或多者:生物组织(例如,结缔组织、肌肉、神经、上皮、植物表皮、血管、地面等)、生物液体或其他生物产物(例如,羊水、胆汁、血液、脑脊液、耵聍、渗出液、粪便、胃液、间质液、细胞内液、淋巴液、牛奶、粘液、瘤胃内容物、唾液、皮脂、精液、汗液、尿液、阴道分泌物、呕吐物等)、微生物悬浮液、空气(包括例如不同的气体含量)、超临界二氧化碳、土壤(包括例如矿物质、有机物质、气体、液体、生物体等)、沉积物(例如,农业、海洋等)、活有机物质(例如植物、昆虫、其他小生物和微生物)、死有机物质、饲料(例如草、豆类、青贮饲料、作物残余物等)、矿物、油或油产品(例如,动物、植物、石化)、水(例如天然淡水、饮用水、海水等)和/或污水(例如卫生、商业、工业和/或农业废水和地表径流)。
图1是示出了根据一些实施例的微制造装置或芯片的透视图。芯片100包括以显微镜载玻片形式成形的基板,在顶部表面102上具有注模特征结构。这些特征结构包括四个单独的微孔阵列(或微阵列)104以及喷射器标记106。每个微阵列中的微孔113(由间隙空间114隔开)以网格图案排列,芯片100的边缘周围以及微阵列104之间具有无孔边缘。图4示出了本公开的微制造装置或芯片的一部分的横截面示意图,其中微孔113的内表面(侧壁和底部)是亲水的,并且微孔之间的间隙空间114是疏水的。
图2A至图2C分别是示出了根据一些实施例的芯片100的尺寸的顶视图、侧视图和端视图。在图2A中,芯片100的顶部约为25.5mm×75.5mm。在图2B中,芯片100的端部约为25.5mm×0.8mm。在图2C中,芯片100的侧面约为75.5mm×0.8mm。
在将样本装载到微制造装置上之后,可将膜施用到微制造装置的至少一部分上。图3A是根据一些实施例从图3B的俯视图所示的微制造装置300的分解图。装置300包括具有孔阵列302的芯片,孔阵列302容纳有例如土壤微生物。膜304放置在孔阵列302的顶部。垫圈306放置在膜304的顶部。具有填充孔310的盖308放置在垫圈306的顶部。最后,将密封带312施用到盖308上。
膜可覆盖包括一个或多个实验单元或微孔的微制造装置的至少一部分。例如,将样本装载到微制造装置上之后,可将至少一张膜施用于高密度微孔阵列的至少一个微孔上。可将多张膜施用于微制造装置的多个部分上。例如,可将单独的膜施用于高密度微孔阵列的单独子区段上。
膜可以连接、附着、部分附着、贴附、密封和/或部分密封至微制造装置以将至少一种生物实体保持在高密度微孔阵列的至少一个微孔中。例如,可使用层压法将膜可逆地贴附到微制造装置上。可将膜刺破、剥离、分离、部分分离、移除和/或部分移除,以接近高密度微孔阵列的至少一个微孔中的至少一个生物实体。
可将至少一个实验单元、孔或微孔中的细胞群的一部分附着于膜(例如通过吸附)。如果是这种情况,可通过剥离膜来取样至少一个实验单元、孔或微孔中的细胞群,使得至少一个实验单元、孔或微孔中的一部分细胞群保持附着于膜。
膜可以是不可渗透的、半渗透的、选择性渗透的、差异渗透的和/或部分渗透的,以允许至少一种营养物扩散到高密度微孔阵列的至少一个微孔中。例如,膜可包含天然材料和/或合成材料。膜可包括水凝胶层和/或滤纸。在一些实施例中,选择孔径足够小的膜,以将至少部分或全部细胞保持在微孔中。对于哺乳动物细胞,孔径可能只有几微米,但仍能保持细胞。然而,在一些实施例中,孔径可小于或等于约0.2μm,诸如0.1μm。不可渗透的膜的孔径接近于零。应当理解,膜可具有复杂的结构,其可能具有也可能不具有限定的孔径。
在一方面,本发明提供一种改性如本文所述的微制造芯片的方法。该芯片由疏水性材料(诸如塑料)制成。该芯片具有包括多个微孔的顶部表面,每个微孔具有底部和侧壁,以及在顶部表面上的微孔之间的间隙空间。该方法包括首先对微制造芯片进行处理,以使微孔的底部的表面和侧壁以及间隙空间具有亲水性(亲水处理步骤);然后选择性地处理间隙空间的表面以使其具有疏水性(疏水性处理步骤)。
对于由疏水性聚合物材料制成的未经处理的芯片,含水样本可能不会简单地进入微孔。相反,液体可以停留在微孔之间的间隙空间。由于微孔的内表面呈现亲水性,同时保留间隙空间的疏水性,因此可以更轻易地将液体样本装载入微孔中。
为了使微制造芯片的整个顶部表面(包括孔内部空间和间隙空间)具有亲水性,可在存在含有氧气的气体(空气或纯氧)下,例如,以30W或更高的功率,通过等离子体对微制造芯片进行处理,处理时间为1分钟或更久。此类处理会在聚合物上产生亲水性官能团,包括羧酸、醛、胺等,这取决于特定的聚合物成分和等离子体处理。可选地,微制造芯片可以进行臭氧处理(例如,1L/min,25分钟,在60℃阶段)、UV/臭氧(UVO)处理、电晕放电或铜增强氧化。在一些实施例中,可在芯片上沉积一薄层金属氧化物。示例为氧化钛或氧化铝,可通过多种方法快捷地沉积氧化钛或氧化铝,包括物理沉积(溅射)。
在一些实施例中,亲水性处理步骤包括在微孔的底部的表面和侧壁以及微制造芯片的间隙空间上形成小分子或聚合物的亲水层。例如,可使用等离子体增强化学气相沉积和/或光化学表面改性。此类小分子或聚合物层可形成在通过等离子体、臭氧或其他处理方式处理过的新鲜“活性”表面的顶部上。例如,可通过铜催化过氧化来改性环烯烃共聚物(COC)表面以引入表面羟基(其可被进一步改性以形成混合表面)。参见Carvalho等人,ACSApplied Materials and Interfaces,2017,9,16644。作为另一个示例,为了增加COC表面的亲水性,可使用两步顺序方法对聚(乙二醇)甲基丙烯酸酯(PEGMA)进行光接枝,该方法包括形成共价结合的表面引发剂,然后从这些引发剂接枝PEGMA聚合。参见Stachowiak等人,J.Sep.Sci.2007,30,1088。
在一些实施例中,疏水性处理步骤包括使物体与间隙空间的表面接触以使间隙空间的表面具有疏水性。该物体可包括PDMS印模的基底。PDMS印模可包括具有剩余未聚合的二甲基硅氧烷或其他硅烷分子(诸如十八烷基三甲氧基硅烷(ODTMS))的平坦表面(用于接触微制造芯片的间隙空间)。在与微制造芯片的顶部表面(但不是微孔的内表面)接触时,未聚合的二甲基硅氧烷或其他硅烷可与间隙空间的活化表面上的羟基或羧基反应,导致在间隙空间的顶部上形成疏水性硅烷层。在一个实施例中,可使用含有疏水性硅酮粘合剂的膜施加到微制造芯片的顶部表面,然后将其剥离,在微孔之间的间隙空间上留下一薄层残留的聚合和/或未聚合的PDMS。
如果之前已经在芯片上(在孔和间隙空间中)沉积了一薄层金属氧化物,则可以使用PDMS印模将疏水性膦酸(诸如十八烷基膦酸(ODPA))转移到间隙空间。
膦酸已被证明与铝氧化物和钛氧化物强烈且选择性地结合。
在一些实施例中,还可以通过选择性去除间隙空间的表面的顶层来完成疏水性处理步骤。例如,可使用有机溶剂(诸如甲苯)来部分蚀刻掉间隙空间上的顶部薄层。可将该溶剂浸渍到PDMS印模中,并且可通过所浸渍的溶剂的量以及PDMS印模和微制造芯片之间的接触时间来控制蚀刻的程度。
在其他实施例中,可从间隙中去除薄层,并且使得间隙可被转化回下面的疏水性基底,从而可以使用化学机械抛光,化学机械抛光是化学蚀刻和自由磨蚀抛光的结合。该工艺使用磨蚀性和腐蚀性化学浆料,结合可以高速旋转的非常平坦的抛光垫。抛光垫的平坦面可通过压力保持紧靠微制造垫的顶部表面,并借助腐蚀性化学品,从微制造芯片的间隙空间中磨掉所需深度的材料。
在一些实施例中,在疏水性处理步骤之前,可以首先在微制造装置上用亲水性液体填充微制造芯片的微孔,以保护微孔的内表面免于进一步处理。可使用载玻片将大量液体散布在微制造芯片的顶部表面上来完成该微孔填充步骤。该步骤可能会造成在间隙空间的顶部留有一些液体。可通过使用软刀片扫过间隙空间表面来去除此类不需要的液体。该软刀片的材料需要足够柔顺以粘附在表面的任何表面拓扑结构,足够亲水以吸引液体并将液体推离间隙,但不能太亲水,不能吸收孔中的所有液体。
可选地或此外,可通过吸收性材料去除不需要的液体。该材料需要足够柔顺以粘附在表面的任何表面拓扑结构,并具有足够的吸附能力以去除间隙中的液体。但它的吸收性不能太强,不能去除孔中的所有液体。
进一步可选地或此外,可通过受控蒸发去除留在间隙空间上的不需要的液体,即通过在微制造芯片周围提供具有受控湿度的密封环境,使得间隙干燥但仍有足够量的液体保留在微孔中。
一旦通过液体保护微孔,转移疏水性材料的方法可用于使微制造芯片的间隙空间具有疏水性,而不是将微制造芯片与印模直接接触。从某种意义上说,充满液体的微孔充当了微孔的掩膜。例如,可以将有机溶剂喷洒在微制造芯片的表面上,以回蚀先前形成在间隙空间上的亲水层。还可以将包含硅烷的溶液或悬浮液喷洒在微制造芯片的顶部表面上以在间隙空间上形成疏水性膜。
也可以通过“长出支链”技术或“嫁接支链”技术使用疏水性膜掩埋先前的亲水性表面,将其他聚合物和小分子喷洒、印刷或平版印刷图案到间隙空间上。CVD或iCVD(引发式化学气相沉积)也可用于在间隙空间的顶部沉积聚合物薄膜。对间隙空间进行处理之后,可去除微孔中的液体。
在另一种方法中,可省略对微制造芯片的整个顶部表面进行的初始疏水性处理步骤。取而代之的是,首先将亲水性液体施用在微制造芯片上,以用亲水性液体填充多个孔中的每个孔的至少一部分。如果在间隙空间上留有亲水性液体的任何部分,则从间隙空间中除去此类不需要的亲水性液体(例如,使用上述方法)。最后,微孔的内表面转换为具有亲水性。填充微孔的亲水性液体可包含适当浓度的表面活性剂,使得表面活性剂的极性尾部基团将吸附到疏水性孔表面,从而亲水头部基团使孔的表面具有亲水性。可以蒸发掉液体,在内孔表面上留下表面活性剂。表面活性剂的浓度足够低,以确保只有孔中的表面活性剂才能形成连续的覆盖层,而在间隙上的残余物不会产生实质性影响。在其他实施例中,亲水性液体可包括可溶性聚合物,诸如聚乙烯醇。当水变干时,聚乙烯醇会形成覆盖微孔的内表面的薄膜。留在间隙上的聚乙烯醇的量不足以形成连续的膜,因此基本上不会改变间隙空间的疏水性。在另外的其他实施例中,亲水性液体可包括可聚合化合物,诸如丙烯酸,以及可通过辐射或其他条件活化的聚合引发剂。可在微孔中引发和进行聚合,以在微孔的内表面形成亲水性薄膜。
在替代实施例中,薄膜掩膜可放置在微制造芯片的顶部表面上,掩膜具有与微制造芯片的微孔的尺寸和相对位置相匹配的通孔,使得在微孔暴露时,微制造芯片上的间隙空间由掩膜覆盖。此后,可通过氧等离子体、平版印刷、喷洒亲水性材料的覆盖层和本文所述的其他技术对暴露的微孔进行处理,以使微孔的内表面具有亲水性。然后去除薄膜掩模,产生具有亲水性微孔和疏水性间隙空间的微制造芯片。
本领域技术人员将理解,在不脱离大体上描述的本发明的精神或范围的情况下,可对如特定实施例中所示的本发明进行多种变化和/或修改。因此,本实施例在所有方面均被视为是说明性的而非限制性的。
Claims (19)
1.一种改性微制造芯片的方法,所述微制造芯片具有包括多个微孔的顶部表面,每个微孔具有底部和侧壁,以及所述微孔之间的间隙空间,所述微制造芯片由疏水材料制成,所述方法包括,按以下顺序:
(a)处理所述微制造芯片,以使所述微孔的所述底部的表面和所述侧壁以及所述间隙空间具有亲水性;以及
(b)选择性地处理所述间隙空间的表面以使其具有疏水性。
2.根据权利要求1所述的方法,其中,(a)包括用等离子体处理所述微制造芯片。
3.根据权利要求1所述的方法,其中,(a)包括用电晕放电、臭氧和铜增强氧化中的一者对所述微制造芯片进行处理。
4.根据权利要求1所述的方法,其中,(a)包括在所述微制造芯片的所述微孔的所述底部的表面和所述侧壁以及所述间隙空间上形成小分子或聚合物的亲水层。
5.根据权利要求4所述的方法,其中,(a)包括光化学表面改性。
6.根据权利要求1所述的方法,其中,(b)包括使物体与所述间隙空间的表面接触以使所述间隙空间的表面具有疏水性。
7.根据权利要求1所述的方法,其中,(b)包括选择性地去除所述间隙空间的表面的顶层。
8.根据权利要求1所述的方法,还包括,在(b)之前:
(c)将亲水性液体施用在所述微制造装置上,以用所述液体填充所述多个孔中的每个孔的至少一部分。
9.根据权利要求8所述的方法,其中,(c)造成所述液体的一部分留在所述间隙空间上,所述方法还包括,在(c)之后和(b)之前:(d)去除留在所述间隙空间上的所述液体的一部分。
10.根据权利要求9所述的方法,其中,(d)包括受控蒸发。
11.根据权利要求9所述的方法,其中,(d)包括使用软刀片扫过所述间隙空间表面。
12.根据权利要求10所述的方法,其中,(d)包括使用吸收性材料通过吸收去除在所述间隙空间上的所述留存的液体。
13.根据权利要求9所述的方法,其中,(b)包括将有机溶剂喷洒到所述间隙空间的表面上。
14.根据权利要求9所述的方法,其中,(b)包括在所述间隙空间的表面上形成疏水性聚合物层。
15.一种改性微制造芯片的方法,所述微制造芯片具有包括多个微孔的顶部表面,每个微孔具有内表面,以及所述微孔之间的间隙空间,所述微制造芯片由疏水性材料制成,所述方法包括,按以下顺序:
(a)将亲水性液体施用在所述微制造芯片上,以用所述亲水性液体填充所述多个孔中的每个孔的至少一部分;
(b)如果在所述间隙空间上留有所述亲水性液体的任何部分,则从所述间隙空间中去除所述亲水性液体的所述部分;和
(c)将所述微孔的所述内表面转换为具有亲水性。
16.根据权利要求15所述的方法,其中,所述亲水性液体是包含水溶性聚合物的水溶液。
17.根据权利要求16所述的方法,其中,所述水溶性聚合物包含聚乙烯醇(PVA)。
18.一种微制造装置,所述微制造装置具有限定微孔阵列的顶部表面,所述微孔阵列的表面密度为至少为750个微孔/平方厘米,每个微孔具有底部和侧壁,以及所述微孔之间的间隙空间,所述微制造装置由疏水性基材制成,其中,所述微孔的所述内表面被改性为具有亲水性,并且所述微孔之间的所述间隙空间是疏水性的。
19.一种使用微制造装置培养和筛选至少一种感兴趣的生物实体的方法,所述微制造装置具有限定微孔阵列的顶部表面,所述微孔阵列的表面密度为至少为750个微孔/平方厘米,其中,每个所述微孔具有底部和侧面壁,以及所述微孔之间的间隙空间,所述微制造装置由疏水性材料制成,其中所述微孔的所述内表面是亲水的并且所述微孔之间的所述间隙空间是疏水的,所述方法包括:
将样本装载到所述微制造装置上,使得所述微孔阵列的至少一个微孔包括至少一个细胞和一定量的营养物;
将膜施用于所述微制造装置以将所述至少一个细胞和所述营养物保留在所述微孔阵列的所述至少一个微孔中;
在不提供额外营养物的情况下,在所述微孔阵列的所述至少一个微孔中从所述至少一个细胞培养出多个细胞;以及
分析所述多个细胞以确定存在或不存在感兴趣的生物实体。
