本发明涉及一种用于化妆品领域的能有效抗氧化的保湿组合物及其制备方法与在护肤品中的应用,具体地,涉及一种用于有效抗氧化保湿的组合物及其制备方法与用途。
背景技术
人体因为与外界的持续接触,包括呼吸(氧化反应)、外界污染、放射线照射等方式摄入氧气,氧气可以维持人体内的各种氧化反应,然而在利用氧气进行代谢反应时,不可避免地会产生一些不稳定物质,而这些不稳定的物质都是氧气的“变身”,它们的个性非常活泼,不喜欢安安稳稳地呆在原地,甚至喜欢去攻击别人,由于它们这种不喜欢受拘束的个性,称之为自由基。自由基或氧化剂会将细胞和组织分解,影响代谢功能,并会引起不同的健康问题。日常生活应摄取足够的抗氧化剂,延缓身体退化速度,防止肌肤衰老,并时刻保持青春神采。目前治疗方法持久性不强,能持久有效抗氧化,保持皮肤湿润,维持皮肤稳态的健康皮肤护肤产品十分稀有。
发明内容
本发明的目的是提供一种适用于所有肤质的能快速有效调节皮肤抗氧化保湿的精油组合物及其制备方法与应用,能够解决现有问题,针对不同原因引起的皮肤氧化干燥,并能有效抗氧化、促进皮肤屏障修复、持久保湿、广谱抗菌及深层抗炎功能等。

神经酰胺是人体天然角质层,可以有效修复皮肤屏障,提高自身免疫系统,消除皮肤炎症,可以快速补充水分,具有较强的抗氧化能力,同时还可以缓解皮肤干燥等症状。
富勒烯具备有超强的抗氧化成份,是维他命C的172倍。可以有效保护肌肤远离自由基的侵害,改善肌肤弹力不足、色斑等各种问题,从根源上阻断肌肤老化;同时具有显著提亮肤色,紧致毛孔的作用,令肌肤再现充满透明感和盈润触感。
棓酸由五倍子制成,对蛋白质有沉淀作用,皮肤、粘膜、溃疡接触棓酸后,其组织蛋白质即被凝固,造成一层被膜,同时小血管也被压迫收缩,血液凝结而奏止血功效;其对金黄色葡萄球菌、链球菌、肺炎球菌等细菌具有明显的抑菌或杀菌作用;鞣酸能和很多重金属离子、生物碱及甙类形成不溶性的复合物,故可以缓解化学物质对皮肤的伤害。

霍霍巴(SIMMONDSIA CHINENSIS)籽油含有丰富的维生素A、B、E和钙、镁等矿物质,具有极强的抗氧化能力,消除自由基;同时能增进皮肤对营养物质的吸收和维持,它对受损的细胞起修护作用;时其可以调节皮肤的油脂分泌,并改善皮肤的瘙痒、发炎等问题。它对过敏性皮肤有超强的疗效与舒缓的功效。
巴西棕榈树(COPERNICIA CERIFERA)蜡是从棕榈树叶上提取的天然植物蜡,主要是棕榈酸蜂酯和蜡酸。棕榈蜡稀薄而黏滞度小,树脂含量低,涂布均匀,流动性能好;具有光泽好、光洁度高、表面没有油腻的优点,同时具有一定的软化与润滑作用。
紫苏(PERILLA OCYMOIDES)籽油的抑菌、防腐能力明显优于尼泊金乙酯和苯甲酸,而且具有用量小、安全性高、不受pH值制约等优点,并兼具防腐和香味的双重效果,对透明质酸酶的生成有很强的促进作用,具有很好的保湿效果;能促进胶原蛋白的生成,消除自由基,具有抗衰、防皱的作用。
黄瓜(CUCUMIS SATIVUS)油可很好地提高角质层的含水量,有保湿作用,可用于干性皮肤的防治;对芳香化酶有活化的作用,芳香化酶可提高局部雌激素水平,因此黄瓜提取物有促进生发的作用也有此关系。
扭刺仙人掌(OPUNTIA STREPTACANTHA)茎提取物具有抗菌性,对雄性激素受体活性的抑制,可用于粉刺类化妆品,对雄性激素偏高引起的粉刺有防治效果;能促进皮肤生长因子的生成,具有活肤的作用,具有抗氧化、消除自由基,抗衰老能力较强。
向日葵(HELIANTHUS ANNUUS)籽油可改善皮肤的水合作用,并保持皮肤外层的完整性。葵花籽油中存在的亚油酸具有出色的润肤性能,可保持皮肤水分。除亚油酸外,它还含有渗入皮肤深处的维生素E,可以有效抗氧化和消除自由基,助于防止皮肤衰老的迹象,例如细纹,皱纹和皮肤松弛。

采用各实施例制备的组合物进行测试及评估,通过测胶原生成的方式,具体如下。
人角质形成细胞(keratinocytes):角质形成细胞是表皮的主要构成细胞,数量占表皮细胞的80%以上,在分化过程中产生角蛋白。根据分化阶段和特点可分为五层,由内至外分别为基底层、棘层、颗粒层、透明层和角质层。
活性氧(ROS):活性氧(ROS)是含氧的化学反应性化学物质。实例包括过氧化物,超氧化物,羟基自由基,单线态氧,和α-氧。在生物学背景下,ROS形成为氧的正常代谢的天然副产物,并且在细胞信号传导和体内平衡中具有重要作用。然而,在环境压力(例如,紫外线或热暴露)期间,ROS水平会急剧增加。这可能会对细胞结构造成严重损害,这被称为氧化应激。
活性氧荧光探针原理:DCFH-DA(2',7'-二氯荧光素二乙酸酯)本身没有荧光,可以自由穿过细胞膜,进入细胞内后,可以被细胞内的酯酶水解生成DCFH。而DCFH不能通透细胞膜,从而使探针很容易被装载到细胞内。细胞内的活性氧可以氧化无荧光的DCFH生成有荧光的DCF。检测DCF的荧光就可以知道细胞内活性氧的水平。通过荧光酶标仪检测,根据荧光值可间接评估人角质形成细胞中活性氧的相对含量。
细胞准备:来自冷冻保存的细胞培养物以一个合适的密度接种到培养基,并在检测前至少传代一次,细胞应以合适的密度接种到培养基中用于试验,细胞接种密度应保证在接种24h后,融合度达45%~60%。
受试物准备:水中溶解度受限的受试物应当溶解在适当的溶剂中,溶剂体积应当在所有培养物中保持一致即在空白对照和受试物中保持一致,并且无细胞毒性。受试物浓度的选择应避免岀现沉淀和溶液呈现云雾状。
使用二甲基亚砜(DMSO)和无水乙醇为溶剂,其他低细胞毒性的溶剂也可使用。
试验步骤包含:
细胞制备:细胞用T75复苏培养4天后,用3ml浓度为0.25%胰酶消化1min,加入等体积的含10%FBS的DMEM中止消化,1200rpm/min离心6min,用适量完全培养基重悬细胞,进行计数;
铺板:取1个96孔板,取32万细胞分别用8ml完全培养基重悬,按每100ul/孔含4000个细胞铺板,具体见96孔板布局,37℃,5%CO2培养箱培养24h±2。布局如下表2所示。

给药:照射时间结束后,吸弃96孔板中的培养基,阴性对照孔中加入正常的细胞培养基,受试物孔中加入含有本发明的培养基,每孔100μl。给药完毕后将96孔板放置在CO2培养箱中继续培养24h±2。
加DCFH-DA:培养结束后,1:1000用无血清培养液稀释DCFH-DA,使终浓度为10微摩尔/升。去除细胞培养液,加入适当体积稀释好的DCFH-DA。37℃细胞培养箱内避光孵育20分钟。孵育结束后,用无血清细胞培养液洗涤细胞三次,以充分去除未进入细胞内的DCFH-DA。检测前,每孔加入100ul的DPBS。
检测:使用荧光酶标仪进行检测,激发光波长488nm,发射光波长525nm。
4.结果:如图1所示。
在试验满足有效性验证的基础上,本发明实验组与阴性对照(未处理)相比,ROS含量下调,且具有显著性差异(P<0.05),说明受试物在该受试浓度下,具有抗氧化能力,能够有效保湿补水,对抗皮肤氧化干燥。
本发明采用的成分中,神经酰胺组合物可以提高自身免疫系统,消除皮肤炎症,从而达到快速补充水分,缓解皮肤干燥脱屑等不适感。同时它还具有超强的保湿能力,可以长效调节皮肤水油平衡。富勒烯可以有效保护肌肤远离自由基的侵害,改善肌肤弹力不足、色斑等各种问题,从根源上阻断肌肤老化;同时具有显著提亮肤色,紧致毛孔的作用。生育酚类具有对抗外界因子引起的各种炎症具有较强的消除能力,它还可以促进皮肤中神经酰胺的合成,形成高保湿皮肤屏障,达到高补水和保湿的双重功效。棓酸对金黄色葡萄球菌、链球菌、肺炎球菌等细菌具有明显的抑菌或杀菌作用;能和很多重金属离子、生物碱及甙类形成不溶性的复合物,故可以缓解化学物质对皮肤的伤害。红没药醇具有抗菌、抗炎的作用,减轻炎症,它还具有杀菌的作用;具有抗刺激、修复的作用,能够保护和治愈皮肤;具有较好稳定性及皮肤相容性,安全性高,可以缓解过敏性皮肤病,具有抗炎、抗菌、舒缓和镇定肌肤的作用。抗坏血酸四异棕榈酸酯具有极强的抗氧化能力、有效消除自由基、抗炎、减少黑色素生成、促进免疫球蛋白合成,防止色素沉着的作用,能美白皮肤,维持皮肤弹性,减轻皱纹,改善皮肤粗糙、苍白、松弛等现象,延缓皮肤自然老化及光老化。西瓜籽油可以具有对皮肤的保护和保湿补水作用。不仅可以软化角质,还具有为皮肤提供营养功效,在炎热和恶劣的环境可以整体维持皮肤的屏障功能,霍霍巴籽油具有极强的抗氧化能力,消除自由基;能增进皮肤对营养物质的吸收和维持,它对受损的细胞起修护作用;其可以调节皮肤的油脂分泌,。它对过敏性皮肤有超强的疗效与舒缓的功效。巴西棕榈树蜡是树脂含量低,涂布均匀,流动性能好;具有光泽好、光洁度高、表面没有油腻的优点,同时具有一定的软化与润滑作用。
本发明提供的用于有效抗氧化保湿的组合物及其制备方法与用途,制备的组合物具备有效抗氧化、消除自由基,快速补水,修复皮肤屏障,保湿锁水,抗炎抑菌等功能,可以达到有效抗氧化保湿皮肤的效果,从而改善肌肤衰老,持久保持皮肤的水润与健康。
尽管本发明的内容已经通过上述优选实施例作了详细介绍,但应当认识到上述的描述不应被认为是对本发明的限制。在本领域技术人员阅读了上述内容后,对于本发明的多种修改和替代都将是显而易见的。因此,本发明的保护范围应由所附的权利要求来限定。
Claims (8)
1.一种用于有效抗氧化保湿的组合物,其特征在于,该组合物包含霍霍巴籽油、紫苏籽油、西瓜籽油、黄瓜油、辛酸/癸酸甘油三酯、碳酸二辛酯、角鲨烷、抗坏血酸四异棕榈酸酯、扭刺仙人掌茎提取物、谷维素、生育酚类、四氢姜黄素、植物甾醇类、红没药醇、二甲基甲氧基苯并二氢吡喃醇、向日葵籽油、棓酸、甘油硬脂酸酯、巴西棕榈树蜡、茶叶提取物、富勒烯、神经酰胺3、硬脂醇甘草亭酸酯。
2.如权利要求1所述的用于有效抗氧化保湿的组合物,其特征在于,所述的组合物中按重量份数计包含:霍霍巴籽油26~34份、紫苏籽油8~12份、西瓜籽油4~6份、黄瓜油4~6份、辛酸/癸酸甘油三酯0.5~1.5份、碳酸二辛酯0.5~1.5份、角鲨烷4~6份、抗坏血酸四异棕榈酸酯0.5~1.5份、扭刺仙人掌茎提取物2~4份、谷维素2~4份、生育酚类2~4份、四氢姜黄素0.5~1.5份、植物甾醇类1~3份、红没药醇4~6份、二甲基甲氧基苯并二氢吡喃醇4~6份、向日葵籽油4~6份、棓酸0.5~1.5份、甘油硬脂酸酯0.5~1.5份、巴西棕榈树蜡0.5~1.5份、茶叶提取物2~4份、富勒烯2~4份、神经酰胺3 4~6份、硬脂醇甘草亭酸酯0.5~1.5份。
3.如权利要求2所述的用于有效抗氧化保湿的组合物,其特征在于,所述的组合物中按质量百分比计包含:霍霍巴籽油30%、紫苏籽油10%、西瓜籽油5%、黄瓜油5%、辛酸/癸酸甘油三酯1%、碳酸二辛酯1%、角鲨烷5%、抗坏血酸四异棕榈酸酯1%、扭刺仙人掌茎提取物3%、谷维素3%、生育酚类3%、四氢姜黄素1%、植物甾醇类2%、红没药醇5%、二甲基甲氧基苯并二氢吡喃醇5%、向日葵籽油5%、棓酸1%、甘油硬脂酸酯1%、巴西棕榈树蜡1%、茶叶提取物3%、富勒烯3%、神经酰胺3 5%、硬脂醇甘草亭酸酯1%。
4.一种如权利要求1~3中任意一项所述的用于有效抗氧化保湿的组合物的制备方法,其特征在于,所述的方法包含:
步骤1、按比例称量各原料,并分为A相、B相;
步骤2、将A相各组分加热并混合均匀;
步骤3、将B相各组分加热并混合均匀;
步骤4、在搅拌条件下将B相加入A相,混合均匀。
5.如权利要求4所述的用于有效抗氧化保湿的组合物的制备方法,其特征在于,所述的A相包含霍霍巴籽油、紫苏籽油、西瓜籽油、黄瓜油、辛酸/癸酸甘油三酯、碳酸二辛酯、角鲨烷、抗坏血酸四异棕榈酸酯、扭刺仙人掌茎提取物、谷维素、生育酚类、四氢姜黄素、植物甾醇类、红没药醇、二甲基甲氧基苯并二氢吡喃醇、向日葵籽油。
6.如权利要求4所述的用于有效抗氧化保湿的组合物的制备方法,其特征在于,所述的B相包含棓酸、甘油硬脂酸酯、巴西棕榈树蜡、茶叶提取物、富勒烯、神经酰胺3、硬脂醇甘草亭酸酯。
7.如权利要求4所述的用于有效抗氧化保湿的组合物的制备方法,其特征在于,所述的步骤2和步骤3中,分别将A相和B相各组分均加热至45℃。
8.一种如权利要求4所述的方法制备的用于有效抗氧化保湿的组合物的用途,其特征在于,所述的组合物用于制备护肤品;所述的护肤品包含精油。
