The invention relates to a compound feed for improving the body color of large yellow croakers and a preparation method thereof, wherein the compound feed comprises a basic feed and carotenoid; the basic feed comprises fish meal, clostridium ethanolate protein, cassava meal, flour, fish oil, soybean lecithin, vitamin premix, mineral premix, choline chloride, calcium dihydrogen phosphate, kelp powder containing glue, microcrystalline cellulose and additives; all percentages are by weight. In the mixed feed containing clostridium ethanolate protein, the yellow values of the skins of a plurality of parts of the large yellow croaker are obviously improved by adding the astaxanthin or the lutein, and the body color of the large yellow croaker is effectively improved under the condition of reducing the using amount of feed fish meal. The preparation method of the compound feed is simple and convenient, the operation is simple, the compound feed is suitable for large-scale and industrial production, and the product is the puffed floating feed, so that the feed waste can be reduced in the using process, and the environmental pollution is avoided.

The large yellow croaker (Larimichthys crocea) is favored by consumers in China due to golden yellow appearance and tender meat quality, and is an economic fish species with extremely high mariculture yield in China. The large yellow croaker belongs to carnivorous fishes, so the content of fish meal in the compound feed is extremely high. In recent years, with the gradual expansion of the culture scale of the large yellow croaker, the popularization rate of the artificial compound feed is gradually improved, and the demand for fish meal is also increased year by year.
In the related art, the golden yellow body color of the large yellow croaker is one of the reasons why the large yellow croaker is well received by consumers, and the quality of the body color has great influence on the market value of the large yellow croaker. However, compared with wild large yellow croakers, the body color of the intensively cultured large yellow croakers is gradually degraded, the market value of the large yellow croakers is greatly reduced, and the key factor for restricting the development of the large yellow croakers culture industry in China is formed, so that the method for improving the body color of the cultured large yellow croakers by using the compound feed has important significance.

In a specific example, different feed large yellow croakers were fed:
cultivation experiment
Before the beginning of the formal culture experiment, pseudosciaena crocea is temporarily cultured in a seawater floating net cage (4 multiplied by 8 multiplied by 4), and expanded commercial feed is fed to adapt to the environment and the feed. After two weeks, starving for 24 hours, selecting large yellow croakers which are strong and have regular specifications (the initial weight: 189.7 +/-0.64 g), randomly dividing the large yellow croakers into floating cages (2 multiplied by 2), feeding the large yellow croakers with 100 tails of each cage in 3 parallel cages corresponding to each feed twice a day, weighing and recording the number of dead fishes and the feed amount. During the culture period, the water temperature is 19.8-28.5 ℃, the salinity is 31.1-34.9, the dissolved oxygen is more than 6mg/L, and the culture period is 90 days.
Body colour measurement
After the culture experiment is finished and 8 o' clock in the evening, 9 fishes are randomly fished out from each net cage, and the body color of the large yellow croaker is measured at 6 different positions (back 1, back site 1, back 2, back site 2, tail, abdomen 1, abdomen site 1, abdomen 2, abdomen site 2 and abdominal bottom) by using a portable color difference meter (CR-400, Konika-Meinenda, Japan) under the condition of keeping out of the sun (the sites are shown in the following figure 1). L represents the brightness value, a represents the red value, b represents the yellow value, measured according to the rules of the international commission on illumination CIE (cie.1976). The larger the value of L, the brighter the color, otherwise, the darker the color; the larger the value of a, the more red the color is, otherwise, the more green the color is; a larger value of b indicates a more yellow color, otherwise the color is more blue. The measurement results are shown in Table 3.

Aiming at the abdominal 1 site of the large yellow croaker, compared with a group without pigments, the yellow value b (P is less than 0.05) can be obviously improved by adding lutein; the addition of astaxanthin slightly increased the yellow value b, but there was no significant difference compared to the group without addition and the group with addition of lutein.
Aiming at 2 sites of the abdomen of the large yellow croaker, compared with a group without pigment, the addition of lutein obviously reduces the brightness value L (P is less than 0.05) and obviously improves the yellow value b (P is less than 0.05); compared with the lutein group, the red value a (P < 0.05) is obviously improved by adding the astaxanthin, and compared with the group without the pigment, the yellow value b (P < 0.05) is obviously improved by adding the astaxanthin.
For the abdominal bottom part of the large yellow croaker, compared with the group without the added pigment, the added astaxanthin or the added lutein obviously reduces the brightness value L, obviously increases the red value a, obviously increases the yellow value b (P < 0.05), and slightly increases the yellow value b by adding the astaxanthin, but has no obvious difference compared with the group without the added pigment and the group with the added lutein.
According to the analysis of the above experimental results, the addition of 80mg/kg astaxanthin to the compound feed containing 22.4% clostridium ethanolate protein can obviously improve the red value a of the abdomen 2 and the bottom of the abdomen of the large yellow croaker, obviously improve the yellow value b of the abdomen 2 and obviously reduce the brightness value L of the bottom of the abdomen. The yellow value b of the back 1, the back 2, the tail, the abdomen 1, the abdomen 2 and the abdominal bottom of the large yellow croaker can be obviously improved by adding 80mg/kg of lutein, the brightness value L of the back 1 is obviously improved, the brightness value L of the abdomen 2 and the abdominal bottom is obviously reduced, and the red value a of the abdominal bottom is obviously improved.
Meanwhile, although the body color of the large yellow croaker can be improved by adding the astaxanthin and the lutein into the basic feed, the body color improvement effect of the lutein is more obvious. Lutein is the preferred choice in view of practical production costs.
Particularly, the clostridium ethanolate protein is a novel mycoprotein with extremely high nutritional value, and as a novel protein source, a few studies show that the clostridium ethanolate protein can replace part of fish meal in certain carnivorous fish compound feeds, but the studies on replacing the fish meal in large yellow croaker compound feeds are rarely reported. Astaxanthin and lutein belong to carotenoid, aquatic animals cannot synthesize the carotenoid, and the body color of large yellow croaker can be improved by adding the astaxanthin or the lutein into feed. In the invention, the astaxanthin and the lutein are creatively added into the large yellow croaker compound feed containing clostridium ethanolate protein, so that the using amount of fish meal in the large yellow croaker compound feed is reduced, and the body color of the cultured large yellow croaker is obviously improved, thereby improving the economic value of the cultured large yellow croaker and reducing the culture cost. Further comparison shows that the lutein has better improvement effect on the yellow value of the skin of the large yellow croaker.
As shown in fig. 2, an embodiment of the present application provides a method for preparing a compound feed for improving body color of large yellow croakers, including:
s101, obtaining solid raw materials of basic feed and preprocessing the solid raw materials;
s102, weighing and uniformly mixing vitamin premix, mineral premix, choline chloride, monocalcium phosphate, kelp powder containing gum, additives and astaxanthin or lutein in the processed solid raw materials according to a mass ratio;
s103, weighing and uniformly mixing the rest solid raw materials according to the mass ratio, and uniformly mixing all the mixtures in a stirrer;
s104, putting the uniformly mixed feed into a bulking machine for bulking and granulating, and then drying;
and S105, uniformly mixing the fish oil, the soybean oil and the soybean lecithin according to the mass ratio, and uniformly spraying the oil mixture on the surface of the feed by using a vacuum oil spraying machine to obtain the pellet feed.
Preferably, the pretreatment of the solid raw material comprises:
the solid feedstock is crushed and sieved.
Specifically, first, the raw material is pretreated, for example, fish meal is crushed and sieved.
Then, accurately weighing and uniformly mixing the vitamin premix with lower content, the mineral premix, choline chloride, monocalcium phosphate, the kelp powder containing the glue, the additive and the astaxanthin or lutein according to the mass ratio.
Then, the rest raw materials are accurately weighed and uniformly mixed according to the mass ratio, and all the mixture is uniformly mixed in a stirrer.
Then, the evenly mixed feed is put into a bulking machine for bulking and granulating, and then is dried.
And then, uniformly mixing the fish oil, the soybean oil and the soybean lecithin according to the mass ratio, and uniformly spraying the oil mixture on the surface of the feed by using a vacuum oil spraying machine to obtain the pellet feed, namely the compound feed provided by the invention.
In summary, the invention provides a compound feed for improving body color of large yellow croakers and a preparation method thereof, wherein the compound feed comprises a basal feed and carotenoids; the basic feed comprises fish meal, clostridium ethanolate protein, cassava meal, flour, fish oil, soybean lecithin, vitamin premix, mineral premix, choline chloride, calcium dihydrogen phosphate, kelp powder containing glue, microcrystalline cellulose and additives; all percentages are by weight. In the mixed feed containing clostridium ethanolate protein, the yellow values of the skins of a plurality of parts of the large yellow croaker are obviously improved by adding the astaxanthin or the lutein, and the body color of the large yellow croaker is effectively improved under the condition of reducing the using amount of feed fish meal. The preparation method of the compound feed is simple and convenient, the operation is simple, the compound feed is suitable for large-scale and industrial production, and the product is the puffed floating feed, so that the feed waste can be reduced in the using process, and the environmental pollution is avoided.

1. A compound feed for improving the body color of large yellow croakers is characterized by comprising a basic feed and carotenoid;
the basic feed comprises the following components in percentage by weight:
34.1 percent of fish meal, 22.44 percent of clostridium ethanolate protein, 5 percent of cassava meal, 9.8 percent of flour, 4.9 percent of fish oil, 2.0 percent of soybean oil, 1.5 percent of soybean lecithin, 0.5 percent of vitamin premix, 0.5 percent of mineral premix, 0.2 percent of choline chloride, 1 percent of calcium dihydrogen phosphate, 1 percent of kelp powder containing glue, 16.8 percent of microcrystalline cellulose and 0.4 percent of additive;
all percentages are by weight.
2. The compound feed for improving body color of large yellow croaker according to claim 1,
the carotenoid is added to the basal feed in an amount of 80 mg/kg.
3. The compound feed for improving body color of large yellow croaker according to claim 1, wherein the carotenoid comprises:
astaxanthin or lutein.
4. The compound feed for improving body color of large yellow croaker according to any one of claims 1 to 3,
the mineral premix comprises FeSO4·H2O,ZnSO4·H2O,NaCl,MnSO4·H2O,CuSO4·5H2O,CoCl2·6H2O,Na2SeO3Potassium iodate and zeolite powder.
5. The compound feed for improving body color of large yellow croaker according to claim 4,
the vitamin premix comprises: inositol, niacinamide, calcium pantothenate, folic acid, biotin, vitamin A, vitamin E, vitamin C ester, vitamin K3, vitamin B2, vitamin B6, vitamin D3, vitamin B1, vitamin B12 and wheat middling.
6. The compound feed for improving body color of large yellow croaker according to claim 5, wherein the additive comprises:
one or two of phagostimulant, mildew preventive, antioxidant and anticaking agent;
wherein the phagostimulant comprises glycine and/or betaine;
the anti-caking agent comprises microcrystalline cellulose;
the mildew preventive comprises calcium propionate and/or fumaric acid.
The antioxidant comprises ethoxyquinoline;
the anti-caking agent comprises microcrystalline cellulose.
7. A preparation method of a compound feed for improving the body color of large yellow croakers is characterized by comprising the following steps:
obtaining solid raw materials of a basic feed and pretreating the solid raw materials;
weighing and uniformly mixing vitamin premix, mineral premix, choline chloride, monocalcium phosphate, kelp powder containing glue, additives and astaxanthin or lutein in the processed solid raw materials according to the mass ratio;
weighing and uniformly mixing the rest solid raw materials according to the mass ratio, and uniformly mixing all the mixtures in a stirrer;
putting the uniformly mixed feed into a bulking machine for bulking and granulating, and then drying;
evenly mixing the fish oil, the soybean oil and the soybean lecithin according to the mass ratio, and evenly spraying the oil mixture on the surface of the feed by using a vacuum oil spraying machine to obtain the pellet feed.
8. The method of claim 7, wherein the pre-treating the solid feedstock comprises:
the solid feedstock is crushed and sieved.
