Abstract
The invention discloses a preparation method of an antioxidant and anti-aging stock solution of incomplete enzyme digestion sericin, which comprises the following steps: firstly, 100g of sericin powder is dissolved in 1L of distilled water to prepare solution A with the concentration of 0.1 g/ml; dissolving 100g protease in 1L distilled water to obtain solution B with pH of 7.0 and concentration of 0.1 g/ml; 10g of trehalose powder, 10g of hyaluronic acid according to 1: dissolving 1L of distilled water in the proportion of 1 to prepare a C solution with the concentration of 0.01 g/ml; heating the solution A to 50 ℃, adding the solution B (the ratio of enzyme to protein is 1: 100) according to the proportion of 1/100 of the solution A to maintain the temperature, and testing the pH value to be 7, wherein the pH value is maintained at 7 by using NAHO if the conditions are changed; after reacting for two hours, heating the solution to 80-100 ℃ to destroy the protease, stopping the hydrolysis reaction of the enzyme, cooling to room temperature, and adding 1/10C solution for stabilization. The present invention can maintain the stability of incompletely hydrolyzed protein component, and thus can reach the stable state of eliminating free radical optimally.
Description
Technical Field
The invention relates to the technical field of hydrolyzed sericin, in particular to a preparation method of antioxidant and anti-aging stock solution of incomplete enzyme digestion sericin.
Background
Free radicals initiate oxidation, so that the oxidation occurs in vitro, active oxidase is easily generated by light and air, the activation of the activity of the tyrosine is promoted, and the possibility of skin wrinkling, skin spots, blackening, aging and skin canceration is caused. Therefore, the skin surface free radical can be removed, the early-stage aging reaction can be effectively prevented, and the effect of resisting the premature aging can be achieved.
Sericin does not react with and remove free radicals because of a macromolecular stable structure, but a sericin incompletely hydrolyzed substance has strong capacity of removing hydroxyl and superoxide radicals; thus, it is intended to provide a hydrolyzed sericin which can stabilize an incompletely hydrolyzed protein component and can improve the radical scavenging ability.
Based on the method, the invention designs a preparation method of the antioxidant and anti-aging stock solution of the incomplete enzyme digestion sericin.

The invention aims to provide a preparation method of an antioxidant and anti-aging stock solution of incomplete enzyme-digested sericin, which aims to solve the problems that sericin proposed in the background art does not react with free radicals and remove the free radicals because of a macromolecular stable structure, but a sericin incomplete hydrolysis substance has strong capacity of removing hydroxyl and superoxide radical; thus, there has been proposed a problem that hydrolyzed sericin, which is capable of stabilizing an incompletely hydrolyzed protein component, can improve its ability to scavenge free radicals.
In order to achieve the purpose, the invention provides the following technical scheme: a preparation method of an antioxidant and anti-aging stock solution of incomplete enzyme-digested sericin comprises the following steps:
the first step is as follows: dissolving 100g of sericin powder in 1L of distilled water to prepare solution A with the concentration of 0.1 g/ml;
the second step is that: dissolving 100g protease in 1L distilled water to obtain solution B with pH of 7.0 and concentration of 0.1 g/ml;
the third step: 10g of trehalose powder, 10g of hyaluronic acid according to 1: dissolving 1L of distilled water in the proportion of 1 to prepare a C solution with the concentration of 0.01 g/ml;
the fourth step: heating the solution A to 50 ℃, adding the solution B (the ratio of enzyme to protein is 1: 100) according to the proportion of 1/100 of the solution A to maintain the temperature, and testing the pH value to be 7, wherein the pH value is maintained at 7 by using NAHO if the conditions are changed;
the fifth step: after reacting for two hours, heating the solution to 80-100 ℃ to destroy the protease, stopping the hydrolysis reaction of the enzyme, cooling to room temperature, and adding 1/10C solution for stabilization.
Compared with the prior art, the invention has the beneficial effects that: trehalose and hyaluronic acid can protect hydrolyzed protein peptide from hydrolysis, trehalose is stable non-reducing disaccharide, has stable property, can form a special protective film on the surface of cells under the severe conditions of high temperature, high cold, drying and water loss, effectively protects the biomolecular structure from being damaged, is a very effective stabilizer and protective agent of bioactive substances, can keep the incompletely hydrolyzed protein components stable, and can achieve the stable state of optimally clearing free radicals, and the stabilizer with the ratio of trehalose to hyaluronic acid of 1:1 has the efficacy of stabilizing and repairing skin injury.

As can be seen from FIG. 3, even if the hydrolysis reaction is stopped by the high-temperature destruction of the hydrolase, a part of the hydrolase continues to undergo the hydrolysis reaction with the sericin, and after the solution is left to stand for 1 hour, the mass spectrum shows that the substance of the hydrolase is changed, and the stable state for optimally scavenging the free radicals cannot be achieved. Therefore, after the stock solution is heated to destroy the enzyme, 1 percent of trehalose and hyaluronic acid stock solution (1: 1) are added to protect the hydrolyzed protein peptide from hydrolysis, the trehalose is stable non-reducing disaccharide, has very stable property, can form a special protective film on the surface of cells under the severe conditions of high temperature, high cold, drying and water loss, effectively protects the biomolecular structure from being destroyed, and is a very effective stabilizer and protective agent for bioactive substances. The stabilizer prepared from trehalose and hyaluronic acid at a ratio of 1:1 has the effects of stabilizing and repairing skin injury, especially sunburn. After addition of the stabilizer of trehalose and hyaluronic acid, the incompletely hydrolyzed protein fraction was again found to remain stable by mass spectrometry.
The hydrolyzed sericin of the invention is added with trehalose and hyaluronic acid as stabilizers, which can maintain the stability of incompletely hydrolyzed protein components, thereby achieving the stable state of optimally clearing free radicals, and the stabilizer with the ratio of trehalose to hyaluronic acid of 1:1 has the efficacy of stabilizing and repairing skin injury.

the second step is that: dissolving 100g protease in 1L distilled water to obtain solution B with pH of 7.0 and concentration of 0.1 g/ml;
the third step: 10g of trehalose powder, 10g of hyaluronic acid according to 1: dissolving 1L of distilled water in the proportion of 1 to prepare a C solution with the concentration of 0.01 g/ml;
the fourth step: heating the solution A to 50 ℃, adding the solution B (the ratio of enzyme to protein is 1: 100) according to the proportion of 1/100 of the solution A to maintain the temperature, and testing the pH value to be 7, wherein the pH value is maintained at 7 by using NAHO if the conditions are changed;
