技术领域
本发明涉及一种灵芝孢子油软胶囊的制备工艺,属于孢子油软胶囊技术领域。
背景技术
灵芝孢子油软胶囊以破壁灵芝孢子粉为原料,经超临界CO2流体萃取后的脂溶性物质精制而成,集中了灵芝孢子粉原生质中的三萜类灵芝酸、不饱和脂肪酸、有机锗和微量元素等多种有效活性成分,是一种主要用于肿瘤患者辅助治疗、日常保健的保健品。现有技术中的灵芝孢子油软胶囊采用超细微粉碎机破壁处理后过滤萃取,由于萃取时孢子粉细化程度较高、随气体进入分离釜易堵塞管路导致萃取失效;软胶囊填充物油粒滴胶于明胶表面、再经重力加速度使下降时胶皮封口、在表面张力作用下形成圆球形逐渐凝固呈软胶囊,明胶胶原蛋白水解的多肽片段分子内与氨基酸分子间发生交联反应、在储存期间存在软胶囊崩解时间延长、囊壳与内容物灵芝包子油接触内壁成膜状物而影响产品稳定性质量的问题。
发明内容
本发明的目的是针对现有技术的缺陷,提供一种灵芝孢子油软胶囊的制备工艺,密封层胶浆、结合层胶浆和缓释层胶浆包裹灵芝孢子油复配软胶囊内容物、制成多层胶壳的灵芝孢子油软胶囊,阻断自氧化或交联作用,避免崩解时间延长、提高产品质量稳定性和生物活性。
本发明是通过如下的技术方案予以实现的:
一种灵芝孢子油软胶囊的制备工艺,其工艺步骤如下:
S1:取灵芝孢子原料经超微粉碎破壁处理后过筛、制粒为灵芝孢子粒;
超微粉碎破壁处理时采用超临界CO2经过喷嘴在压力25-45MPa、温度-10-15℃下携带灵芝孢子原料撞击硬质靶材进行射流粉碎、降压至8-10Mpa分离获得灵芝孢子粉,过筛细度大于等于200目,灵芝孢子粉经制粒机制为40-120目的灵芝孢子粒并真空干燥;
S2:取灵芝孢子粒经超临界萃取、分离获得灵芝孢子油,灵芝孢子油与灵芝孢子粉、大豆磷脂、活性肽、二十二碳六烯酸、维生素按照质量比:75-90:6-8:0.1-5:0.05-2:0.05-2:0.05-0.5均匀混合配制为软胶囊内容物;
超临界萃取时在萃取釜中、压力28~42MPa、温度40-55℃、超临界CO2流量30-45Kg/h、超声频率为30~60kHz、超声功率为100~800W下、进行超临界CO2萃取3-6h,分离时在一级或两级分离釜中降温5-10℃、在压力8-10Mpa下分离收集灵芝孢子油;
所述活性肽为苦瓜肽、海参肽、海藻肽、大豆肽、核桃肽中的一种或多种,所述维生素为脂溶性维生素A、维生素D、维生素E、维生素K中的一种或多种;
软胶囊内容物还添加占总质量比为0.01-0.5%的丁基羟基茴香醚、2,6-二叔丁基-4-甲基苯酚、叔丁基对苯二酚、没食子酸丙酯中的一种或多种;
S3:将软胶囊胶壳材料在60-80℃化胶、均匀搅拌、保温1-2h静置消泡制成密封层胶浆、结合层胶浆和缓释层胶浆,密封层胶浆的软胶囊壳材料为羟丙基甲基纤维素、增塑剂、去离子水按质量比50-60:15-20:20-40组成,结合层胶浆的软胶囊壳材料为高分子化合物、增稠剂、去离子水按质量比40-60:10-15:20-30组成,缓释层胶浆的软胶囊壳材料为普鲁兰多糖、明胶、增塑剂、崩解剂、遮光剂和去离子水按质量比20-30:25-35:15-25:7-12:0.1-0.5:15-20组成;
所述增塑剂为甘油、琼脂、聚乙二醇400、邻苯二甲酸酯中的一种或多种,所述高分子化合物为瓜尔豆胶、壳多糖、果胶、阿拉伯树胶中的一种或多种,所述增稠剂为海藻酸丙二醇酯、柠檬酸钠中的一种或多种;
所述崩解剂为淀粉、海藻酸钠中的一种或多种,所述遮光剂为胡萝卜素、姜黄素、胭脂红色素、亮蓝色素中的一种或多种;
取软胶囊内容物和密封层胶浆滴胶成型、制成灵芝孢子油软胶囊,密封层胶浆包裹软胶囊内容物的厚度为80-100μm,冷却、干燥定型、再通过流化床喷雾依次与结合层胶浆和缓释层胶浆包衣,结合层胶浆的包衣厚度为4-8μm,缓释层胶浆包衣厚度为50-150μm,干燥定型、清洗获得成品。
本发明的有益效果为:
(1)采用超临界CO2射流粉碎、降压分离获得灵芝孢子粉对灵芝孢子进行超微粉碎破壁处理、提高破壁率,制粒避免萃取时灵芝孢子粉过细而堵塞管路导致萃取失效问题,优选超临界萃取条件、结合超声辅助超临界CO2萃取,提高提取率,灵芝孢子油与灵芝孢子粉、大豆磷脂、活性肽、二十二碳六烯酸、维生素复配,使复配后的软胶囊内容物生物活性更高、抑制氧化断解为小分子链与囊壳交联;
(2)密封层胶浆、结合层胶浆和缓释层胶浆,经滴胶成型、流化床喷雾结合层胶浆和缓释层胶浆包衣,制成多层胶壳的灵芝孢子油软胶囊成品,密封层胶浆阻断软胶囊内容物与明胶中的氨基酸残基自氧化或交联作用,结合层胶浆增加结合性、避免分层,缓释层胶浆抗氧化、释放可控,避免储存期间存在软胶囊崩解时间延长、囊壳与内容物灵芝包子油接触内壁成膜状物而影响产品稳定性质量的问题,使灵芝孢子油软胶囊成品可以应用于制备抗肿瘤食品、保健食品或药物。

实施例1:
一种灵芝孢子油软胶囊的制备工艺,其工艺步骤如下:
S1:取灵芝孢子原料经超微粉碎破壁处理后过筛、制粒为灵芝孢子粒;
超微粉碎破壁处理时采用超临界CO2经过喷嘴在压力35MPa、温度-12℃下携带灵芝孢子原料撞击硬质靶材进行射流粉碎、降压至8Mpa分离获得灵芝孢子粉,过筛细度为300目,灵芝孢子粉经制粒机制为80目的灵芝孢子粒并真空干燥;
S2:取灵芝孢子粒经超临界萃取、分离获得灵芝孢子油,灵芝孢子油与灵芝孢子粉、大豆磷脂、活性肽、二十二碳六烯酸、维生素按照质量比:85:6:3:0.1:0.2:0.3均匀混合配制为软胶囊内容物;
超临界萃取时在萃取釜中、压力34MPa、温度35℃、超临界CO2流量32Kg/h、超声频率为50kHz、超声功率为600W下、进行超临界CO2萃取4h,分离时在一级中降温8℃、在压力8Mpa下分离收集灵芝孢子油;
所述活性肽为苦瓜肽、海参肽、海藻肽按质量比1:1:1组成,所述维生素为脂溶性维生素A、维生素D按质量比1:1组成;
软胶囊内容物还添加占总质量比为0.05%的丁基羟基茴香醚和0.05%的2,6-二叔丁基-4-甲基苯酚;
S3:将软胶囊胶壳材料在62℃化胶、均匀搅拌、保温1.5h静置消泡制成密封层胶浆、结合层胶浆和缓释层胶浆;

取软胶囊内容物和密封层胶浆滴胶成型、制成灵芝孢子油软胶囊,密封层胶浆包裹软胶囊内容物的厚度为85μm,冷却、干燥定型、再通过流化床喷雾依次与结合层胶浆和缓释层胶浆包衣,结合层胶浆的包衣厚度为6μm,缓释层胶浆包衣厚度为100μm,干燥定型、清洗获得成品。
实施例2:
一种灵芝孢子油软胶囊的制备工艺,其工艺步骤如下:
S1:取灵芝孢子原料经超微粉碎破壁处理后过筛、制粒为灵芝孢子粒;
超微粉碎破壁处理时采用超临界CO2经过喷嘴在压力40MPa、温度5℃下携带灵芝孢子原料撞击硬质靶材进行射流粉碎、降压至9Mpa分离获得灵芝孢子粉,过筛细度为300目,灵芝孢子粉经制粒机制为60目的灵芝孢子粒并真空干燥;
S2:取灵芝孢子粒经超临界萃取、分离获得灵芝孢子油,灵芝孢子油与灵芝孢子粉、大豆磷脂、活性肽、二十二碳六烯酸、脂溶性维生素E按照质量比:86:6:0.1:0.05:2:0.3均匀混合配制为软胶囊内容物;
超临界萃取时在萃取釜中、压力33MPa、温度40℃、超临界CO2流量35Kg/h、超声频率为45kHz、超声功率为500W下、进行超临界CO2萃取4h,分离时在一级分离釜中降温8℃、在压力10Mpa下、二级分离釜中降压为8Mpa下,分离收集灵芝孢子油;
所述活性肽为海参肽、海藻肽、大豆肽按质量比1:1:1组成;
软胶囊内容物还添加占总质量比为0.08%的2,6-二叔丁基-4-甲基苯酚、0.08%的叔丁基对苯二酚;
S3:将软胶囊胶壳材料在68℃化胶、均匀搅拌、保温1h静置消泡制成密封层胶浆、结合层胶浆和缓释层胶浆,密封层胶浆的软胶囊壳材料为羟丙基甲基纤维素、增塑剂、去离子水按质量比55:18:32组成,所述增塑剂为聚乙二醇400、邻苯二甲酸酯按质量比1:1组成;
结合层胶浆的软胶囊壳材料为高分子化合物、增稠剂、去离子水按质量比58:12:27组成,所述高分子化合物为壳多糖和果胶按质量比1:1组成,增稠剂为海藻酸丙二醇酯、柠檬酸钠按质量比1:1组成;
缓释层胶浆的软胶囊壳材料为普鲁兰多糖、明胶、增塑剂、崩解剂淀粉、遮光剂和去离子水按质量比25:32:19:10:0.4:18组成,增塑剂甘油、琼脂按质量比1:1组成,增稠剂为姜黄素、胭脂红色素按质量比3:1组成;
取软胶囊内容物和密封层胶浆滴胶成型、制成灵芝孢子油软胶囊,密封层胶浆包裹软胶囊内容物的厚度为90μm,冷却、干燥定型、再通过流化床喷雾依次与结合层胶浆和缓释层胶浆包衣,结合层胶浆的包衣厚度为5μm,缓释层胶浆包衣厚度为60μm,干燥定型、清洗获得成品。
实施例3:
一种灵芝孢子油软胶囊的制备工艺,其工艺步骤如下:
S1:取灵芝孢子原料经超微粉碎破壁处理后过筛、制粒为灵芝孢子粒;
超微粉碎破壁处理时采用超临界CO2经过喷嘴在压力26MPa、温度10℃下携带灵芝孢子原料撞击硬质靶材进行射流粉碎、降压至8Mpa分离获得灵芝孢子粉,过筛细度为300目,灵芝孢子粉经制粒机制为100目的灵芝孢子粒并真空干燥;
S2:取灵芝孢子粒经超临界萃取、分离获得灵芝孢子油,灵芝孢子油与灵芝孢子粉、大豆磷脂、活性肽、二十二碳六烯酸、维生素按照质量比:76:8:4:1:1.3:0.18均匀混合配制为软胶囊内容物;
超临界萃取时在萃取釜中、压力35MPa、温度42℃、超临界CO2流量44Kg/h、超声频率为50kHz、超声功率为600W下、进行超临界CO2萃取6h,分离时一级分离釜中降温8℃、在压力10Mpa下、二级分离釜中再降温8℃下,分离收集灵芝孢子油;
所述活性肽为海藻肽、大豆肽、核桃肽按质量比1:1:2组成,所述维生素为脂溶性维生素A、维生素E、维生素K按质量比1:1:1组成;
软胶囊内容物还添加占总质量比为0.03%的叔丁基对苯二酚、0.06%的没食子酸丙酯;
S3:将软胶囊胶壳材料在75℃化胶、均匀搅拌、保温2h静置消泡制成密封层胶浆、结合层胶浆和缓释层胶浆;密封层胶浆的软胶囊壳材料为羟丙基甲基纤维素、增塑剂、去离子水按质量比55:18:36组成,所述增塑剂为琼脂、邻苯二甲酸酯按质量比1:2组成;
结合层胶浆的软胶囊壳材料为高分子化合物、增稠剂柠檬酸钠、去离子水按质量比56:14:25组成,所述高分子化合物为瓜尔豆胶、壳多糖、阿拉伯树胶按质量比1:1:2组成;
缓释层胶浆的软胶囊壳材料为普鲁兰多糖、明胶、增塑剂、崩解剂海藻酸钠、遮光剂和去离子水按质量比28:33:17:11:0.3:18组成,增塑剂为甘油、聚乙二醇400按质量比1:2组成,遮光剂为胡萝卜素、亮蓝色素2:1组成;
取软胶囊内容物和密封层胶浆滴胶成型、制成灵芝孢子油软胶囊,密封层胶浆包裹软胶囊内容物的厚度为85μm,冷却、干燥定型、再通过流化床喷雾依次与结合层胶浆和缓释层胶浆包衣,结合层胶浆的包衣厚度为6μm,缓释层胶浆包衣厚度为130μm,干燥定型、清洗获得成品。
本发明的机理为:
(1)采用超临界CO2射流粉碎、降压分离获得灵芝孢子粉对灵芝孢子进行超微粉碎破壁处理,避免流体粘性增大导致扩散下降问题,保证加速、形成多孢子的相互碰撞、穿透作用,具有无掺杂易分离的特点,破壁率达到88%以上,提高破壁效率、促进人体对营养物的吸收、利于孢子油高效萃取、过筛细度大于等于200目后粉经制粒机制为40-120目的灵芝孢子粒并真空干燥,避免萃取时灵芝孢子粉过细而堵塞管路导致萃取失效问题;
(2)优选超临界萃取条件、结合超声辅助超临界CO2萃取,一级或两级分离釜中降温5-10℃、在压力8-10Mpa下分离收集高效灵芝孢子油,促进对灵芝孢子三萜类灵芝酸、不饱和脂肪酸、有机锗和微量元素等活性成分吸收提高提取率,将灵芝孢子油与灵芝孢子粉、大豆磷脂、活性肽、二十二碳六烯酸、维生素复配,大豆磷脂是由甘油、脂肪酸、胆碱或胆胺所组成的酯,无药物副作用、对防治预防疾病由积极作用;
包括但不限于苦瓜肽、海参肽、海藻肽、大豆肽、核桃肽的活性肽,由至少两个以上的氨基酸以肽键,具有抗病毒、抗高血压、免疫调节、激素调节、抑菌、降胆固醇等作用,二十二碳六烯酸有利于促进细胞活性、包括但不限于脂溶性维生素A、维生素D、维生素E、维生素K的维生素促进机体的新陈代谢、抗氧化,还添加丁基羟基茴香醚、2,6-二叔丁基-4-甲基苯酚、叔丁基对苯二酚、没食子酸丙酯中的一种或多种抗氧化,使复配后的软胶囊内容物生物活性更高、抑制氧化断解为小分子链与囊壳交联;
(3)将软胶囊胶壳材料化胶、均匀搅拌消泡制成密封层胶浆、结合层胶浆和缓释层胶浆,经滴胶成型、流化床喷雾结合层胶浆和缓释层胶浆包衣,并控制灵芝孢子油软胶囊的密封层胶浆包裹软胶囊内容物的厚度为80-100μm,结合层胶浆的包衣厚度为4-8μm,缓释层胶浆包衣厚度为50-150μm,制成多层胶壳的灵芝孢子油软胶囊成品;
密封层胶浆的软胶囊壳材料采用羟丙基甲基纤维素、具有表面活性,透明度高、性能稳定、成膜性、分散性和粘结性的特点,包括但不限于甘油、琼脂、聚乙二醇400、邻苯二甲酸酯的增塑剂增加成膜性,通过将软胶囊内容物滴制包过形成密封层胶皮,在重力加速度下降作用下封口形成软胶囊、干燥定型,阻断软胶囊内容物与明胶中的氨基酸残基自氧化或交联作用,避免存储过程中囊壳变性、溶出时间延长和交联引起胶囊内容物降解失活影响稳定性、营养价值的问题;
结合层胶浆的软胶囊壳材料采用包括但不限于瓜尔豆胶、壳多糖、果胶、阿拉伯树胶高分子化合物、包括但不限于海藻酸丙二醇酯、柠檬酸钠的增稠剂,尔豆胶是从豆科植物瓜尔豆的胚乳中提取出的一种非离子型半乳甘露聚糖,壳多糖为N-乙酰葡糖胺通过β连接聚合而成的结构同多糖,果胶羟基被不同程度甲酯化的线性聚半乳糖醛酸和聚L-鼠李糖半乳糖醛酸,阿拉伯树胶包括树胶醛糖、半乳糖、葡萄糖醛酸等,与海藻酸丙二醇酯、柠檬酸钠乳化稳定,用于增加密封层胶浆层与密封层胶浆层的结合性能,避免分层;
缓释层胶浆软胶囊壳材料采用普鲁兰多糖、明胶、增塑剂、崩解剂、遮光剂,普鲁兰多糖是出芽短梗霉发酵所产生的类似葡聚糖、黄原胶的胞外水溶性粘质多糖,成膜性、阻气性、可塑性、粘性均较强,起到抗氧化作用,崩解剂淀粉、海藻酸钠在胃肠液中迅速裂碎成细小颗粒迅速溶解吸收,遮光剂降低紫外作用、在存储过程中抑制自氧化或交联作用,提高胶囊内容物的释放可控性;
将实施例1-3制得的灵芝孢子油软胶囊与市购灵芝孢子油软胶囊(仙芝楼牌)作为对比例检测对比,结果如下:

使灵芝孢子油软胶囊成品可以应用于制备抗肿瘤食品、保健食品或药物,释放可控,避免储存期间存在软胶囊崩解时间延长、囊壳与内容物灵芝包子油接触内壁成膜状物而影响产品稳定性质量的问题。

Claims (10)
1.一种灵芝孢子油软胶囊的制备工艺,其特征在于,其工艺步骤如下:
S1:取灵芝孢子原料经超微粉碎破壁处理后过筛、制粒为灵芝孢子粒;
S2:取灵芝孢子粒经超临界萃取、分离获得灵芝孢子油,灵芝孢子油与灵芝孢子粉、大豆磷脂、活性肽、二十二碳六烯酸、维生素配制为软胶囊内容物;
S3:将软胶囊胶壳材料化胶、均匀搅拌消泡制成密封层胶浆、结合层胶浆和缓释层胶浆,取软胶囊内容物和密封层胶浆滴胶成型、制成灵芝孢子油软胶囊,冷却、干燥定型、再通过流化床喷雾依次与结合层胶浆和缓释层胶浆包衣,干燥定型、清洗获得成品。
2.根据权利要求1所述的一种灵芝孢子油软胶囊的制备工艺,其特征在于,超微粉碎破壁处理时采用超临界CO2经过喷嘴在压力25-45MPa、温度-10-15℃下携带灵芝孢子原料撞击硬质靶材进行射流粉碎、降压至8-10Mpa分离获得灵芝孢子粉,过筛细度大于等于200目,灵芝孢子粉经制粒机制为40-120目的灵芝孢子粒并真空干燥。
3.根据权利要求1所述的一种灵芝孢子油软胶囊的制备工艺,其特征在于,超临界萃取时在萃取釜中、压力28~42MPa、温度40-55℃、超临界CO2流量30-45Kg/h、超声频率为30~60kHz、超声功率为100~800W下、进行超临界CO2萃取3-6h,分离时在一级或两级分离釜中降温5-10℃、在压力8-10Mpa下分离收集灵芝孢子油。
4.根据权利要求1所述的一种灵芝孢子油软胶囊的制备工艺,其特征在于,软胶囊内容物由灵芝孢子油、灵芝孢子粉、大豆磷脂、活性肽、二十二碳六烯酸、维生素按照质量比:75-90:6-8:0.1-5:0.05-2:0.05-2:0.05-0.5均匀混合配制而成。
5.根据权利要求1所述的一种灵芝孢子油软胶囊的制备工艺,其特征在于,所述活性肽为苦瓜肽、海参肽、海藻肽、大豆肽、核桃肽中的一种或多种,所述维生素为脂溶性维生素A、维生素D、维生素E、维生素K中的一种或多种。
6.根据权利要求1所述的一种灵芝孢子油软胶囊的制备工艺,其特征在于,软胶囊内容物还添加占总质量比为0.01-0.5%的丁基羟基茴香醚、2,6-二叔丁基-4-甲基苯酚、叔丁基对苯二酚、没食子酸丙酯中的一种或多种。
7.根据权利要求1所述的一种灵芝孢子油软胶囊的制备工艺,其特征在于,密封层胶浆的软胶囊壳材料为羟丙基甲基纤维素、增塑剂、去离子水按质量比50-60:15-20:20-40组成,结合层胶浆的软胶囊壳材料为高分子化合物、增稠剂、去离子水按质量比40-60:10-15:20-30组成,缓释层胶浆的软胶囊壳材料为普鲁兰多糖、明胶、增塑剂、崩解剂、遮光剂和去离子水按质量比20-30:25-35:15-25:7-12:0.1-0.5:15-20组成。
8.根据权利要求7所述的一种灵芝孢子油软胶囊的制备工艺,其特征在于,所述增塑剂为甘油、琼脂、聚乙二醇400、邻苯二甲酸酯中的一种或多种,所述高分子化合物为瓜尔豆胶、壳多糖、果胶、阿拉伯树胶中的一种或多种,所述增稠剂为海藻酸丙二醇酯、柠檬酸钠中的一种或多种。
9.根据权利要求8所述的一种灵芝孢子油软胶囊的制备工艺,其特征在于,所述崩解剂为淀粉、海藻酸钠中的一种或多种,所述遮光剂为胡萝卜素、姜黄素、胭脂红色素、亮蓝色素中的一种或多种。
10.根据权利要求1-9任意一项所述的一种灵芝孢子油软胶囊的制备工艺,其特征在于,滴胶时灵芝孢子油软胶囊的密封层胶浆包裹软胶囊内容物的厚度为80-100μm,结合层胶浆的包衣厚度为4-8μm,缓释层胶浆包衣厚度为50-150μm。
