And (3) purifying the phage: the plaques isolated initially are often of inconsistent size and morphology and require further purification. Picking single independent, uniform-shaped, clear and transparent plaques on a double-layer plate with plaques, soaking the single independent, uniform-shaped and clear and transparent plaques in 1ml of physiological saline for 10 minutes, taking clear liquid, filtering the clear liquid by using a 0.22 micron microporous filter, properly diluting filtrate (single plaques exist when the double-layer plate is laid), laying the filtrate on the double-layer plate together with host bacterium suspension, repeating the step for 3-4 times, and obtaining purified phage when the size, the shape and the definition of each plaque on the double-layer plate are consistent.
(3) Determination of phage titer
Spreading purified bacteriophage on double-layer plate, adding single plaque into LB broth, adding host bacteria suspension, proliferating bacteriophage, centrifuging proliferation liquid at 10000rpm for 5min, filtering with 0.22 micrometer microporous filter, and diluting filtrate by 10 times gradient to 108Taking 0.1 ml of phage diluent and 0.1 ml of host bacterial suspension in the last 3 dilutionsMl, double plates were plated and phage titer was determined. Phage titer (pfu/mL) ═ plaque number dilution factor ÷ 0.1. The result shows that the plaque has consistent shape, size and definition and has typical lytic phage characteristics. The titer of the bacteriophage can reach 1013pfu/mL or more.
(4) Preservation of phages
Phage multiplication fluid was mixed with 60% glycerol according to 1: 1, and storing in liquid nitrogen.
EXAMPLE 2 Electron microscopy of phages
Phage liquid was dropped on a paraffin plate (titer 10)10pfu/m L), placing the film-coated side of the copper mesh on the phage droplet, taking off after 10min, and naturally drying in air for 2-3 min. And then, dripping a drop of 2% phosphotungstic acid (PTA) aqueous solution on a copper net for dyeing, taking down after 10min, drying in the air for 10-15 min, observing by using an electron microscope, and selecting a clear phage image for photographing.
As can be seen from the electron micrograph shown in FIG. 1, the phage head is in the shape of a regular hexagon, the diameter of the head is about 55nm, and the phage head has a tail.
Example 3 phage genome sequencing
After enrichment culture of a single phage, centrifuging for 15 minutes at 8000g under the condition of 4 ℃, adding 10% PEG8000 and 0.5M NaCl, standing overnight, adding equal amount of chloroform, mixing uniformly, standing for layering, centrifuging for 10 minutes at 5000g, removing a chloroform layer and a PEG layer, adding restriction endonuclease for digestion treatment, suspending the phage under the condition of gradient density cesium chloride, dialyzing for 3 times by TM buffer solution at the later stage, and sending the phage to a Huada gene biological sequencing company for whole genome sequencing after 30 minutes each time. The results are shown in the sequence listing.
Example 4: experiment for optimal multiplicity of infection
According to a multiplicity of infection of 1: 1. 1: 10. 10: 1. 100, and (2) a step of: 1. 1000: 1 ratio the phage multiplication solution and the host were added to the LB broth and the total volume of the culture system was ensured to be the same. After shaking culture at 37 ℃ and 200rpm for 4-6h, centrifuging at 12000r/min at normal temperature for 5min, taking supernatant, spreading on a double-plate, and measuring the titer. The results are shown in Table 1.

As can be seen from the data in Table 1, the optimal multiplicity of infection for bacteriophage RDP-VP-21007 is 100: 1.
example 5: effect of bacteriophage on growth curve of Vibrio parahaemolyticus BV-21001
The lysis curve of the phage on the host is obtained by adding the phage to the bacterial suspension, and the OD600 value is changed due to the lysis effect of the phage on the host. The host bacterial suspension cultured for 6h is inoculated into 100mL LB culture medium according to the proportion of 2 percent, phage multiplication liquid is added according to the optimal multiplicity of infection, and 3 groups of parallel controls are made by taking the control without adding phage, and the result is taken as the average value.
As can be seen from FIG. 2, the OD600 of the host without phage gradually increases with the increment of the host, and reaches the equilibrium at 6.5h, while the host with phage increases with the phage being in the adsorption and infection stage for the host in the first 1h, the OD600 increases, and with the exponential increase of the number of phage, after 1h, the phage lyses the host, the OD600 begins to decrease, but with the time being longer, the OD600 gradually fluctuates.
The vibrio parahaemolyticus phage RDP-VP-21007 is added into the host bacterium BV-21001 bacterial suspension, a certain cracking effect appears after 1h, compared with a control group, the host OD600 without the phage is obviously increased, which shows that the phage has stronger cracking capability on the host and can continuously crack the host bacterium. As the phage is in the adsorption and infection stage of the host bacteria in the first 1h, only a small part of the phage starts to crack the host, and therefore, the amount of the cracked host is far smaller than the increment of the host, the OD600 still keeps a certain rising trend in the first 1h, the number of the cracked host is larger than the multiplication speed of the host along with the exponential increase of the number of the phage, the OD600 value starts to be gradually reduced, and the mixed bacteria liquid is clearest. After continuous action for 4h, the mixed bacteria liquid turns turbid, the original sensitive bacteria are cracked by the bacteriophage, and the flora replaced by the mutant tolerant bacteria is alternated. Therefore, in order to obtain the phage with high titer, the mixed bacteria liquid should be controlled within the time range from the highest resolution to the beginning of the turbidity returning phenomenon, so as to avoid the influence on the titer of the phage.
EXAMPLE 6 determination of acid-base stability
Adjusting pH of normal saline with dilute hydrochloric acid and dilute NaOH solution to obtain buffer solution with pH of 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, and diluting bacteriophage to 5 x 10 with prepared buffer solution10pfu/mL, the dilution was incubated at 37 ℃ for 1h in a water bath, diluted 10-fold with physiological saline and plated on double plates. Counting after inverted culture at 37 ℃ for 4-6 h. The results are shown in FIG. 3.
As can be seen from FIG. 3, RDP-VP-21007 has an optimum growth pH for a neutral and alkaline environment.
Example 7 determination of thermal stability
Subpackaging the phage stock solution into EP tubes, incubating at 40 deg.C, 50 deg.C, 60 deg.C and 70 deg.C for 30min and 60min, diluting with physiological saline 10 times, spreading on double plates, and counting. The results are shown in FIG. 4.
As can be seen from FIG. 4, the activity of the phage decreased with increasing temperature and with increasing time. After the phage is acted for 60min at 70 ℃, the titer can still reach 7.7 multiplied by 10^4pfu/mL, which indicates that the phage has certain temperature tolerance.
Example 9: cracking rate test
Under the aseptic condition, 1mL of sample and 1mL of host bacterium liquid (1 × 105CFU/mL) are respectively taken, incubated for 15 minutes at 37 ℃, uniformly mixed, diluted to 10-1-10-3 gradients by using physiological saline, 100 μ L of each gradient is taken and coated on an LB agar plate, the mixture is placed at 37 ℃, cultured for 24 hours, and each gradient is repeated twice. Simultaneously, 1mL of physiological saline and 1mL of host bacterium liquid (1X 105CFU/mL) are taken as blank control, and the steps are repeated. Plates with 30-300 colonies were picked for enumeration. The test was repeated 3 times and the average was taken. The phage lysis rate was (1-number of treated colonies/number of control colonies) × 100%.
The calculation shows that the cracking rate of RDP-VP-21007 reaches 99.3%, has very good cracking effect on a host, and is suitable for being used in the culture process.
It should be noted that the specific embodiments are merely representative examples of the present invention, and it is obvious that the technical solution of the present invention is not limited to the above examples, and many variations are possible.
