white gourd: sweet and light taste, slightly cold nature; clearing away heat and toxic materials, promoting diuresis, eliminating phlegm, relieving restlessness, quenching thirst, eliminating dampness, and relieving summer-heat;
orange peel: bitter and pungent taste, warm in nature; entering lung and spleen meridians; regulating qi-flowing, invigorating spleen, eliminating dampness and phlegm, activating qi-flowing, removing food stagnation, activating spleen, and invigorating stomach.
White hyacinth bean: sweet in taste and slightly warm in nature, and has the effects of invigorating spleen, eliminating dampness, inducing diuresis, relieving swelling, clearing liver and improving eyesight; has effects in invigorating qi and spleen.
Chinese yam: sweet and neutral. To invigorate the spleen, tonify the lung, strengthen the kidney and replenish vital essence. Fructus alpiniae oxyphyllae: pungent flavor and warm nature. It enters spleen and kidney meridians. The main treatment is as follows: deficiency-cold in spleen and stomach, vomiting, diarrhea, cold pain in abdomen, excessive saliva, enuresis due to kidney deficiency, frequent micturition, spermatorrhea, and whitish and turbid urine.
Balloon flower: bitter and pungent in flavor and mild in nature. Can be used for treating cough with excessive phlegm, chest distress, pharyngalgia, hoarseness, lung carbuncle, and purulent vomiting.
Corn silk: sweet and bland in flavor and mild in nature. It enters bladder, liver and gallbladder meridians. Promoting urination and relieving swelling; clearing liver heat and promoting bile flow.
Fingered citron: pungent, bitter and sour in flavor and warm in nature. It enters liver, spleen, stomach and lung meridians. Can be used for treating stagnation of qi of liver and stomach, distending pain in chest and hypochondrium, fullness and distention in stomach, anorexia, emesis, cough, and excessive phlegm.

Compared with the prior art, the invention has the beneficial effects that:
(1) the dampness-clearing spleen-tonifying traditional Chinese medicine composition disclosed by the invention is prepared by compounding poria cocos, lotus leaves, wax gourd, orange peel, white hyacinth beans, Chinese yam, gordon euryale seed, sharpleaf galangal fruit, platycodon grandiflorum, corn stigma, fingered citron, mulberry and houttuynia cordata according to a reasonable proportion, so that the drug effect and the food effect of each component generate a synergistic effect, the effect of each component is maximized, the dampness-clearing dampness-excreting traditional Chinese medicine composition has the effects of clearing heat, excreting water and promoting diuresis and improving the damp-heat physique, and the common damp-heat physique of southern people can be well improved;
(2) according to the invention, in the preparation process, lotus leaves, white gourds, Chinese yams, white hyacinth beans, mulberries and fingered citrons are steamed, effective ingredients in the components can be dissolved out to a greater extent by controlling the steaming pressure, temperature and time, the release amount of the effective ingredients is improved, the product efficacy is ensured, and the traditional Chinese medicine composition prepared by the method has high stability and cannot be solidified or layered after being placed for a long time;

The experimental procedures in the following examples are all conventional ones unless otherwise specified. The raw materials, reagent materials and the like used in the following examples are all commercially available products unless otherwise specified. The difference from example 4 is that: the weight ratio of the poria cocos to the Chinese yam to the white gourd is 6:1:1, namely 81 parts of poria cocos, 13.5 parts of Chinese yam and 13.5 parts of white gourd, the lotus leaves and the white gourd are not steamed in the preparation process, and are directly decocted by adding water, and other operations and preparation methods are the same as those in example 4.

The difference from example 4 is that: the mass ratio of the corn stigma to the houttuynia cordata is 1:1, namely 17.5 parts of the corn stigma and 17.5 parts of the houttuynia cordata, and other steps and operations are the same as those in example 4.

The difference from example 4 is that: all the components are not steamed in the preparation process, and are directly decocted by adding water, namely (1) poria cocos, lotus leaves, wax gourd, orange peel, white hyacinth beans, Chinese yam, gordon euryale seed, fructus alpiniae oxyphyllae, platycodon grandiflorum, corn stigma, fingered citron, mulberry and houttuynia cordata are cleaned and dried for later use;

1. detecting dioscin: precisely weighing 0.5000 g of sample, adding 10ml of 70% methanol, soaking overnight, weighing, performing reflux extraction for 1h in water bath at 80 ℃, shaking once every 10min, cooling, supplementing 70% methanol to the weight, sufficiently shaking, centrifuging sample liquid for 2min at 3000r/min, centrifuging supernatant fluid for 13min at 13000r/min, and filtering through a 0.45-micron microporous membrane to obtain the product. And precisely weighing 1mg of dioscin reference substance, placing into a 10mL volumetric flask, adding chromatographic methanol solution, and fixing the volume to a scale to obtain 0.1mg/mL dioscin reference substance solution. The chromatographic conditions are that the chromatographic column is a sinoChrom ODS-BP chromatographic column (250mm multiplied by 4.6mm, 5 μm), the mobile phase is methanol-water (84:16), the flow rate is 1mL/min, the detection wavelength is 206nm, the column temperature is 30 ℃, the sample injection amount is 20 μ L, and the chromatographic peak is measured.
2. Detection of pachyman: precisely weighing 0.5000 g of sample, performing reflux extraction for 2h by using 100mL of 90% ethanol, filtering, drying, dripping 300mL of 0.5mol/L sodium hydroxide solution, standing for 10h, filtering, placing in a 250mL volumetric flask, adding water to dilute to a scale mark, and shaking uniformly for later use. Precisely weighing 20mg of glucose reference substance dried to constant weight, placing the reference substance in a 100mL volumetric flask, adding water to dissolve the reference substance and diluting the reference substance to a scale, and shaking up for later use. Precisely sucking 1mL of sample solution and 1mL of reference solution, respectively dropping into a test tube, respectively dropping into 5% phenol solution, then slowly dropping into 5mL of concentrated sulfuric acid, and quickly shaking up. Scanning between wavelengths of 300-700nm, both of which have maximum absorption values at 490nm, was selected by an ultraviolet-visible spectrophotometer, so 490nm was selected as the measurement wavelength for this experiment.

According to the detection data in table 1 above, it can be seen that the contents of dioscin and pachyman in the dampness-eliminating and spleen-strengthening traditional Chinese medicine compositions prepared in examples 1-4 are improved to a certain extent compared with those in comparative examples 1-5, and the contents of dioscin and pachyman in example 4 are the highest, so that the contents of the medicinal components can influence the dissolution rate of the effective components in the medicine to a certain extent, so that the contents of the effective components can fluctuate to a certain extent, and the contents of the medicinal components and the effective components are not in a direct relationship, so that the effective components can be better dissolved out only by controlling the medicinal components within a reasonable range, and a better treatment effect can be achieved.
On the other hand, the detection of the effective components is only performed according to the content of the dioscin and the pachyman, but the traditional Chinese medicine composition does not act on one effective component, a plurality of components are mixed to achieve a better treatment effect through interaction, each effective component can achieve a better treatment effect only within a certain content, and the treatment effect is not better when the content of one effective component is higher.
Second, Effect experiment
The anti-oxidation effect of the traditional Chinese medicine composition for tonifying spleen and eliminating dampness in each example and comparative example (the experiment refers to the method disclosed in the prior art)
1. Measurement of hydroxyl radical scavenging activity:
taking 1mL of 0.75mmol/L o-diazaphenanthrene solution into a 10mL test tube with a plug, sequentially adding 2mL of 0.2mmol/L PBS buffer solution with the pH value of 7.5 and 1mL of 0.75mmol/L ferrous sulfate, uniformly mixing, adding 1mL of sample solution, uniformly mixing, adding 1mL of 0.01% hydrogen peroxide, fixing the volume with deionized water, heating in a water bath at 37 ℃ for 60min, and measuring the absorbance value at 510 nm.
The scavenging rate of hydroxyl radicals is calculated as follows: clearance rate ═ [ (a1-a0)/(a2-a0) ] × 100%, where a1 is the absorbance when hydrogen peroxide and the sample were added; a0 is the absorbance when hydrogen peroxide was added but no sample was added; a2 is the absorbance when neither hydrogen peroxide nor the sample is added.
In addition, the preparation method of the sample solution comprises the following steps: 5g of the spleen-tonifying and dampness-eliminating traditional Chinese medicine composition for each example and each comparative example is respectively put into 9 beakers, 100mL of deionized water is added, microwave heating is carried out at 45 ℃ for 60min, and the sample solution required by the invention is obtained after cooling to room temperature.
2. Measurement of DPPH clearance:
and (3) taking 2mL of each sample solution, adding 2mL of 0.1mmol/L DPPH solution, uniformly mixing, reacting for 30min in a dark place, and measuring an absorbance value A1 at 217 nm. According to the detection data in table 2 above, it can be seen that the pharmaceutical compositions provided by the present invention all have certain effects of scavenging hydroxy radicals and DPPH, but the scavenging effects of examples 1 to 4 are better, especially the pharmaceutical composition prepared in example 4 has the best scavenging effect, even though the content of dioscin and pachymaran in example 4 is not the highest, the pharmaceutical composition prepared by the method provided in example 4 has the best effect of scavenging hydroxy radicals and DPPH, i.e. has better effects of clearing damp and invigorating spleen, and the present invention claims a damp-clearing and spleen-invigorating traditional Chinese medicine composition, so the technical scheme of the present invention functions as a whole, while the content of individual pharmaceutical components in the comparative example is changed, and the preparation method affects the overall performance of the traditional Chinese medicine composition, thereby affecting the therapeutic effect.
Third, evaluation of Using Effect
90 volunteers 25-30 years old are collected, and the clinical symptoms of the patients are inappetence, thick and greasy tongue, abdominal distention and energy deficiency, etc. with different degrees. Dividing the volunteers into 9 groups, 10 persons in each group; the traditional Chinese medicine compositions for invigorating spleen and eliminating dampness prepared in examples 1-4 and comparative examples 1-5 are respectively used, 15 g is taken every day, and two months are a treatment course. Physical symptoms were recorded for each volunteer and the specific data are shown in table 3. It should be noted that during the period of taking the spleen invigorating and dampness eliminating composition provided by the present invention, each volunteer suspends the use of other drugs.
The curative effect is judged as follows:
the effect is shown: symptoms such as bad appetite, inappetence, thick and greasy tongue coating, abdominal distention and energy deficiency are obviously improved;
the method has the following advantages: bad appetite, inappetence, thick and greasy tongue coating, abdominal distention, and poor energy;
and (4) invalidation: no improvement in symptoms.

A traditional Chinese medicine composition for eliminating dampness and strengthening spleen is characterized in that: the composition comprises the following components in parts by weight: 20-80 parts of poria cocos, 10-50 parts of lotus leaves, 15-40 parts of wax gourd, 10-30 parts of orange peel, 15-35 parts of white hyacinth beans, 20-50 parts of Chinese yam, 15-30 parts of gordon euryale seed, 20-40 parts of fructus alpiniae oxyphyllae, 10-30 parts of platycodon grandiflorum, 15-35 parts of corn stigma, 20-40 parts of fingered citron, 15-25 parts of mulberry, 5-15 parts of houttuynia cordata and 10-30 parts of maltose;
the preparation method of the traditional Chinese medicine composition for clearing damp and tonifying spleen comprises the following steps:
(1) cleaning Poria, folium Nelumbinis, fructus Benincasae, pericarpium Citri Tangerinae, semen lablab album, rhizoma Dioscoreae, semen euryales, fructus Alpinae Oxyphyllae, radix Platycodi, stigma Maydis, fructus Citri Sarcodactylis, Mori fructus and herba Houttuyniae, and drying;
(2) steaming lotus leaves, white gourds, Chinese yams, white hyacinth beans, mulberries and fingered citron according to the formula dosage respectively to obtain steamed lotus leaves, Chinese yams, white hyacinth beans, mulberries and fingered citron; pulverizing Poria, semen euryales, fructus Alpinae Oxyphyllae and radix Platycodi into coarse particles; shredding pericarpium Citri Tangerinae;
(3) decocting steamed folium Nelumbinis, fructus Benincasae, rhizoma Dioscoreae, semen lablab album, Mori fructus, and fructus Citri Sarcodactylis in water, and filtering to obtain extractive solution and residue;
(4) decocting the filter residue obtained in the step (3) and poria cocos, orange peel, gorgon fruit, fructus alpiniae oxyphyllae, platycodon grandiflorum, corn stigma and houttuynia cordata in water for 2 times, adding 6-7 times of water for the first time, and decocting for 2-3 hours; adding 3-4 times of water for the second time, decocting for 1-2 hr, filtering, mixing filtrates, concentrating, adding maltose, and concentrating to obtain the Chinese medicinal composition;
the steaming conditions of the lotus leaves in the step (2) are as follows: the pressure is 10-15KPa, the temperature is 100-105 ℃, and the steaming time is 20-30 min;
the steaming conditions of the white gourd and the Chinese yam are as follows: steaming at 95-100 deg.C under 6-8KPa for 15-20 min;
the steaming condition of the white hyacinth beans is as follows: the pressure is 10-12KPa, the temperature is 100-103 ℃, and the steaming time is 30-40 min;
the steaming conditions of the mulberry and the fingered citron are as follows: the pressure is 9-10KPa, the temperature is 90-95 deg.C, and the steaming time is 15-25 min.
