Abstract
The invention relates to the technical field of biological materials and medical treatment, in particular to injection equipment for detoxifying injection for snake bite emergency treatment and a preparation method thereof, wherein the injection equipment comprises a suction cup, an outer needle cylinder, an outer piston shaft, an outer piston handle, an inner needle cylinder, a needle head, an inner piston rod and an inner piston handle, the front end of the outer needle cylinder is communicated with the rear end of the suction cup through a connecting channel, and the outer piston slides in a matching way with the inner cavity of the outer needle cylinder; the detoxification injection comprises calcium oxalate hydrogel and a metal organic framework; the calcium oxalate hydrogel is prepared from Sodium Alginate (SA), calcium carbonate and Gluconolactone (GDL); the metal organic framework is prepared from 2-methylimidazole and zinc nitrate hexahydrate, N, N-dimethylformamide. The injection makes up the defects of other detoxification products such as serum and the like, can delay the time that the toxin enters blood circulation for nearly 1h, and strives for precious time for patients to obtain professional treatment.

Snake bites are a problem affecting public health and safety around the world. According to statistics, about 400 million people suffer from snake bites every year all over the world, wherein about 10 million people die, about 40 million people are permanently disfigured or disabled, and 180 to 270 million people have serious clinical symptoms such as neuromuscular toxicity, tissue injury, acute kidney injury and the like. At present, the most effective treatment method in China is the traditional antivenin, although the antivenin has certain treatment effect on some common snake bites, the storage and transportation conditions of the bioprotein products are strictly limited, and timely and accurate treatment is difficult to obtain after the bioprotein products are bitten by the poisonous snakes in the field. And domestic anti-snake venom serum production companies meeting the standards are few and few, the annual yield is low, and the current situation of short supply and short demand appears on the market.

a through hole for the inner needle cylinder to pass through is axially formed in the outer piston, the inner piston slides in a matched manner with the inner cavity of the inner needle cylinder, the front end of the inner piston rod is axially connected with the rear wall of the inner piston, and the rear end of the inner piston rod passes through the outer piston handle and then is connected with the inner piston handle; the needle is arranged at the front end of the inner needle cylinder and communicated with the inner cavity of the inner needle cylinder, and the inner cavity of the inner needle cylinder is internally provided with a detoxifying injection.
The connecting channel is of a tubular structure, the inner diameter of the connecting channel is smaller than the outer diameter of the inner needle cylinder, and the outer diameter of the inner needle cylinder is smaller than the inner diameter of the suction cup.
The invention discloses a preparation method of detoxification injection for snake bite emergency treatment, which comprises calcium oxalate hydrogel and a metal organic framework ZIF-8,
the synthesis steps of the calcium oxalate hydrogel are as follows:
a) Weigh 2.45g of a 2wt% sodium alginate solution (2 g dissolved in 100mL ddH2O);
b) 0.05g of calcium carbonate is added;
c) After stirring, adding 10mg of D- (+) -gluconic acid delta-lactone (GLD) and stirring for 10s;
the synthesis steps of the metal organic framework ZIF-8 are as follows:
(1) weighing 20mmol of Zn (NO 3) 2.6H 2O and 70mmol of 2-methylimidazole;
(2) dissolving the two solutions in 150mL of methanol (MeOH) respectively to form two clear solutions;
(3) mixing the two transparent solutions in the step (2);
(4) mixing and stirring at room temperature for 24 h to obtain white precipitate, centrifuging, washing with methanol for several times, and oven drying at 60 deg.C.
The invention has the beneficial effects that:
1. the calcium halloysite hydrogel can play a role in blocking through the characteristics of the hydrogel, so that blood can be rapidly prevented from diffusing to the far end, and then the metal organic framework solution can be adsorbed for a long time.
2. The injection is easy to store and carry, and is convenient for self-rescue after snake bite.
3. The injection can delay the toxin from entering the blood of the whole body for nearly 1 hour, strive for precious time for the patient to go to the hospital for treatment and save the life of the patient.

The invention discloses injection equipment for detoxifying injection for snake bite emergency treatment and a preparation method thereof, and the injection equipment comprises an absorption cup 1, an outer needle cylinder 2, an outer piston 3, an outer piston shaft 4, an outer piston handle 5, an inner needle cylinder 6, a needle head 7, an inner piston 8, an inner piston rod 9 and an inner piston handle 10, wherein the outer needle cylinder 2 is of a hollow cylindrical structure with an opening at the rear end, the front end of the outer needle cylinder is communicated with the rear end of the absorption cup 1 through a connecting channel 11, and the absorption cup 1 is of a hollow cylindrical structure with an opening at the front end; the outer piston 3 is matched with the inner cavity of the outer needle cylinder 2 to slide, the outer piston shaft 4 is of an annular structure, the front end of the outer piston shaft is connected with the rear wall of the outer piston 3, and the rear end of the outer piston shaft is connected with the outer piston handle 5;
a through hole for the inner needle cylinder 6 to pass through is axially formed in the outer piston 3, the inner piston 8 and the inner cavity of the inner needle cylinder 6 slide in a matched mode, the front end of the inner piston rod 9 is axially connected with the rear wall of the inner piston 8, and the rear end of the inner piston rod passes through the outer piston handle 5 and then is connected with the inner piston handle 10; the needle head 7 is arranged at the front end of the inner needle cylinder 6 and is communicated with the inner cavity of the inner needle cylinder 6, and a detoxifying injection is arranged in the inner cavity of the inner needle cylinder 6, so that a person bitten can independently operate; the injection is easy to store and carry, and is convenient for self-rescue after snake bite.
The connecting channel 11 is of a tubular structure, the inner diameter of the connecting channel is smaller than the outer diameter of the inner needle cylinder 6, and the outer diameter of the inner needle cylinder 6 is smaller than the inner diameter of the suction cup 1.
The injection method comprises the following steps: opening the injection external package, pressing the suction cup 1 at the front part on the bite part with force, pulling the outer piston handle 5 at the rear part to suck out the superficial toxin, and then pulling out the suction cup 1; then, the inner piston handle 10 is pressed to slide the inner needle cylinder 6 along the axial through hole of the outer piston 3 until the front end of the inner needle cylinder 6 is contacted with the front wall of the inner cavity of the outer needle cylinder 2, and the inner calcium oxalate hydrogel and the metal organic frame injection are injected into the bite part through the needle head 7 by continuously pressing the inner piston handle 10.
The invention discloses a preparation method of detoxification injection for snake bite emergency treatment, which comprises calcium oxalate hydrogel and a metal organic framework ZIF-8,
the synthesis steps of the calcium oxalate hydrogel are as follows:

35 KM mice (SJA laboratory animals Co., ltd., hunan, china) were selected for this experiment. The amount of the bungarus multicinctus or cobra injected into each mouse was 10 or 3 mg/kg. SA 0.1mL was injected and ZIF-8 was twice as injected as the venom.
Selecting mouse tail veins as an experimental model, setting 10 groups (at least 3 in each group) as (1) SA injection, (2) ZIF-8 injection, (3) SA and ZIF-8 injection as negative control groups, (4) golden trepang (venom) intravenous injection as positive control groups, (5) SA and ZIF-8 intravenous injection only at the upstream of snake venom for 1 min, (6) SA and ZIF-8 intravenous injection only at the upstream of venom for 1 min, (7) SA and ZIF-8 intravenous injection only at the upstream of snake venom for 5min, (8) SA and ZIF-8 intravenous injection only at the upstream of snake venom for 5min after snake venom injection, (9) SA and ZIF-8 intravenous injection only at the upstream of snake venom before injection, (10) SA and ZIF-8 intravenous injection at the upstream of snake venom before injection. After injection, the survival time of each mouse was observed and recorded.
The calcium oxalate hydrogel can block toxins in blood and slow down the speed of the toxins flowing to the heart; the metal organic framework can adsorb toxins in blood and reduce the amount of toxins flowing to the heart; the injection has longer quality guarantee period, and the preservation condition has no strict limit; the experimental result shows that the calcium sea oxalate hydrogel has good biocompatibility with the metal organic framework; the detoxification injection can promote the healing of wounds while detoxifying, blocking poison and detoxifying; the experimental result shows that the novel detoxification injection for the first aid of snake bite can delay the toxin from entering the whole body through blood for nearly 1 hour; the synthesis process of the calcium sea oxalate hydrogel and the metal organic framework is simple and convenient, and the yield is high; the detoxification injection has the functions of adsorbing and detoxifying one toxin, and is generally applicable to at least three common biological protein toxins (neurotoxin, blood toxin and toxin mixture).
