Abstract
本发明属于免疫荧光法检测技术领域,尤其是一种恙虫病东方体感染IgM抗体检测用免疫荧光法及其检测试剂盒,解决了现有技术中应用的巢氏PCR检测方法对实验仪器要求高、操作繁琐、耗时,扩增完成后需要通过电泳、显影等步骤对产物进行判别,不适合基层医院及现场快速检测的要求的问题,所述试剂盒由12个孔组成,每孔滴加2μL细胞悬液铺孔爬片,待孔内细胞悬浮液挥发后,用冷丙酮固定;将患者血清用PBS稀释后加入抗原片孔中,每片抗原片设置一孔做空白对照,同时设置阳性对照和阴性对照。本发明检测恙虫病东方体病原体的特异性及敏感性相对较高,检测准确率高、检测效率高,检测成本低。

技术领域
本发明涉及免疫荧光法检测技术领域,尤其涉及一种恙虫病东方体感染IgM抗体检测用免疫荧光法及其检测试剂盒。
背景技术
恙虫病东方体以前称为恙虫病立克次体,是恙虫病的病原体,属自然疫源性疾病。恙虫病是由恙虫病东方体(Orient ia tsutsug amushi,Ot)通过恙螨幼虫叮咬而传播的一种自然疫源性疾病。恙虫病的疫区主要分布在亚太地区的热带和亚热带国家。在我国主要分布在东南沿海的省市和地区,我国东北和华北等也报道有该病的流行,疫区有进一步扩大的趋势。
恙虫病为自然疫源性疾病,主要流行于啮齿类动物。野鼠和家鼠感染后多无症状,但病原体可在其体内长期保留,为该病的主要传染源。恙螨既是传播媒介又是储存宿主,立克次体由恙螨幼虫叮咬而侵入人体。人被感染了恙虫病东方体的恙螨幼虫叮咬后,立克次体由叮咬部位侵入人体,在局部繁殖后入血,随血流到达全身各组织器官,主要在小血管内皮细胞中增殖,引起血管栓塞。立克次体死亡后释放的毒素样物质是主要致病因素,可引起发热、头痛等全身中毒症状及各组织脏器血管炎。恙虫病的潜伏期为10~14天,多突然起病,发热可达39℃以上。临床表现除高热外尚有皮疹、皮肤焦痂与溃疡、淋巴结肿大、肝脾肿大等,危重病例可呈严重的多器官损害,如肝炎、肺炎、心脏病变、肾功能衰竭、循环衰竭与出血现象等,少数病例还可发生DIC和脑炎。因此,建立简洁高效、同时惠及患者价格低廉的恙虫病东方体感染IgM抗体快速检测方法及其检测试剂盒来协助临床及患者的快速诊断至关重要。目前,国内外广泛应用的巢氏PCR检测方法则因实验仪器要求高、操作繁琐、耗时,扩增完成后需要通过电泳、显影等步骤对产物进行判别,不适合基层医院及现场快速检测的要求。因此,开发一款能够适用于临床疾控快速筛查要求,且灵敏性高、特异性强、价格低廉的恙虫病东方体感染IgM抗体快速检测方法及其检测试剂盒对于疾病感染的控制和临床诊治显得尤其重要。基于上述陈述,本发明提出了一种恙虫病东方体感染IgM抗体检测用免疫荧光法及其检测试剂盒。
发明内容
本发明的目的是为了解决现有技术中应用的巢氏PCR检测方法对实验仪器要求高、操作繁琐、耗时,扩增完成后需要通过电泳、显影等步骤对产物进行判别,不适合基层医院及现场快速检测的要求的问题,而提出的一种恙虫病东方体感染IgM抗体检测用免疫荧光法及其检测试剂盒。
一种恙虫病东方体感染IgM抗体检测用免疫荧光法,包括以下步骤:
(1)消化已培养好的感染恙虫病东方体菌种的细胞;
(2)完成步骤(1)中消化操作后,用含3%血清的DMEM液体吹散细胞;
(3)收集步骤(2)中完成吹散处理的细胞于ep管中进行离心,设置离心转速为300g,离心时间为15min,收集离心后的沉淀;
(4)用无菌PBS清洗步骤(3)中收集的沉淀2-3次;
(5)将步骤(4)清洗完成的沉淀在显微镜下边观察边用PBS调节细胞浓度至其可以在载玻片孔内铺成单层,即得细胞悬浮液;
(6)将步骤(5)中的细胞悬浮液滴加入12孔载玻片中,每孔滴加2μL细胞悬液铺孔爬片;
(7)待步骤(6)中孔内细胞悬浮液挥发后,用冷丙酮(-20℃-4℃)固定5-10min,晾干冻存玻片备用;
(8)将需要检测的患者血清用PBS于无菌状态下以1:80的比例进行稀释;
(9)将步骤(8)中稀释好的血清加入抗原片孔中,每孔20μL,每片抗原片设置一孔加20μL PBS做空白对照,同时设置阳性对照和阴性对照;
(10)将步骤(9)中的抗原片放入湿盒,置于37℃温箱内孵育45min;
(11)完成步骤(10)中的孵育反应后,将抗原片置于玻片染色缸中,加入PBS浸润抗原片,将玻片染色缸置于微量振荡器上震荡5min,重复3次,以清洗抗原片,清洗完成后室温吹干;
(12)以步骤(8)中相同的比例将二抗进行稀释,然后加入到经步骤(11)处理后的抗原片孔中,同样放置在湿盒中,然后置于37℃温箱孵育45min;
(13)完成步骤(12)中的孵育反应后,将抗原片置于玻片染色缸中,加入PBS浸润抗原片,将玻片染色缸置于微量振荡器上震荡5min,重复3次,以清洗抗原片,清洗完成后室温吹干;
(14)滴加甘油封片剂于步骤(13)清洗吹干后的抗原片上,盖上盖玻片封片,避光保存,荧光显微镜下观察,即可进行患者检测。
优选的,所述步骤(1)的具体操作如下:将恙虫病东方体菌种感染至L929细胞中,感染3天后,每天对细胞进行涂片,并进行吉姆萨染色,在显微镜下观察细胞质内菌体数量,直到胞质内充满菌体即可将带有恙虫病东方体感染的贴壁细胞全部刮下备用;通过前期预实验发现,恙虫病东方体在小鼠成纤维细胞(L929细胞)中生长状态良好,恙虫病东方体为胞内寄生菌,生长在其所感染的L929细胞的胞质内,吉姆萨染色呈紫色短杆状;用PBS洗带有恙虫病东方体感染的贴壁细胞2-3次,并用0.25%的胰蛋白酶消化细胞。
优选的,所述步骤(9)中的阳性对照为已经确诊恙虫病东方体患者血清滴加的孔;阴性对照为滴加正常人血清的孔。
本发明还提出了一种恙虫病东方体感染IgM抗体检测试剂盒,其包括:细胞悬浮液、患者血清和二抗;所述试剂盒由12个孔组成,每孔滴加2μL细胞悬液铺孔爬片,待孔内细胞悬浮液挥发后,用冷丙酮固定;将患者血清用PBS稀释后加入抗原片孔中,每孔20μL,每片抗原片设置一孔加20μL PBS做空白对照,同时设置阳性对照和阴性对照;将抗原片放入湿盒孵育;完成孵育,清洗、吹干;将二抗稀释后加入抗原片孔中,同样放置在湿盒中孵育;完成孵育,清洗、吹干,滴加甘油封片剂盖上盖玻片封片,即得检测试剂盒。
本发明提出的一种恙虫病东方体感染IgM抗体检测用免疫荧光法及其检测试剂盒,具有以下有益效果:
1、本发明检测恙虫病东方体病原体的特异性及敏感性相对较高,发明人进行假阳性率及假阴性率的检测(即采用多例健康人血清和确诊患者血清分别用该方法进行测定,荧光显微镜下观察),假阳性率小于0.01,同时假阴性率可忽略不计,检测准确率高、检测效率高,检测成本低。
2、现有技术方案中,还未有人利用抗原片方法来检测该种病原体,本方案不仅提高了临床检验的特异性及灵敏性,并且充分考虑到患者的经济水平,检测方法操作简便,成本低。

具体实施方式
下面结合具体实施例对本发明作进一步解说。
实施例一
本发明提出的一种恙虫病东方体感染IgM抗体检测用免疫荧光法,包括以下步骤:
(1)将恙虫病东方体菌种感染至L929细胞中,感染3天后,每天对细胞进行涂片,并进行吉姆萨染色,在显微镜下观察细胞质内菌体数量,直到胞质内充满菌体即可将带有恙虫病东方体感染的贴壁细胞全部刮下备用;通过前期预实验发现,恙虫病东方体在小鼠成纤维细胞(L929细胞)中生长状态良好,恙虫病东方体为胞内寄生菌,生长在其所感染的L929细胞的胞质内,吉姆萨染色呈紫色短杆状;用PBS洗带有恙虫病东方体感染的贴壁细胞2次,并用0.25%的胰蛋白酶消化细胞;
(2)完成步骤(1)中消化操作后,用含3%血清的DMEM液体吹散细胞;
(3)收集步骤(2)中完成吹散处理的细胞于ep管中进行离心,设置离心转速为300g,离心时间为15min,收集离心后的沉淀;
(4)用无菌PBS清洗步骤(3)中收集的沉淀2次;
(5)将步骤(4)清洗完成的沉淀在显微镜下边观察边用PBS调节细胞浓度至其可以在载玻片孔内铺成单层,即得细胞悬浮液;
(6)将步骤(5)中的细胞悬浮液滴加入12孔载玻片中,每孔滴加2μL细胞悬液铺孔爬片;
(7)待步骤(6)中孔内细胞悬浮液挥发后,用冷丙酮(-20℃)固定5min,晾干冻存玻片备用;
(8)将需要检测的患者血清用PBS于无菌状态下以1:80的比例进行稀释;
(9)将步骤(8)中稀释好的血清加入抗原片孔中,每孔20μL,每片抗原片设置一孔加20μL PBS做空白对照,同时设置阳性对照和阴性对照,阳性对照为已经确诊恙虫病东方体患者血清滴加的孔;阴性对照为滴加正常人血清的孔;
(10)将步骤(9)中的抗原片放入湿盒,置于37℃温箱内孵育45min;
(11)完成步骤(10)中的孵育反应后,将抗原片置于玻片染色缸中,加入PBS浸润抗原片,将玻片染色缸置于微量振荡器上震荡5min,重复3次,以清洗抗原片,清洗完成后室温吹干;
(12)以步骤(8)中相同的比例将二抗进行稀释,然后加入到经步骤(11)处理后的抗原片孔中,同样放置在湿盒中,然后置于37℃温箱孵育45min;
(13)完成步骤(12)中的孵育反应后,将抗原片置于玻片染色缸中,加入PBS浸润抗原片,将玻片染色缸置于微量振荡器上震荡5min,重复3次,以清洗抗原片,清洗完成后室温吹干;
(14)滴加甘油封片剂于步骤(13)清洗吹干后的抗原片上,盖上盖玻片封片,避光保存,荧光显微镜下观察,即可进行患者检测。
实施例二
本发明提出的一种恙虫病东方体感染IgM抗体检测用免疫荧光法,包括以下步骤:

(11)完成步骤(10)中的孵育反应后,将抗原片置于玻片染色缸中,加入PBS浸润抗原片,将玻片染色缸置于微量振荡器上震荡5min,重复3次,以清洗抗原片,清洗完成后室温吹干;
(12)以步骤(8)中相同的比例将二抗进行稀释,然后加入到经步骤(11)处理后的抗原片孔中,同样放置在湿盒中,然后置于37℃温箱孵育45min;
(13)完成步骤(12)中的孵育反应后,将抗原片置于玻片染色缸中,加入PBS浸润抗原片,将玻片染色缸置于微量振荡器上震荡5min,重复3次,以清洗抗原片,清洗完成后室温吹干;
(14)滴加甘油封片剂于步骤(13)清洗吹干后的抗原片上,盖上盖玻片封片,避光保存,荧光显微镜下观察,即可进行患者检测。
以上所述,仅为本发明较佳的具体实施方式,但本发明的保护范围并不局限于此,任何熟悉本技术领域的技术人员在本发明揭露的技术范围内,根据本发明的技术方案及其发明构思加以等同替换或改变,都应涵盖在本发明的保护范围之内。
Claims (4)
1.一种恙虫病东方体感染IgM抗体检测用免疫荧光法,其特征在于,包括以下步骤:
(1)消化已培养好的感染恙虫病东方体菌种的细胞;
(2)完成步骤(1)中消化操作后,用含3%血清的DMEM液体吹散细胞;
(3)收集步骤(2)中完成吹散处理的细胞于ep管中进行离心,设置离心转速为300g,离心时间为15min,收集离心后的沉淀;
(4)用无菌PBS清洗步骤(3)中收集的沉淀2-3次;
(5)将步骤(4)清洗完成的沉淀在显微镜下边观察边用PBS调节细胞浓度至其可以在载玻片孔内铺成单层,即得细胞悬浮液;
(6)将步骤(5)中的细胞悬浮液滴加入12孔载玻片中,每孔滴加2μL细胞悬液铺孔爬片;
(7)待步骤(6)中孔内细胞悬浮液挥发后,用冷丙酮固定5-10min,晾干冻存玻片备用;
(8)将需要检测的患者血清用PBS于无菌状态下以1:80的比例进行稀释;
(9)将步骤(8)中稀释好的血清加入抗原片孔中,每孔20μL,每片抗原片设置一孔加20μL PBS做空白对照,同时设置阳性对照和阴性对照;
(10)将步骤(9)中的抗原片放入湿盒,置于37℃温箱内孵育45min;
(11)完成步骤(10)中的孵育反应后,将抗原片置于玻片染色缸中,加入PBS浸润抗原片,将玻片染色缸置于微量振荡器上震荡5min,重复3次,以清洗抗原片,清洗完成后室温吹干;
(12)以步骤(8)中相同的比例将二抗进行稀释,然后加入到经步骤(11)处理后的抗原片孔中,同样放置在湿盒中,然后置于37℃温箱孵育45min;
(13)完成步骤(12)中的孵育反应后,将抗原片置于玻片染色缸中,加入PBS浸润抗原片,将玻片染色缸置于微量振荡器上震荡5min,重复3次,以清洗抗原片,清洗完成后室温吹干;
(14)滴加甘油封片剂于步骤(13)清洗吹干后的抗原片上,盖上盖玻片封片,避光保存,荧光显微镜下观察,即可进行患者检测。
2.根据权利要求1所述的一种恙虫病东方体感染IgM抗体检测用免疫荧光法,其特征在于,所述步骤(1)的具体操作如下:将恙虫病东方体菌种感染至L929细胞中,感染3天后,每天对细胞进行涂片,并进行吉姆萨染色,在显微镜下观察细胞质内菌体数量,直到胞质内充满菌体即可将带有恙虫病东方体感染的贴壁细胞全部刮下,用PBS洗带有恙虫病东方体感染的贴壁细胞2-3次,并用0.25%的胰蛋白酶消化细胞。
3.根据权利要求1所述的一种恙虫病东方体感染IgM抗体检测用免疫荧光法,其特征在于,所述步骤(9)中的阳性对照为已经确诊恙虫病东方体患者血清滴加的孔;阴性对照为滴加正常人血清的孔。
4.根据权利要求1-3任一项所述的一种恙虫病东方体感染IgM抗体检测试剂盒,其特征在于,包括:细胞悬浮液、患者血清和二抗;所述试剂盒由12个孔组成,每孔滴加2μL细胞悬液铺孔爬片,待孔内细胞悬浮液挥发后,用冷丙酮固定;将患者血清用PBS稀释后加入抗原片孔中,每孔20μL,每片抗原片设置一孔加20μL PBS做空白对照,同时设置阳性对照和阴性对照;将抗原片放入湿盒孵育;完成孵育,清洗、吹干;将二抗稀释后加入抗原片孔中,同样放置在湿盒中孵育;完成孵育,清洗、吹干,滴加甘油封片剂盖上盖玻片封片,即得检测试剂盒。
