Description

Steamed dried orange peel and preparation method thereof
Technical Field
The invention relates to the technical field of traditional Chinese medicines, in particular to steamed tangerine peel and a preparation method thereof.
Background
The pericarpium Citri Tangerinae is derived from dried mature pericarp of Citrus reticulata Blanco of Rutaceae and its cultivar. The medicinal materials are divided into pericarpium Citri Tangerinae and pericarpium Citri Tangerinae. It is warm in nature, bitter in taste and pungent in flavor, such as spleen and lung meridian. Has the functions of regulating qi, strengthening spleen, eliminating dampness and phlegm. Can be used for treating anorexia, vomiting, diarrhea, cough, excessive phlegm, etc. The main active ingredients in the dried orange peel are flavonoids and volatile oil, and in recent epidemic disease investigation reports, the flavonoids can reduce blood pressure and blood fat and prevent and improve cardiovascular and cerebrovascular diseases, and the volatile oil active ingredients can remove free radicals in vivo and play roles in resisting oxidation, aging and the like. The water extract or volatile oil can block or relieve tracheal smooth muscle contraction caused by acetylcholine chloride, and the volatile oil has protective effect on drug-induced asthma of guinea pig. The dried orange peel alcohol extract can completely resist spastic contraction of bronchial smooth muscle caused by histamine, is one of people with high asthma-relieving potency, and has remarkable cough-relieving effect.
The prior art discloses a processing method of traditional Chinese medicine dried orange peel, and provides a method of moistening and steaming to improve the pungency and dryness of the dried orange peel. But the method obviously reduces the content of volatile oil active ingredients in the dried orange peel, thereby affecting the efficacy of the dried orange peel; the prior art discloses a method for processing pericarpium citri reticulatae with high efficiency and quick fragrance, which adopts the methods of wine stewing, steaming and long-term storage to remove the pungent taste and improve the mellow fragrance of the pericarpium citri reticulatae. The method also removes a large amount of volatile active ingredients in pericarpium Citri Tangerinae, and has long shelf life and difficulty in controlling product quality. At present, the content of active ingredients of the dried orange peel manufactured on the market only reaches the standard specified in Chinese pharmacopoeia, and the efficacy of dried orange peel manufacturing is far from being fully exerted.
The quality control standard of dried orange peel in 'Chinese pharmacopoeia' of 2015 edition stipulates the content determination of hesperidin, and meanwhile, researches show that flavanones are one of the main categories of flavone contained in dried orange peel, including narirutin, hesperidin and the like. Hesperidin has a complex structure and is difficult to prepare in a full-synthesis manner. Generally, narirutin with a similar structure to the narirutin is extracted from plants and used as a starting raw material, and then the hesperidin is prepared by conversion through a chemical synthesis method, wherein the process conditions are harsh, and the post-treatment of the product is troublesome. The hesperidin biotransformation method can successfully transform the hesperidin into the neohesperidin under the combined action of glucosyltransferase and rhamnose-7-O-glucoside, and the requirements on process conditions are very strict. Therefore, the invention aims to find an enzyme existing in the plant as a catalyst, which is used for catalyzing the conversion of the dried orange peel to obtain the hesperidin and promoting the dissolution of the volatile oil from the viewpoint.

Preparation of control solutions: taking appropriate amount of hesperidin control, precisely weighing, and adding methanol to obtain solution containing 0.4mg per 1 ml.
Measuring by high performance liquid chromatography (general rule 0512).
Chromatographic conditions and system applicability test: octadecylsilane chemically bonded silica is used as a filling agent; methanol-acetic acid-water (35: 4: 61) is used as a mobile phase; the detection wavelength was 283 nm. The number of theoretical plates is not less than 2000 calculated according to hesperidin peak.
The determination method comprises the following steps: precisely sucking 5 μ l of each of the reference solution and the sample solution, injecting into liquid chromatograph, and measuring.
The test analysis was repeated 3 times for each sample, and the average was taken as the final data, and the results are shown in Table 1.
(2) Determination of the type and content of volatile oil
Pulverizing pericarpium Citri Tangerinae, and sieving to obtain pericarpium Citri Tangerinae powder. Accurately weighing 50g of tangerine peel powder in a round-bottom flask, adding 100mL of petroleum ether, heating and refluxing for extraction twice, combining extracting solutions, cooling, filtering, concentrating filtrate under reduced pressure to obtain a paste, mixing the extract with anhydrous yellow wine in a volume ratio of 1: 10, stirring, and freezing at-20 deg.C for 4 h; filtering with filter paper, and concentrating the filtrate under reduced pressure to obtain pericarpium Citri Tangerinae volatile extract; the extract was analyzed by gas chromatography-mass spectrometry. Chromatographic conditions are as follows: HP-INNOWAX capillary chromatography column (30m × 250 μm × 0.25 μm) with carrier gas He at flow rate of 1.0 mL/min; the programmed heating mode is adopted, the initial temperature is 50 ℃, the temperature is kept for 5min, the temperature is increased to 160 ℃ at the speed of 2 ℃/min, the temperature is kept for 5min, and then the temperature is increased to 250 ℃ at the speed of 12 ℃/min, and the temperature is kept for 5 min. The injection port temperature is 250 ℃, the split ratio is 40: 1. mass spectrum conditions: signals are collected in a full scanning mode, an ionization source is EI, and ionization energy is 70 eV; the interface temperature is 280 ℃, the ion source temperature is 200 ℃, the quadrupole rod temperature is 150 ℃, and the scanning mass range is 35 amu-500 amu.
The test analysis was repeated 3 times for each sample and averaged to give the final data.
The results before storage are shown in Table 1.

Figure BDA0003014062480000101
Note: the relative deviation of the data in table 1 is within 2.5%.
As can be seen from the data in Table 1, compared with the existing method for processing steamed tangerine peel, the steamed tangerine peel obtained by the method of the invention has the advantages that the types of volatile oil, hesperidin and the content of volatile oil are greatly improved, the types and the content of active ingredients are more, and the effect of steaming tangerine peel is more favorably exerted. Compared with comparative examples 2-3, the content of effective components and the storage time of the steamed orange peel obtained by the method are far higher than those of the prior art. Compared with the comparative example 1, the components in the plant extracting solution act together, so that the content of hesperidin in the dried orange peel and the type and content of volatile oil are improved, and the storage time of the dried orange peel is greatly prolonged. Compared with the comparative example 4, the pH is adjusted to a proper range, the conversion generation of the hesperidin is facilitated, but the influence on the type and content of the volatile oil and the storage time of the dried orange peel is little.
Data of examples 1 to 6 and comparative example 5 after half a year storage are shown in Table 2.
TABLE 2
Figure BDA0003014062480000102
