Abstract

Background
China is the largest tobacco producing country and consuming country in the world, wherein the yield of tobacco leaves in Yunnan province accounts for about 40 percent of the whole country. Due to the high standard of tobacco leaves for flue-cured tobacco, about 100 ten thousand tons of waste fresh tobacco raw materials are generated in tobacco fields after the tobacco leaves for flue-cured tobacco are harvested every year. The abandoned fresh tobacco raw materials mainly refer to top leaves and bottom leaves generated in the flue-cured tobacco planting process, fireworks, tobacco stems and tobacco stems, and fresh tobacco leaves which need to be treated in the field and have poor development, rigid structure and low industrial availability. If the natural fermentation time is too long, the method occupies a factory and influences the capital turnover of a tobacco plant. Therefore, by utilizing proper technical means such as extraction and separation, endogenous total enzymes of waste fresh tobacco raw materials are developed, and the method has good market prospect for improving the quality of tobacco leaves and promoting alcoholization.
The published documents on the development and application of endogenous total enzymes of waste fresh tobacco raw materials are very rare, and part of related patents are basically limited to single biological enzymes and traditional complex processes. For example, in the separation technology of tobacco polyphenol oxidase disclosed in chinese patent CN102477413A, acetone precipitation and ammonium sulfate precipitation are used to prepare polyphenol oxidase, acetone has high toxicity and is not suitable for use in cigarettes, and ammonium sulfate requires a salting-out process which is too complicated and is not suitable for industrial production. A tobacco additive from fresh tobacco leaf and its preparation method and application are disclosed in Chinese patent CN105795505A, which is prepared by taking out main vein from mature middle tobacco leaf, adding phosphoric acid buffer solution and preparing with hollow fiber column for reducing cigarette tar. However, the mature middle tobacco leaves are tobacco leaves for flue-cured tobacco, are high in price, complicated in operation of removing the main veins, complex in product components, contain a plurality of pigments and the like, and external impurities are introduced into the phosphoric acid buffer solution, so that the method belongs to the application direction of harm reduction and tar reduction. The technical scheme adopted by the patents is not suitable for the development of endogenous total enzymes of waste fresh tobacco raw materials. At present, no report is found about a technology which integrates a low-temperature extraction and separation process by taking waste fresh tobacco as a raw material, develops tobacco endogenous total enzymes and is applied to the aspects of quality improvement and alcoholization of tobacco leaves.

The invention aims to fill the blank of the prior art and provides a method for preparing endogenous total enzyme by taking waste fresh tobacco as a raw material and integrating a low-temperature extraction and separation process. 4. The method for preparing endogenous total enzymes from fresh tobacco according to claim 1 or 2, wherein the freezing time for freezing the wet enzymes at a temperature of 0 ℃ or lower in the step (4) is 2 to 24 hours.
5. The application of the endogenous total enzyme of any one of claims 1 to 4, which is to apply the endogenous total enzyme as a tobacco leaf quality-improving agent to the feeding process of the cigarette tobacco-making line, wherein the addition amount is 0.1 to 2.0 percent of the mass of tobacco leaves.
6. The application of the endogenous total enzyme of any one of claims 1 to 4, which is to apply the endogenous total enzyme as a tobacco mellowing agent to a second moistening link or an export feeding process of a threshing and redrying line, wherein the addition amount is 0.1 to 2.0 percent of the mass of tobacco leaves.
