Abstract
本发明提供了一种利用MK‑5108阻断AURKA来减轻肾纤维化的方法,其特征在于,包括肾脏组织病理学分析、免疫组织化学分析、细胞培养研究、细胞增殖和细胞毒性测定、实时定量PCR检测、蛋白质测定、统计分析。本发明证明了AURKA在CKD患者和UUO小鼠的纤维化肾脏中高表达,表明AURKA在病理条件下起一定作用。更重要的是,用MK‑5108阻断AURKA的药理作用可抑制与肾纤维发生有关的若干事件,并显著减轻肾纤维化。本发明还证实了抑制AURKA可以通过抑制间质成纤维细胞的增殖和活化以及肾小管细胞的部分表型转变来延迟CKD肾纤维化的进展。在此基础上,阻断AURKA将是治疗肾纤维化的一种有前途的治疗策略。
Description
技术领域
本发明涉及生物领域,具体涉及一种利用MK-5108阻断AURKA来减轻肾纤维化的方法。
背景技术
慢性肾脏病(CKD)是全球增长最快的死亡原因之一,预计到2040年将成为第五大死亡原因。当病情发展为终末期肾病(ESRD)时,患者通常需要通过透析或肾移植来替代肾功能。由于肾纤维化是CKD演变为ESRD的最终共同途径,因此研究和鉴定介导肾纤维化进展的关键机制对于开发新的CKD策略至关重要。肾纤维化通常特征在于细胞外基质(ECM)成分的过度产生和沉积。
一般认为,受损的肾小管细胞经过适应不良性修复后,会转变成分泌型,并产生多种促纤维化因子,这些因子参与活跃的上皮-间充质沟通(EMC)并促进间质成纤维细胞的活化。
过去十年的研究表明,活化的成肌纤维细胞是疤痕组织形成中ECM的主要来源,是肾纤维化的明确影响者和驱动者,而常驻成纤维细胞是其重要来源之一;极光激酶-A(AURKA)是丝氨酸/苏氨酸激酶,是进入有丝分裂并通过有丝分裂所必需的。已经证明其活性突变和过表达与高度增殖和恶性癌症有关。MK-5108是一种功能强大且高度选择性的AURKA抑制剂,已在临床前和临床研究中显示出有效的活性。然而,尚未研究AURKA在肾纤维发生中的作用。
AURKA被认为是一种癌基因,它是在各种肿瘤中扩增的已知热点。因此,抑制AURKA是抗癌治疗的有效策略,目前正在进行的临床试验中评估几种此类药物。
发明内容
在体内,AURKA在CKD患者的纤维化肾脏和单侧输尿管梗阻(UUO)小鼠模型中高表达。MK-5108可能通过抑制成纤维细胞的增殖和活化以及肾小管细胞的表型转变,显著减轻了UUO诱导的肾纤维化。同时,梗塞性肾脏中炎性因子的增加也被抑制。
在体外,MK-5108预处理抑制转化生长因子-β1(TGF-β1)诱导的肾细胞的促纤维化反应和部分表型改变。
两者合计,MK-5108对AURKA的抑制作用可以抑制与纤维化相关的多个事件,并显著减轻肾纤维化。靶向AURKA的MK-5108有望成为对抗肾纤维化并延缓CKD进展的治疗策略。
为实现上述目的,本发明提供的具体技术方案如下:
一种利用MK-5108阻断AURKA来减轻肾纤维化的方法,其特征在于,包括肾脏组织病理学分析、免疫组织化学分析、细胞培养研究、细胞增殖和细胞毒性测定、实时定量PCR提取、蛋白质测定、统计分析。
所述肾脏组织病理学分析具体包括如下步骤:
步骤1.1:将肾脏固定在4%多聚甲醛(PFA)中过夜,脱水并包埋在石蜡中,然后横向切成3μm切片;
步骤1.2:脱石蜡后,肾脏切片用Masson三色染色和Sirius红染色染色。
所述免疫组织化学分析具体包括如下步骤:
步骤2.1:将肾脏固定在4%多聚甲醛中,并包埋在石蜡中;
步骤2.2:将切片(3微米厚)安装在载玻片上;
步骤2.3:将每个样品的载玻片脱蜡,并使用二甲苯和梯度乙醇按标准方法再水化;
步骤2.4:将这些切片在3%过氧化氢中孵育20分钟,在改良的柠檬酸盐抗原提取溶液(Beyotime,中国上海,P0086)中煮沸1分钟,然后冷却至室温;
步骤2.5:将玻片与抗AURKA(1:250),PDGFR-β(1:200),p-PDGFR-β(1:250),α-SMA(1:10000)和单核细胞趋化蛋白1(MCP1)(1:250)的一抗在4℃孵育过夜;
步骤2.6:用磷酸盐缓冲盐水(PBS)洗涤后,应用生物素偶联的二抗,并使用DakoREALTMEnVisionTM检测系统K5007(丹麦)可视化信号。
所述细胞培养研究具体包括如下步骤:
步骤3.1:将正常的大鼠肾成纤维细胞(NRK-49F)和小鼠肾近端肾小管上皮细胞(mPTC)在添加有10%FBS的细胞培养基中生长,并在37℃,5%CO2的培养箱中培养;
步骤3.2:使用0.25%胰蛋白酶-0.02%EDTA(Invitrogen)以60-80%汇合将细胞传代培养;
步骤3.3:让细胞在无血清培养基中饥饿6-8h,然后用MK-5108(200nM或400nM)预处理1h;
步骤3.4:然后将重组人转化生长因子-β1(TGF-β1)(15ng/ml)添加到培养基中;
步骤3.5:在重组人TGF-β1(15ng/ml)刺激6-8小时后,用AURKA质粒转染的细胞;
步骤3.6:24小时后收集细胞进行qRT-PCR分析,或48小时后收集细胞进行WB分析。
所述细胞增殖和细胞毒性测定具体步骤如下:
步骤4.1:细胞活力通过CCK-8测定试剂盒(MedChemExpress,中国)确定;
步骤4.2:将NRK-49F和mPTCs以104-105个细胞/孔的密度接种在100μL培养基中的孔板上,并用各种浓度的MK-5108(0-1000nM)处理;
步骤4.3:温育24小时后,将每孔CCK-8试剂10μL加入培养基中,温育1-4小时;
步骤4.4:使用酶标仪在450nm处检测吸光度。
所述实时定量PCR具体包括如下步骤:
步骤5.1:使用RNA isoPlus试剂(TaKaRa生物技术有限公司,中国大连)根据制造商的方案从组织或细胞中提取总RNA;
步骤5.2:使用PrimeScriptTMRT Master Mix(Takara,草津,日本,RR036)对cDNA进行反转录;
步骤5.3:通过SYBR Green PCR Master Mix(Vazyme,中国南京)和ABI7500实时PCR检测系统(Applied Biosystems,加利福尼亚州福斯特市)进行实时PCR扩增;
其中,温度循环条件为95℃10分钟,95℃40循环15s,60℃1分钟;
其中,将mRNA的相对表达水平相对于GAPDH标准化,并使用2-ΔΔCt方法计算;
步骤5.4:所需的引物由Primer 3.0软件(http://Frodo.wi.mit.edu)设计,其引物序列列于表2。

所述蛋白质测定包括具体步骤如下:
步骤6.1:使用含有1x蛋白酶抑制剂(Roche)和1x磷酸酶抑制剂(Roche)的RIPA缓冲液匀浆组织或细胞裂解物;
步骤6.2:然后将样品以5000rpm离心30分钟;
步骤6.3:使用BCA蛋白质测定试剂盒(Thermofisher,Waltham,MA,USA,23225)测定蛋白质浓度;
步骤6.4:等效样品用于针对FN1(1:1000),胶原III(1:1000),PDGFRβ(1:1000),p-PDGFRβ(1:1000),AURKA(1:1000)和GAPDH(1:2000),然后添加HRP标记的二抗(1:5000);
步骤6.5:免疫反应条带通过Amersham Biosciences ECL检测系统(Amersham,Little Chalfont,英国)可视化。
步骤6.6:通过测量条带的强度进行光密度分析,并使用Quantity One软件(Bio-Rad,Hercules,CA)将其归一化为相应的GAPDH条带。
所述统计分析包括如下内容:
步骤7.1:所有定量值均表示为平均值±平均值标准误差(SEM);
步骤7.2:使用GraphPad Prism(6.0版,GraphPad Software,La Jolla,CA,美国),通过两尾Student t检验分析两组之间的统计学差异;
步骤7.3:单因素方差分析用于多个组之间的比较;P<0.05在统计学上被认为是有统计学差异的。
本发明提供的一种利用MK-5108阻断AURKA来减轻肾纤维化的方法,可显著抑制间质成纤维细胞的增殖和活化,肾小管细胞的部分表型转化以及阻塞性肾脏中的炎症浸润。
本发明证明了AURKA在CKD患者和UUO小鼠的纤维化肾脏中高表达,表明AURKA在病理条件下起一定作用。更重要的是,用MK-5108阻断AURKA的药理作用可抑制与肾纤维发生有关的若干事件,并显著减轻肾纤维化。本发明还证实了抑制AURKA可以通过抑制间质成纤维细胞的增殖和活化以及肾小管细胞的部分表型转变来延迟CKD肾纤维化的进展。在此基础上,阻断AURKA将是治疗肾纤维化的一种有前途的治疗策略。
鉴于MK-5108具有很强的临床前和临床效果,应进一步研究以阐明AURKA在不同肾脏疾病动物模型中的作用和机制,并评估靶向抑制AURKA在延缓CKD到ESRD进程中的功效。

实施例
试剂和抗体来源:
Dulbecco改良的Eagle培养基或营养混合物F12,即DMEM/F-12,胎牛血清FBS购自GIBCO。
抗纤连蛋白(FN1),α平滑肌肌动蛋白(α-SMA)和AURKA的抗体购自Abcam(Cambridge,MA)。
Anti-Collagen III和GAPDH购自Bioss(中国北京)。
抗血小板衍生的生长因子受体β(PDGFRβ),磷酸化PDGFRβ(p-PDGFRβ)和成纤维细胞特异性蛋白1(FSP1)抗体来自Abclonal(武汉,中国)。
MK-5108购自Selleck(中国上海)。
参与试验的患者来源:
肾活检标本取自在南京医科大学附属儿童医院肾内科接受诊断评估的CKD患者。
根据可用于组织学切片的石蜡包埋的组织样本中至少有十个肾小球的标准收集肾脏活检样本。
总共招募了10名受试者(年龄范围为1-14岁)并经病理诊断为硬化性肾小球肾炎,ANCA相关血管炎(AAV),过敏性紫癜性肾炎(HSPN),肾小管间质病变(TIL)或局灶性节段性肾小球硬化症(FSGS)。患者信息列于表1。
表1.CKD患者的基本信息和诊断
将来自患有良性肾脏肿瘤并接受部分肾切除术的患者的非肿瘤肾脏组织用作正常对照。
南京医科大学附属儿童医院人类实验委员会批准使用患者的活检样本和肾切除组织。
每位患者均提供书面知情同意书。
参与试验的动物来源:
由南京医科大学实验动物中心提供成年雄性C57BL/6J小鼠(20-25g),并在温度和湿度受控的房间中保持12:12h的黑暗-黑暗周期,饲喂标准啮齿动物饮食并允许免费获得饮用水。
为了阐明AURKA在CKD中的作用,构建单侧输尿管闭塞(UUO)动物模型。
将成年雄性小鼠随机分为以下各组(每组N=10):假手术组,UUO组和UUO+MK-5108组。
在UUO实验中,小鼠在麻醉后,通过正中腹侧切口用4-0丝线将左输尿管结扎在输尿管骨盆连接处。
将MK-5108溶解在盐水中至2μg/μl的工作浓度,然后以20mg/kg/d腹膜内注射到小鼠中。
在UUO手术后3至7天注射MK-5108。UUO 7天后,对所有组的小鼠实施安乐死,并收集肾脏组织用于进一步分析。
所有动物程序均经南京医科大学动物保护与使用委员会批准。
对实验对象进行本发明所述如下方法的试验:
一种利用MK-5108阻断AURKA来减轻肾纤维化的方法,其特征在于,包括肾脏组织病理学分析、免疫组织化学分析、细胞培养研究、细胞增殖和细胞毒性测定、实时定量PCR提取、蛋白质测定、统计分析。
所述肾脏组织病理学分析具体包括如下步骤:
步骤1.1:将肾脏固定在4%多聚甲醛(PFA)中过夜,脱水并包埋在石蜡中,然后横向切成3μm切片;
步骤1.2:脱石蜡后,肾脏切片用Masson三色染色和Sirius红染色染色。
所述免疫组织化学分析具体包括如下步骤:
步骤2.1:将肾脏固定在4%多聚甲醛中,并包埋在石蜡中;
步骤2.2:将切片(3微米厚)安装在载玻片上;
步骤2.3:将每个样品的载玻片脱蜡,并使用二甲苯和梯度乙醇按标准方法再水化;
步骤2.4:将这些切片在3%过氧化氢中孵育20分钟,在改良的柠檬酸盐抗原提取溶液(Beyotime,中国上海,P0086)中煮沸1分钟,然后冷却至室温;
步骤2.5:将玻片与抗AURKA(1:250),PDGFR-β(1:200),p-PDGFR-β(1:250),α-SMA(1:10000)和单核细胞趋化蛋白1(MCP1)(1:250)的一抗在4℃孵育过夜;
步骤2.6:用磷酸盐缓冲盐水(PBS)洗涤后,应用生物素偶联的二抗,并使用DakoREALTMEnVisionTM检测系统K5007(丹麦)可视化信号。
所述细胞培养研究具体包括如下步骤:
步骤3.1:将正常的大鼠肾49F(NRK-49F)和小鼠肾近端肾小管上皮细胞(mPTC)在添加有10%FBS的无血清角质形成细胞培养基中生长,并在37℃,5%CO2的培养箱中培养;
步骤3.2:使用0.25%胰蛋白酶-0.02%EDTA(Invitrogen)以60-80%汇合将细胞传代培养;
步骤3.3:让细胞在无血清培养基中饥饿6-8h,然后用MK-5108(200nM或400nM)预处理1h;
步骤3.4:然后将重组人转化生长因子-β1(TGF-β1)(15ng/ml)添加到培养基中;
步骤3.5:在重组人TGF-β1(15ng/ml)刺激6-8小时后,用AURKA质粒转染的细胞;
步骤3.6:24小时后收集细胞进行qRT-PCR分析,或48小时后收集细胞进行WB分析。
所述细胞增殖和细胞毒性测定具体步骤如下:
步骤4.1:细胞活力通过CCK-8测定试剂盒(MedChemExpress,中国)确定;
步骤4.2:将NRK-49F和mPTCs以104-105个细胞/孔的密度接种在100μL培养基中的孔板上,并用各种浓度的MK-5108(0-1000nM)处理;
步骤4.3:温育24小时后,将每孔CCK-8试剂10μL加入培养基中,温育1-4小时;
步骤4.4:使用酶标仪在450nm处检测吸光度。
所述实时定量PCR具体包括如下步骤:
步骤5.1:使用RNA isoPlus试剂(TaKaRa生物技术有限公司,中国大连)根据制造商的方案从组织或细胞中提取总RNA;
步骤5.2:使用PrimeScriptTMRT Master Mix(Takara,草津,日本,RR036)对cDNA进行反转录;
步骤5.3:通过SYBR Green PCR Master Mix(Vazyme,中国南京)和ABI 7500实时PCR检测系统(Applied Biosystems,加利福尼亚州福斯特市)进行实时PCR扩增;
其中,温度循环条件为95℃10分钟,95℃40循环15s,60℃1分钟;
其中,将mRNA的相对表达水平相对于GAPDH标准化,并使用2-ΔΔCt方法计算;
步骤5.4:所需的引物由Primer 3.0软件(http://Frodo.wi.mit.edu)设计,其引物序列列于表2。

所述蛋白质测定包括具体步骤如下:
步骤6.1:使用含有1x蛋白酶抑制剂(Roche)和1x磷酸酶抑制剂(Roche)的RIPA缓冲液匀浆组织或细胞裂解物;
步骤6.2:然后将样品以5000rpm离心30分钟;
步骤6.3:使用BCA蛋白质测定试剂盒(Thermofisher,Waltham,MA,USA,23225)测定蛋白质浓度;
步骤6.4:等效样品用于针对FN1(1:1000),胶原III(1:1000),PDGFRβ(1:1000),p-PDGFRβ(1:1000),AURKA(1:1000)和GAPDH(1:2000),然后添加HRP标记的二抗(1:5000);
步骤6.5:免疫反应条带通过Amersham Biosciences ECL检测系统(Amersham,Little Chalfont,英国)可视化。
步骤6.6:通过测量条带的强度进行光密度分析,并使用Quantity One软件(Bio-Rad,Hercules,CA)将其归一化为相应的GAPDH条带。
所述统计分析包括如下内容:
步骤7.1:所有定量值均表示为平均值±平均值标准误差(SEM);
步骤7.2:使用GraphPad Prism(6.0版,GraphPad Software,La Jolla,CA,美国),通过两尾Student t检验分析两组之间的统计学差异;
步骤7.3:单因素方差分析用于多个组之间的比较;P<0.05在统计学上被认为是具有统计学差异的。
试验结果如下:
1)AURKA在CKD患者和UUO小鼠的纤维化肾脏中的表达。
通过免疫组织化学和蛋白质印迹法测定AURKA的表达,如图1A所示,在CKD患者的肾小管中,AURKA几乎不在正常的肾组织中表达,并且AURKA蛋白的表达水平显著上调;在动物模型中,通过Western印迹和qRT-PCR分析进一步确定(如图1C),AURKA主要在肾小管细胞中表达,其蛋白表达水平在阻塞性肾脏中也有所增加(如图1B)。上述数据暗示AURKA可能与肾纤维化的发生和发展有关。
2)MK-5108改善了UUO小鼠阻塞性肾脏的肾脏病理。
为了阐明AURKA在CKD的肾脏病理中的作用,在UUO后3天对小鼠施用AURKA抑制剂MK-5108,直到处死小鼠。Masson和天狼星红染色表明,与假手术组小鼠相比,MK-5108显著减少了阻塞性肾脏间质中纤维化病变和胶原的大量积累(如图2A-D)。
3)MK-5108改善了UUO损伤后的肾间质纤维化。
通过qRT-PCR进一步检测了胶原蛋白I,胶原蛋白III和FN1的表达。与假手术组相比,UUO小鼠明显诱发了阻塞性肾脏的肾纤维化。在用MK-5108处理后,这些因子的mRNA水平被明显抑制。与这些数据一致,蛋白质印迹分析进一步证实了MK-5108处理后FN1和胶原III的下调,这表明AURKA的靶向抑制作用可能减弱UUO小鼠阻塞性肾脏的ECM沉积(图3A,B和C)。这些结果为MK-5108改善UUO诱导的肾间质纤维化提供了有力的证据。
4)MK-5108抑制UUO诱导的成纤维细胞增殖和活化以及肾小管细胞的部分表型转化。
FSP1,也称为S100A4,是检测组织驻留成纤维细胞的可靠标志物;α-SMA通常被认为是将成纤维细胞活化为成肌纤维细胞的标志物。
如图4A和B所示,Fsp1和α-SMA的mRNA表达降低,并且α-SMA的蛋白质水平也被轻微下调(如图4C)。同时,免疫组织化学分析进一步证实,MK-5108处理后FSP1和α-SMA阳性成纤维细胞明显减少(如图4D和E)。
另外,PDGFRβ也被认为是间充质标记,介导肾成纤维细胞的活化和纤维化。使用免疫印迹分析来检测MK-5108对PDGFRβ的表达和磷酸化水平的影响,在假手术肾脏中,Tyr751的PDGFRβ和p-PDGFR-β的表达较弱。然而,UUO增加了阻塞性肾脏中PDGFRβ的表达及其磷酸化水平,这明显受到MK-5108的抑制(如图4C)。波形蛋白在肾纤维化中表达水平倾向于显著增加,qRT-PCR分析显示,MK-5108处理后Snail1,Twist1和波形蛋白的mRNA表达水平显著下调,这表明AURKA的靶向抑制作用可以抑制肾小管细胞的表型变化(如图4F)。
5)MK-5108抑制UUO诱导的肾脏炎症。
炎症和纤维化是CKD的两个主要特征。炎症在CKD的进展中也起着至关重要的作用。检查用或不用MK-5108治疗的UUO小鼠阻塞性肾脏的炎症状态,如qRT-PCR分析所示,UUO明显诱导炎症因子的表达,包括白介素-1β(IL-1β),白介素6(IL-6)和肿瘤坏死因子-α(TNF-α),这些因子明显变钝通过施用MK-5108(如图5A)。同时,免疫组化进一步证实了MK-5108治疗后阻塞性肾脏中MCP1的减少(如图5B),表明MK-5108可以在一定程度上改善UUO诱导的肾脏炎症浸润。
6)MK-5108对NRK-49F细胞和mPTC没有促纤维化作用。
为了确定单独的MK-5108是否对肾细胞有一定的作用,将MK-5108加入NRK-49F细胞(肾成纤维细胞的细胞系)和mPTCs(小鼠近端肾小管细胞)24小时。通过CCK8分析,我们监测了不同浓度的MK-5108的细胞生存力,并将200nM和400nM定义为以下实验的适当浓度(如图6A和C)。Western blotting显示,MK-5108处理后,肾细胞中FN1和胶原III的蛋白水平没有改变,表明MK-5108不会干扰NRK-49F细胞分泌ECM,也不会干扰NRK-49F细胞分泌ECM,或促进了mPTCs向纤维化表型的转化(如图5B和D)。
7)MK-5108阻断了TGF-β1诱导的NRK-49F细胞活化。
为了进一步阐明MK-5108对处于损伤状态的肾细胞的直接影响,在TGF-β1刺激的NRK-49F细胞中添加了MK-5108。如上所述,选择200nM和400nM作为实验的适当浓度。来自qRT-PCR和Western Blot的数据表明,MK-5108处理明显抑制了NRK-49F细胞的FN1,胶原I和胶原III的表达(如图7A-C)。这些数据表明,MK-5108可通过抑制成纤维细胞的活化来减轻肾纤维化。
8)MK-5108降低了mPTC中TGF-β1诱导的促纤维化反应。
检测MK-5108对TGF-β1刺激的mPTCs的作用,CCK8实验表明200nM和400nM这两种浓度对细胞无明显影响。如图8A-C所示,MK-5108显著降低了FN1,胶原蛋白I或胶原蛋白III的mRNA和蛋白表达,并显著抑制了mPTC中α-SMA表达的上调。这些数据表明,在肾纤维化过程中,MK-5108可以显著抑制肾小管细胞转换为分泌表型和产生ECM。
9)AURKA促进了TGF-β1诱导的NRK-49F细胞活化。
鉴于以上数据表明AURKA抑制剂MK-5108具有显著的抗纤维化功能,我们进一步探讨了AURKA对处于损伤状态的NRK-49F细胞的作用。如图9A-D所示,仅AURKA的过表达对细胞没有促成纤维作用。但是,当细胞对TGF-β1作出反应时,AURKA的过度表达显著增加了包括胶原蛋白I和胶原蛋白III在内的ECM成分的产生。而且,α-SMA表达也显著增强。此外,蛋白质印迹显示,在过量表达AURKA后,NRK-49F细胞中FN1和胶原III的蛋白水平增加了(如图9E)。这些数据表明,AURKA可以促进TGF-β1诱导的肾成纤维细胞的活化和基质的产生。
本实施例研究表明,MK-5108显著减轻了UUO诱导的肾纤维化。FN1,胶原I和III的表达在受阻动物和TGF-β1处理的肾细胞中增加,而MK-5108处理则显著减少。在分别用MK5108治疗的UUO小鼠中,FSP1和α-SMA通常分别被认为是间质成纤维细胞和成肌纤维细胞的标志物。据报道,PDGFRβ介导肾成纤维细胞的活化和纤维化,并且我们证明药理学抑制AURKA也明显降低了其表达和磷酸化水平。这些结果表明,MK-5108可以抑制成纤维细胞的增殖和活化。此外,肾损伤导致肾细胞表型发生明显变化,从而导致大量纤维生成因子释放增加,并促进间质成纤维细胞活化。
本实施例指出,受阻的肾脏中蜗牛家族锌指1(Snail1),扭曲基本螺旋-环-螺旋转录因子1(Twist1)和波形蛋白的mRNA水平显著降低。同时,体外实验证实,MK-5108明显减少了由TGF-β1诱导的近端肾小管细胞向分泌表型的转变。这些结果表明,MK-5108可以适度减轻肾细胞的表型转变,从而进一步减轻肾纤维化。CKD通常伴随着不同类型的炎症细胞的活化和浸润。纤维化和炎症构成了有害的循环,并且随着肾功能的恶化,纤维化的发展常常伴随着肾功能的丧失。
本实施例证明了MK-5108治疗可明显降低促炎细胞因子(如IL-6,IL-1β和TNF-α)的mRNA水平,并显著下调了肾脏阻塞患者的MCP-1蛋白水平。随后对缓解的肾纤维化的反应可能会加剧这种现象。当前的研究数据表明,MK-5108阻断AURKA可以抑制与纤维发生有关的多种事件,并显著改善肾纤维化。
