本发明提供一种水稻种子高效催芽及快速育苗方法,包括:(1)将含水率≤8%的超干水稻种子于高温湿环境下炼种;(2)晒种,通风;(3)将稻种置于30~32℃的含0.1~0.5%海蒿子提取液的浸种液中浸种,沥干,再置于35~36℃的含0.8~1.0%海蒿子提取液的浸种液中浸种;(4)清洗种子,沥干,平铺于蒸汽温室中催芽;定期于表面均匀喷施含L‑苹果酸与反丁烯二酸的混合水溶液,稻种露白后,降至28~30℃继续催芽;本发明通过采用高温湿炼种、不同浓度的海蒿子提取液的浸种液浸种联合蒸汽温室的催芽方法,可更加适用于低水分的水稻种子的高效催芽,提高种子萌发效果,保证水稻育苗质量。

背景技术
水稻作为主要粮食作物,其对全球粮食安全发挥重要作用。水稻种子的催芽技术是水稻生产过程中关键的环节,是培育壮苗的基础,高质量的水稻育苗质量,是水稻高产和优质的保证。由于为了保证水稻种子的保存期限,往往需要对鲜种子进行干燥处理,随着水分的降低,保存期限延长,但不同种子状态下的浸种催芽效果差异悬殊,现有低水分的稻种在浸种催芽过程中,普遍存在不均衡萌芽现象,尤其是对于含水率≤8%的超干水稻种子的浸种催芽,易出现稻种内的养分溶解损失,种子萌发率低,出芽势和整齐度差,难以达到理想的“快、齐、匀、壮”的效果,导致水稻苗期的营养缺失或失衡,移栽后生长速率慢,后期出现发育不完全,降低水稻产量的问题。因此,提出一种可更加适用于低水分的水稻种子的高效催芽及快速育苗方法,以提高种子萌发效果,保证水稻育苗质量,为水稻的高效生产提供重要基础。
发明内容
鉴于此,本发明提出一种水稻种子高效催芽及快速育苗方法。
本发明的技术方案是这样实现的:
本发明提供一种水稻种子高效催芽方法,包括如下步骤:
步骤1:选择含水率≤8%的超干水稻种子,置于湿度为96~98%,温度为37.5~38.0℃的高温湿环境下炼种3~5h;
步骤2:将炼种后的水稻种子进行晒种6~8h后,放置阴凉干燥处通风2~3h;
步骤3:将晒种后的水稻种子先置于30~32℃的含0.1~0.5%质量比的海蒿子提取液的浸种液中进行浸种5~6h,取出沥干,再置于35~36℃的含0.8~1.0%质量比的海蒿子提取液的浸种液中进行浸种10~12h;水稻种子与浸种液均按1:(6~8)的质量体积比混合;
步骤4:用清水清洗浸种后的水稻种子,沥干,平铺于湿度为80~85%,温度为35~36℃的蒸汽温室中催芽,平铺厚度为8~10cm;每隔1~2h进行翻动,并在每2次翻动后,在稻种表面均匀喷施含质量比为0.01~0.03%L-苹果酸与0.06~0.08%反丁烯二酸的混合水溶液,待出现稻种露白后,将温度降至28~30℃进行继续催芽;
步骤5:待种芽长至≥谷粒的1/2长且生有一根时,即可用于播种育苗。本发明的催芽时间至18h后均可出齐芽。本发明通过对超干水稻种子利用高温湿炼种与晒种组合唤醒杀菌的基础上,采用了添加有不同浓度的海蒿子提取液的浸种液结合有效稳定的蒸汽温室的催芽方法,从而充分保证了低含水量水稻种子的发芽势和发芽率,实现更好的稻种“快、齐、匀、壮”的萌发效果,充分提高水稻苗期的栽培和生长质量。
进一步说明,步骤1中,所述超干水稻种子的含水率为7~8%。
进一步说明,步骤3中,所述含0.1~0.5%质量比的海蒿子提取液的浸种液的配方为:10~20%聚乙二醇-6000、0.03~0.06%赤霉素、0.05~0.06%水杨酸、0.3~0.5%壳聚糖、0.1~0.5%海蒿子提取液,水补足100%。
进一步说明,步骤3中,所述含0.8~1.0%质量比的海蒿子提取液的浸种液的配方为:28~36%聚乙二醇-6000、0.02~0.04%赤霉素、0.03~0.05%水杨酸、0.6~0.8%壳聚糖、0.8~1.0%海蒿子提取液,水补足100%。采用一定量的海蒿子提取液与壳聚糖复配,有利于对稻种形成保护屏障作用,并保证种子吸收有效成分,对种子萌发具有良好的促进作用。
进一步说明,所述海蒿子提取液的制备方法,包括如下步骤:
(1)将海蒿子用清水清洗,除去藻体表面的泥沙,干燥后于-20℃下冷冻保存过夜;
(2)称取海蒿子进行捣碎后,由组织匀浆机进行匀浆3~5次,每次匀浆2~3min,每次间隔3~5min,得到海蒿子匀浆;
(3)向海蒿子匀浆中加水混合,海蒿子与水的料液比为1:(3~4)g/ml,加热至35~36℃浸提2~3h后,进行离心后收集浸提液,即得到海蒿子提取液。

(2)苗期管理:播种初期,控制育苗温度为28~30℃,当出苗>35%后,降低育苗温度为22~24℃,并定期喷施肥水,保证出全苗,待秧苗出齐后,配合育苗肥进行追肥,施肥量为25~35g/m2;下田插秧的秧苗株高≤15cm。基于快速的稻种萌发的基础上,通过采用由膨胀珍珠岩与泥炭混合而成的育苗基质,搭配合理的播种和苗期管理,有利保证低含水量水稻种子的成出苗活率和稻苗的生长质量。
与现有技术相比,本发明的有益效果是:本发明提出的水稻种子高效催芽方法及快速育苗方法,首先在晒种前对含水率≤8%的超干水稻种子在高温湿环境下炼种,有利于增强超干种子呼吸和提高酶的活性,并利于增强超干种子浸种时的吸水性,提高种子的发芽势;在浸种的过程中,通过采用了含有不同含量海蒿子提取液的浸种液,在一定温度下进行浸种处理,不仅对稻种形成了一定的包衣作用,并可显著缩短了浸种时间,减少稻种内的养分溶解损失,提高种子的发芽势、发芽率和萌发指数,最后在催芽时,采用了蒸汽温室的催芽方法,并在催芽的过程中,联合采用含有L-苹果酸与反丁烯二酸的混合水溶液进行表面喷施,可降低种子催芽的温度敏感性,有效避免烧苗现象的同时实现了更加稳定的出芽势,提高稻种的出芽效率,减少养分的损失,有效达到了更好的稻种“快、齐、匀、壮”的萌发效果,提高成苗率,进而有利于促进水稻苗期的栽培和生长质量。

步骤1:选择含水率为7%的超干水稻种子,置于湿度为96%,温度为37.5℃的高温湿环境下炼种3~5h;
步骤2:将炼种后的水稻种子进行晒种6h后,放置阴凉干燥处通风2h;
步骤3:将晒种后的水稻种子先置于30℃的含0.1%质量比的海蒿子提取液的浸种液中进行浸种5h,取出沥干,再置于35℃的含0.8%质量比的海蒿子提取液的浸种液中进行浸种10h;水稻种子与浸种液均按1:6的质量体积比混合;
步骤4:用清水清洗浸种后的水稻种子,沥干,平铺于湿度为80%,温度为35℃的蒸汽温室中催芽,平铺厚度为8cm;每隔1h进行翻动,并在每2次翻动后,在稻种表面均匀喷施含质量比为0.01%L-苹果酸与0.06%反丁烯二酸的混合水溶液,喷施量为150ml/m2,待出现稻种露白后,将温度降至28℃,进行继续催芽;
步骤5:待种芽长至≥谷粒的1/2长且生有一根时,即可用于播种育苗。
实施例2-一种水稻种子高效催芽方法,包括如下步骤:
步骤1:选择含水率为8%的超干水稻种子,置于湿度为98%,温度为38.0℃的高温湿环境下炼种5h;
步骤2:将炼种后的水稻种子进行晒种8h后,放置阴凉干燥处通风3h;
步骤3:将晒种后的水稻种子先置于32℃的含0.5%质量比的海蒿子提取液的浸种液中进行浸种6h,取出沥干,再置于36℃的含1.0%质量比的海蒿子提取液的浸种液中进行浸种12h;水稻种子与浸种液均按1:8的质量体积比混合;
步骤4:用清水清洗浸种后的水稻种子,沥干,平铺于湿度为85%,温度为36℃的蒸汽温室中催芽,平铺厚度为10cm;每隔2h进行翻动,并在每2次翻动后,在稻种表面均匀喷施含质量比为0.03%L-苹果酸与0.08%反丁烯二酸的混合水溶液,喷施量为250ml/m2,待出现稻种露白后,将温度降至30℃,进行继续催芽;

步骤4:用清水清洗浸种后的水稻种子,沥干,平铺于湿度为83%,温度为35.5℃的蒸汽温室中催芽,平铺厚度为9cm;每隔1.5h进行翻动,并在每2次翻动后,在稻种表面均匀喷施含质量比为0.02%L-苹果酸与0.07%反丁烯二酸的混合水溶液,喷施量为200ml/m2,待出现稻种露白后,将温度降至29℃,进行继续催芽;

(1)将海蒿子用清水清洗,除去藻体表面的泥沙,干燥后于-20℃下冷冻保存过夜;
(2)称取海蒿子进行捣碎后,由组织匀浆机进行匀浆4次,每次匀浆2.5min,每次间隔4min,得到海蒿子匀浆;
(3)向海蒿子匀浆中加水混合,海蒿子与水的料液比为1:3.5g/ml,加热至35.5℃浸提2.5h后,进行离心后收集浸提液,即得到海蒿子提取液。
其中,上述实施例1~3中初次浸泡的海蒿子提取液的浸种液的配方为:15%聚乙二醇-6000、0.04%赤霉素、0.055%水杨酸、0.4%壳聚糖、0.3%海蒿子提取液,水补足100%;上述二次浸泡的海蒿子提取液的浸种液的配方为:32%聚乙二醇-6000、0.03%赤霉素、0.04%水杨酸、0.7%壳聚糖、0.9%海蒿子提取液,水补足100%。
实施例4-如实施例3的一种水稻种子高效催芽方法,区别在于:步骤3中,将晒种后的水稻种子先置于31℃的含0.6%质量比的海蒿子提取液的浸种液中进行浸种5.5h后,升高温度至35.5℃,继续浸种11h。
实施例5-如实施例3的一种水稻种子高效催芽方法,区别在于:步骤3中,先采用31℃的含0.9%质量比的海蒿子提取液的浸种液中进行浸种5.5h后,再采用35.5℃的含0.3%质量比的海蒿子提取液的浸种液中进行浸种11h。
对比例1-如实施例3的一种水稻种子高效催芽方法,区别在于:步骤1中,将含水率为7.5%的超干水稻种子未进行炼种,直接进行晒种。
对比例2-如实施例3的一种水稻种子高效催芽方法,区别在于:步骤3中,分别在31℃下浸种5.5h和在35.5℃下浸种11h中所采用均未含有海蒿子提取液的浸种液,其余组分与实施例3相同。
对比例3-如实施例3的一种水稻种子高效催芽方法,区别在于:步骤4中,在对水稻种子翻动后,未采用L-苹果酸与反丁烯二酸的混合水溶液喷施于稻种表面。
1、水稻种子的催芽和育苗实验
(1)水稻种子的催芽:根据上述的实施例1~5和对比例1~3的水稻种子的催芽方法,选择籼型H优518稻种作为供试样品,进行相应的催芽培养,对应设定8个处理,每个处理200粒种子,每个处理重复4次,催芽培养光周期为光照12h/黑暗12h,每天观察水稻种子的发芽情况,以幼芽达到种子长度一半,根长与种子等长作为发芽指标,分别记录发芽数,计算发芽率、发芽势、发芽指数和活力指数,测定各处理的水稻种子发芽指标,其结果如下表1:
发芽率(%)=种子总发芽数/供试种子数×100.
发芽势(%)=3d内种子发芽数/供试种子数×100.
发芽指数:Gi=Σ(Gt/Dt).
式中:Gt为t日的发芽数;Dt为相应的发芽日数.
活力指数:Vi=Gi×S.
式中:Gi为发芽指数;S为发芽结束时单株芽的平均干质量(mg)。

由上表可以看出,根据本发明的水稻种子高效催芽方法,可有效获得较高的发芽势、发芽率、发芽指数和活力指数,表明本发明通过对超干水稻种子利用高温湿炼种与晒种组合唤醒杀菌的基础上,采用添加有不同浓度的海蒿子提取液的浸种液结合有效稳定的蒸汽温室的催芽方法,课充分保证了低含水量水稻种子的发芽势和发芽率,实现更好的稻种“快、齐、匀、壮”的萌发效果。而在实施例4、5和对比例1~3中,稻种的萌发效果降低,在对比例1~3中,其稻种的发芽势、发芽率、萌发指数和活力指数明显降低。
(2)水稻的育苗:将由实施例1~5的水稻种子的催芽方法所获得水稻种子,进行如下育苗方法:
播种:将水稻种子均匀播撒于由膨胀珍珠岩与泥炭按质量比3:2混合而成的苗床基质表面,播种量为0.6kg/m2,并采用基质与1.5mm细沙按照1:3混合而成的混合物覆盖种子,盖种厚度为0.8cm;
苗期管理:播种初期,控制育苗温度为29℃,当出苗>35%后,降低育苗温度为23℃,并定期喷施水溶肥营养液,以保证出全苗,待秧苗出齐后,配合市售育苗肥(梅塔多)进行定期追肥,施肥量为30g/m2;并分别统计育苗的株高、根长和根数,其结果如下表:

由上表可以看出,根据实施例1~5的水稻种子的催芽方法所获得水稻种子的育苗效果稳定,其成活率均可在88.5%以上,而在实施例4和5的稻苗生长效果与实施例3相比相对降低,表明高质量的稻种催芽效果,有利于保证稻苗的生长质量。
以上所述仅为本发明的较佳实施例而已,并不用以限制本发明,凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。

Claims (4)

1.一种水稻种子高效催芽方法,其特征在于:包括如下步骤:
步骤1:选择含水率7~8%的超干水稻种子,置于湿度为96~98%,温度为37.5~38.0℃的高温湿环境下炼种3~5h;
步骤2:将炼种后的水稻种子进行晒种6~8h后,放置阴凉干燥处通风2~3h;
步骤3:将晒种后的水稻种子先置于30~32℃的含0.1~0.5%质量比的海蒿子提取液的浸种液中进行浸种5~6h,取出沥干,再置于35~36℃的含0.8~1.0%质量比的海蒿子提取液的浸种液中进行浸种10~12h;水稻种子与浸种液均按1:(6~8)的质量体积比混合;
所述含0.1~0.5%质量比的海蒿子提取液的浸种液的配方为:10~20%聚乙二醇-6000、0.03~0.06%赤霉素、0.05~0.06%水杨酸、0.3~0.5%壳聚糖、0.1~0.5%海蒿子提取液,水补足100%;
所述含0.8~1.0%质量比的海蒿子提取液的浸种液的配方为:28~36%聚乙二醇-6000、0.02~0.04%赤霉素、0.03~0.05%水杨酸、0.6~0.8%壳聚糖、0.8~1.0%海蒿子提取液,水补足100%;
步骤4:用清水清洗浸种后的水稻种子,沥干,平铺于湿度为80~85%,温度为35~36℃的蒸汽温室中催芽,平铺厚度为8~10cm;每隔1~2h进行翻动,并在每2次翻动后,在稻种表面均匀喷施含质量比为0.01~0.03%L-苹果酸与0.06~0.08%反丁烯二酸的混合水溶液,喷施量为150~250ml/m2,待出现稻种露白后,将温度降至28~30℃进行继续催芽;
步骤5:待种芽长至≥谷粒的1/2长且生有一根时,即可用于播种育苗。
2.根据权利要求1所述的一种水稻种子高效催芽方法,其特征在于:所述海蒿子提取液的制备方法,包括如下步骤:
(1)将海蒿子用清水清洗,除去藻体表面的泥沙,干燥后于-20℃下冷冻保存过夜;
(2)称取海蒿子进行捣碎后,由组织匀浆机进行匀浆3~5次,每次匀浆2~3min,每次间隔3~5min,得到海蒿子匀浆;
(3)向海蒿子匀浆中加水混合,海蒿子与水的料液比为1:(3~4)g/ml,加热至35~36℃浸提2~3h后,进行离心后收集浸提液,即得到海蒿子提取液。
3.根据权利要求1所述的一种水稻种子高效催芽方法,其特征在于:步骤4中,所述混合溶液中L-苹果酸与反丁烯二酸的质量比为2:7。
4.一种根据权利要求1~3中任意一项所述的水稻种子高效催芽方法进行水稻快速育苗的方法,其特征在于:包括如下步骤:
(1)播种:将经过催芽后的水稻种子均匀播撒于由膨胀珍珠岩与泥炭按质量比3:2混合而成的苗床基质表面,播种量为0.5~0.8kg/m2,并采用基质与1~2mm细沙按照1:3混合而成的混合物覆盖种子,盖种厚度为0.5~1cm;
(2)苗期管理:播种初期,控制育苗温度为28~30℃,当出苗>35%后,降低育苗温度为22~24℃,并定期喷施肥水,保证出全苗,待秧苗出齐后,配合育苗肥进行追肥,施肥量为25~35g/m2;下田插秧的秧苗株高≤15cm。
