The fineness of the sea cucumber slurry obtained after the grinding is preferably below 5 mu m.
Mixing the obtained sea cucumber slurry with neutral protease for enzymolysis to obtain a first sea cucumber enzymolysis liquid, wherein the first sea cucumber enzymolysis liquid mainly contains sea cucumber polysaccharide and sea cucumber saponin; the dosage of the neutral protease accounts for 0.3-0.5 percent of the total mass of the sea cucumber slurry, and is preferably 0.4 percent. The source of the neutral protease is not particularly limited in the present invention, and a conventional commercially available product may be used. In the present example, the neutral protease is preferably obtained from Novoxin (China) Biotechnology, Inc., produced in Denmark, under the product number of 16. In the invention, the enzymolysis temperature is preferably 50-52 ℃, and more preferably 51 ℃; the enzymolysis time is preferably 2-3 h, and more preferably 2.5 h.
The first sea cucumber enzymolysis liquid is subjected to enzyme deactivation and sterilization to obtain enzyme deactivation and sterilization sea cucumber enzymolysis liquid; the temperature for enzyme inactivation and sterilization is 85-90 ℃, and is preferably 88 ℃; the time for enzyme deactivation and sterilization is 15-30 min, preferably 20 min. The method for killing the enzyme and sterilizing is not particularly limited, and a conventional enzyme and sterilizing method is adopted.
The obtained enzyme-deactivated and sterilized sea cucumber enzymatic hydrolysate is filtered to obtain the sea cucumber enzymatic hydrolysate; the particle size of the sea cucumber enzymolysis liquid is 80-120 meshes. The invention has no special limitation on the type of the filter, and the filter can be a conventional filter.
The invention also provides a preparation method of the royal jelly sea cucumber enzymolysis peptide freeze-dried powder, which comprises the following steps: mixing royal jelly with the first water for dissolving, mixing with the sea cucumber enzymolysis liquid prepared by the preparation method of the technical scheme and the second water, and carrying out vacuum freeze drying to obtain royal jelly sea cucumber enzymolysis peptide freeze-dried powder; the mass ratio of the royal jelly to the first water is (70-90) to (70-90), and preferably 80: 80; the mass ratio of the royal jelly to the sea cucumber enzymolysis liquid is (70-90) to (20-30), and preferably 80: 25; the mass ratio of the first water to the second water is (70-90): 60-80, preferably 80: 71. The mixing mode is not particularly limited, and the conventional mixing method can be adopted. In the present invention, the vacuum freeze-drying includes quick freezing and dehydration drying; the quick-freezing temperature is preferably-45 ℃ to-40 ℃, and the quick-freezing time is preferably 2.5 to 3.5 hours, and more preferably 3 hours; the vacuum degree of the dehydration drying is preferably 10-100 Pa, and more preferably 30-60 Pa; the dehydration drying temperature is preferably-45-35 ℃, and more preferably-35-25 ℃; the dehydration drying time is preferably 75-85 h, and more preferably 80 h.
The invention also provides the lyophilized royal jelly sea cucumber enzymolysis peptide powder prepared by the preparation method of the technical scheme.
The invention also provides a food, which comprises the lyophilized royal jelly sea cucumber enzymolysis peptide powder and auxiliary materials. In the invention, the auxiliary materials preferably comprise one or more of medicinal and edible raw materials, new resource food, new food raw materials, common food, food additives, nutrition enhancers and edible strains. The adjuvants include one or more of bee pollen, Mel, fructus Hippophae, Cordyceps militaris, Euglena, Bifidobacterium bifidum, vitamin E, taurine and Ginseng radix. In the invention, the freeze-dried powder accounts for 50-99.9% of the total mass of the food. The formulation of the food of the present invention is not particularly limited, and preferably includes powders, granules, capsules and tablets.
The invention has no special limitation on the use method of the prepared royal jelly-containing sea cucumber enzymolysis peptide freeze-dried powder and the food prepared by using the freeze-dried powder, and preferably adopts the use method of instant food after being dissolved in water.
The technical solution of the present invention will be clearly and completely described below with reference to the embodiments of the present invention. It is to be understood that the described embodiments are merely exemplary of the invention, and not restrictive of the full scope of the invention. All other embodiments, which can be derived by a person skilled in the art from the embodiments given herein without making any creative effort, shall fall within the protection scope of the present invention.
Example 1
The preparation method of the sea cucumber enzymolysis liquid comprises the following steps:
(1) cutting 20kg of fresh and alive sea cucumber for 3 years from the upper part of the tail part of the sea cucumber along the abdomen, removing lime in the mouth of the sea cucumber, washing away silt, and washing with water to obtain cleaned sea cucumber;
(2) mixing cleaned sea cucumber with 10kg of water, and then grinding to obtain sea cucumber pulp, wherein the grinding fineness of the sea cucumber pulp is 5 mu m;
(3) mixing 30kg of the obtained sea cucumber slurry with 0.09kg of protease for enzymolysis at 50 ℃ for 3h to obtain a first sea cucumber enzymolysis liquid;
(4) carrying out enzyme deactivation and sterilization on the obtained first sea cucumber enzymolysis liquid at 85 ℃ for 30min to obtain a second sea cucumber enzymolysis liquid;
(5) and filtering the obtained second sea cucumber enzymolysis liquid by using a filter with the particle size of 80 meshes to obtain 25kg of sea cucumber enzymolysis liquid.
Example 2
The preparation method of the sea cucumber enzymolysis liquid comprises the following steps:
(1) cutting 10kg of fresh and alive sea cucumber for 3 years from the upper part of the tail part of the sea cucumber along the abdomen, removing lime in the mouth of the sea cucumber, washing away silt, and washing with water to obtain cleaned sea cucumber;
(2) mixing the cleaned sea cucumber with 5kg of water, and then grinding the mixture into thick liquid to obtain sea cucumber thick liquid, wherein the grinding fineness of the sea cucumber thick liquid is 5 microns;
(3) mixing 15kg of the obtained sea cucumber slurry with 0.045kg of protease for enzymolysis, wherein the enzymolysis temperature is 50 ℃, and carrying out enzymolysis for 3h to obtain a first sea cucumber enzymolysis liquid;
(4) carrying out enzyme deactivation and sterilization on the obtained first sea cucumber enzymolysis liquid at 85 ℃ for 30min to obtain a second sea cucumber enzymolysis liquid;
(5) and filtering the obtained second sea cucumber enzymolysis liquid by using a filter with the particle size of 80 meshes to obtain 10kg of sea cucumber enzymolysis liquid.
Example 3
The preparation method of the sea cucumber enzymolysis liquid comprises the following steps:
(1) cutting 30kg of live sea cucumber for 3 years from the upper part of the tail part of the sea cucumber along the abdomen, removing lime in the mouth of the sea cucumber, washing away silt, and washing with water to obtain cleaned sea cucumber;
(2) mixing cleaned sea cucumber with 15kg of water, and grinding to obtain sea cucumber pulp with the grinding fineness of 5 mu m;
(3) mixing 45kg of the obtained sea cucumber slurry with 0.135kg of protease for enzymolysis, wherein the enzymolysis temperature is 50 ℃, and the enzymolysis is carried out for 3 hours to obtain a first sea cucumber enzymolysis liquid;
(4) carrying out enzyme deactivation and sterilization on the obtained first sea cucumber enzymolysis liquid at 85 ℃ for 30min to obtain a second sea cucumber enzymolysis liquid;
(5) and filtering the obtained second sea cucumber enzymolysis liquid by using a filter with the particle size of 80 meshes to obtain 40kg of sea cucumber enzymolysis liquid.
Example 4
The preparation method of the sea cucumber enzymolysis liquid comprises the following steps:
(1) cutting 29kg of live sea cucumber for 3 years from the upper part of the tail part of the sea cucumber along the abdomen, removing lime in the mouth of the sea cucumber, washing away silt, and washing with water to obtain cleaned sea cucumber;
(2) mixing cleaned sea cucumber with 14.5kg of water, and grinding to obtain sea cucumber pulp with the grinding fineness of 5 mu m;
(3) mixing 43.5kg of the obtained sea cucumber slurry with 0.1305kg of protease for enzymolysis at 50 ℃ for 3h to obtain a first sea cucumber enzymolysis liquid;
(4) carrying out enzyme deactivation and sterilization on the obtained first sea cucumber enzymolysis liquid at 85 ℃ for 30min to obtain a second sea cucumber enzymolysis liquid;
(5) and filtering the obtained second sea cucumber enzymolysis liquid by using a filter with the particle size of 80 meshes to obtain 38.5kg of sea cucumber enzymolysis liquid.
Example 5
The preparation method of the sea cucumber enzymolysis liquid comprises the following steps:
(1) cutting 20kg of fresh and alive sea cucumber for 3 years from the upper part of the tail part of the sea cucumber along the abdomen, removing lime in the mouth of the sea cucumber, washing away silt, and washing with water to obtain cleaned sea cucumber;
(2) mixing cleaned sea cucumber with 10kg of water, and then grinding to obtain sea cucumber pulp, wherein the grinding fineness of the sea cucumber pulp is 5 mu m;
(3) mixing 30kg of the obtained sea cucumber slurry with 0.09kg of protease for enzymolysis at 52 ℃ for 2h to obtain a first sea cucumber enzymolysis liquid;
(4) carrying out enzyme deactivation and sterilization on the obtained first sea cucumber enzymolysis liquid at 90 ℃ for 15min to obtain a second sea cucumber enzymolysis liquid;
(5) and filtering the obtained second sea cucumber enzymolysis liquid by using a filter with the particle size of 120 meshes to obtain 25kg of sea cucumber enzymolysis liquid.
Example 6
A preparation method of lyophilized royal jelly sea cucumber enzymolysis peptide powder comprises the following steps: mixing 80kg of royal jelly with 80kg of partial water for dissolving, then mixing with 25kg of the sea cucumber enzymolysis liquid prepared in the embodiment 1 and 71kg of the residual water, filling the mixed liquid into a freeze-drying bottle in an automatic filling and sealing machine, covering a glue cover, semi-sealing, and drying by dehydration for 80 hours at the vacuum degree of 10Pa and the vacuum degree of 45 ℃ for 16 ml/bottle, wherein the semi-sealed freeze-drying bottle is quickly frozen in a vacuum freeze-drying machine, and the water content of the freeze-drying powder is lower than 3%.
Example 7
A preparation method of lyophilized royal jelly sea cucumber enzymolysis peptide powder comprises the following steps: mixing and dissolving 60kg of royal jelly and 60kg of partial water, mixing with 18.75kg of the sea cucumber enzymolysis liquid prepared in the embodiment 2 and 53.25kg of the rest water, filling the mixed liquid into a freeze-drying bottle in an automatic filling and sealing machine, covering a glue cover, semi-sealing, and drying in 16ml per bottle, quickly freezing the semi-sealed freeze-drying bottle in a vacuum freeze-drying machine at the temperature of minus 45 ℃ for 3 hours, and then dehydrating and drying at the vacuum degree of 10Pa, the temperature of minus 45 ℃ and the time of 80 hours until the water content of the freeze-dried liquid is lower than 3%.
Example 8
A preparation method of lyophilized royal jelly sea cucumber enzymolysis peptide powder comprises the following steps: mixing 50kg of royal jelly with 50kg of partial water for dissolution, mixing with 15.625kg of the sea cucumber enzymolysis liquid prepared in the embodiment 3 and 44.375kg of the residual water, filling the mixed liquid into a freeze-drying bottle in an automatic filling and sealing machine, covering a glue cover, semi-sealing, and drying in 16 ml/bottle, quickly freezing the semi-sealed freeze-drying bottle in a vacuum freeze-drying machine at the temperature of minus 45 ℃ for 3 hours, and then dehydrating and drying at the vacuum degree of 10Pa, the temperature of minus 45 ℃ and the time of 80 hours, wherein the water content of the freeze-dried powder is lower than 3%.
Example 9
A preparation method of lyophilized royal jelly sea cucumber enzymolysis peptide powder comprises the following steps: 70kg of royal jelly and 70kg of partial water are mixed and dissolved, then the mixture is mixed with 21.875kg of sea cucumber enzymolysis liquid prepared in the embodiment 4 and 62.125kg of residual water, the mixed liquid is filled into a freeze-drying bottle in an automatic filling and sealing machine, a glue cover is covered on the freeze-drying bottle, the freeze-drying bottle is semi-sealed and 16 ml/bottle, the semi-sealed freeze-drying bottle is quickly frozen in a vacuum freeze-drying machine, the freezing time is 3 hours, then dehydration drying is carried out at the vacuum degree of 10Pa, the temperature of minus 45 ℃ and the time of 80 hours, and the water content of freeze-drying component is lower than 3%.

(3) carrying out enzyme deactivation and sterilization on the first sea cucumber enzymolysis liquid obtained in the step (2) to obtain enzyme-deactivated and sterilized sea cucumber enzymolysis liquid; the temperature for enzyme inactivation and sterilization is 85-90 ℃; the time for enzyme deactivation and sterilization is 15-30 min;
(4) filtering the enzyme-deactivated and sterilized sea cucumber enzymatic hydrolysate obtained in the step (3) to obtain a sea cucumber enzymatic hydrolysate; the particle size of the sea cucumber enzymolysis liquid is 80-120 meshes.
2. The preparation method according to claim 1, wherein the temperature of enzymolysis in the step (2) is 50-52 ℃, and the time of enzymolysis is 2-3 h.
3. A preparation method of royal jelly sea cucumber enzymolysis peptide freeze-dried powder is characterized by comprising the following steps: mixing royal jelly with first water for dissolving, mixing with the sea cucumber enzymolysis liquid prepared by the preparation method of claim 1 or 2 and second water, and performing vacuum freeze drying to obtain royal jelly sea cucumber enzymolysis peptide freeze-dried powder; the mass ratio of the royal jelly to the first water is (70-90) to (70-90); the mass ratio of the royal jelly to the sea cucumber enzymolysis liquid is (70-90) to (20-30); the mass ratio of the first water to the second water is (70-90) to (60-80).
4. The preparation method according to claim 3, wherein the vacuum freeze drying comprises quick freezing and dehydration, the temperature of the quick freezing is-45 ℃ to-40 ℃, and the time of the quick freezing is 2.5 to 3.5 hours.
5. The preparation method according to claim 4, wherein the vacuum degree of the dehydration is 10-100 Pa, the temperature of the dehydration is-45-35 ℃, and the time of the dehydration is 75-85 h.
6. The lyophilized royal jelly sea cucumber enzymolysis peptide powder prepared by the preparation method of any one of claims 3 to 5.
7. A food, which is characterized by comprising the lyophilized powder of royal jelly sea cucumber zymolytic peptide of claim 6 and auxiliary materials.
8. The food according to claim 7, wherein the auxiliary materials comprise one or more of medicinal and edible raw materials, new resource food, new food raw materials, common food, food additives, nutrition enhancers and edible strains.
9. The food of claim 8, wherein the adjuvant comprises one or more of bee pollen, honey, sea buckthorn, cordyceps militaris, euglena, bifidobacterium bifidum, vitamin E, taurine and ginseng.
10. The food product of claim 7, wherein the food product is in a dosage form selected from the group consisting of a powder, a granule, a capsule, and a tablet.
