Abstract

本申请公开了一种具有持续抗菌能力的水凝胶,所述水凝胶由含锌的有机物、凝胶基质和水制成。其优点表现在:本申请的水凝胶中的抗菌物质能发出一定波长、频率的电磁波,电磁波对细菌的DNA、细胞膜等结构产生破坏,从而起到杀菌抑菌的作用,在抗菌过程中不会因为抗菌作用而消耗,且不为人体所吸收,故在皮肤或创面上使用后能在其表面形成保护膜,避免外界细菌侵袭。本申请的水凝胶具有杀菌抑菌作用,特别是在皮肤或创面使用后能有效治疗和预防感染,并能通过保湿作用进一步促进创面愈合。

具有持续抗菌能力的水凝胶
技术领域
本申请涉及医疗器械技术领域,尤其是涉及一种具有持续抗菌能力的水凝胶。
背景技术
水凝胶,是一种能够在水中溶胀,吸收并保持大量水分而又不溶解于水的亲水性的网状高分子溶胀体,表面光滑,生物相容性好,在生物和医学工程领域得到了广泛的研究和应用。传统敷料由于吸液量低,需要厚厚地包扎并经常更换,敷料吸液后易干燥,并与伤口发生粘连而造成二次创伤等,水凝胶制备的敷料具有加快伤口愈合速度、容易更换、创面没有敷料残留屑等优点,正在逐渐取代传统敷料。
现有的水凝胶产品虽能促进创面愈合,但容易发生感染;部分具有抗菌功能的水凝胶产品如普朗特,虽能抗菌,但不具有长效性,不能在皮肤或创面上形成一个长效的抗菌保护层,且以化学抗菌机制为主要作用方式,难免会产生一定的生物毒性。
发明内容
本申请提供一种具有长效抗菌能力、能在皮肤或创面形成长效物理抗菌作用的水凝胶。
本申请采用下述技术方案:
一种具有持续抗菌能力的水凝胶,所述水凝胶由含锌的有机物、凝胶基质和水制成。
进一步地,所述含锌的有机物为甲基酸锌。
进一步地,甲基酸锌的添加量为0.02%-1%。
进一步地,所述凝胶基质由甘油和明胶组成。
进一步地,甘油的添加量为6%-30%,明胶的添加量为1%-3%。
进一步地,所述凝胶基质为纤维素衍生物。
进一步地,所述纤维素衍生物包括羟乙基纤维素、羟丙基纤维素、羟丙甲纤维素、羧甲基纤维素钠。
进一步地,纤维素衍生物的添加量为2%-6%。
本申请的有益效果如下:
本申请的水凝胶中的抗菌物质能发出一定波长、频率的电磁波,电磁波对细菌的DNA、细胞膜等结构产生破坏,从而起到杀菌抑菌的作用,在抗菌过程中不会因为抗菌作用而消耗,且不为人体所吸收,故在皮肤或创面上使用后能在其表面形成保护膜,避免外界细菌侵袭。本申请的水凝胶具有杀菌抑菌作用,特别是在皮肤或创面使用后能有效治疗和预防感染,并能通过保湿作用进一步促进创面愈合。

以下实施例中所用的甲基酸锌采购自中船重工(沈阳)抗微生物科技有限公司。
实施例1
将0.4g甲基酸锌溶解于159.6g去离子水中,加入甘油30g,搅拌均匀,加入10g明胶,60℃溶解,搅拌2h,制成0.2%的水凝胶制剂。
实施例2
将2g甲基酸锌溶解于192g去离子水中,加入羧甲基纤维素钠6g,搅拌均匀,制成1%的水凝胶制剂。
实施例3
将0.04g甲基酸锌溶解于159.96g去离子水中,加入甘油30g,搅拌均匀,加入10g明胶,60℃溶解,搅拌2h,制成0.02%的水凝胶制剂。
实施例4
将1g甲基酸锌溶解于189g去离子水中,加入羟乙基纤维素10g,搅拌均匀,制成0.5%的水凝胶制剂。
实施例5
将0.2g甲基酸锌溶解于159.8g去离子水中,加入甘油30g,搅拌均匀,加入10g明胶,60℃溶解,搅拌2h,制成0.1%的水凝胶制剂。
对比例1
159.6g去离子水中加入甘油30g,搅拌均匀,加入10g明胶,60℃溶解,搅拌2h,制成水凝胶制剂。
实施例6
实验方法:
取20只SD大鼠,随机分为实验组和对照组,每组10只。无菌条件下于大鼠背部做直径20mm的全层创面,造模后立即取106cfu/ml浓度鲍氏不动杆菌标准菌株细菌悬液1mL滴注于创面,常规包扎,造模后24h后打开创面再次滴注1mL细菌悬液,造模后48h打开创面,观察创面是否出现明显感染症状,确认感染模型制备是否成功。将上述实施例1样品和对比例1样品分别按照组别覆盖于创面上,常规包扎。每4天进行换药1次,咽拭子取创面分泌物进行细菌培养,结合创面分泌物、气味、创周炎症表现,判断创面是否感染,计算感染率,第20天取全层创面组织,行细菌定量实验。
实验结果:
①创面感染率:对照组创面无明显改善,实验期间感染率维持在100%,细菌培养结果均提示鲍曼不动杆菌感染,创面较多脓性分泌物,创周炎症明显,有明显异味。实验组在第1次(4天)换药时已有7个样本无明显感染症状,细菌培养阴性,余下3个仍有感染症状的创面细菌培养结果提示鲍曼不动杆菌感染,创面少量分泌物,有特殊气味,创周炎症不明显,感染率降为30%;第2次换药(8天)时,前一次换药是感染转阴的样本未再次感染,余下3个感染的创面中有2个创面感染症状消失,细菌培养结果阴性,剩余1个仍有少量分泌物,无明显异味和创周炎症,感染率降为10%;第3次换药(12天),所有创面均无明显感染症状,细菌培养结果阴性,感染率0%。感染率变化见图1。
②细菌定量结果:第20天,对照组创面组织细菌含量高达(2.79±0.74)×1012/g,实验组创面组织细菌含量仅为(6.07±0.34)×104/g,明显少于对照组,P<0.05。

Claims (9)

1.一种具有持续抗菌能力的水凝胶,其特征在于,所述水凝胶由含锌的有机物、凝胶基质和水制成。
2.如权利要求1所述的水凝胶,其特征在于,所述含锌的有机物为甲基酸锌。
3.如权利要求2所述的水凝胶,其特征在于,甲基酸锌的添加量为0.02%-1%。
4.如权利要求1所述的水凝胶,其特征在于,所述凝胶基质由甘油和明胶组成。
5.如权利要求4所述的水凝胶,其特征在于,甘油的添加量为6%-30%,明胶的添加量为1%-3%。
6.如权利要求1所述的水凝胶,其特征在于,所述凝胶基质为纤维素衍生物。
7.如权利要求6所述的水凝胶,其特征在于,所述纤维素衍生物包括羟乙基纤维素、羟丙基纤维素、羟丙甲纤维素、羧甲基纤维素钠。
8.如权利要求6所述的水凝胶,其特征在于,纤维素衍生物的添加量为2%-6%。
9.如权利要求4或5所述的水凝胶的制备方法,其特征在于,所述制备方法包括以下步骤:将甲基酸锌溶解于水中,加入甘油,搅拌均匀,加入明胶,60℃溶解,搅拌,制成水凝胶。
