1.3 Primary vaccines and reagents
Newcastle disease live vaccine (ND, La Sota strain), provided by Qingdao Yibang bioengineering, ltd; avian influenza (subtype H5), provided by Luoyang Hui Zhong Biotechnology, Inc. Antigen, negative serum and positive serum of the newcastle disease hemagglutination inhibition test are purchased from Qingdao Yibang bioengineering limited company; avian influenza virus H5 subtype hemagglutination inhibition test antigen, negative serum and positive serum were purchased from Harbin veterinary institute.
1.4 Main Instrument
An electronic balance; a high speed refrigerated centrifuge; a multifunctional microplate reader; constant temperature incubators, low temperature refrigerators, etc.
2 method
2.1 test animals and groups
800 healthy day-old black-bone chicks were randomly divided into 5 groups of 4 replicates each, 40 replicates each. Wherein the control group was fed basal diet; the astragalus polysaccharide group is fed with basic ration and 0.1% astragalus polysaccharide powder as a positive drug control; the low-dose group is fed with a basic ration and 0.1 percent of a traditional Chinese medicine composition for improving the growth performance and the immune function of the black-bone hen (example 1); the medium-dose group is fed with a basic ration and 0.3 percent of a traditional Chinese medicine composition for improving the growth performance and the immune function of the black-bone hen (example 2); the high-dose group was fed with a basic diet plus 0.5% of a Chinese medicinal composition for improving the growth performance and immune function of the black-bone hen chicks (example 3). The test started with the age of 1 day of the chicks and the test period was 28 days.
2.2 Breeding management
The feeding mode adopts single-layer cage culture, and chicks can freely drink water. The indoor temperature is: 36-38 ℃ at 1-3 days old, 32-35 ℃ at 4-7 days old, 30-33 ℃ at 8-14 days old, and then keeping the indoor temperature constant (25-28 ℃). The daily ration was gradually changed from 6 times daily (1 time every 4 h) to 3 times daily (1 time every 8 h), 8:00 feeding 1 st time. The composition and nutritional level of the basal diet are shown in table 1.

Note: the premix can provide Cu 8mg, Fe 90mg, Zn 50mg, Mn 80mg, I0.30 mg, Se 0.15mg, vitamin A10000 IU, and vitamin D for each kilogram of basic daily ration32100 IU, vitamin E10mg, vitamin K30.6mg, vitamin Bl2.0mg, vitamin B24.0nag, vitamin Bl20.01mg, 30.0mg of nicotinic acid, 0.6mg of folic acid, 0.15mg of biotin, 1mg of D-pantothenic acid l and 700U of phytase. The nutrient levels are calculated values.
The immunization procedure was as follows: at 7 days of age, dripping two times of vaccine of Newcastle disease infectious bronchitis into nose for eye drop; when the patient is 7 days old, the injection is subcutaneously injected with 0.3mL of newcastle disease and avian influenza (H5) per neck.
2.3 measurement of growth Performance
Observing and recording the basic conditions of daily activities, drinking water, ingestion and the like of each group of chicks every day, and recording the death condition of each group of chicks; the weight of the chicks was weighed on the 28 th day of the experiment, the average daily gain, the average daily feed intake and the feed-meat ratio were calculated.
2.4 immune organ index determination
At 8:00 days 28 of the test, 3 animals were randomly selected per each replicate, and the thymus, spleen and bursa of Fabricius were dissected and removed, and the surrounding fat was stripped off, and then the whole was weighed after the water was removed by suction with filter paper. Calculated according to the following formula: organ index (g/kg) is organ weight (g)/body weight (kg).
2.5 antibody and immune factor detection
The antibody titer of Newcastle disease and avian influenza (H5 subtype) is detected by referring to the technical Specification for diagnosing Newcastle disease (GB 16550-1996) and the technical method for diagnosing highly pathogenic avian influenza (GB/T18936-2003).
2.6 statistical analysis of data
The experimental data were analyzed for the significance of differences between groups by SPSS19.0 One-way ANO-VA with Duncan's method multiple test, the data are expressed as "mean. + -. standard deviation", and P <0.05 as the criterion for significance of differences.
3 results and analysis
3.1 cultivation Observation
In the test process, the chicks drink and eat water normally and are vigorous without infectious diseases, and the stool formability of the test group is superior to that of the control group; control dead chicks were mainly due to weakness.
3.2 influence of Chinese medicinal composition for improving growth performance and immunologic function of gallus Domesticus
The results are shown in Table 2.

1-28 days, wherein the average feed intake difference of each group is not significant (P is more than 0.05); compared with the average daily gain, the astragalus polysaccharide group, the medium dose group (example 2) and the high dose group (example 3) are obviously higher than the control group (P <0.05), and the low dose group (example 1) has no significant difference (P >0.05) from each group; compared with the feed-meat ratio, the astragalus polysaccharide group, the medium dose group (example 2) and the high dose group (example 3) are obviously lower than the control group (P <0.05), and the low dose group (example 1) has no obvious difference (P >0.05) from each group; the survival rate was compared, with the lowest in the control group and the highest in the high dose group (example 3).
3.2 Effect of micropowder on immune organ index of chicken at each stage
The results are shown in Table 3.

As can be seen from table 3, at day 28, the thymus index comparison shows that the medium dose (example 2) and the high dose (example 3) were significantly higher than the control (P < 0.05); the astragalus polysaccharides group, the low dose group (example 1) did not differ significantly from each group (P > 0.05); spleen index comparison, the astragalus polysaccharide group and the medium dose (example 2) and the high dose (example 3) are significantly higher than the control group (P <0.05), and the low dose (example 1) is not significantly different from each group (P > 0.05); in the bursa of Fabricius index comparison, the astragalus polysaccharide group and the low dose (example 1), the medium dose (example 2) and the high dose (example 3) are all significantly higher than the control group (P <0.05), and the high dose group is significantly higher than the low dose group (P < 0.05).
3.3 Effect of micropowder on serum antibody titer and immune factor of chicken
The results are shown in Table 4.

As can be seen from Table 4: on day 28, the astragalus polysaccharides group and the low dose (example 1), medium dose (example 2), high dose group (example 3) had significantly higher ND antibody titers than the control group (P < 0.05); the antibody titer of the astragalus polysaccharide group and the medium dose (example 2) and high dose (example 3) H5 is significantly higher than that of the control group (P <0.05), and the difference between the low dose group (example 1) and other groups is not significant (P > 0.05).
Discussion 4
The good growth performance of the chicks in the brooding stage can improve the later economic benefit, and the traditional Chinese medicine can improve the growth performance of the chicks. The traditional Chinese medicine composition for improving the growth performance and the immune function of the black-bone hen chicks prepared in the embodiments 1 to 3 has the effects of clearing heat and removing toxicity, protecting liver and benefiting gallbladder, promoting urination and activating blood circulation and regulating qi. The test result shows that the addition of 0.3-0.5% of the traditional Chinese medicine composition for improving the growth performance and the immune function of the black-bone hen chick in the basic ration, namely the feed prepared in the embodiments 2 and 3 of the invention, can enable the chick to have the average daily gain, the feed conversion ratio and the survival rate which are obviously superior to those of a control group in each growth stage.
The immune organ index has important significance for improving the immunity of the organism. The chicken immune organ index is increased, and a material foundation is laid for the organism to generate more immune cells and improve the vaccine antibody level. At day 28, the thymus index was significantly increased in the medium (example 2) and high (example 3) dose groups; the spleen index of the astragalus polysaccharide group, the medium dose group (example 2) and the high dose group (example 3) is obviously improved; the bursal disease index of the astragalus polysaccharide group and each dosage group can be obviously improved, and no obvious difference exists.
The current prevention of the newcastle disease and the avian influenza is mainly carried out by adopting a method of immunizing by using an attenuated vaccine and an inactivated vaccine. The body can reflect the specific humoral immunity function of the chicken by receiving the antigen to generate the antibody titer. The astragalus polysaccharide can promote the generation of antibodies by enhancing cellular immunity and humoral immunity, and can improve the antibody titer by assisting a vaccine. At 28 days, the antibody titer of ND was improved in each administration group, and the H5 antibody titer was significantly improved in the medium dose group (example 2) and the high dose group (example 3).

Claims (9)

1. A traditional Chinese medicine composition for improving growth performance and immune function of black-bone hen chicks is characterized by comprising the following components in parts by mass: 1-3 parts of field horsetail, 1-3 parts of tylophora floribunda, 1-3 parts of usnea, 1-2 parts of gnetum, 1-2 parts of poria cocos, 1-2 parts of lespedeza, 0.5-1.5 parts of magnolia officinalis and 0.5-1.5 parts of throat grass.
2. The traditional Chinese medicine composition according to claim 1, which is characterized by comprising the following components in parts by mass: 2 parts of field horsetail, 2 parts of tylophora floribunda, 2 parts of usnea, 1.5 parts of gnetum montanum, 1.5 parts of poria cocos, 1.5 parts of lespedeza virgata, 1 part of mangnolia officinalis and 1 part of laryngopharynx.
3. A feed for improving the growth performance and the immune function of a black-bone hen chick is characterized by comprising a basic ration and the traditional Chinese medicine composition of claim 1, wherein the traditional Chinese medicine composition accounts for 0.1-0.5% of the total mass of the basic ration.

The feed according to claim 3, wherein the basal diet consists of the following components in parts by mass: 59.20 parts of corn, 29.50 parts of soybean meal, 2.07 parts of rape oil, 5.00 parts of rapeseed cake, 1.70 parts of calcium hydrophosphate, 0.15 part of lysine, 0.22 part of methionine, 0.30 part of premix, 0.36 part of salt, 1.30 parts of stone powder and 0.20 part of choline.
5. The feed according to claim 3, characterized in that the formula of the premix is: cu 8mg, Fe 90mg, Zn 50mg, Mn 80mg, I0.30 mg, Se 0.15mg, vitamin A10000 IU, vitamin D32100 IU, vitamin E10mg, vitamin K30.6 mg, vitamin Bl 2.0mg, vitamin B24.0 nag, vitamin B120.01 mg, nicotinic acid 30.0mg, folic acid 0.6mg, biotin 0.15mg, D-pantothenic acid 1mg and phytase 700U.
6. A preparation method of a feed for improving the growth performance and the immune function of a black-bone hen chick is characterized by comprising the following steps:
step 1, preparing basic daily ration;
step 2, preparing a traditional Chinese medicine composition for improving the growth performance and the immune function of the black-bone hen;
and 3, preparing the feed for improving the growth performance and the immune function of the black-bone chicken chick.
7. The preparation method according to claim 6, wherein the preparing of the basic ration in step 1 is specifically:
step 1.1, premix preparation: weighing 8mg of Cu, 90mg of Fe, 50mg of Zn, 80mg of Mn, 0.30mg of I, 0.15mg of Se, 10000 IU of vitamin A, 32100 IU of vitamin D, 10mg of vitamin E, 30.6 mg of vitamin K, 2.0mg of vitamin Bl, 24.0 nag of vitamin B, 20.01 mg of vitamin Bl, 30.0mg of nicotinic acid, 0.6mg of folic acid, 0.15mg of biotin, 1mg of D-pantothenic acid l and 700U of phytase, and mixing the weighed components to prepare a premix;
step 1.2, weighing the following components in parts by mass: 59.20 parts of corn, 29.50 parts of soybean meal, 2.07 parts of rape oil, 5.00 parts of rapeseed cakes, 1.70 parts of calcium hydrophosphate, 0.15 part of lysine, 0.22 part of methionine, 0.30 part of premix, 0.36 part of salt, 1.30 parts of stone powder and 0.20 part of choline, and the weighed components are mixed to prepare the basic ration.
