Abstract
本发明提供一种柔性基底的细胞细菌互作模型的制备方法,包括:S1,丙烯酰胺和双丙烯酰胺在四甲基乙二胺和过硫酸铵的作用下,聚合形成一定基质刚度的聚丙烯酰胺凝胶,基质刚度模拟自生物组织的刚度;S2,在聚丙烯酰胺凝胶上孵育微环境物质,用于模拟细胞所处的微环境;S3,在聚丙烯酰胺凝胶界面上接种宿主细胞,加入互作的细菌建立细胞细菌互作模型。本发明考虑了人体力学微环境因素调控的细胞细菌互作模型,基于聚丙烯酰胺水凝胶细胞外微环境系统,建立了界面适于细胞生长柔性基底刚度的水凝胶。

技术领域
本发明涉及细胞感染技术领域,具体涉及一种柔性基底的细胞细菌互作模型的制备方法及应用。
背景技术
细胞细菌互作的体外模型一般建立于已经商品化的塑料底或者玻璃底的细胞培养板/皿中。然而真正的体内细菌感染宿主细胞的微环境是受到多因素调控的,包括细胞外基质(Extracellular matrix,ECM),ECM是细胞外许多大分子组成的复杂网络,包括胶原蛋白、非胶原蛋白、弹性蛋白、蛋白聚糖与氨基聚糖,其中胶原蛋白是ECM结构中的主要蛋白。这些蛋白形成了ECM的生理功能和机械特性,作用于组织上的机械负荷是基质刚度形成的基础,基质刚度施加压力于ECM并且在ECM组分、ECM受体和细胞内结构中传递力刺激源。各组织细胞所处环境的硬度均低于100kPa,如图2所示,脑组织中的细胞弹性模量为0.3~1kPa,皮肤组织为3~7kPa,心肝脾肺肾脏器组织均小于20kPa,肠组织也只有10~20kPa,牙龈组织也只在100kPa左右。然而常规用于研究细胞病理生理作用的细胞板类玻璃底或塑料底的材料的硬度却高达GPa级别,约等于106kPa,与宿主体内组织相差10万倍。近期也研究表明,基质刚度可以调节细菌感染。例如在不同基质刚度作用下,内皮细胞对李斯特菌的摄入不同,细胞基质的刚度越强的细胞越容易感染李斯特菌。然而,细胞细菌互作中与ECM刚度之间的调控关系并不明确,常规的在普通细胞板/皿材料上研究细菌和细胞互作并不能真实表征体内细胞所处的力学微环境。
发明内容
(一)要解决的技术问题
针对上述问题,本发明提供了一种柔性基底的细胞细菌互作模型的制备方法及应用,用于至少部分解决传统方法对于细胞细菌互作的体外研究中缺失了基质刚度因素这一重要物理条件的技术问题。
(二)技术方案
本发明一方面提供了一种柔性基底的细胞细菌互作模型的制备方法,包括:S1,丙烯酰胺和双丙烯酰胺在四甲基乙二胺和过硫酸铵的作用下,聚合形成一定基质刚度的聚丙烯酰胺凝胶,基质刚度模拟自生物组织的刚度;S2,在聚丙烯酰胺凝胶上孵育微环境物质,用于模拟细胞所处的微环境;S3,在聚丙烯酰胺凝胶界面上接种宿主细胞,加入互作的细菌建立细胞细菌互作模型。
进一步地,一定基质刚度的聚丙烯酰胺凝胶包括单一基质刚度的聚丙烯酰胺凝胶和不同刚度变化的聚丙烯酰胺凝胶。
进一步地,S2之后还包括:在聚丙烯酰胺凝胶表面装载荧光粒子,荧光粒子的直径范围为200~500nm。
进一步地,丙烯酰胺的浓度范围为10%~20%,双丙烯酰胺的浓度范围为0.2%~6%。
进一步地,聚丙烯酰胺凝胶基质刚度的范围为0~150kPa。
进一步地,S3之前还包括:在聚丙烯酰胺凝胶表面建立不同的微图案。

本发明另一方面提供了一种根据前述柔性基底的细胞细菌互作模型的制备方法得到的柔性基底的细胞细菌互作模型在确定细胞细菌互作与细胞外基质刚度之间的调控关系的应用。
本发明还有一方面提供了一种根据前述柔性基底的细胞细菌互作模型的制备方法得到的柔性基底的细胞细菌互作模型在抗菌药物治疗胞内细菌感染药效的检测方法的应用。
(三)有益效果
本发明实施例提供的一种柔性基底的细胞细菌互作模型的制备方法及应用,不同于以往的毫不考虑物理力学因素的坚硬的塑料或玻璃底的细胞细菌互作模型,该细胞细菌互作模型建立于单一基质刚度变化和连续基质刚度梯度变化的聚丙烯酰胺水凝胶界面上,充分展现了细胞外基质刚度的变化对于细胞细菌互作研究中的作用。

具体实施方式
为使本发明的目的、技术方案和优点更加清楚明白,以下结合具体实施例,并参照附图,对本发明进一步详细说明。
本发明的第一实施例提供了一种柔性基底的细胞细菌互作模型的制备方法,包括:S1,丙烯酰胺和双丙烯酰胺在四甲基乙二胺和过硫酸铵的作用下,聚合形成一定基质刚度的聚丙烯酰胺凝胶,基质刚度模拟自生物组织的刚度;S2,在聚丙烯酰胺凝胶上孵育微环境物质,用于模拟细胞所处的微环境;S3,在聚丙烯酰胺凝胶界面上接种宿主细胞,加入互作的细菌建立细胞细菌互作模型。
通过不同浓度配比聚丙烯酰胺水凝胶形成单一基质刚度,如表1所示,丙烯酰胺单体(Acrylamide)和少量交联剂双丙烯酰胺(Bis-acrylamide)通过化学催化剂过硫酸铵,加速剂四甲基乙二胺(TEMED)聚合形成三维空间的高聚物,聚合后的聚丙烯酰胺凝胶形成网状结构。不同丙烯酰胺配比形成的对应刚度如表1所示(0-150kPa),该基底刚度用以模拟脏器组织力学微环境(如图3)。基质上孵育生物学微环境物质,如基质蛋白(胶原蛋白I或纤维蛋白等),多聚赖氨酸,多巴胺等用以模拟不同生物学微环境。

如表1所示,想形成如第一列所示的弹性模量的聚丙烯酰胺(PAAm)水凝胶,例如,想制作10kPa的柔性基底PAAm水凝胶,则按照10.14kPa的所对应的那一行的数据,分别加入纯水丙烯酰胺,少量交联剂双丙烯酰胺,10%APS和TEMED混合,滴加到玻片上,形成柔性水凝胶基底。
柔性水凝胶基底形成后,按照S2步骤说述,首先按照图5的步骤,在水凝胶上装载荧光粒子,然后进一步在表面孵育细胞外基质蛋白,如胶原蛋白I,纤连蛋白等,模拟类组织的微环境。荧光粒子的载入是为了后期测定细胞力所用。按照S3步骤所述,进一步在完成装载的水凝胶上接种上皮细胞系,如大鼠肠隐窝上皮细胞系(IEC-6),人肠上皮细胞系(HIEC)等,待细胞形成单层后,加入GFP荧光标记的细菌,如蜡样芽孢杆菌(NVH0075/95),大肠杆菌(ATCC25922)或金黄色葡萄球菌(ATCC29213)等菌感染细胞2h,感染后进行下一步的试验分析。
在上述实施例的基础上,一定基质刚度的聚丙烯酰胺凝胶包括单一基质刚度的聚丙烯酰胺凝胶和不同刚度变化的聚丙烯酰胺凝胶。
本发明制备的细胞细菌互作模型中的力学微环境不仅包括了单一基质刚度变化的水凝胶基质还包括了不同刚度变化的水凝胶基质,以及不同微图案模型的基质。可以先建立单一基质刚度变化的水凝胶界面,该基质刚度模拟宿主体内细胞基质刚度(<100kPa),研究细菌和细胞互作过程中,细菌侵染细胞的规律;还可以建立另一具有不同刚度变化的聚丙烯酰胺水凝胶界面,此外通过ECM蛋白的孵育,还可以改变该界面上细胞细菌互作位点。不同刚度变化的水凝胶界面更接近于细胞细菌互作时所处的细胞外基质刚度环境,例如肠道组织某些部分刚度截然不同,肠道隐窝处的刚度比其他肠道细胞部分要硬。在病理条件下,炎症组织表面比炎症组织表面更硬。故不同刚度界面上建立的细胞细菌互作模型可以更精准的研究细菌在体内力学环境控制下与细胞互作的规律。以水凝胶表面建立微图案为基础的细胞细菌互作模型,可进一步研究不同微图案中细胞单层形成对于细胞细菌互作的影响,也可以检测到互作过程中细胞力学的变化。
基于不同刚度的聚丙烯酰胺水凝胶界面的细胞细菌互作模型,可以有序性的调控基质刚度梯度的变化跨度,更有益于建立以细胞外基质刚度变化为基础的细胞细菌互作模型。这些都为一种以细胞外基质刚度变化为特性的细胞细菌互作模型的制备和应用奠定了夯实的基础。
在上述实施例的基础上,在聚丙烯酰胺凝胶表面装载荧光粒子,荧光粒子的直径范围为200~500nm。
聚丙烯酰胺水凝胶形成后,在表面修饰荧光粒子。如图5所示,主要操作步骤为取1μl 200nm直径的荧光粒子(羧基修饰)加入1000μl纯水。每一块胶上加250μl,孵育40min,纯水清洗2次。取122μl Sulfo-NHS(N-羟基硫代琥珀酰亚胺);152μl EDC(二氯乙烷);726μlMES【2-(N-吗啉)乙磺酸】缓冲液,混匀,每一块胶上加250μl,孵育2h,PBS清洗3次,加PBS浸泡2h后,备用即可。
凝胶基质装载荧光粒子用以测定细胞力大小。简单的来说,在检测细胞牵引力的时候,我们通过荧光显微镜,同时观测荧光粒子的变化,拍摄一段时间的荧光粒子的图片,这时候如果细胞的牵引力有变化,荧光粒子则会随着细胞的力的变化而发生位移场的改变,最后,利用氢氧化钠或者胰酶将柔性基底上的细胞除掉,恢复原始的荧光粒子的位移场的图。根据这张图来对比之前的每一张图,通过位移场的变化,通过metlab软件,进一步计算牵引力的大小。
首先使用甲醇清洁盖玻片以去除灰尘,然后用溶解在乙酸中的终浓度为4%(v/v)的3-(三甲氧基甲硅烷基)甲基丙烯酸丙酯活化20分钟,然后在去离子水中进行浸泡洗涤三次,对吼在化学通风橱中风干20分钟。将活化的玻片在0.5%的戊二醛溶液中固定30分钟,用去离子水洗涤3次,最后后在室温下干燥20分钟备用。在制备水凝胶之前,事先准备好包含丙烯酰胺,双丙烯酰胺过硫酸铵(APS),四甲基乙二胺(TEMED)(Sigma-Aldrich)的预聚物溶液,在前面已经活化并固定好的干燥玻片上,滴加25~30μl的预聚物,用印有pattern的干净的玻片,盖上预聚物,形成水凝胶。最后按图5所示的方式将直径200nm的红色荧光微珠(稀释至1∶125,F8810,ThermoFisher)装载在水凝胶表面。
在上述实施例的基础上,丙烯酰胺的浓度范围为10%~20%,双丙烯酰胺的浓度范围为0.2%~6%。
聚丙烯酰胺水凝胶的刚度主要与丙烯酰胺和双丙烯酰胺的浓度的浓度有关,在该浓度范围内可以得到0~150kPa的类组织刚度的水凝胶。通常情况下,丙烯酰胺的浓度越高,水凝胶的刚度越大,丙烯酰胺的浓度与具体杨氏模量刚度之间的关系参照表1。
在上述实施例的基础上,聚丙烯酰胺凝胶基质刚度的范围为0~150kPa。
细菌感染是一项涉及细胞与细菌相互作用的复杂力学生物学过程。细胞外基质不仅具有生物学调控特性更兼具力学调控能力。细菌细胞互作过程中,基质刚度对宿主细胞屏障功能的调节是调控细菌感染的关键。然而,以往的对于细胞细菌互作的体外研究中缺失了基质刚度因素这一重要物理条件。本发明结合宿主细胞在体内各组织脏器的刚度大小,例如脑组织中的细胞弹性模量为0.3~1kPa,皮肤组织为3~7kPa,心肝脾肺肾脏器组织均小于20kPa,肠组织为10~20kPa,牙龈组织为100kPa左右,设定了聚丙烯酰胺凝胶基质刚度的范围为0~150kPa,以更贴近于细菌感染宿主细胞时所处的力学微环境。
在上述实施例的基础上,S3之前还包括:在聚丙烯酰胺凝胶表面建立不同的微图案。
按照表1所示,用聚丙烯酰胺(PAAm)水凝胶制备具有不同刚度的弹性基材。图6为同一水凝胶界面上实现连续基质刚度梯度变化的细胞细菌互作模型。a,建立不同微图案的形成,有助于细胞形成不同形状。b,在不同刚度水凝胶界面上接种宿主细胞,以形成规则的微图案单层细胞。c,加入互作的细菌完成细胞细菌互作模型,同时检测细胞细菌互作中细胞的牵引力的变化。d,在连续基质刚度梯度水凝胶界面上建立的细胞细菌互作模型中加入治疗的抗菌药物,然后采用LC-MS/MS的方法检测抗菌药物的分布,用以评估抗菌药物的药效。
在微阵列上形成高通量水凝胶基底,按照图6a的步骤,采用微接触印刷技术可形成圆形水凝胶基底,接入细胞后则形成如图6b所示的高通量的圆形细胞单层,具体操作步骤如下:
(1)准备有100,300,600μm直径圆形patterns的PDMS印章;
(2)将印章有pattern的一面向上,UV下处理7~10min(亲水处理);
(3)PDMS印章上加胶原蛋白(0.2mg/ml)与高碘酸钠(20mM)(1∶1)常温下孵育两个小时;
(4)将孵育后PDMS印章印在晾干后的聚丙烯酰胺凝胶上,盖印至少一个小时,勿动;
(5)浸入0.3%聚醚(Polyethylene-polypropylene glycol,F127),中固定,并在紫外下照射30min,即可用于细胞培养。
基于已建立好的微阵列上皮细胞单层,用转载内源性荧光蛋白(GFP)或酸性指示剂(pHrodo)着色的细菌侵染上皮细胞单层,设定不同的侵染比率和侵染时间,以构建细胞细菌感染互作模型(如图6b和6c)。通过流式细胞术(FACS)检测被感染的细胞数以及通过平板计数法检测细菌入侵的总量(CFU/ml),同时拟合细菌和细胞感染受基质刚度调控的数学关系,计算感染的上皮细胞比率(细菌数比上细胞数量),分析细菌侵染规律。
在上述实施例的基础上,S3之后还包括:通过流式细胞术检测被感染的细胞数以及通过平板计数法检测细菌入侵的总量。
图3为不同基质刚度下细胞细菌感染互作模型及感染检测手段。通过聚丙烯酰胺凝胶建立不同基质刚度的上皮细胞培养模型,利用流式细胞术(FACS)和细胞内菌落计数(CFU)分别检测细胞感染总数(Numbers of infected cells,NIC)和细菌侵染总量(Numbersof internalized bacteria,NIB)。
图4为基质刚度调控细菌感染细胞的规律。a,柔性基底刚度pattern形成示意图。b,感染的上皮细胞数量(Numbers of infected cells,NIC)。检测上皮细胞在不同硬度基质上。将IEC-6细胞置于水凝胶中,并孵育I型胶原蛋白。然后,以MOI为100,在单层膜上感染细菌(B.cereus NVH0075/95)2小时。侵袭到细胞内的细菌总数(Numbers of invasivebacteria,NIB、log10CFU/ml)。
通过用于检测细胞细菌互作模型中的揭示细胞感染率的流式细胞术以及评估感染细菌总量的菌落平板计数法进一步为细胞细菌互作规律展开研究。例如细胞外基质刚度可以有效地调控细胞细菌互作模式,越硬的基质刚度越有利于感染细胞的扩散,而越软的基质刚度能够促进入侵细胞的细菌总量增多,这两种规律都能和细胞外基质刚度值建立良好的数学线性关系(如图4)。进而不同刚度的变化对于细胞细菌互作模型是为了更贴近于细菌感染宿主细胞时所处的力学微环境。因为不同组织细胞的基质刚度并不是均一的,例如处于肠道隐窝处的肠道上皮细胞所处的基质刚度就比其他细胞要硬的多,而炎症组织处的细胞基质刚度也是较硬的。因此本发明通过可控性的调控连续基质刚度梯度,为建立更精准的力学微环境下的细胞细菌互作平台奠定基础。
在上述实施例的基础上,S3之后还包括:采用共聚焦激光扫描显成像确定细胞形态和牵引力显微镜分析法测定细胞牵引力。
对微阵列细胞进行荧光染色和细胞力测定分别分析微阵列上的单层上皮细胞的细胞形态和牵引力大小以确保微阵列上皮细胞单层建立成功。同时,采用共聚焦激光扫描显成像系统(Confocal laser scanning microscopy,CLSM)确定细胞形态和牵引力显微镜分析法(Traction force microscopy,TFM)测定细胞牵引力,以确保成单一基质刚度变化的细胞细菌互作模型的建立。单一基质刚度变化的细胞单层的形成,主要依赖于检测细胞是否在微pattern上面形成了,如印章图案一样的紧密的细胞单层。基底刚度越大,细胞的牵引力越大,细胞形态也是随刚度增加而增大。
本发明的第二实施例提供了使用柔性基底的细胞细菌互作模型在确定细胞细菌互作与细胞外基质刚度之间的调控关系的应用。如图4所示,通过该体系建立的细菌细胞互作模型,结果发现,随着细胞外基质刚度的增加,感染的细胞数量逐渐增多,而感染的细菌量减少,进一步的说明,组织刚度的增强,会导致细菌的感染从聚集状态逐步向扩散态转化。
比较不同软硬基质刚度上的单层上皮细胞,比较细胞遭受细菌侵染前后的细胞牵引力(Traction force)的变化。采用TFM(细胞牵引力显微镜)测定细胞牵引力,并且分析软硬基质对细胞牵引力的影响,细胞牵引力随感染时间的变化关系,以及感染点和未感染点细胞牵引力的变化。除此之外同时分析单细胞牵引力的变化。最终揭示基质刚度影响细菌侵染细胞的时空变化规律。通过显微成像技术定性的观测不同基质刚度上感染的细胞的形态学变化,进一步比较分析不同基质上感染细胞和未感染细胞的差别。定量检测分析细胞骨架力相关蛋白,如actin的动态变化(F-actin和G-actin)。利用骨架蛋白抑制和稳定药物,单因素分析基质刚度和骨架蛋白actin的相互作用,阐明基质刚度调控细菌侵染细胞的力学机制。
本发明的第三实施例提供了使用柔性基底的细胞细菌互作模型在抗菌药物治疗胞内细菌感染药效的检测方法的应用。我们通过该细菌细胞互作感染模型,检测了环丙沙星和四环素,对于细胞内化菌的治疗。如图6所示,在完成a和b步骤后,基底上已经形成了上皮细胞单层,这时按照c步骤所示,感染细菌,感染2h后,按照d的步骤加入治疗的10倍该细菌最小抑菌浓度(MIC)的环丙沙星和四环素,治疗6h,然后通过图3所示的两种方法检测感染细胞数量NIC和感染细菌量NIB,同时如图6d的右侧所示,检测细胞内的抗生素浓度。共同评估治疗效果,结果发现,无论是环丙沙星还是四环素,相比于硬的基底93.46kPa,都是在10.14kPa的基底上的治疗效果更好。
单一变化的基质刚度界面的细胞细菌互作模型和不同刚度变化的细胞细菌互作模型,均可以进一步扩展为研究抗菌药物在以基质刚度变化的水凝胶界面为载体的细胞细菌互作模型中的药效作用。本发明为抗菌药物治疗细胞内细菌感染提供了实效的细胞细菌互作平台,为筛选有效抗胞内菌入侵和增殖的抗菌药物提供筛选平台。
基于建立的高通量微阵列细胞感染模型,选取有效的抗生素药物进行治疗,根据细菌入侵量和细胞感染比率评估抗生素治疗效能,并给出药物有效治疗时间和给药浓度浓度。采用液相色谱质谱联用技术(LC-MS/MS)进一步分析抗生素药物分布,评价抗菌药物的治疗效能(如图6d)。
基于细菌侵染规律评估抗菌药物药效并制定合理的给药方案结合细胞外基质刚度调控细菌入侵细胞的力学规律和生物学机制的研究,以及对不同抗菌药物药效评价,并结合给药浓度和给药时间合理制定不同脏器细胞感染的的抗菌治疗策略。
本发明优于其他建立在坚硬的塑料底或玻璃底的细胞培养板/皿上的细胞细菌互作模型,更接近于宿主细胞所处的真实力学微环境。单一基质刚度变化的水凝胶界面建立的细胞细菌互作模型,可以通过有效且直接的建立基质刚度与细菌侵染之间的数学关系;不同基质刚度变化的水凝胶界面建立的细胞细菌互作模型是基于宿主细胞体内生存的微环境的刚度变化来设定。以细胞外基质刚度变化为特性的细胞细菌互作模型,更有益于抗菌药物的筛选,在该模型中不仅可以精准的检测到在模型中药物浓度的分布,还可以通过药物浓度进入细胞的药效来评估和筛选抗菌药物。

1.一种柔性基底的细胞细菌互作模型的制备方法,包括:
S1,丙烯酰胺和双丙烯酰胺在四甲基乙二胺和过硫酸铵的作用下,聚合形成一定基质刚度的聚丙烯酰胺凝胶,所述基质刚度模拟自生物组织的刚度;
S2,在所述聚丙烯酰胺凝胶上孵育微环境物质,用于模拟细胞所处的微环境;
S3,在所述聚丙烯酰胺凝胶界面上接种宿主细胞,加入互作的细菌建立细胞细菌互作模型。
2.根据权利要求1所述的柔性基底的细胞细菌互作模型的制备方法,其特征在于,所述一定基质刚度的聚丙烯酰胺凝胶包括单一基质刚度的聚丙烯酰胺凝胶和不同刚度变化的聚丙烯酰胺凝胶。
3.根据权利要求1所述的柔性基底的细胞细菌互作模型的制备方法,其特征在于,所述S2之后还包括:在所述聚丙烯酰胺凝胶表面装载荧光粒子,所述荧光粒子的直径范围为200~500nm。
4.根据权利要求1所述的柔性基底的细胞细菌互作模型的制备方法,其特征在于,所述丙烯酰胺的浓度范围为10%~20%,所述双丙烯酰胺的浓度范围为0.2%~6%。

9.一种根据权利要求1~8任一所述的柔性基底的细胞细菌互作模型的制备方法得到的柔性基底的细胞细菌互作模型在确定细胞细菌互作与细胞外基质刚度之间的调控关系的应用。
10.一种根据权利要求1~8任一所述的柔性基底的细胞细菌互作模型的制备方法得到的柔性基底的细胞细菌互作模型在抗菌药物治疗胞内细菌感染药效的检测方法的应用。
