Abstract
本发明公开了复合Fe3O4‑GO‑ASA的PLA微泡及其制备方法;属于生物医疗领域。本发明提供了一种集靶向造影、辅助抑制血栓功能于一体的多功能造影剂及其制备方法。本发明是由PLA微泡以及PVA附着在PLA微泡表面形成壳层构成的;所述PLA微泡是由PLA膜层构成的微泡壁,Fe3O4‑GO‑ASA复合物镶嵌于PLA膜层內以及PLA膜层的内壁上,并且PLA微泡內包裹有惰性气体,其中,所述Fe3O4‑GO‑ASA复合物是在GO表面沉积Fe3O4后利用π‑π吸附负载ASA得到的。本发明制得的复合Fe3O4‑GO‑ASA的PLA微泡可以有效延长凝血时间,阻止血栓的形成,可以达到抗凝血的辅助治疗效果。

技术领域
本发明属于生物医疗领域;具体涉及复合Fe3O4-GO-ASA的PLA微泡及其制备方法。
背景技术
随着人口老龄化的加剧,血栓性疾病的患者日渐增多。血栓性疾病有很多,通常分为动脉性的血栓性疾病、静脉性的血栓性疾病,还有毛细血管性的血栓性疾病。其中常见的冠心病、脑梗塞、中风等,都是人类生命安全健康的“头号杀手”,据统计,全球每年有超过一千万人因血栓疾病而死。在诊断血栓性疾病的过程中,时间是非常宝贵的,因此,就需要一种集快捷、安全、准确于一体的诊断方法。超声成像(CEUS)是血栓症状评估的一线方法,它的广泛应用基于许多优点,除了必须条件,超声造影剂的优点还包括成本低、可重复性好、耐受性好、无禁忌症等。CEUS不仅可用于检测血栓症状,还可用于溶解血栓。有研究表明,CEUS治疗可有效溶解大血栓阻塞形成的冠状动脉、大脑大动脉和外周动脉的血栓,然而这种溶解方法是否会导致新的栓塞或者引起血管硬化区域破损等问题还有待进一步研究。
化疗是主要的癌症治疗方法,大多数癌症疾病都需要依赖化疗进行治疗。然而,由于其中细胞毒性药物的全身性副作用,如脱发、恶心、心脏毒性和肝损伤等,当今化疗的效率还有待提升,这大大限制了其在癌症治疗中的应用。因此,研发一种改善细胞摄取和减少副作用的药物传递方式成了该领域的首要问题。近年来研究表明,基于纳米材料的药物载体是最具潜力的药物载体,因为它们具有更高的负载能力和释放能力,经过修饰后还可以完成高效的靶向递送。基于氧化石墨烯的纳米药物载体已经成为一种可行的、可控的药物传递系统。GO作为传递系统的优势在于较大的表面积比,以及表面所含有大量的含氧官能团,如羧基、羟基和环氧基等都可以增加药物载荷和水溶性。
Hussien等人开发出了一种果胶修饰的磁性GO递药体系,用于紫杉醇的靶向传递。实验结果表明,研发得到的GO递药体系具有良好的生物相容性和优越的载药性能,而且在癌细胞系中的药物释放率大于正常细胞系中的药物释放率。此外,细胞毒性试验表明,合成的纳米复合物具有生物相容性,给药后的细胞具有很高的相对细胞活力。根据这些发现,这种果胶修饰的磁性GO递药体系有望在未来的癌症药物载体配方中发挥重要作用。
基于以上所述,氧化石墨烯以其优秀的性能、独特的结构和稳定的性质在生物研究领域受到广泛的重视,有关氧化石墨烯的科学研究也在世界范围内火热开展。在最近十年,氧化石墨烯的科学研究已经取得了一定的进展,然而将氧化石墨烯应用于生物医疗领域的研究还仅仅停留于初步阶段,若想将其应用于实际使用,仍有大量的实验工作需要完成。
发明内容
本发明目的是提供一种集靶向造影、辅助抑制血栓功能于一体的多功能造影剂及其制备方法。
为解决上述技术问题,本发明中复合Fe3O4-GO-ASA的PLA微泡是由PLA微泡以及PVA附着在PLA微泡表面形成壳层构成的;所述PLA微泡是由PLA膜层构成的微泡壁,Fe3O4-GO-ASA复合物镶嵌于PLA膜层內以及PLA膜层的内壁上,并且PLA微泡內包裹有惰性气体,其中,所述Fe3O4-GO-ASA复合物是在GO表面沉积Fe3O4后利用π-π吸附负载ASA得到的;具体是按通过下述步骤实现的:
步骤一、采用原位沉积法在GO表面沉积Fe3O4,得到Fe3O4-GO复合物;
步骤二、然后利用π-π吸附负载ASA,得到Fe3O4-GO-ASA复合物;
步骤三、然后通过双乳化溶剂挥发法将Fe3O4-GO-ASA复合物包裹于PLA微泡中,第一次超声乳化时,PLA溶液破坏成包裹着Fe3O4-GO-ASA内水相的小液泡,由于超声破碎过程中持续通入惰性气体,所以小液泡中也会含有惰性气体,然后转入PVA溶液中搅拌均匀并进行二次超声乳化,PVA会附着在所形成的PLA微泡表面形成坚硬的壳层,经过真空冻干,即得到复合Fe3O4-GO-ASA的PLA微泡。
进一步限定,所述惰性气体为氮气;
进一步限定,步骤一所述原位沉积法在GO表面沉积Fe3O4是按下述操作进行的:
将Fe2(SO4)3和FeSO4·7H2O按照Fe2+:Fe3+摩尔浓度比为1:1.5的比例混合,在20mL双蒸水中配制成铁盐总浓度为0.3mol·L-1的溶液,分别将质量为0.058~0.29g的GO超声分散于30mL双蒸水,在搅拌条件下加到上述铁盐溶液中,加热至体系温度达到60℃时,滴加0.25mol·L-1NaOH溶液直至过量,搅拌1h,反应完成后,将沉淀物反复水洗抽滤后,真空冻干,得到Fe3O4-GO复合物。
进一步限定,步骤二中Fe3O4-GO-ASA复合物的制备是按下述步骤进行的:将0.5gFe3O4-GO复合物置于50mL无水乙醇得到Fe3O4-GO分散液,将0.5g ASA加入到该分散液中,超声处理1min~2min,使其充分溶解,在20℃下搅拌2h,微孔滤膜抽滤,沉淀经无水乙醇洗涤、抽滤、真空冻干,即得Fe3O4-GO-ASA复合物。
进一步限定,步骤三具体步骤如下:
称取3.5g聚乙烯醇(PVA)于100mL双蒸水中浸泡3h,之后90℃水浴条件搅拌至完全溶解,保持搅拌状态备用,得到PVA溶液;
将0.6g聚乳酸(PLA)加到12mL二氯甲烷(DCM)中,浸泡5h至完全溶解,得到有机相;
取Fe3O4-GO-ASA复合物0.1g加到0.2mL乙醇水溶液,作为内水相;
向有机相加入内水相,以25kHz频率、33W功率,在一定开关频率下进行超声乳化,在反应过程中向体系中持续通入惰性气体,制得W/O乳液(初乳);
将初乳逐滴加到PVA溶液中,搅拌3min;以25kHz频率、55W功率,在一定开关频率下进行二次超声乳化,制得W/O/W乳液(复乳);
向复乳中加入5.5%异丙醇溶液100mL,室温搅拌5h,挥发有机溶剂,待挥发完毕后,用双蒸水反复离心洗涤剩余溶液,收集下层微泡,真空冻干后即为复合Fe3O4-GO-ASA的PLA微泡。
进一步限定,PLA微泡最佳内水相与有机相体积比为1:60,初乳制备过程中最佳开关时间为4s开2s关,复乳制备过程中最佳开关时间为4s开2s关。
进一步限定,上述氧化石墨烯的制备是通过下述步骤完成的:
先将100mL三颈烧瓶洗净、烘干、封口并置于冰箱中冷却;
取鳞片石墨粉1.5g,置于冷却后的三口瓶中,并加入30mL浓硫酸,在冰浴条件下缓慢搅拌,混合均匀,该体系温度保持在5℃~8℃,向体系中缓慢分次加入高锰酸钾4.5g,持续搅拌2.5h,混合液呈墨绿色;
然后将体系转移到38℃水浴条件下继续搅拌1h,再将体系转入到装有280mL去离子水烧杯中,搅拌5min,此时溶液变为褐色,最后加入60mL质量浓度为30%的H2O2溶液,此时混合液剧烈沸腾并伴随大量气泡,溶液颜色由褐色转变为金黄色,反应停止后在室温下继续搅拌24h;
将上步得到产物在5000r·min-1条件下反复离心水洗,除去残酸,洗至pH>6,并用BaCl2溶液检测无硫酸根离子为止;
将所得沉淀分散于去离子水中,在超声功率为30%条件下破碎10min,50℃下烘干即得到氧化石墨烯。
本发明方法所制备的Fe3O4纳米粒子磁学性能优良,可以作为磁性靶头使用。
本发明制得的复合Fe3O4-GO-ASA的PLA微泡具有良好的磁学性能、稳定性和GO包封率,尺寸满足超声造影剂的体循环需求。
本发明复合Fe3O4-GO-ASA的PLA微泡呈规则球体,内部为中空结构,个体边界明显,无畸形或互相融合现象。
本发明制得的复合Fe3O4-GO-ASA的PLA微泡可以有效地增强超声造影图像的辨识度,并可以在外加磁场作用下富集在目标区域,进一步增加超声造影图像的清晰度,具有良好的增强超声成像效果,且其中的Fe3O4和GO不影响超声造影效果。
本发明制得的复合Fe3O4-GO-ASA的PLA微泡可以在磁场作用下在目标区域富集,从而增强目标区域的超声成像效果,达到靶向造影和靶向辅助抑制的效果。
本发明制得的复合Fe3O4-GO-ASA的PLA微泡可以有效延长凝血时间,阻止血栓的形成,可以达到抗凝血的辅助治疗效果。
本发明制得的复合Fe3O4-GO-ASA的PLA微泡安全范围大,最大耐受剂量为临床成人日用量的1406.25倍,其在体内的代谢过程符合一般生物相容性材料在体内的降解过程,不具有致敏原性,也不导致溶血和红细胞凝集,与机体组织无不良反应,无致敏性,无溶血现象。

实施例1:本实施例中的复合Fe3O4-GO-ASA的PLA微泡的制备方法是按通过下述步骤实现的:
步骤一、Fe3O4-GO复合物的制备:
将Fe2(SO4)3和FeSO4·7H2O按照Fe2+:Fe3+摩尔浓度比为1:1.5的比例混合,在20mL双蒸水中配制成铁盐总浓度为0.3mol·L-1的溶液,将质量为0.174g的GO超声分散于30mL双蒸水,在搅拌条件下加到上述铁盐溶液中,加热至体系温度达到60℃时,滴加0.25mol·L- 1NaOH溶液直至过量,搅拌1h,反应完成后,将沉淀物反复水洗抽滤后,真空冻干,得到在不同铁盐和GO比例下制备的Fe3O4-GO复合物。
步骤二、Fe3O4-GO-ASA复合物的制备
将0.5gFe3O4-GO复合物置于50mL无水乙醇得到Fe3O4-GO分散液,将0.5g ASA加入到该分散液中,超声处理1min~2min,使其充分溶解。在20℃下搅拌2h,微孔滤膜抽滤,沉淀经无水乙醇洗涤、抽滤、真空冻干,即得Fe3O4-GO-ASA复合物。
步骤三、复合Fe3O4-GO-ASA的PLA微泡的制备:称取3.5g聚乙烯醇(PVA)于100mL双蒸水中浸泡3h,之后90℃水浴条件搅拌至完全溶解,保持搅拌状态备用,得到PVA溶液。
将0.6g聚乳酸(PLA)加到12mL二氯甲烷(DCM)中,浸泡5h至完全溶解,得到有机相;
取Fe3O4-GO-ASA复合物0.1g加到0.2mL乙醇水溶液,作为内水相,乙醇的水溶液是由乙醇与蒸馏水按1:4的体积比配置的;
以25kHz频率、33W功率,在一定开关频率下进行超声乳化,在反应过程中向体系中持续通入N2,制得W/O乳液(初乳)。
将初乳逐滴加到PVA溶液中,搅拌3min。以25kHz频率、55W功率,在一定开关频率下进行二次超声乳化,制得W/O/W乳液(复乳)。
向复乳中加入5.5%异丙醇溶液100mL,室温搅拌5h,挥发有机溶剂。待挥发完毕后,用双蒸水反复离心洗涤剩余溶液,收集下层微泡,真空冻干后即为PLA微泡,
在初乳制备过程中,控制初乳超声开关时间为4s开2s关;在复乳制备过程中,控制超声开关时间为4s开2s关。
其中,本实施例所用的氧化石墨烯的制备是通过下述步骤完成的:
先将100mL三颈烧瓶洗净、烘干、封口并置于冰箱中冷却;
精密称取鳞片石墨粉1.5g,置于冷却后的三口瓶中,并加入30mL浓硫酸,在冰浴条件下缓慢搅拌,混合均匀。该体系温度保持在5℃~8℃,向体系中缓慢分次加入高锰酸钾4.5g,持续搅拌2.5h,混合液呈墨绿色。然后将体系转移到38℃水浴条件下继续搅拌1h,再将体系转入到装有280mL去离子水烧杯中,搅拌5min,此时溶液变为褐色,最后加入60mL浓度为30%的H2O2溶液,此时混合液剧烈沸腾并伴随大量气泡,溶液颜色由褐色转变为金黄色,反应停止后在室温下继续搅拌24h;
将上步得到产物在5000r·min-1条件下反复离心水洗,除去残酸,洗至pH>6,并用BaCl2溶液检测无硫酸根离子为止;
将所得沉淀分散于去离子水中,在超声功率为30%条件下破碎10min,50℃下烘干即得到氧化石墨烯。
关于PLA微泡的制备
称取3.5g聚乙烯醇(PVA)于100mL双蒸水中浸泡3h,之后90℃水浴条件搅拌至完全溶解,保持搅拌状态备用。将0.6g聚乳酸(PLA)加到12mL二氯甲烷(DCM)中,浸泡5h至完全溶解,得到有机相。向有机相加入一定量内水相,即乙醇的水溶液(乙醇与蒸馏水体积比为1:4)。以25kHz频率、33W功率,在一定开关频率下进行超声乳化,在反应过程中向体系中持续通入N2,制得W/O乳液(初乳)。将初乳逐滴加到PVA溶液中,搅拌3min。以25kHz频率、55W功率,在一定开关频率下进行二次超声乳化,制得W/O/W乳液(复乳)。向复乳中加入5.5%异丙醇溶液100mL,室温搅拌5h,挥发有机溶剂。待挥发完毕后,用双蒸水反复离心洗涤剩余溶液,收集下层微泡,取少量沉淀分散于去离子水中,用于测定粒度和Zeta电位,其余部分真空冻干后即为PLA微泡。
在初乳制备过程中,分别加入0.05mL、0.10mL、0.15mL、0.20mL和0.25mL蒸馏水作为内水相,控制初乳超声开关时间分别为4s开3s关、4s开2s关和4s开1s关;在复乳制备过程中,控制超声开关时间分别为4s开3s关、4s开2s关和4s开1s关。
本实施例制得的Fe3O4纳米粒子形貌为球形,平均粒径为65.15nm,饱和磁强度为63.5emu·g-1。对于原位沉积法制备的Fe3O4-GO复合物,GO表面的Fe3O4纳米粒子分布均匀且不占据过多空间。Fe3O4-GO-ASA复合物的ASA负载率为7.3%,其饱和磁强度为37.7emu·g-1。
图1、2、3分别为复合Fe3O4-GO-ASA的PLA微泡SEM、粒径分布和Zeta电位。由图1可见,所制得的复合Fe3O4-GO-ASA的PLA微泡表面光滑、大小较为均一,没有互相融合或者残缺现象。由图2可见,制得的复合Fe3O4-GO-ASA的PLA微泡平均粒径为479.1nm,分布区间为200nm~700nm,满足临床上对微泡超声造影剂的体循环尺寸要求。其中,微泡呈不规则球体,这可能是因为复合微泡在冷冻干燥过程中引起的,在溶液中则更接近球形。由图3可知,复合Fe3O4-GO-ASA的PLA微泡的Zeta电位为(-36.5±10.0)mV,说明复合微泡水溶液具有较好的分散体稳定性。
本实施例制备的PLA微泡平均粒径为252.7nm;复合Fe3O4-GO-ASA的PLA微泡的平均粒径为479.1nm,Zeta电位为(-36.5±10.0)mV,饱和磁强度为19.4emu·g-1,对GO的包封率为75.5%;复合Fe3O4-GO-ASA的PLA微泡呈规则球体,内部为中空结构,边界明显,无畸形或互相融合现象。
图4为制得的复合Fe3O4-GO-ASA的PLA微泡的能谱图。除了能谱图无法扫描出的H元素,复合Fe3O4-GO-ASA的PLA微泡由C、O和Fe元素组成,这是因为GO、ASA、PLA、PVA和异丙醇等提供C、O元素,Fe3O4提供Fe元素。由图4(a)(b)(c)可见,C、O和Fe元素成散点状分布,分散良好,无团聚现象。
图5为Fe3O4、Fe3O4-GO复合物和复合Fe3O4-GO-ASA的PLA微泡的磁滞曲线。其中,复合Fe3O4-GO-ASA的PLA微泡具有19.4emu·g-1的饱和磁强度。由图5可见,Fe3O4、Fe3O4-GO复合物和复合Fe3O4-GO-ASA的PLA微泡磁滞曲线类似,且矫顽力均为0Oe,说明三者磁性行为类似,且在零点处均未见矫顽磁场和剩磁,这表明三者均具有顺磁性特征,可以在外加磁场条件下富集。另外,Fe3O4、Fe3O4-GO复合物和复合Fe3O4-GO-ASA的PLA微泡比饱和磁化强度依次减弱,这是因为相同质量的三种物质中Fe3O4的含量依次减少,说明Fe3O4-GO复合物和复合Fe3O4-GO-ASA的PLA微泡的磁性均来源于内部的Fe3O4。
复合Fe3O4-GO-ASA的PLA微泡体内超声造影的最佳浓度为60mg·mL-1,复合微泡中的Fe3O4和GO不影响超声造影效果。复合Fe3O4-GO-ASA的PLA微泡可以在磁场作用下在目标区域富集,从而增强目标区域的超声成像效果,达到靶向造影和靶向辅助抑制血栓形成的效果。
注射复合Fe3O4-GO-ASA的PLA微泡后,兔血样的PT、TT、APTT和INR指标均随复合微泡浓度的上升不同程度地增加,这说明复合微泡可以提升血栓因子的游离性,具有抗凝血的辅助治疗作用。
复合Fe3O4-GO-ASA的PLA微泡安全范围大,最大耐受剂量为临床成人日用量的1406.25倍,其在体内的代谢过程符合一般生物相容性材料在体内的降解过程,不具有致敏原性,也不导致溶血和红细胞凝集。
采用下述实验验证发明效果:
实验一、复合Fe3O4-GO-ASA的PLA微泡超声成像效果
通过体内外超声造影实验考察复合Fe3O4-GO-ASA的PLA超声造影剂微泡的造影效果,并通过对比不同浓度复合微泡在兔子体内的超声造影效果寻找复合Fe3O4-GO-ASA的PLA微泡的适宜的使用浓度。通过PLA微泡中负载不同复合物(GO、Fe3O4-GO和Fe3O4-GO-ASA),考察不同复合微泡的超声造影特点。通过在造影区域外是否添加磁场,考察复合Fe3O4-GO-ASA的PLA微泡的磁靶向效果。本实验使用的新西兰大白兔均由佳木斯大学动物实验中心按照标准条件饲养,所有动物实验程序均符合伦理道德规范。

1.2实验方法
1.2.1PLA微泡的体外造影实验
按照实施例1的方法制备方法,分别制备PLA微泡、复合GO-ASA的PLA微泡和复合Fe3O4-GO-ASA的PLA微泡的冻干粉样品;用脱气生理盐水将PLA微泡冻干粉分别配制浓度为0.2mg·mL-1、0.6mg·mL-1和1mg·mL-1的悬浮液,备用;取50mL一次性注射器,缓慢抽取不同浓度微泡悬浮液后备用。取一个底部附有海绵消音层的水槽,将硅胶管放入,加脱气双蒸水模拟体内环境,液面外留出硅胶管两端,一端固定并于高出水面10cm,出口向上。用注射器从硅胶管的另一端反复缓慢抽吸微泡悬浮液,以模拟超声造影剂微泡在血流中的状态。检测不同浓度的PLA微泡悬浮液和复合Fe3O4-GO-ASA的PLA微泡的显像效果,超声检查参数在整个实验过程中保持不变。
1.2.2不同微泡的体内超声成像实验
用脱气生理盐水将复合GO-ASA的PLA微泡和复合Fe3O4-GO-ASA的PLA微泡的冻干粉分别配制浓度为60mg·mL-1的悬浮液,备用。将体重约为2kg的新西兰大白兔仰卧位固定于手术台,耳缘静脉建立静脉通道,腹部剃毛并备皮,清除实验台残余毛发,以防对实验产生影响。按照0.5mL·kg-1体重且不高于2.5mL的剂量进行推注,推注后即刻推注生理盐水0.5mL。经兔耳缘静脉分别注射浓度为60mg·mL-1的PLA微泡悬浮液、复合GO的PLA微泡悬浮液,复合Fe3O4-GO的PLA微泡悬浮液和复合Fe3O4-GO-ASA的PLA微泡悬浮液,每次推注间隔为5min。推注后注射生理盐水冲洗并立刻对兔的腹主动脉进行造影。观察不同微泡对白兔腹主动脉的超声成像效果,对比复合GO的PLA微泡、复合Fe3O4-GO的PLA微泡和复合Fe3O4-GO-ASA的PLA微泡的超声造影图像,从而探究GO、Fe3O4-GO和Fe3O4-GO-ASA对造影效果的影响。
1.2.3复合Fe3O4-GO-ASA的PLA微泡的体内超声成像实验
将新西兰大白兔仰卧位固定于手术台,耳缘静脉建立静脉通道,腹部剃毛并备皮。分别推注生理盐水和浓度为20mg·mL-1、60mg·mL-1和100mg·mL-1复合Fe3O4-GO-ASA的PLA微泡悬浮液。推注微泡后即刻推注生理盐水0.5mL,每次推注间隔为5min,推注后立刻对兔的腹主动脉进行造影,考察得出复合Fe3O4-GO-ASA的PLA的最佳造影浓度。
1.2.4复合Fe3O4-GO-ASA的PLA微泡的磁靶向实验
将新西兰大白兔用丙泊酚进行肌肉麻醉,仰卧位固定于手术台。建立耳缘静脉通道,腹部剃毛并备皮。于兔下腹的剑突正中切口,打开腹腔,游离腹主动脉,剥开腹主动脉壳,选取约1cm血管在近心端和远心端分别用手术线结扎,在该段血管上敷浸泡过80%浓度FeCl3溶液的滤纸片,并将滤纸片用保鲜膜包裹,防止污染其他区域。15min后取出滤纸片和保鲜膜,解开结扎线并对兔进行缝合,准备进行超声检查。
从家兔耳缘静脉推注之前所得最佳造影浓度的复合Fe3O4-GO-ASA的PLA微泡,生理盐水冲洗后立即开始记录超声造影图像。采集完毕后立即在兔的腹主动脉区域附加磁场,10s后记录超声造影图像,通过前后对照,比较有无磁场条件下复合Fe3O4-GO-ASA的PLA微泡的超声造影效果,从而评价复合Fe3O4-GO-ASA的PLA微泡在外加磁场条件下所展现的磁靶向性。
1.3结果与讨论
1.3.1PLA微泡的体外造影效果
不同浓度的PLA微泡在医用硅胶管中的超声造影照片见图8。由图8(a)可见,单注入生理盐水时,医用硅胶管内几乎完全黑暗,仅有下层边缘发亮,可能是液体流动产生的少量气泡导致的信号增强;当PLA微泡浓度为0.2mg·mL-1时(图8(b)),可以看见硅胶管内超声信号明显增强,超声信号显著提升;当PLA微泡浓度为0.6mg·mL-1时(图8(c)),超声信号继续增强,此时可以清晰地辨识硅胶管的轮廓,超声造影效果得到显著提升;由图8(d)可见,随着PLA微泡浓度达到1mg·mL-1,硅胶管内超声信号继续提升,但这样强烈信号提升导致管内完全被超声信号充斥,反而给辨识硅胶管轮廓带来困难。由此可见,PLA微泡与生理盐水相比具有显著的增强超声造影信号功能,而且随着浓度提升,其增强超声造影效果越明显。
1.3.2不同微泡的体内超声成像效果
PLA微泡、复合GO的PLA微泡、复合Fe3O4-GO的PLA微泡悬浮液和复合Fe3O4-GO-ASA的PLA微泡在兔腹主动脉处超声造影照片见图9。可见,在不附加磁场的情况下,PLA微泡、复合GO的PLA微泡、复合Fe3O4-GO的PLA微泡悬浮液和复合Fe3O4-GO-ASA的PLA微泡的超声信号没有显著的区别,说明Fe3O4和GO均不会对超声成像产生明显影响。
1.3.3复合Fe3O4-GO-ASA的PLA微泡的体内超声成像效果
不同浓度的复合Fe3O4-GO-ASA的PLA微泡在兔的主动脉超声造影照片见图10。由图10(a)可见,单注入生理盐水时,图像较为模糊,腹主动脉轮廓灰暗且不明显,较难与其他组织区分;当复合Fe3O4-GO-ASA的PLA微泡浓度为20mg·mL-1时(图10(b)),兔腹主动脉超声信号增强,图像对比度和辨识度均有较大提升;当复合Fe3O4-GO-ASA的PLA微泡浓度大于60mg·mL-1时(图10(c)),兔腹主动脉部分超声造影效果得到显著提升,此时可以清晰辨别腹主动脉;当复合Fe3O4-GO-ASA的PLA微泡浓度达到100mg·mL-1时(图10(d)),超声图像的清晰度没有更加明显的提升。因此,复合Fe3O4-GO-ASA的PLA微泡最佳超声造影浓度选择60mg·mL-1。
1.3.4复合Fe3O4-GO-ASA的PLA微泡的磁靶向效果
在兔腹主动脉血栓造模后经兔耳缘静脉注入复合Fe3O4-GO-ASA的PLA微泡后,分别在有和无磁场条件下的超声造影图像见图11。由图11(a)可见,血栓右侧出现强且鲜明的蓝色信号,说明该处存在因血栓栓塞而回流的血液;同时,照片中兔腹主动脉下方也出现了弱且面积较大的深蓝色信号,这是由于复合Fe3O4-GO-ASA的PLA微泡随血液全身流动,在经过组织或器官时也会出现超声信号。由图11(b)可见,当在腹主动脉区域附加磁场后,该区域超声信号强度明显增强,其他区域信号显著减弱,这说明复合Fe3O4-GO-ASA的PLA微泡能够有效富集在磁场区域。可见,复合Fe3O4-GO-ASA的PLA微泡在外加磁场作用下具有磁靶向效果,实现了在不增加复合微泡剂量前提下使所观察部位图像清晰度显著增强。
在体外造影实验中,当PLA微泡浓度为0.6mg·mL-1时就能明显增强超声造影效果,说明本次制备的PLA微泡可以很好的增强超声造影效果。复合Fe3O4-GO-ASA的PLA微泡的体内最佳造影浓度为60mg·mL-1。复合Fe3O4-GO-ASA的PLA微泡中的Fe3O4和GO对超声造影无影响。复合Fe3O4-GO-ASA的PLA的微泡具有优良的磁靶向性,可以在目标区域通过附加磁场的方式有效富集,并能够增强该区域的超声造影效果。
实验二、复合Fe3O4-GO-ASA的PLA微泡抗凝血性能
凝血四项属于检验科临检检查项目之一,包括凝血酶时间(TT)、活化部分凝血活酶时间(APTT)、凝血酶原时间(PT),以及用凝血活酶所测得的参比血浆与正常血浆的PT比值和所用试剂标出的ISI值计算出的国际标准化比率(INR),属于血栓性疾病检查的重要指标。本实验通过在血样中加入复合微泡,测定TT、APTT、PT和INR的数值,从而确定复合Fe3O4-GO-ASA的PLA微泡对凝血参数的影响;采用干重湿重法,通过测定加入复合微泡破碎液的血样形成血凝块的干重和湿重,评价复合Fe3O4-GO-ASA的PLA微泡的抗血栓效果。

2.2.1复合Fe3O4-GO-ASA的PLA微泡的抗凝血实验
取复合Fe3O4-GO-ASA的PLA微泡冻干粉适量,于生理盐水中超声破碎2h,将破碎液分别稀释至20mg·mL-1、40mg·mL-1、60mg·mL-1、80mg·mL-1和100mg·mL-1,并设置生理盐水为空白对照组。将一次性真空抗凝管插入家兔耳缘静脉,待取血完成后将上步备好的各组微泡破碎液分别加入血样,轻微震荡,使微泡破碎液充分与血样接触。将混合好的血样各取1mL加入ep管,分别加入相同长度四号手术线,并浸没至相同深度。将血样于37℃水浴1.5h后取出血凝块,观察形态,测量质量,记为湿重。将滤纸连同得到的血凝块共同烘干,称重后减去滤纸质量,记为干重。通过比较加入不同浓度复合微泡形成血凝块干重和湿重,评价复合Fe3O4-GO-ASA的PLA微泡的抗凝血效果。
2.2.2复合Fe3O4-GO-ASA的PLA微泡的凝血因子实验
取复合Fe3O4-GO-ASA的PLA微泡适量,于生理盐水中超声破碎2h,将破碎液分别稀释至10mg·mL-1、20mg·mL-1、40mg·mL-1、80mg·mL-1和160mg·mL-1备用,并设置生理盐水为空白对照组。将一次性真空抗凝管插入家兔耳缘静脉,取血完成后,将上步准备好的各组溶液分别注入采集器,轻微震荡摇匀。待微泡破碎液与家兔血作用1h后,采用半自动凝血分析仪分别测量血样的PT、TT、APTT和INR数值。
2.3实验结果
2.3.1复合Fe3O4-GO-ASA的PLA微泡的抗凝血实验结果
不同浓度复合Fe3O4-GO-ASA的PLA微泡与血样作用时所得凝血块湿重与干重质量见表2-1。

由表2-1可见,随复合Fe3O4-GO-ASA的PLA微泡浓度的上升,凝血块湿重和干重均明显下降。ASA可以通过抑制环氧合酶的合成,减少前列腺素及抑制血小板血栓素A2的生成,导致血小板凝聚力降低,最终有效抑制血栓的形成[6]。对于血栓的形成,血栓湿重与干重越小,说明血液中血栓因子游离性越强,血栓因子聚集导致血栓形成的速度越慢。ASA对血栓因子的游离有促进作用,随着复合微泡浓度的上升,血样中ASA的浓度也在上升,最终导致血栓因子游离性的提升。可见,复合Fe3O4-GO-ASA的PLA微泡具有抗血栓能力且其抗血栓能力随其浓度的提升而增强。
2.3.2复合Fe3O4-GO-ASA的PLA微泡的凝血因子实验结果
不同浓度复合Fe3O4-GO-ASA的PLA微泡与血样作用时凝血参数的变化见表2-2。

由表2-2可见,随复合Fe3O4-GO-ASA的PLA微泡浓度的增加,血样的PT、TT、APTT和INR数值呈现增加趋势,说明凝血酶时间,活化部分凝血活酶时间和凝血酶原时间均不同程度地延长了。具体地,APTT值在微泡浓度为20mg·mL-1时显著增加,PT和TT值在复合浓度为40mg·mL-1时显著增加,INR值在给药浓度为80mg·mL-1时显著增加,可见复合微泡在浓度为80mg·mL-1时能有效地延长PT、TT、APTT和INR时间,达到抗血栓效果。
含有ASA的复合Fe3O4-GO-ASA的PLA多功能微泡能够通过延长TT,APTT,PT和INR时间,促进血栓因子游离等方式,抑制血栓形成,达到辅助抑制的目的。
实验三、复合Fe3O4-GO-ASA的PLA微泡安全性初步评价
本发明复合Fe3O4-GO-ASA的PLA微泡需注入血管使用,所以必须确保其作为注射剂的安全性。本实验分别采取急性毒性实验、生物相容性实验、过敏实验和溶血实验四个方面初步评价复合Fe3O4-GO-ASA的PLA微泡的安全性。

3.2.1复合Fe3O4-GO-ASA的PLA微泡的急性毒性实验
3.2.1.1预实验
选择健康3周龄CL级昆明小鼠80只,雌雄各半,体重均为(14±2)g,适应性饲养一周,记录动物的行为活动、饮食、体重及精神状况。在实验过程中,严格控制动物环境管理,每隔一天换一次垫料,确保其能够自由饮食饮水。经过多次预实验,操作技术娴熟,在更换垫料,喂食等实验过程中对小鼠不产生系统性实验影响。
取复合Fe3O4-GO-ASA的PLA微泡冻干粉适量,用脱气生理盐水将复合微泡配制成悬浮液,4℃条件下无菌密闭保存于冰箱中,备用。为计算LD50,需进行预实验以确定给药剂量。本发明复合Fe3O4-GO-ASA的PLA使用浓度为60mg·mL-1,经折算后临床日用量为40mg·kg-1体重,预计安全限度预实验中复合微泡给药量至少为4000mg·kg-1体重,以此为依据找出引起0%致死给药剂量和100%致死给药剂量。5.2.1.2最大耐受剂量的测定
由于复合Fe3O4-GO-ASA的PLA微泡的安全范围较大,以最高浓度给药也无法找到LD100,解剖后也没有发现实验小鼠各脏器的明显病变或者变化,故无法测算LD50。为了进一步评价其安全性,将继续对小鼠的最大耐受剂量(MTD)进行测定。
取小鼠雌雄各半随机分成低中高三个剂量组,每组5只,分别按照给药剂量为2000mg·kg-1、4000mg·kg-1和6000mg·kg-1体重,体积为0.3mL,经腹腔注射一次,注射后观察72h,记录小鼠的行为状态和死亡数。在无死亡组和与其相邻的更高浓度组之间按剂量比重复试验两次,最后确定的最大不致死给药量(Dn),基于试验客观必要性和人道主义原则,将其近似看作MTD。
由于本试验中复合Fe3O4-GO-ASA的PLA微泡作为诊断试剂的特殊用途,选取近似MTD值作为小鼠的最大给药量进行安全限度试验。随机选取无孕昆明小鼠20只,雌雄各半,以近似MTD不计体重按每只0.3mL注射一次,记录毒性反应、摄食情况和行为体重变化,观察14d,于最后一日处死解剖,观察脏器变化。用最大耐受倍数公式进行换算。
3.2.2复合Fe3O4-GO-ASA的PLA微泡的生物相容性实验
考虑到复合Fe3O4-GO-ASA的PLA微泡在体内的代谢情况和组织反应,复合Fe3O4-GO-ASA的PLA微泡的完整性对于实验结果也有一定的影响,所以于小鼠背部直接皮下注射复合Fe3O4-GO-ASA的PLA微泡溶液,以考察复合微泡在体内的实际代谢情况。
取23g~25g昆明小鼠15只,分为5组。小鼠俯卧位固定于实验台上,于背部皮下注射150mg·mL-1复合Fe3O4-GO-ASA的PLA微泡溶液0.1mL。对照组以同样方式注射生理盐水0.1mL。分别生理盐水组和第1日、第3日、第9日和第21日的复合Fe3O4-GO-ASA的PLA微泡组的小鼠脱颈处死,观察注射部位的局部组织。同时,取注射部位复合微泡所接触的肌肉,置于10%福尔马林溶液中固定保存,常规脱水,每个标本由中央切开,石蜡包埋,并以3μm厚度切片,按照说明步骤进行HE染色。
3.2.3复合Fe3O4-GO-ASA的PLA微泡的过敏实验
选择健康、经观察确定行为正常的昆明小鼠20只,雌雄各半,腹腔注射复合Fe3O4-GO-ASA的PLA微泡破碎液0.2mL,隔日一次,共致敏四次,致敏期间记录每日小鼠状态,初次和末次致敏及激发当日称定各鼠的体重。于末次致敏7d后一次腹腔注射激发,剂量均为每只0.4mL。激发后在30min内持续观察状态,包括是否明显躁动现象、休克、颤抖、瘙痒以及反复抓鼻(或鼠前爪抓鼻等行为5min内不超过3次)。
3.2.4复合Fe3O4-GO-ASA的PLA微泡的溶血实验
取志愿者血液50mL,置于锥形烧中磁力搅拌10min,除去纤维蛋白原,得到脱纤血液。加入生理盐水至50mL摇匀,1500r·min-1离心15min,除去上清液,取沉淀反复洗涤至上清液澄清且不显红色为止,即得到红细胞溶液。将所得的红细胞用生理盐水配成2%混悬液,备用。取Fe3O4-GO-ASA的PLA微泡冻干粉适量,与生理盐水按照1:3比例超声破碎2h,分别稀释至20mg·mL-1(1号)、40mg·mL-1(2号)、80mg·mL-1(3号)、160mg·mL-1(4号)和320mg·mL-1(5号),备用。
分别取七只洁净试管,加入内容物如表3-1所示,1~5号试管为测试组,6号试管只装入生理盐水,为阴性对照组,7号试管只装入蒸馏水,为阳性对照组。分别按顺序依次加入内容物后轻微震荡摇匀,转移至37℃条件下温浴3h。待反应完成后观察1~5号试管上清液,若上清液呈红色,颜色与阳性对照组接近,且与阴性对照组相比管底红细胞数量减少,则说明发生溶血现象。若颜色与阴性对照组相同,红细胞全部沉淀且上清液清澈透明,则说明无溶血现象,此时轻微震荡沉淀,若发现沉淀为絮状且震荡后不分散,则说明发生了红细胞凝聚现象。

3.3结果与讨论
3.3.1复合Fe3O4-GO-ASA的PLA微泡的急性毒性实验结果
通过预实验,在复合Fe3O4-GO-ASA的PLA微泡的给药浓度大于4000mg·kg-1的条件下寻找0%致死给药剂量和100%致死给药剂量,由于复合Fe3O4-GO-ASA的PLA微泡安全范围较大,在微泡最大给药浓度下仍不能达到100%致死,于是复合微泡的给药浓度在2000mg·kg-1~6000mg·kg-1区间内寻找MTD。实验结果表明,小鼠的近似MTD为4500mg·kg-1,代入最大耐受倍数公式计算可得,复合Fe3O4-GO-ASA的PLA微泡的最大耐受倍数(小鼠)=(1350mg/0.024kg)*(60kg/2400mg)=1406.25倍。小鼠经腹腔注射复合Fe3O4-GO-ASA的PLA微泡给药后,14d内无死亡,日常行为无明显异常,摄食饮水量、体重等参数与对空白对照组相比没有明显变化;处死后对小鼠进行解剖发现,各主要脏器无明显异常改变,具体情况见表3-2。本实验证明,小鼠对复合Fe3O4-GO-ASA的PLA微泡的最大耐受剂量为临床成人日用量的1406.25倍,说明复合Fe3O4-GO-ASA的PLA微泡安全性良好,可以进行下一步的研究。

3.3.2复合Fe3O4-GO-ASA的PLA微泡的生物相容性实验结果
皮下注射生理盐水和复合Fe3O4-GO-ASA的PLA微泡不同时间后小鼠肌肉细胞HE染色照片见图12。由于保存过程中使用的是10%的福尔马林溶液,在切片过程中样本过于坚硬,切片受到较大阻力,导致得到的HE染色照片不够好。如图12(a)所见,生理盐水组的肌肉细胞结构完整,可见组织发生没有病理改变或细胞浸润现象。注射复合Fe3O4-GO-ASA的PLA微泡1d后,解剖后可见注射局部形成包壁较薄的包裹,包裹内含微泡溶液及组织液。由图12(b)可见,与生理盐水组相比注射部位出现少量炎性细胞浸润现象,且基本为中性粒细胞。注射复合Fe3O4-GO-ASA的PLA微泡3d后,解剖发现注射局部包裹明显减小,打开包裹后可以看见未分解的复合微泡。由图12(c)可见,注射部位炎性细胞浸润现象有所减弱,但并未完全消失,这可能是由于PLA降解过程中产生的少量碎屑导致人体组织难以将降解产物及时排除或吸收,因而导致局部炎症反应。注射复合Fe3O4-GO-ASA的PLA微泡9d后,注射局部包裹基本消失,打开包裹后基本上没有可见的微泡残骸,但仍有少量黑色物质,可能是未降解的GO。由图12(d)可见,炎性细胞浸润明显减少。注射复合Fe3O4-GO-ASA的PLA微泡21d后,由图12(e)可以看出,炎性细胞浸润现象基本消失。从开始注射复合Fe3O4-GO-ASA的PLA微泡到完全代谢,深层肌肉结构始终保持完整,未见明显病变。
由此可见,复合Fe3O4-GO-ASA的PLA微泡注射给药后仅引起轻微的炎症反应,没有组织渗透液蓄积,并且没有观察到明显血管与纤维的增生,说明复合Fe3O4-GO-ASA的PLA微泡在体内的代谢过程符合一般生物相容性材料在体内的降解过程,具有良好的生物相容性。
3.3.3复合Fe3O4-GO-ASA的PLA微泡的过敏实验结果
小鼠由于具有易于过敏的特性,所以是过敏实验的理想动物。但是由于其个体差异较大,同时伴随着个体小而又过于活泼,为行为学判断造成了一定困难。所以,基于小鼠的过敏性实验只能作为复合Fe3O4-GO-ASA的PLA微泡的初级安全性实验。表3-3是注射复合Fe3O4-GO-ASA的PLA微泡30min内小鼠的行为表现。

由表3-3可见,在注射后30min内,20只小鼠均无明显的躁动、休克、颤抖、瘙痒、反复抓鼻等过敏现象,这说明复合Fe3O4-GO-ASA的PLA微泡不具有致敏原性。
33.3.4复合Fe3O4-GO-ASA的PLA微泡的溶血实验结果
待试管内血样水浴3h后,发现阴性对照组未发生溶血现象,阳性对照组发生了溶血现象,浓度为20mg·mL-1、40mg·mL-1、80mg·mL-1、160mg·mL-1和320mg·mL-1的复合微泡破碎液试管中均无溶血现象。1至5号试管内均无红细胞凝集现象,浓度为20mg·mL-1、40mg·mL-1、80mg·mL-1、160mg·mL-1和320mg·mL-1的复合Fe3O4-GO-ASA的PLA微泡破碎液试管中均无溶血现象。实验证明,复合Fe3O4-GO-ASA的PLA微泡在不同浓度下3h内不引发溶血和凝聚,可以作为注射剂使用。
本发明复合Fe3O4-GO-ASA的PLA微泡的安全范围大,最大耐受剂量为临床成人日用量的1406.25倍。复合Fe3O4-GO-ASA的PLA微泡在体内的代谢过程符合一般生物相容性材料在体内的降解过程,肌肉注射仅导致轻微的中性细胞炎性浸润现象,同时,复合Fe3O4-GO-ASA的PLA微泡不会导致溶血和红细胞凝集,也不具有致敏原性。复合Fe3O4-GO-ASA的PLA微泡具有良好的生物相容性,可以作为注射剂安全使用。
Claims (8)
1.复合Fe3O4-GO-ASA的PLA微泡,其特征在于所述微泡是由PLA微泡以及PVA附着在PLA微泡表面形成壳层构成的;所述PLA微泡是由PLA膜层构成的微泡壁,Fe3O4-GO-ASA复合物镶嵌于PLA膜层內以及PLA膜层的内壁上,并且PLA微泡內包裹有惰性气体,其中,所述Fe3O4-GO-ASA复合物是在GO表面沉积Fe3O4后利用π-π吸附负载ASA得到的。
2.根据权利要求1所述复合Fe3O4-GO-ASA的PLA微泡,其特征在于所述惰性气体为氮气。
3.如权利要求1或2所述的复合Fe3O4-GO-ASA的PLA微泡的制备方法,其特征在于所述制备方法是按通过下述步骤实现的:
步骤一、采用原位沉积法在GO表面沉积Fe3O4,得到Fe3O4-GO复合物;
步骤二、然后利用π-π吸附负载ASA,得到Fe3O4-GO-ASA复合物;
步骤三、然后通过双乳化溶剂挥发法将Fe3O4-GO-ASA复合物包裹于PLA微泡中,第一次超声乳化时,PLA溶液破坏成包裹着Fe3O4-GO-ASA内水相的小液泡,然后转入PVA溶液中搅拌均匀并进行二次超声乳化,PVA会附着在所形成的PLA微泡表面形成坚硬的壳层,经过真空冻干,即得到复合Fe3O4-GO-ASA的PLA微泡。
4.根据权利要求3所述的复合Fe3O4-GO-ASA的PLA微泡的制备方法,其特征在于步骤一所述原位沉积法在GO表面沉积Fe3O4是按下述操作进行的:
将Fe2(SO4)3和FeSO4·7H2O按照Fe2+:Fe3+摩尔浓度比为1:1.5的比例混合,在20mL双蒸水中配制成铁盐总浓度为0.3mol·L-1的溶液,分别将质量为0.058~0.29g的GO超声分散于1h,反应完成后,将沉淀物反复水洗抽滤后,真空冻干,得到Fe3O4-GO复合物。30mL双蒸水,在搅拌条件下加到上述铁盐溶液中,加热至体系温度达到60℃时,滴加0.25mol·L-1NaOH溶液直至过量,搅拌。
5.根据权利要求4所述的复合Fe3O4-GO-ASA的PLA微泡的制备方法,其特征在于步骤二中Fe3O4-GO-ASA复合物的制备是按下述步骤进行的:
将0.5gFe3O4-GO复合物置于50mL无水乙醇得到Fe3O4-GO分散液,将0.5g ASA加入到该分散液中,超声处理1min~2min,使其充分溶解,在20℃下搅拌2h,微孔滤膜抽滤,沉淀经无水乙醇洗涤、抽滤、真空冻干,即得Fe3O4-GO-ASA复合物。
6.根据权利要求5所述的复合Fe3O4-GO-ASA的PLA微泡的制备方法,其特征在于步骤三具体步骤如下:
称取3.5g聚乙烯醇(PVA)于100mL双蒸水中浸泡3h,之后90℃水浴条件搅拌至完全溶解,保持搅拌状态备用,得到PVA溶液;
将0.6g聚乳酸(PLA)加到12mL二氯甲烷(DCM)中,浸泡5h至完全溶解,得到有机相;
取Fe3O4-GO-ASA复合物0.1g加到0.2mL乙醇水溶液,作为内水相;
向有机相加入内水相,以25kHz频率、33W功率,进行超声乳化,在反应过程中向体系中持续通入惰性气体,制得W/O乳液(初乳);
将初乳逐滴加到PVA溶液中,搅拌3min;以25kHz频率、55W功率,进行二次超声乳化,制得W/O/W乳液(复乳);
向复乳中加入5.5%异丙醇溶液100mL,室温搅拌5h,挥发有机溶剂,待挥发完毕后,用双蒸水反复离心洗涤剩余溶液,收集下层微泡,真空冻干后即为复合Fe3O4-GO-ASA的PLA微泡。
7.根据权利要求6所述的复合Fe3O4-GO-ASA的PLA微泡的制备方法,其特征在于步骤三中最佳内水相与有机相体积比为1:60,初乳制备过程中最佳开关时间为4s开2s关,复乳制备过程中最佳开关时间为4s开2s关。
8.根据权利要求1所述的复合Fe3O4-GO-ASA的PLA微泡的制备方法,其特征在于所述氧化石墨烯的制备是通过下述步骤完成的:
先将100mL三颈烧瓶洗净、烘干、封口并置于冰箱中冷却;
取鳞片石墨粉1.5g,置于冷却后的三口瓶中,并加入30mL浓硫酸,在冰浴条件下缓慢搅拌,混合均匀,该体系温度保持在5℃~8℃,向体系中缓慢分次加入高锰酸钾4.5g,持续搅拌2.5h,混合液呈墨绿色;
然后将体系转移到38℃水浴条件下继续搅拌1h,再将体系转入到装有280mL去离子水烧杯中,搅拌5min,此时溶液变为褐色,最后加入60mL质量浓度为30%的H2O2溶液,此时混合液剧烈沸腾并伴随大量气泡,溶液颜色由褐色转变为金黄色,反应停止后在室温下继续搅拌24h;
将上步得到产物在5000r·min-1条件下反复离心水洗,除去残酸,洗至pH>6,并用BaCl2溶液检测无硫酸根离子为止;
将所得沉淀分散于去离子水中,在超声功率为30%条件下破碎10min,50℃下烘干即得到氧化石墨烯。
