the preparation method of the agastache rugosus extract comprises the following steps of:
s1, weighing agastache rugosus, cleaning to remove impurities, and crushing into coarse powder;
s2, adding 6 times of distilled water into the coarse powder obtained in the step S1, soaking for 20min, and performing reflux extraction for 50 min;
s3, adding 5 times of distilled water into the extract obtained in the step S2, and performing 500W microwave extraction for 30 min;
s4, centrifuging the extract obtained in the step S3 to obtain supernatant, namely the agastache rugosus extract;

s1, weighing agastache rugosus, cleaning to remove impurities, and crushing into coarse powder;

s5, slowly adding 70% ethanol into the mixed extract B at the temperature of 60 ℃, stirring while adding, and adding the agastache rugosus extract B after the temperature is reduced to room temperature;
and S6, combining the mixture obtained in the step S4 and the mixture obtained in the step S5, and then purifying to obtain the disinfectant.
Experiment 1: effect test of agastache rugosus extract
And (3) testing a sample: agastache rugosa extracts obtained in example 1 and comparative example 1
The experimental method comprises the following steps:
treating the filter disc: preparing filter disc with diameter of 7mm, sterilizing the filter disc, drying, soaking in herba Agastaches extract for 13 hr, drying, and standing at 4 deg.C for use. Wherein the filter disc was immersed in sterile water as a negative control and the filter disc was immersed in an ampicillin solution as a positive control.
Preparing bacterial liquid: inoculating activated escherichia coli, staphylococcus aureus, bacillus cereus and salmonella in a liquid culture medium for culture, culturing at 37 ℃ for 24 hours, and diluting with sterile water; ② placing the activated streptococcus mutans in an anaerobic box for culturing, culturing for 24h at 37 ℃, and then diluting with sterile water.
And (3) effect measurement: respectively and uniformly inoculating equal amounts of escherichia coli, staphylococcus aureus, bacillus cereus, salmonella and streptococcus mutans to different culture plates. The treated filter discs were placed on a culture plate with 3 parallel of each extract of Agastache rugosa, the plate was placed upside down after the experiment was finished, and after one day of culture at 37 ℃, the bacteriostatic diameter was measured. The experiment is repeated three times, no obvious difference exists between experimental data of each time, and the average value of the experimental data of each time is taken as the final detection result. The final measurement results are shown in table 1 below.

As can be seen from table 1: the agastache rugosus extract has no obvious inhibition effect on salmonella and escherichia coli, the agastache rugosus extract has obvious inhibition effect on bacillus cereus, staphylococcus aureus and streptococcus mutans, and the agastache rugosus extract obtained in the example 1 of the invention has equivalent antibacterial effect to the agastache rugosus extract obtained in the comparative example 1.
Experiment 2: effect verification of disinfectants
And (3) testing a sample: disinfectants obtained in examples 2-4 and comparative examples 2-7
The experimental method comprises the following steps:
preparing a strain solution: inoculating activated escherichia coli, staphylococcus aureus, bacillus cereus and salmonella in a liquid culture medium for culture, culturing at 37 ℃ for 24 hours, and diluting by 30 times with sterile water; ② placing the activated streptococcus mutans in an anaerobic box for culturing, culturing for 24h at 37 ℃, and diluting by 30 times with sterile water.
The antibacterial effect of the disinfectant is verified by adopting a test tube method, the disinfectant obtained in the examples 2-4 and the comparative examples 2-7 is diluted to 100 mu g/mL by broth, then the diluted disinfectant is diluted to 10 test tubes in a descending manner, the numbers of the test tubes are respectively tube 1, tube 2, tube 3, tube 4, tube 5, tube 6, tube 7, tube 8, tube 9 and tube 10, then the strain liquid is respectively added to 0.5mL, the culture is carried out at 37 ℃ for 24h, the antibacterial effect is measured, and the measurement result is shown in the following table 2.

Likewise, to further illustrate the beneficial effects of the present invention, the following comparative examples are provided:
comparative example 2.1 is provided, which is distinguished from example 4 in that: no dandelion extract is added;

comparative example 2.3 is provided, which comparative example 2.3 differs from example 4 in that: no houttuynia cordata extract is added;
comparative example 2.4 is provided, which comparative example 2.4 differs from example 4 in that: no pericarpium Citri Tangerinae extract is added;
comparative example 2.5 is provided, which is distinguished from example 4 by the following: no addition of auxiliary agent;
the disinfectant prepared by the preparation method disclosed by the invention is verified for bacteriostasis, and the test is carried out according to the method for testing the effect verification of the disinfectant. The antibacterial effects of the disinfectants prepared in comparative example 2.1, comparative example 2.2, comparative example 2.3, comparative example 2.4 and comparative example 2.5 were observed. The results were similar to those in comparative example 2 described above.
Likewise, to further illustrate the beneficial effects of the present invention, the following comparative examples are provided:
comparative example 3.1 is provided, which comparative example 3.1 differs from example 4 in that: no polypropylene glycol was added;
comparative example 3.2 is provided, which comparative example 3.2 differs from example 4 in that: carboxymethyl chitosan is not added;
the disinfectant prepared by the preparation method disclosed by the invention is verified for bacteriostasis, and the test is carried out according to the method for testing the effect verification of the disinfectant. The antibacterial effects of the disinfectants prepared in the comparative examples 3.1 and 3.2 were observed. The results were similar to those in comparative example 3 above.
As can be seen from table 2: the disinfectant has obvious bacteriostatic action on staphylococcus aureus, bacillus cereus and streptococcus mutans in gram-positive bacteria, and also has certain bacteriostatic action on escherichia coli and salmonella in gram-negative bacteria; the antibacterial effect of the disinfectant on escherichia coli and salmonella in gram-negative bacteria is improved by further screening the using amount of each component in the disinfectant. The disinfectant without the formula system has poor bacteriostatic action on staphylococcus aureus, bacillus cereus and streptococcus mutans in gram-positive bacteria and escherichia coli and salmonella in gram-negative bacteria.

Claims (10)

1. A preparation method of a wrinkled giant hyssop extract is characterized by comprising the following steps:
s1, weighing agastache rugosus, cleaning to remove impurities, and crushing into coarse powder;
s2, adding 6-8 times of distilled water into the coarse powder obtained in the step S1, soaking, and performing reflux extraction;
s3, adding 2-5 times of distilled water into the extract obtained in the step S2, and then performing microwave extraction;
s4, centrifuging the extract obtained in the step S3 to obtain supernatant, namely the agastache rugosus extract.
2. The method as claimed in claim 1, wherein the soaking time of the distilled water in step S2 is 20-30min, and the extraction time of the reflux extraction in step S2 is 50-60 min.
3. The method of claim 1, wherein the microwave extraction in step S3 has a microwave power of 500W and a microwave time of 30 min.
4. An agastache rugosa extract, which is prepared by the preparation method of any one of claims 1 to 3.
5. A disinfectant, characterized in that it comprises the Agastache rugosa extract of claim 4.
6. The disinfectant according to claim 5, wherein the disinfectant further comprises honeysuckle extract, dandelion extract, apple peel extract, houttuynia cordata extract, dried orange peel extract and auxiliaries, wherein the auxiliaries are prepared from corn starch, polypropylene glycol and carboxymethyl chitosan.
7. The disinfectant as recited in claim 6, wherein the weight portion of said corn starch is 3-8, the weight portion of said polypropylene glycol is 5-10, and the weight portion of said carboxymethyl chitosan is 3-5.
8. The disinfectant as recited in claim 6, wherein the honeysuckle extract is 5-15 parts by weight, the dandelion extract is 12-20 parts by weight, the apple peel extract is 13-18 parts by weight, the houttuynia cordata extract is 10-20 parts by weight, the dried orange peel extract is 8-12 parts by weight, and the agastache rugosus extract is 6-15 parts by weight.
9. A method of preparing a disinfectant as claimed in any one of claims 6 to 8, characterised in that said method of preparing comprises the steps of:
s1, preparing a honeysuckle extract, a dandelion extract, an apple peel extract, a houttuynia cordata extract and a tangerine peel extract, mixing the prepared extracts, and dividing the mixed extracts into two parts, wherein one part is a mixed extract A, and the other part is a mixed extract B;
s2, preparing a wrinkled gianthyssop herb extract, and equally dividing the wrinkled gianthyssop herb extract into two parts, wherein one part is a wrinkled gianthyssop herb extract A, and the other part is a wrinkled gianthyssop herb extract B;
s3, preparing an auxiliary agent, namely dividing the auxiliary agent into two parts, wherein one part is an auxiliary agent A, and the other part is an auxiliary agent B;
s4, mixing the mixed extract A and the auxiliary A, slowly adding 60% ethanol at 50-60 ℃, stirring while adding, cooling to room temperature, and adding the agastache rugosus extract A;
s5, mixing the mixed extract B and the auxiliary agent B, slowly adding 70% ethanol at the temperature of 60-70 ℃, stirring while adding, cooling to room temperature, and adding the agastache rugosus extract B;
and S6, combining the mixture obtained in the step S4 and the mixture obtained in the step S5, and then purifying to obtain the disinfectant.
10. Use of a disinfectant according to any one of claims 5 to 8 for disinfection and sterilization and disease control.
