背景技术
甜菜根属藜科,发源于亚洲和欧洲,在全球,藜科共约130个属1500余种。它是一种二年生的开花植物,颜色多为红色和黄色。甜菜根一年四季都可以种植,并且耐高温。

本发明的目的是提供一种甜菜根提取物的制备方法,充分利用生物技术,环保高效,可以高效提取出生物活性高的甜菜根提取物,并且尽可能保持活性,以获得最佳保健效果的甜菜根提取物。
本发明所需要解决的技术问题可以通过以下技术方案来实现。
一种甜菜根提取物的制备方法,首先将甜菜根进行干燥、粉碎,接着采用多种方法协同破壁,之后透析、干燥即得甜菜根提取物。
具体的,所述制备方法包括如下步骤:
(1)冷冻干燥:选取新鲜红甜菜根,清洁后冷冻干燥、粉碎;
(2)多效破壁:称取冷冻干燥后的干粉,加水浸润,再在-20±2℃冷冻处理,再依次加水、纤维素酶,甜菊甙,50±5℃水浴并同时处以超声处理;再在-20±2℃冷冻处理,之后40±5℃水浴并同时处以超声处理30±5min;合并破壁滤液减压浓缩;
(3)低温破壁处理:将浓缩液再加入甜菊甙,将其输入破壁机进行低温破壁处理,充分破壁;
(4)用2000Da截留分子量的透析袋流水透析以去除小分子杂质,冷冻干燥,获得甜菜根提取物粉末。
作为本申请的优选技术方案,所述步骤(1)冷冻干燥具体方法如下:选取新鲜红甜菜根,清洁后置于冷冻干燥机冷冻干燥后,粉碎过80目筛。
更优选的,冷冻干燥时间为48h。

(1)冷冻干燥:选取新鲜红甜菜根,清洁后置于冷冻干燥机冷冻干燥48h后,粉碎过80目筛;
(2)多效破壁:按照重量份,称取冷冻干燥后的干粉100g,加1000g水浸润,置于-20℃冰箱中冷冻处理后,再加500g水,加入每克干粉6U的纤维素酶,50℃水浴并同时处以超声处理25min;再次置于-20℃冰箱中冷冻处理后,40℃水浴并同时处以超声处理30min;合并破壁滤液减压浓缩;
(3)低温破壁处理:将浓缩液再加入2g的甜菊甙,将其输入破壁机进行低温破壁处理,依次保持温度10℃10min,5℃15min,以充分破壁;
(4)用2000Da截留分子量的透析袋流水透析12h以去除小分子杂质,冷冻干燥,获得甜菜根提取物粉末5.36g。
实施例5增强免疫效果测试:
用本发明实施例1-4的制备方法制备得到的甜菜根提取物粉末,对猪瘟疫苗进行免疫效果试验。试验分为:空白对照组、疫苗组、口服甜菜根提取物+疫苗组,每组均采用健康猪10头。
1.材料本发明口服甜菜根提取物;动物:30日龄猪仔;猪瘟兔化弱毒疫苗。
2.方法试验方法:①空白对照组。②疫苗对照组:疫苗注射。③口服甜菜根提取物组,疫苗注射+每日二次喂入本发明甜菜根提取物粉末(每公斤体重0.03g)。
3.疫苗抗体水平检测:疫苗免疫前、疫苗免疫后的7天、14天、30天、60天,分别采血分离血清,采用猪瘟抗体检测试剂盒测定,结果如下(log2):
上表数据表明,本发明制得的甜菜根提取物粉末具有显著的增强免疫力的效果,并且甜菊甙具有显著的作用。
本发明的目的是制得高生物活性的甜菜根提取物复合物粉末,无需高度纯化节约成本。可见,本发明选择多种手段协同破壁,破壁效果好,虽然甜菜根提取物得率有所下降,但是在本发明的优化工艺下,甜菜根提取物的生物活性有显著增强。无有机溶剂加入,无有害杂质。
需要说明的是,以上所述仅为本发明的优选具体的实施例,若依本发明的构想所作变动,其产生的功能作用,仍未超出说明书所涵盖的精神时,均应在本发明的范围内。

Claims (9)
1.一种甜菜根提取物的制备方法,其特征在于,首先将甜菜根进行干燥、粉碎,接着采用多种方法协同破壁,之后透析、干燥即得甜菜根提取物。
2.根据权利要求1所述的甜菜根提取物,其特征在于,包括如下具体步骤:
(1)冷冻干燥:选取新鲜红甜菜根,清洁后冷冻干燥、粉碎;
(2)多效破壁:称取冷冻干燥后的干粉,加水浸润,再在-20±2℃冷冻处理,再依次加水、纤维素酶,甜菊甙,50±5℃水浴并同时处以超声处理;再在-20±2℃冷冻处理,之后40±5℃水浴并同时处以超声处理30±5min;合并破壁滤液减压浓缩;
(3)低温破壁处理:将浓缩液再加入甜菊甙,将其输入破壁机进行低温破壁处理,充分破壁;
(4)用2000Da截留分子量的透析袋流水透析以去除小分子杂质,冷冻干燥,获得甜菜根提取物粉末。
3.根据权利要求1所述的一种甜菜根提取物的制备方法,其特征在于:所述步骤(1)冷冻干燥具体方法如下:选取新鲜红甜菜根,清洁后置于冷冻干燥机冷冻干燥后,粉碎过80目筛。
4.根据权利要求3所述的一种甜菜根提取物的制备方法,其特征在于,冷冻干燥时间为48h。
5.根据权利要求1-4任一所述的一种甜菜根提取物的制备方法,其特征在于:所述步骤(2)多效破壁具体方法如下:按照重量份,称取冷冻干燥后的干粉1份,加10份水浸润,置于-20±2℃冰箱中冷冻处理后,再加5份水,加入每克干粉6-10U的纤维素酶,每克干粉甜菊甙0.01份,50±5℃水浴并同时处以超声处理;再次置于-20±2℃冰箱中冷冻处理后,40±5℃水浴并同时处以超声处理30±5min;合并破壁滤液减压浓缩。
6.根据权利要求5所述的一种甜菜根提取物的制备方法,其特征在于:步骤(2)中,50℃水浴并同时处以超声波处理25±5min。
7.根据权利要求1-4任一项所述的一种甜菜根提取物的制备方法,其特征在于:所述步骤(3)低温破壁处理具体方法如下:将浓缩液按照每克干粉0.02份的甜菊甙再次加入甜菊甙,将其输入破壁机进行低温破壁处理,依次保持温度10℃ 10min,5℃ 15min,以充分破壁。
8.根据权利要求1-4任一项所述的一种甜菜根提取物的制备方法,其特征在于:步骤(4)用2000Da截留分子量的透析袋流水透析以去除小分子杂质,冷冻干燥,获得甜菜根提取物粉末。
9.根据权利要求8任一项所述的一种甜菜根提取物的制备方法,其特征在于:透析时间为12h。
