Abstract
本发明公开miR‑588及其靶基因在胃癌中的应用。本发明发现:miR‑588在胃癌患者血清中的表达显著低于健康人群,miR‑588靶基因CXCL5、CXCL9或CXCL10在胃癌组织中的表达显著低于正常组织,在区别胃癌患者和健康人群方面,miR‑588比传统胃癌肿瘤标志物CEA和CA19‑9具有更好的敏感性和特异性,miR‑588可用作胃癌分子标志物;过表达miR‑588通过上调靶基因CXCL5、CXCL9或CXCL10的表达进而显著性抑制胃癌细胞在体内外增殖和体外侵袭,最终miR‑588、CXCL5、CXCL9或CXCL10的高水平表达与胃癌患者更长的总生存期显著相关。

技术领域
本发明属于肿瘤分子生物学及生物检测领域,具体涉及miR-588及其靶基因在胃癌中的应用。
背景技术
全球恶性肿瘤患者死亡率中胃癌高居第2位,每年全球因胃癌死亡人数约100余万。我国是胃癌的发病大国,发病率约占全世界的50%,并且胃癌患者5年生存率只达到20-30%。对于早期胃癌患者来说,外科手术切除是最有效的治疗和延长了病人生存时间的手段,而晚期胃癌经常在淋巴结、血液和腹膜反复出现;此外,对于患者的预后诊断水平还是十分低下。因此,胃癌的早期诊断意味着可以提早干预治疗,对患者的预后意义重大,通过改进治疗可以增加患者的生存时间。不幸的是,大多数患者在患病早期没有症状而难以被发现。
目前,用于胃癌诊断主要依赖于内镜活检,这种方法有创,给患者带来痛苦,且对实施此项技术的医师有较高要求。胃癌的无创检查方法主要是血清肿瘤标志物,目前临床常用的胃癌肿瘤标志物有癌胚抗原(CEA)、糖类抗原125(CA125)、糖类抗原199(CA199)等,但是它们对于胃癌诊断的特异性和敏感性都不强。因此,寻找具有较高灵敏度和特异性的胃癌血清肿瘤标志物,从而更加准确及时地对胃癌做出早期诊断,对胃癌患者具有十分重要的意义。
microRNA(miRNA)是一类由19-25个核苷酸组成的单链非编码RNA。据报道,miRNA通过调控一系列分子机制在人类病理和生理过程中扮演至关重要的角色。另外,异常表达的miRNA可以通过与靶基因mRNA的3'-非翻译区(3'UTR)的完全或不完全配对,降解靶基因mRNA或抑制其翻译,从而参与调控机体发育、细胞凋亡、增殖及分化等生命活动。已有研究发现许多miRNA通过调控增殖或侵袭相关靶蛋白的表达参与细胞增殖、细胞分化的发生,与癌症的发生发展具有密切的关系,具有重要的研究价值和临床意义。对于miRNA来说,其调控方式的特点决定了它的靶基因不会只有一个,而是一对多的方式,这就使得miRNA的功能研究内容非常丰富。有些miRNA具有癌基因功能,有些miRNA具有抑癌基因功能。甚至,同一种miRNA在一种癌症中发挥抑癌作用,在另一种癌症中发挥癌基因作用。
例如,Qian L等在题为“MicroRNA-588suppresses tumor cell migration andinvasion by targeting GRN in lung squamous cell carcinoma”(Mol Med Rep,2016,14(4):3021-3028)的论文中报道,miR-588在肺鳞癌组织中的表达明显的低于非肿瘤组织,与肿瘤的分期、淋巴结侵袭呈负相关,并且通过靶向调控颗粒体蛋白(granulin,GRN)抑制肺鳞状细胞癌的侵袭和转移,指出miR-588在肺鳞状细胞癌中发挥抑癌作用;ZHAO N等在题为“MicroRNA-588is upregulated in human prostate cancer with prognostic andfunctional implications”(J Cell Biochem,2019,120(7):12070)的论文中报道,miR-588在前列腺癌中高表达,具有促进肿瘤细胞增殖作用而表现为促癌基因。但是,miR-588与胃癌的关系尚未见报道,miR-588在胃癌的发生发展的作用尚不清楚,尤其miR-588的异常表达与胃癌的诊断、预后评估尚不清楚。
发明内容
有鉴于此,本发明的目的在于提供miR-588及其靶基因在胃癌中的应用。
本发明是通过如下技术方案实现的:
本发明的第一个方面,提供miR-588在制备胃癌分子标志物中的应用。
进一步地,所述应用中,所述胃癌分子标志物用于诊断胃癌或预测胃癌预后。
进一步地,所述应用中,所述胃癌分子标志物用于液体活检诊断胃癌。
本发明的第二个方面,提供miR-588靶基因在制备用于胃癌诊断或预测胃癌预后的药物中的应用,所述miR-588靶基因包括CXCL5基因及其表达产物、CXCL9基因及其表达产物、CXCL10基因及其表达产物。
本发明的第三个方面,提供一种用于胃癌诊断或预测胃癌预后的组合物,包含:基于RT-PCR方法和/或基于蛋白质印迹方法,对样品中miR-588表达、和/或CXCL5基因及其表达产物、和/或CXCL9基因及其表达产物、和/或CXCL10基因及其表达产物进行检测的药物或试剂盒。
在本发明的一些具体实例中,一种用于胃癌诊断或预测胃癌预后的组合物,包含:基于RT-PCR方法和/或基于蛋白质印迹方法,对血清样品中miR-588表达进行检测的药物或试剂盒。
在本发明的一些具体实例中,一种用于胃癌诊断或预测胃癌预后的组合物,包含:基于RT-PCR方法和/或基于蛋白质印迹方法,对组织样品中CXCL5基因及其表达产物、和/或CXCL9基因及其表达产物、和/或CXCL10基因及其表达产物进行检测的药物或试剂盒。
进一步地,所述组合物中,所述基于RT-PCR方法进行检测的药物或试剂盒,包括RT-PCR扩增引物对,具体如下:
miR-588引物对,序列分别如SEQ ID NO:2和SEQ ID NO:3所示,U6引物对的序列分别如SEQ ID NO:4和SEQ ID NO:5所示;

CXCL5引物对,序列如SEQ ID NO:6和SEQ ID NO:7所示,扩增片段为56bp;

本发明的第四个方面,提供过表达miR-588的物质、和/或上调CXCL5基因及其表达产物的物质、和/或上调CXCL9基因及其表达产物的物质、和/或上调CXCL10基因及其表达产物的物质在如下(I)~(II)至少一种中的应用:
(I)抑制胃癌细胞的增殖,或制备用于抑制胃癌细胞增殖的产品;
(II)抑制胃癌细胞的侵袭,或者制备用于抑制胃癌细胞侵袭的产品。
在本发明的一些具体实例中,所述过表达miR-588的物质包括:miR-588上调慢病毒、质粒GV268-miR-588或miR-588-mimics
在本发明的一些具体实例中,所述上调CXCL5基因及其表达产物的物质包括质粒GV268-CXCL5。
在本发明的一些具体实例中,所述上调CXCL9基因及其表达产物的物质包括质粒GV268-CXCL9。
在本发明的一些具体实例中,所述上调CXCL10基因及其表达产物的物质包括质粒GV268-CXCL10。
在本发明的一些具体实例中,所述胃癌细胞为SGC-7901细胞或MNK28细胞。
本发明通过检测和比较胃癌患者和健康人群中血清miR-588的表达,发现:胃癌患者中血清miR-588表达显著低于健康人群中血清miR-588表达,且ROC曲线显示:在区别胃癌患者和健康人群方面,miR-588比传统胃癌肿瘤标志物CEA和CA19-9具有更好的敏感性和特异性。因此,miR-588能够用作胃癌分子标志物区别胃癌患者和健康人群,适合用于制备胃癌诊断的药物,尤其适合用于制备液体活检诊断胃癌的药物。
同时,通过检测和比较在胃癌组织和旁边非癌组织中CXCL5、CXCL9或CXCL10的表达,发现CXCL5、CXCL9或CXCL10在胃癌组织中的表达显著低于正常组织,这意味着CXCL5、CXCL9或CXCL10可用于制备胃癌诊断的药物。
此外,体外细胞实验结果显示:过表达miR-588能够上调CXCL5、CXCL9或CXCL10的表达,而过表达miR-588、CXCL5、CXCL9或CXCL10均可抑制胃癌细胞的增殖,过表达miR-588能够抑制胃癌细胞在体外的侵袭。动物实验结果显示:过表达miR-588能够抑制体内胃癌细胞的增殖、抑制肿瘤生长。这意味着CXCL5、CXCL9或CXCL10是抑癌因子,也是miR-588的靶基因,是miR-588调控胃癌增殖的主要途径。过表达miR-588通过上调靶基因CXCL5、CXCL9或CXCL10的表达进而显著性抑制胃癌细胞在体内外增殖和体外侵袭。
最后,统计显示:miR-588、CXCL5、CXCL9或CXCL10的高水平表达与胃癌患者更长的总生存期显著相关。这意味着miR-588、CXCL5、CXCL9或CXCL10可用于制备预测胃癌预后的药物,而过表达miR-588或上调靶基因CXCL5、CXCL9或CXCL10的表达,对胃癌患者预后有良性影响。
相对于现有技术,本发明具有以下有益的技术效果:
本发明提供了用于诊断早期胃癌的非侵入性标志物miRNA-588,且在区别胃癌患者和健康人群方面,miR-588比传统胃癌肿瘤标志物CEA和CA19-9具有更好的敏感性和特异性。
同时,本发明首次提出过表达miR-588能够上调CXCL5/9/10表达,miR-588通过上调CXCL5/9/10表达抑制肿瘤细胞活性,可以实现对肿瘤细胞在体内外的生长和侵袭的显著抑制,miR-588、CXCL5、CXCL9或CXCL10的高水平表达与胃癌患者更长的总生存期显著相关,CXCL5、CXCL9或CXCL10是抑癌因子,也是胃癌中miR-588的直接和功能相关靶点。

具体实施方式
下面结合附图和具体实施例对本发明的实施方案进行详细描述。应理解,这些实施例仅用于说明本发明,而不应视为限定本发明的范围。
以下实施例中,高速离心机、高速冷冻离心机购自德国Eppendorf公司;普通PCR反应仪和实时荧光PCR仪购自美国Roche公司;超低温冰箱购自海尔集团。mirVanaTM parisTMkit购自美国Ambion公司;Agilent RNA 6000Pico Kit购自美国Agilent公司;Trizol总RNA提取试剂购自美国Invitrogen公司;TaqMan MicroRNAReverse Transcription Kit、TaqMan Universal PCRMaster Mix、cDNA合成试剂盒购自美国赛默飞世尔ThermoScientific公司;引物、探针委托生工生物工程(上海)股份有限公司合成。质粒GV268-miR-588、GV268-CXCL5、GV268-CXCL9、GV268-CXCL10购于上海吉凯基因化学技术公司,miR-588抑制剂、miR-588-mimics购于生工生物工程(上海)股份有限公司。Lipofectamine 2000转染试剂(11668019)购自Thermo Fisher。实施例中未注明的仪器和试剂均为本领域技术人员常规使用的仪器和试剂,且可通过商业途径获得。实施例中未注明具体试验条件和试验方法的按照常规条件或者制造厂商所建议的条件实施。
胃癌患者和健康志愿者自2018年9月至2019年9月期间于浙江省肿瘤医院收集,取血经患者同意并通过浙江省肿瘤医院伦理委员会审批。测试集:胃癌患者45例,健康对照37例,样本年龄在30-65岁之间,且胃癌患者和健康对照人群年龄无统计学差异;所有胃癌患者和健康对照均经过组织病理确证,无既往肿瘤病史,且血常规正常。
早晨空腹抽取测试集胃癌患者和健康人群外周血3mL。将抽取的血液放入EDTA抗凝管内并轻轻上下颠倒混匀,同时使血与管内抗凝物质充分接触,防止血细胞破裂。将EDTA管放入4℃冰箱,在2小时内分离血浆:首先,1000g转速离心10分钟,将上层液相小心吸出,避免吸取中间白色细胞层;然后,将上层液相转入事先预冷的1.5mL离心管,16000g转速继续离心10分钟,以便进一步分离血浆和血细胞。将第二步离心以后得到的上层液相(血浆),以500μL体积分装,转入-80℃冰箱长期保存,作为待测血浆样本。
收集外周血到未加抗凝剂的采血管中,室温(25℃)静置30分钟,1500rpm离心5分钟,收集上清,得到待测血清样本。
实施例1:miR-588作为胃癌诊断分子标志物的统计分析
1、胃癌患者和健康人群中血清miR-588表达的比较与统计分析
以上述胃癌患者和健康人群的血清为对象,按照以下方法进行实时荧光定量PCR,检测胃癌患者和健康人群中的血清miR-588的表达(以U6基因作为内参对照)水平,并通过散点图来分析和比较。
实时荧光定量PCR方法:
以总RNA为逆转录模板,使用带有茎环结构的特异性引物将microRNA逆转录为cDNA。将RT的产物cDNA使用TaqMan探针法进行定量PCR检测,PCR扩增釆用特异性前引物及通用后引物(miR-588引物对的序列分别如SEQ ID NO:2和SEQ ID NO:3所示,U6引物对的序列分别如SEQ ID NO:4和SEQ ID NO:5所示),并使用特异性的TaqMan探针检测目的片段。具体参照试剂盒TaqMan MicroRNA Reverse Transcription Kit、TaqMan MicroRNAAssay、TaqMan Universal PCR Master Mix的说明书,每个样品上样量为100ng,反应步骤如下:
100ng RNA标本加入每个RT反应试管中,Nuclease-free water补足体积;加入各反应成分后稍作离心混匀,在PCR反应仪执行如下程序:
参照TaqMan MicroRNA Assay说明书在实时荧光定量PCR仪上进行,体系和条件如下:
加入各反应成分并充分混匀后稍作离心,每个孔10μL,每个样本做三个复孔。在实时荧光定量PCR仪上执行如下程序:
总共40个循环。60℃时进行荧光数据采集,吸收波长为490nm,释放波长为530nm。
检测结果:
检测血清miR-588表达的散点图如图1A所示,PCR检测结果显示:相对于内参U6,胃癌患者的血清miRNA-588表达是下调,健康个体的血清miRNA-588表达是上升;胃癌患者的血清miRNA-588表达水平明显低于健康人群的血清miRNA-588表达水平,差异性显著(**:p<0.01)。图中长横线代表中位值。采用Mann-Whitney U检验来计算统计学差异。
为了与传统胃癌肿瘤标志物CEA、CA19-9进行比较,按照目前医院常用的化学发光方法,检测胃癌患者和健康人群中血清CEA、CA19-9的表达水平,并通过散点图来分析和比较。检测血清CEA表达的散点图如图1B所示,检测结果显示:胃癌患者的血清CEA表达较之健康人群升高,有一定的差异性(*:p<0.05)。检测血清CA19-9表达的散点图如图1C所示,检测结果显示:胃癌患者的血清CA19-9表达较之健康人群升高,有一定的差异性(*:p<0.05)。

由图1A~图1C及表1可见,血清miR-588与血清CEA、CA19-9一样具有成为胃癌肿瘤标志物的潜力。
2、血清miR-588检测作为胃癌诊断工具的可行性分析
利用SPSS软件,采用ROC曲线分析来评价使用血清miR-588检测来作为胃癌诊断工具的可行性,同时,还比较了miR-588与传统胃癌肿瘤标志物CEA、CA19-9在区分胃癌患者和健康人群的特异性和敏感性的差异性,如图1D所示。
从图1D可以看出,在区别胃癌患者和健康人群方面,miR-588在ROC曲线以下的面积为0.796;CEA在ROC曲线以下的面积为0.694;CA19-9在ROC曲线以下的面积为0.658。比较发现,miR-588具有更高的敏感性和特异性。可见,ROC曲线分析显示:在区别胃癌患者和健康人群方面,miR-588比传统胃癌肿瘤标志物CEA和CA19-9具有更好的敏感性和特异性,miR-588能够作为胃癌潜在分子标志物区别胃癌患者和健康人群。
由此可见,可以将miR-588作为胃癌分子标志物,并将血清miR-588检测用于胃癌诊断:检测胃癌患者与健康人群对照组的血清miR-588的表达水平的差异,当检测结果显示血清miR-588水平低于对照组中的水平,则提示所述对象胃癌患病风险。
实施例2:miR-588调控的靶基因的预测及验证
为探究miR-588(序列如SEQ ID NO:1所示)对胃癌细胞进行调控的机制,寻找其下游关键靶点基因,我们使用miRWalk数据库寻找到上千条潜在靶基因,经过分析,我们筛选出CXCL5(NCBI ID:6374),CXCL9(NCBI ID:4283)和CXCL10(NCBI ID:3627)基因。进一步,还发现:miR-588在1171-1177bp的位点直接结合CXCL5,如图2A所示;miR-588在344-350bp和914-920bp的位点直接结合CXCL9,如图2B所示。我们推测miR-588是通过与CXCL5,CXCL9和CXCL10相互作用,从而降低了胃癌细胞的活力。
为了验证上述对于miR-588的靶基因的预测,进行了以下实验:
1、免疫组化实验检测胃肿瘤组织和相应的非肿瘤邻近胃组织中CXCL5,CXCL9和CXCL10的表达水平
选择测试集中胃癌患者的胃肿瘤组织和对应的非肿瘤邻近胃组织样本,免疫组化染色试剂盒购自北京中山金桥生物技术有限公司。
按常规方法进行免疫组化实验,检测CXCL5,CXCL9和CXCL10的表达水平:
将石蜡切片依次进行去石蜡,再水化和煮沸以回收抗原。然后,在室温下,将主要的CXCL5(D263012-0025,BBI),CXCL9(ab202961,Abcam),CXCL10(D220389-0025,BBI)(1:500)添加到每个切片中2小时。将切片在PBS中轻轻洗涤3次后,在室温下加入400μL组织化学聚合物增强剂20分钟。在PBS中轻轻洗涤3次后,添加二抗并孵育20分钟,然后进行洗涤,DAB染色,复染和固定。
免疫组化结果如图2C所示。免疫组织化学染色显示CXCL5,CXCL9和CXCL10蛋白在非肿瘤邻近胃组织(N)中大量且均匀表达,但在所有肿瘤样品中这些蛋白的表达均显著下调(C)。这与前述在胃癌患者和健康人群中血清miR-588表达差异是完全一致的,表明CXCL5,CXCL9和CXCL10和miR-588存在正向的遗传关系。
2、细胞株的来源说明:
(1)SGC-7901和MNK28细胞的培养:
胃癌细胞系SGC-7901和MNK28从中国典型培养物保藏中心(中国武汉)购买。将细胞在含有10%FBS和抗生素(100U/mL链霉素和100U/mL青霉素)的RPMI-1640培养基中于37℃在含有5%CO2培养。
(2)miR-588上调胃癌稳定株的构建:
miR-588上调慢病毒是由上海吉凯有限公司构建的。将MNK-28和SGC-7901细胞分别接种在6孔板中(2×105细胞/孔),并在补充有10%FBS的RPMI-1640培养基中于37℃培养16-24小时。然后,向每个孔中加入20μl miR-588慢病毒和3μL Polybrene(Solarbio,H8761),并孵育16小时。从孔中小心除去培养基,并将细胞在补充有10%FBS的RPMI-1640培养基中培养。
(a)48小时后,将SGC-7901细胞在含有10%FBS和10mg/ml新霉素(Sigma-Aldrich,CAS:1404-04-2)的RPMI-1640培养基中培养,用以构建高表达miR-588的SGC-7901稳转株。
(b)48小时后,将MNK-28细胞在含有10%FBS和2mg/ml新霉素的RPMI-1640培养基中培养,每三天更换一次培养基,并添加新霉素,用以构建高表达miR-588的MNK-28稳转株。
(3)转染构建过表达miR-588的胃癌细胞
用质粒GV268-miR-588转染MNK28细胞,得到过表达miR-588的MNK28细胞。用质粒GV268-miR-588转染SGC-7901细胞,得到过表达miR-588的SGC-7901细胞。
3、实时荧光定量PCR
以检测结果中最稳定的U6基因作为内参对照,按照下述方法实时荧光定量PCR检测MNK28细胞(control)和过表达miR-588的MNK28细胞(miR-588)中CXCL5、CXCL9和CXCL10的表达。同样的实时荧光定量PCR方法检测SGC-7901细胞(control)和过表达miR-588的SGC-7901细胞(miR-588)中CXCL5,CXCL9和CXCL10的表达。
实时荧光定量PCR的条件具体如下:
细胞中总RNA提取采用索莱宝总RNA提取试剂盒(R1200)。RNA浓度通过ThermoScientificTM NanoDrop Lite分光光度计测定。1ug总RNA通过TaKaRa逆转录试剂盒(RR037A)反转录成cDNA。荧光定量PCR反应采用BIOMIGA荧光定量PCR试剂盒(RT0411-01)。PCR反应参数:95℃30s,95℃5sec;60℃30sec共40个循环。60℃至95℃绘制溶解曲线。以上实验重复三次。用于PCR扩增的引物由上海生工公司合成。CXCL5引物对的序列如SEQ IDNO:6和SEQ ID NO:7所示,扩增片段为56bp;CXCL9引物对的序列如SEQ ID NO:8和SEQ IDNO:9所示,扩增片段为58bp;CXCL10引物对的序列如SEQ ID NO:10和SEQ ID NO:11所示,扩增片段为118bp。
检测结果如图3A和图3B所示。
图3A显示实时荧光定量PCR检测过表达miR-588的SGC-7901细胞和对照的SGC-7901细胞中miR-588、CXCL5,CXCL9和CXCL10的表达差异。由图3A可见:过表达miR-588的SGC-7901细胞中,miR-588、CXCL5、CXCL9和CXCL10均为高表达。与SGC-7901细胞(control)相比,过表达miR-588的SGC-7901细胞中CXCL5,CXCL9和CXCL10的表达均显著增加,该区别具有统计学差异(p<0.05)。
图3B显示过表达miR-588的MNK28细胞和对照细胞中miR-588、CXCL5,CXCL9和CXCL10的表达差异,图3B显示:过表达miR-588的MNK28细胞中,miR-588、CXCL5、CXCL9和CXCL10均为高表达。与MNK28细胞(control)相比,过表达miR-588的MNK28细胞中CXCL5,CXCL9和CXCL10的表达均显著增加,该区别具有统计学差异(p<0.05)。
4﹑蛋白质印迹
蛋白质印迹检测MNK28细胞(control)和过表达miR-588的MNK28细胞(miR-588)中CXCL5、CXCL9和CXCL10的表达。蛋白质印迹检测SGC-7901细胞(control)和过表达miR-588的SGC-7901细胞(miR-588)中CXCL5、CXCL9和CXCL10的表达。蛋白质印迹检测方法如下:
通过RIPA缓冲液(P0013C,Beyotime)裂解细胞,并以10,000×g离心10分钟。然后通过十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)分离提取的蛋白质。将蛋白质印迹与适当的抗体一起孵育,并通过增强的化学发光(ECL)或ECLPLUS(Amersham,Piscataway,NJ,USA)观察蛋白质条带。使用的抗体分别是CXCL5(D263012-0025,BBI),CXCL9(ab202961,Abcam),CXCL10(D220389-0025,BBI)和抗微管蛋白(Tubulin,#T8203,Sigma-Aldrich,St.Louis,MO,USA)。GeneGnome HR系统(Syngene,Cambridge,UK)用于扫描印迹。
结果如图3C和图3D所示。图3C显示蛋白质印迹检测过表达miR-588的SGC-7901细胞和对照SGC-7901细胞中CXCL5,CXCL9和CXCL10的表达差异,由图3C可见:过表达miR-588的SGC-7901细胞中,CXCL5、CXCL9和CXCL10表达均有所上调。图3D显示蛋白质印迹检测过表达miR-588的MNK28细胞和对照细胞中CXCL5,CXCL9和CXCL10的表达差异,由图3D可见:过表达miR-588的MNK28细胞中,CXCL5、CXCL9和CXCL10表达均有所上调。
图3A和图3B、以及图3C和图3D的结果表明:miR-588能够上调CXCL5/9/10的表达。
5﹑MTT测定
将胃癌细胞(SGC-7901或MNK28细胞)以1×104细胞/mL的密度接种在96孔板上。将细胞在37℃下温育24小时,并分为六组:分别用Lipofectamine 2000转染试剂(记为1-对照组)、miR-588抑制剂(记为2-抑制组)、miR-588-mimics(记为3-过表达组)、GV268-CXCL5(记为4-CXCL5组),GV268-CXCL9(记为5-CXCL9组)或GV268-CXCL10(记为6-CXCL10组)转染胃癌细胞,每组的不同孔分别转染24小时、48小时、72小时,随后,向每个孔中加入10μL MTT(5mg/ml),并将细胞在37℃,5%CO2下培养3小时。将二甲基亚砜(DMSO,150μL)添加到每个孔中。使用酶标仪(Bio-Rad Laboratories,Inc.,Hercules,CA,USA)在490nm处测定光密度。计算各组细胞增殖率,绘制曲线。
miR-588抑制剂的序列如SEQ ID NO:12所示,miR-588-mimics的序列如SEQ IDNO:13所示。
细胞增殖曲线图如图3E和图3F所示。图3E显示各组中胃癌细胞SGC-7901增殖的情况,结果显示:3-过表达组、4-CXCL5组、5-CXCL9组和6-CXCL10组中SGC-7901胃癌细胞的增殖明显不如1-对照组和2-抑制组,说明miR-588、CXCL5、CXCL9和CXCL10可以抑制SGC-7901胃癌细胞的增殖(或者说,过表达miR-588、上调CXCL5/9/10可以抑制SGC-7901胃癌细胞的增殖)。图3F显示各组中胃癌细胞MNK28增殖的情况,结果显示:3-过表达组、4-CXCL5组、5-CXCL9组和6-CXCL10组中SGC-7901胃癌细胞的增殖明显不如1-对照组和2-抑制组,说明miR-588、CXCL5、CXCL9和CXCL10可以抑制MNK28胃癌细胞的增殖(或者说,过表达miR-588、上调CXCL5/9/10可以抑制MNK28胃癌细胞的增殖)。
6、结晶紫测定
分别用Lipofectamine 2000转染试剂(记为对照组)、miR-588抑制剂(记为miR-588inhibitor)、miR-588-mimics(记为miR-588)、GV268-CXCL5(记为CXCL5),GV268-CXCL9(记为CXCL9)或GV268-CXCL10(记为CXCL10)转染胃癌细胞(SGC-7901或MNK28细胞,1×105细胞/孔),并一式三份接种在6孔板。转染12小时后,在37℃下培养48小时后,小心除去培养基。用3mL的磷酸盐缓冲盐水(PBS;P5368,Sigma-Aldrich)轻轻洗涤细胞后,添加750μL结晶紫溶液(ECM101,Sigma-Aldrich)。将细胞在室温下孵育10分钟,然后用自来水洗涤两次。最后将培养板倒置在纸巾上照相。
结晶紫测定单克隆形成实验结果通过奥林巴斯倒置显微镜观察,如图3G所示。图3G显示:用miR-588-mimics、GV268-CXCL5,GV268-CXCL9或GV268-CXCL10转染的胃癌细胞(SGC-7901或MNK28细胞)的数量显著少于用miR-588抑制剂和Lipofectamine 2000转染的胃癌细胞数量。这说明:(1)miR-588、CXCL5、CXCL9和CXCL10可以抑制SGC-7901胃癌细胞的增殖。(2)miR-588、CXCL5、CXCL9和CXCL10可以抑制MNK28胃癌细胞的增殖。
图3E和图3F、以及图3G的结果表明:在抑制胃癌细胞(SGC-7901或MNK28细胞)的增殖上,miR-588与CXCL5/9/10表现出相同的趋势。
7﹑Transwell侵袭测定
将基质胶(BD matrigelTM,354234)以1:8稀释(即,Matrigel胶与RPMI-1640培养基的体积比为1:8),并涂覆于Transwell腔室底部膜的上腔室表面(Corning,3412)。在没有FBS的RPMI-1640培养基中培养胃癌细胞(SGC-7901细胞或MNK-28细胞)。24小时后,分别将miR-588过表达组(高表达miR-588的SGC-7901稳转株或高表达miR-588的MNK-28稳转株)和对照组(被慢病毒载体感染的SGC-7901细胞或MNK-28细胞)接种到Transwell室中,每组各接种24小时、48小时。移走Transwell室并弃去培养基。将腔室用无钙PBS洗涤两次,用甲醇固定30分钟,然后干燥。将细胞用0.1%结晶紫染色20分钟,然后用棉签轻轻擦拭上部未迁移的细胞,并将剩余的细胞用PBS洗涤3次。立即在100倍放大下的三个视野中观察细胞,然后计数。在奥林巴斯显微镜下观察图像,如图3H~3K所示。
图3H显示的是在奥林巴斯显微镜下观察的miR-588过表达组和对照组中SGC-7901细胞的侵袭能力图,由图3H可见:与对照组相比,过表达miR-588的SGC-7901细胞的侵袭能力显著下降,即,miR-588过表达显著性抑制SGC-7901细胞的侵袭能力。
图3J显示的是在100倍放大下的三个视野中观察到的miR-588过表达组和对照组中SGC-7901细胞的数量统计对比图,由图3J可见:与对照组相比,过表达组的SGC-7901细胞数量明显少得多,这说明过表达miR-588的SGC-7901细胞的侵袭能力显著下调(**p<0.01)。
图3I显示的是在奥林巴斯显微镜下观察的miR-588过表达组和对照组中MNK-28细胞的侵袭能力图,由图3I可见:与对照组相比,过表达miR-588的MNK-28细胞的侵袭能力显著下降,即miR-588过表达显著性抑制MNK-28细胞的侵袭能力。
图3K显示的是在100倍放大下的三个视野中观察到的miR-588过表达组和对照组中MNK-28细胞的数量统计对比图,由图3K可见:与对照组相比,过表达组的MNK-28细胞数量明显少得多,这说明过表达miR-588的MNK-28细胞的侵袭能力显著下调(**p<0.01)。
图3H~3K说明:miR-588(或者说过表达miR-588)可以抑制胃癌细胞(SGC-7901细胞或MNK-28细胞)在体外的侵袭。
综上,用GV268-miR-588/CXCL5/CXCL9/CXCL10转染后,MNK28和SGC-7901细胞的增殖明显降低。结晶紫染色结果与MTT分析的结果一致。此外,miR-588的过表达显著降低了MNK28和SGC-7901细胞的侵袭能力。由此,可以推测miR-588是通过与CXCL5,CXCL9和CXCL10相互作用,从而降低了胃癌细胞的活力。
实施例3:动物实验证实miR-588与体内胃癌细胞的增殖显著相关
六周大的裸鼠(n=15)购自SLAC实验动物有限公司(中国上海),随机分为三组(每组n=5):SGC-7901细胞(1×107细胞/mL,对照组)、被慢病毒载体感染的SGC-7901细胞(1×107细胞/mL,载体组)和被miR-588慢病毒感染的SGC-7901细胞(1×107细胞/mL,过表达miR-588组),皮下注射到每只裸鼠(0.2mL)的背部。每周测量三次平均肿瘤体积。实验结束时(第35天),处死小鼠并切除肿瘤进行体积测量。
图4A给出了各组小鼠最后的肿瘤组织的照片,图4B给出了每7天测一次小鼠肿瘤体积大小数据。图4A和图4B显示:过表达miR-588组切除的肿瘤明显小于对照组和载体组。对照组、载体组的裸鼠瘤子明显大于miR-588过表达组。说明:miR-588抑制体内胃癌细胞的增殖。
实施例4:miR-588与胃癌患者的整体存活率(OS)的关系
为了调查miR-588、CXCL5、CXCL9和CXCL10是否与胃癌患者的预后相关,我们挖掘了规模最大的与生存相关的临床数据集Kaplan-Meier(KM)绘图仪(http://www.kmplot.com),计算了总生存期(OS)。该图评估了54,675个基因对癌症患者生存期的影响。使用Kaplan-Meier(KM)绘图仪分析了总共10,188个癌症样品,包括1,648个卵巢癌,4,142个乳腺癌,1,065个胃癌和2,437个肺样品微阵列表达谱。在这项研究中,我们分析了miR-588,CXCL5,CXCL9和CXCL10(高表达与低表达)对胃癌患者OS的影响,如图4C~4F所示,其中,对数p值和危险比(HR)为95%置信区间(CI)。由图4C~4F可见,miR-588、CXCL5、CXCL9和CXCL10的高水平表达与胃癌患者更长的总生存期显著相关(p<0.05),这说明miR-588是抑癌因子,说明miR-588持续低表达对于胃癌发生发展来说是个危险信号,也反应miR-588可以作为胃癌肿瘤标志物。同时,CXCL5、CXCL9和CXCL10与miR-588具有相同的趋势。这些与体外实验结论一致。
综上,本实施例阐述了一种靶向miR-588抑制胃癌侵袭迁移及增殖的分子机制,并表明miR-588可作为一种胃癌的分子标志物,同时也是一个胃癌潜在的治疗靶点。

1.miR-588在制备胃癌分子标志物中的应用。
2.如权利要求1所述的应用,其特征在于,所述胃癌分子标志物用于诊断胃癌或预测胃癌预后。
3.如权利要求2所述的应用,其特征在于,所述胃癌分子标志物用于液体活检诊断胃癌。
4.miR-588靶基因在制备用于胃癌诊断或预测胃癌预后的药物中的应用,所述miR-588靶基因包括CXCL5基因及其表达产物、CXCL9基因及其表达产物、CXCL10基因及其表达产物。
5.一种用于胃癌诊断或预测胃癌预后的组合物,其特征在于,包含:基于RT-PCR方法和/或基于蛋白质印迹方法,对样品中miR-588表达、和/或CXCL5基因及其表达产物、和/或CXCL9基因及其表达产物、和/或CXCL10基因及其表达产物进行检测的药物或试剂盒。
6.如权利要求5所述的组合物,其特征在于,包含:基于RT-PCR方法和/或基于蛋白质印迹方法,对血清样品中miR-588表达进行检测的药物或试剂盒;
和/或,基于RT-PCR方法和/或基于蛋白质印迹方法,对组织样品中CXCL5基因及其表达产物、和/或CXCL9基因及其表达产物、和/或CXCL10基因及其表达产物进行检测的药物或试剂盒。。
7.如权利要求5或6所述的组合物,其特征在于,所述基于RT-PCR方法进行检测的药物或试剂盒,包括RT-PCR扩增引物对,具体如下:
miR-588引物对,序列分别如SEQ ID NO:2和SEQ ID NO:3所示,
U6引物对,序列分别如SEQ ID NO:4和SEQ ID NO:5所示;
和/或,
CXCL5引物对,序列如SEQ ID NO:6和SEQ ID NO:7所示,扩增片段为56bp;
和/或,
CXCL9引物对,序列如SEQ ID NO:8和SEQ ID NO:9所示,扩增片段为58bp;
和/或,
CXCL10引物对,序列如SEQ ID NO:10和SEQ ID NO:11所示,扩增片段为118bp。
8.过表达miR-588的物质、和/或上调CXCL5基因及其表达产物的物质、和/或上调CXCL9基因及其表达产物的物质、和/或上调CXCL10基因及其表达产物的物质在如下(I)~(II)至少一种中的应用:
(I)抑制胃癌细胞的增殖,或制备用于抑制胃癌细胞增殖的产品;
(II)抑制胃癌细胞的侵袭,或者制备用于抑制胃癌细胞侵袭的产品。
9.如权利要求8所述的应用,其特征在于,所述过表达miR-588的物质包括:miR-588上调慢病毒、质粒GV268-miR-588或miR-588-mimics。

所述上调CXCL5基因及其表达产物的物质包括:质粒GV268-CXCL5;
