Abstract

本发明提供一种高效阻隔病菌口罩,为层叠结构,由外向内依次包括:第一无纺布层、阻隔病菌层及第二无纺布层;所述阻隔病菌层由聚丙烯纤维和抗菌明胶复合材料组成,其中,聚丙烯纤维占85‑95重量份,抗菌明胶复合材料占5‑15重量份。本发明制备的口罩对病菌具有很好的过滤效果,实验结果显示,对金黄色葡萄球菌的过滤效果可达到97%以上。

技术领域
本发明属于医用卫生材料技术领域,具体涉及一种口罩,特别涉及一种高效阻隔病菌口罩。
背景技术
2020年以来,新型冠状病毒逐渐在全球范围爆发,牵动着世人敏感的神经。在医护专家的不断呼吁下,人们对口罩的认知日渐重视。口罩是一种医用卫生材料用品,是覆盖在佩戴者口鼻部位用于阻挡如灰尘、有害气体、气味、飞沫病毒进出佩戴者口鼻的用具。在呼吸道传染病流行时,在粉尘等污染的环境中作业时,戴口罩具有非常好的作用。而如今,随着新型冠状病毒(COVID-19)的持续发酵,防止通过呼吸道侍郎该病毒的口罩需求也不断增加。
一般来说,大多数病毒的大小约为10nm至300nm,而此次疫情的新型冠状病毒(COVID-19)的大小约为125nm。目前,尽管口罩对阻隔病菌防止交叉感染起到了很好的作用。但是,目前即便是由世界卫生组织(WHO)推广的N95口罩所能阻挡的微粒的大小也仅仅是大约300nm.,因而也就无法彻底阻挡包括此次COVID-19在内的大多数病毒进入佩戴者口鼻。因此,开发一种可高效阻隔病菌的新型复合材料作为防疫口罩的过滤层使用,是有效阻止病毒传播开来的关键。
发明内容
本发明的目的是为了解决上述现有技术问题而提供的一种高效阻隔病菌口罩。
为了实现上述目的,本技术方案提供的技术方案为:提供的一种高效阻隔病菌口罩,为层叠结构,由外向内依次包括:第一无纺布层、阻隔病菌层及第二无纺布层;所述阻隔病菌层由聚丙烯纤维和抗菌明胶复合材料组成,其中,聚丙烯纤维占85-95重量份,抗菌明胶复合材料占5-15重量份;
所述抗菌明胶复合材料的制备过程包括如下步骤:
S1:将大麻二酚溶于乙醇-水溶液中,配制质量浓度为2~5%的大麻二酚溶液,然后将其与质量浓度为5~10%的甘油异硬脂酸酯水溶液在65~75℃条件下共混搅拌30~60min,然后加入共混溶液质量20~30%的羧甲基纤维素,之后超声处理30min,得到混合液A;
S2:配制质量浓度为10%的明胶水溶液,然后加入占明胶干重2~8%的羟丙基瓜尔胶,室温下搅拌40~60min,之后将其与混合液A等体积共混,室温下搅拌60~80min,调节混合液的pH值为7~9,得混合液B;
S3:将等体积的矢车菊素-3-芸香糖苷溶液、赖氨酸溶液、混合液B充分混合,并在室温下继续搅拌40~60min,最后将其注入模具中干燥即制备得到抗菌明胶复合材料。
优选地,所述步骤(1)中大麻二酚溶液与甘油异硬脂酸酯水溶液的体积比为1:(1~2);
优选地,所述步骤(3)中矢车菊素-3-芸香糖苷溶液的质量分数为2~5%,所述赖氨酸溶液的质量分数为6~10%。
优选地,所述的第一无纺布层和第二无纺布层由壳聚糖纤维、活性炭纤维及聚丙烯纤维中一种或几种构成。
更进一步地,所述的阻隔病菌层与第二无纺布层之间,还设置有相变纤维材料层。
优选地,步骤(1)所述乙醇-水溶液的体积比为1:1~1.5。
本发明至少具有如下的有益效果:
(1)矢车菊素-3-芸香糖苷上的羟基与赖氨酸上的羰基结合形成氢键,同时保护了赖氨酸上氨基,从而使得该材料具有很好的抑制病菌的效果;
(2)作为天然植物提取物的大麻二酚引入到明胶中,可增强其稳定性,而大麻二酚的分子两端具有两个羟基,其在碱性条件下会发生电子云偏离的共轭效应,进而促使其更好地发挥其抗炎抗菌的作用,在这种双重抗病菌因素的联合作用下,使得该材料的抗病菌作用得到进一步增强;
(3)亲水性材料羧甲基纤维素与矢车菊素-3-芸香糖苷分子之间的羟基形成了强烈的氢键,有利于材料形成一个整体,使得复合材料的机械性能增强,从而使材料性能更稳定;另一方面,可赋予材料一定的吸湿效果,从而形成一种有利于阻隔抑制病菌繁殖或穿透的微环境,有效保护佩戴者免受病菌侵袭。相变纤维材料层可吸湿吸热。
具体实施方式
下面给出本发明的实施例,通过实施例对本发明进行具体描述。有必要指出的是,实施例只用于对本发明做进一步的说明,不能理解为对本发明保护范围的限制,该领域的技术熟练人员可根据本发明的内容做出一些非本质的改进和调整。
预备实施例1:
提供一种明胶复合膜,其制备过程包括如下步骤:
(1)将大麻二酚溶于体积比为1:1的乙醇-水溶液中,配制质量浓度为4%的大麻二酚溶液,然后将其与质量浓度为5%的甘油异硬脂酸酯水溶液在75℃条件下共混搅拌30min,然后加入共混溶液质量20%的羧甲基纤维素,之后超声处理30min,得到混合液A;
(2)配制质量浓度为10%的明胶水溶液,然后加入占明胶干重4%的羟丙基瓜尔胶,室温下搅拌50min,之后将其与混合液A等体积共混,室温下搅拌60min,调节混合液的pH值为X,得混合液B;
(3)将等体积的矢车菊素-3-芸香糖苷溶液、赖氨酸溶液、混合液B充分混合,并在室温下继续搅拌40min,最后将其注入模具中干燥即制备得到抗菌明胶复合材料。
其中:
步骤(1):大麻二酚溶液与甘油异硬脂酸酯水溶液的体积比为1:1;
步骤(3):中矢车菊素-3-芸香糖苷溶液的质量分数为5%,所述赖氨酸溶液的质量分数为10%。
抑菌效果测定:本发明的效果测试选用大肠杆菌为灭菌对象和抑菌圈试验方法研究其抗菌性能。采用用米尺测定样品周围的抑菌圈大小,为保证数据的准确性,每隔60°在不同的方向对每个样品的抑菌圈宽度各测量6次,抑菌圈大小为各样品抑菌圈宽度的平均值,测量的抑菌圈大小。选取一片同样大小的白纸,作为空白对照。
为了准确测量,每组样品平行培养三组试样,将这三组试样再测一次平均值,得到的最终平均值的试验结果均列于表1中。

从表1的结果看,当步骤(2)中混合液的pH值大于9时,明胶复合膜的对大肠杆菌的抑菌能力在逐渐减弱,当pH<7时,明胶复合膜的抑菌能力一般。当pH在7~9时,即在适当的碱性条件下,由于大麻二酚的分子两端具有两个羟基,该羟基会发生电子云偏离的共轭效应,进而促使其更好的发挥抗菌抗作用。
预备实施例2:
将预备实施例1中步骤(3)中的矢车菊素-3-芸香糖苷溶液直接去除,或者用茶多酚、双醛黄原胶、京尼平、单宁酸替代,步骤(2)中pH值为7,其他条件与预备实施例1完全相同,制备得到的明胶复合膜按上述同样的方法测试其对大肠杆菌的抑菌性,结果如下表2所示。
预备实施例3:
将预备实施例1中步骤(1)羧甲基纤维素溶液去除,步骤(2)中pH值为7,其余与预备实施例1的条件完全相同,制备得到的明胶复合膜按上述同样的方法测试其对大肠杆菌的抑菌性,结果如下表2所示。

从上表中可以看出,用其他交联剂替换车菊素-3-芸香糖苷,其对大肠杆菌的抑菌效果下降明显,而制备条件中有无羧甲基纤维素对抑菌能力也有一定的影响。这主要是因为有矢车菊素-3-芸香糖苷时,一方面,其分子结构中的羟基与赖氨酸上的羰基结合形成氢键,同时保护了赖氨酸上氨基,从而使得该材料具有很好的抑制病菌的效果;另一方面,亲水性材料羧甲基纤维素与矢车菊素-3-芸香糖苷分子之间的羟基形成了强烈的氢键,有利于材料形成一个整体,使得复合材料的机械性能增强,从而使材料性能更稳定;而且羧甲基纤维素可赋予材料一定的吸湿效果,从而形成了一种有利于阻隔抑制病菌繁殖或穿透的微环境,有效保护佩戴者免受病菌侵袭。
实施例1
一种高效阻隔病菌口罩,为层叠结构,由外向内依次包括:第一无纺布层、阻隔病菌层及第二无纺布层;阻隔病菌层由聚丙烯纤维和抗菌明胶复合材料组成,其中,聚丙烯纤维占85重量份,抗菌明胶复合材料占15重量份;
其中,抗菌明胶复合材料的制备过程包括如下步骤:
(1)将大麻二酚溶于体积比为1:1的乙醇-水溶液中,配制质量浓度为4%的大麻二酚溶液,然后将其与质量浓度为5%的甘油异硬脂酸酯水溶液在75℃条件下共混搅拌30min,然后加入共混溶液质量20%的羧甲基纤维素,之后超声处理30min,得到混合液A;

一种高效阻隔病菌口罩,为层叠结构,由外向内依次包括:第一无纺布层、阻隔病菌层及第二无纺布层;阻隔病菌层由聚丙烯纤维和抗菌明胶复合材料组成,其中,聚丙烯纤维占90重量份,抗菌明胶复合材料占10重量份;
其中,抗菌明胶复合材料的制备过程包括如下步骤:
(1)将大麻二酚溶于体积比为1:1.5的乙醇-水溶液中,配制质量浓度为2%的大麻二酚溶液,然后将其与质量浓度为6%的甘油异硬脂酸酯水溶液在65℃条件下共混搅拌50min,然后加入共混溶液质量30%的羧甲基纤维素,之后超声处理30min,得到混合液A;
(2)配制质量浓度为10%的明胶水溶液,然后加入占明胶干重2%的羟丙基瓜尔胶,室温下搅拌45min,之后将其与混合液A等体积共混,室温下搅拌75min,调节混合液的pH值为8,得混合液B;
(3)将等体积的矢车菊素-3-芸香糖苷溶液、赖氨酸溶液、混合液B充分混合,并在室温下继续搅拌45min,最后将其注入模具中干燥即制备得到抗菌明胶复合材料。
其中:步骤(1)中大麻二酚溶液与甘油异硬脂酸酯水溶液的体积比为1:2;步骤(3)中矢车菊素-3-芸香糖苷溶液的质量分数为2%,所述赖氨酸溶液的质量分数为7%。
上述第一无纺布层由活性炭纤维构成、第二无纺布层由壳聚糖纤维构成。
阻隔病菌层与第二无纺布层之间,还夹杂一层相变纤维材料层。
实施例3
一种高效阻隔病菌口罩,为层叠结构,由外向内依次包括:第一无纺布层、阻隔病菌层及第二无纺布层;阻隔病菌层由聚丙烯纤维和抗菌明胶复合材料组成,其中,聚丙烯纤维占92重量份,抗菌明胶复合材料占8重量份;
其中,抗菌明胶复合材料的制备过程包括如下步骤:
(1)将大麻二酚溶于体积比为1:1.2的乙醇-水溶液中,配制质量浓度为3%的大麻二酚溶液,然后将其与质量浓度为10%的甘油异硬脂酸酯水溶液在70℃条件下共混搅拌50min,然后加入共混溶液质量25%的羧甲基纤维素,之后超声处理30min,得到混合液A;
(2)配制质量浓度为10%的明胶水溶液,然后加入占明胶干重8%的羟丙基瓜尔胶,室温下搅拌40min,之后将其与混合液A等体积共混,室温下搅拌70min,调节混合液的pH值为7.5,得混合液B;
(3)将等体积的矢车菊素-3-芸香糖苷溶液、赖氨酸溶液、混合液B充分混合,并在室温下继续搅拌50min,最后将其注入模具中干燥即制备得到抗菌明胶复合材料。

其中,抗菌明胶复合材料的制备过程包括如下步骤:
(1)将大麻二酚溶于体积比为1:1.3的乙醇-水溶液中,配制质量浓度为5%的大麻二酚溶液,然后将其与质量浓度为8%的甘油异硬脂酸酯水溶液在65℃条件下共混搅拌60min,然后加入共混溶液质量28%的羧甲基纤维素,之后超声处理30min,得到混合液A;
(2)配制质量浓度为10%的明胶水溶液,然后加入占明胶干重6%的羟丙基瓜尔胶,室温下搅拌60min,之后将其与混合液A等体积共混,室温下搅拌80min,调节混合液的pH值为9,得混合液B;
(3)将等体积的矢车菊素-3-芸香糖苷溶液、赖氨酸溶液、混合液B充分混合,并在室温下继续搅拌60min,最后将其注入模具中干燥即制备得到抗菌明胶复合材料。
其中:步骤(1)中大麻二酚溶液与甘油异硬脂酸酯水溶液的体积比为1:1.2;步骤(3)中矢车菊素-3-芸香糖苷溶液的质量分数为4%,所述赖氨酸溶液的质量分数为10%。

实施例5口罩的对细菌的过滤效率测试
上述实施例1~4中口罩对病菌的阻隔效果通过细菌过滤效率来表征(选用的菌种为金黄色),具体检测步骤如下:
(1)取金黄色葡萄球菌24h新鲜肉汤培养物,用含蛋白胨的磷酸盐缓冲液将其稀释配制成试验浓度的菌悬液,备用。试验前将口罩样品放置在温度为(21±5)℃,相对湿度为(85±5)%的环境中预处理至少4h;
(2)将气溶胶发生器与蠕动泵和气源相连,发生器喷雾口与管道进气口相连,管道有两个采样口。一个采样口与测试口罩相连,经空气微生物采样器采样后作为实验组;另一个采样口没有口罩,直接经空气微生物采样器采样后作为阳性质控。
(3)将菌悬液置于气溶胶发生器菌液瓶内,启动气溶胶发生器,按照流量5L/min,喷雾压力1.8kg/cm2进行喷雾发生气溶胶喷雾染菌1min,将直径90mm采样平板装入六级采样器,按采样流量28.3L/min,对测试口罩样品采样2min,对阳性质控采样1min。计算出细菌气溶胶的平均颗粒直径(MPS)应为(3.0±0.3)μm,细菌气溶胶分布的几何标准差应不超过1.5;
(4)将采样平板放入37C恒温箱中培养48h,计数每个采样平板菌落数(cfu),适用采样第4、5级的结果符合标准中的粒径要求,并使用校正表进行校正,细菌气溶胶的浓度换算为cfu/m3,根据过滤前后细菌气溶胶的浓度计算过滤效率。口罩细菌过滤效率计算公式:BFE=(C-T)/C×100%(式中C:阳性质控气溶胶浓度;T:测试样品气溶胶浓度)。
并选用豫北卫材生产的普通的医用外科口罩产品作为对照,其结果如表3所示:

从表3的结果可以看出,相对于豫北卫材生产的普通医用外科口罩,本发明条件下制造的口罩,其对病菌的过滤效果有较大提升,达到97%以上,具有显著的进步。
应当知道地是,本发明并不限定于上述实施方式。上述实施方式为示例,具有与本发明的权利要求书所述的技术思想实质上相同的构成并发挥相同作用效果的所有发明均包含在本发明的技术范围内。

Claims (6)

1.一种高效阻隔病菌口罩,其特征在于,为层叠结构,由外向内依次包括:第一无纺布层、阻隔病菌层及第二无纺布层;所述阻隔病菌层由聚丙烯纤维和抗菌明胶复合材料组成,其中,聚丙烯纤维占85-95重量份,抗菌明胶复合材料占5-15重量份;
所述抗菌明胶复合材料的制备过程包括如下步骤:
(1)将大麻二酚溶于乙醇-水溶液中,配制质量浓度为2~5%的大麻二酚溶液,然后将其与质量浓度为5~10%的甘油异硬脂酸酯水溶液在65~75℃条件下共混搅拌30~60min,然后加入共混溶液质量20~30%的羧甲基纤维素,之后超声处理30min,得到混合液A;
(2)配制质量浓度为10%的明胶水溶液,然后加入占明胶干重2~8%的羟丙基瓜尔胶,室温下搅拌40~60min,之后将其与混合液A等体积共混,室温下搅拌60~80min,调节混合液的pH值为7~9,得混合液B;
(3)将等体积的矢车菊素-3-芸香糖苷溶液、赖氨酸溶液、混合液B充分混合,并在室温下继续搅拌40~60min,最后将其注入模具中干燥即制备得到抗菌明胶复合材料。
2.根据权利要求1所述的高效阻隔病菌口罩,其特征在于,所述步骤(1)中大麻二酚溶液与甘油异硬脂酸酯水溶液的体积比为1:(1~2)。
3.根据权利要求1所述的高效阻隔病菌口罩,其特征在于,所述步骤(3)中矢车菊素-3-芸香糖苷溶液的质量分数为2~5%,所述赖氨酸溶液的质量分数为6~10%。
4.根据权利要求1所述的高效阻隔病菌口罩,其特征在于,所述的第一无纺布层和第二无纺布层由壳聚糖纤维、活性炭纤维及聚丙烯纤维中一种或几种构成。
5.根据权利要求1所述的高效阻隔病菌口罩,其特征在于,所述的阻隔病菌层与第二无纺布层之间,还设置有相变纤维材料层。
6.根据权利要求1所述的高效阻隔病菌口罩,其特征在于,步骤(1)所述乙醇-水溶液的体积比为1:1~1.5。
