Abstract
The invention discloses a peucedanum cluster bud culture medium with bud tips as explants, wherein each 1000mL of the culture medium contains 1.5-2.5mg of 6-benzylamino adenine, 0.5-1.5mg of kinetin, 0.2-0.4mg of indoleacetic acid, 160mg of coconut milk, 3-5g of agar powder, 28-34g of cane sugar and the balance of a basic culture medium. The peucedanum praeruptorum clustered bud culture medium taking the bud tips as the explants can greatly improve the induction rate of the peucedanum praeruptorum clustered bud induction taking the bud tips as the explants, improve the production efficiency and save the production cost.

Technical Field
The invention relates to the technical field of peucedanum praeruptorum tissue culture, in particular to a peucedanum praeruptorum cluster bud culture medium taking bud tips as explants.
Background
Radix Peucedani is a perennial herb of Umbelliferae, and has irregular cylindrical, conical or spindle-shaped root, and dark brown or gray yellow surface. Preferably, each 1000mL of the culture medium contains 2.0mg of 6-benzylamino adenine, 1.0mg of kinetin, 0.3mg of indoleacetic acid, 140mg of coconut milk, 4.0g of agar powder, 31g of cane sugar and the balance of a basal culture medium.
Preferably, the coconut milk is collected from immature coconut fruit grown for 5-7 months.
The basic culture medium can be a conventional basic culture medium for plant tissue culture, and preferably, the basic culture medium is an MS culture medium.

Test examples
The induction of multiple shoots was carried out using the medium of examples 1 to 11 and comparative examples 1 to 4, with the shoot tips as explants. The method comprises the following specific steps: washing strong terminal bud of radix Peucedani with running water for 0.5h, soaking in 75% ethanol for 5s under aseptic condition, soaking in 0.1% mercuric chloride solution for 10-15min, washing with sterile water for 3-5 times, stripping 0.8-1.0mm bud tip, inoculating on culture medium, and culturing under 24-26 deg.C, illumination intensity of 1200lx, and illumination time of 12h per day. Subculture for 1 time for 30 days, and recording the induction condition of cluster buds of the sterilized test-tube plantlet of radix peucedani when culturing for 60 days. The induction rate calculation method comprises the following steps: the induction rate is the number of bud tips inducing the cluster buds/the total number of inoculated bud tips.
The induction test results are shown in table 2:

From the test results in table 2, it can be seen that the peucedanum cluster bud culture medium using bud tips as explants according to the present invention can successfully induce cluster buds using bud tips as explants, and the induction rate can reach 96.8%, wherein the induction rate of peucedanum cluster buds using bud tips as explants can be greatly improved by adding coconut milk in the culture medium.

A peucedanum cluster bud culture medium taking bud tips as explants is characterized in that every 1000mL of the culture medium contains 2.0mg of 6-benzylamino adenine, 1.0mg of kinetin, 0.3mg of indoleacetic acid, 140mg of coconut milk, 4.0g of agar powder, 31g of cane sugar and the balance of basal culture medium; and (4) stripping bud tips, inoculating the bud tips on a culture medium, and performing cluster bud induction.
2. The culture medium for the clustered shoots of peucedanum praeruptorum with shoot tips as explants according to claim 1, wherein the coconut milk is collected from immature coconut fruits growing for 5-7 months.
3. The culture medium for the clustered shoots of peucedanum praeruptorum with shoot tips as explants according to claim 1, wherein the basic culture medium is MS culture medium.
