Abstract
A health food for inhibiting tumor and improving immunity and a preparation method thereof are provided, the health food is prepared from the following raw materials in parts by weight: 13-25 parts of cordyceps militaris powder, 6-15 parts of pine pollen, 5-15 parts of selenium-rich malt powder, 3-12 parts of naked algae powder, 20-70 parts of fruit and vegetable coarse cereal powder and 0-5 parts of stevioside. The health food has dual anticancer effects, can inhibit tumor cell proliferation, and improve immunity, and is a nutritional partner for tumor patients during operation, radiotherapy, chemotherapy and rehabilitation.

Technical Field
The invention provides a health food for inhibiting tumor and improving immunity, belonging to the technical field of health foods.
Background
The number of new cancer cases is about 380 million each year, the number of deaths is about 229 million, the average annual incidence rate of the cancer is increased by about 3.9 percent, and the incidence rate and the mortality rate are on the increasing trend year by year. Cancer becomes one of four chronic diseases in China, and the health of people in China is seriously influenced.
Aiming at the treatment of cancer, China advocates multidisciplinary and standardized comprehensive treatment. In the comprehensive treatment of tumors, the traditional Chinese medicine not only assists the treatment of western medicine operations and chemoradiotherapy, but also can dialectically treat different cancer species at different stages.
At present, some cordyceps products and selenium-rich products are used for the auxiliary treatment of cancer prevention and cancer resistance, but the curative effect of the cordyceps products and the selenium-rich products is still to be further improved.
Disclosure of Invention
Aiming at the problems in the prior art, the invention provides the health food with the effects of inhibiting tumor and improving immunity, has double anti-cancer effects, can inhibit the proliferation of tumor cells and improve the immunity of the organism, and is a nutrition partner of tumor patients during operations, chemoradiotherapy and rehabilitation.
In order to achieve the purpose, the invention comprises the following technical scheme:
a health food for inhibiting tumor and improving immunity is prepared from the following raw materials in parts by weight: 13-25 parts of cordyceps militaris powder, 6-15 parts of pine pollen, 5-15 parts of selenium-rich malt powder, 3-12 parts of naked algae powder, 20-70 parts of fruit and vegetable coarse cereal powder and 0-5 parts of stevioside.
The health food for inhibiting tumor and improving immunity is preferably prepared from the following raw materials in parts by weight: 17-20 parts of cordyceps militaris powder, 10-12 parts of pine pollen, 9-11 parts of selenium-rich malt powder, 6-8 parts of euglena powder, 50-60 parts of fruit and vegetable coarse cereal powder and 0-2 parts of stevioside.
The health food for inhibiting tumor and improving immunity as described above is preferably prepared from the cordyceps militaris powder by the following method: the artificially cultured fresh cordyceps militaris sporocarp is prepared by drying, crushing and sieving processes, wherein the content of cordycepin is 1 wt%.
The health food for inhibiting tumor and improving immunity is preferably prepared from the following raw materials by the following steps: manually collected pollen pini is sieved by a 120-mesh sieve, and the pollen pini is subjected to jet milling to break the wall, wherein the content of crude polysaccharide is 1.5 wt%. Preferably, the frequency of the wall breaking machine is 55 Hz-60 Hz, and the pressure of the air source is 0.7-1.0 MPa.
The health food for inhibiting tumor and improving immunity is preferably prepared from selenium-rich malt sprouts by cleaning, drying and pulverizing, wherein the selenium content is 30-50 mg/kg.
Preferably, the gymnocyanine powder is prepared by culturing, concentrating, drying and sieving gymnocyanine seeds, and the content of beta-glucan is 50 wt%.
Preferably, the fruit, vegetable and coarse cereal powder is prepared from one or more of orange powder, pumpkin powder, carrot powder and malt powder, and the fruit and vegetable powder is an aqueous extract of corresponding fruits and vegetables.
In another aspect, the present invention provides a method for preparing a health food for inhibiting tumor and enhancing immunity, the method comprising the steps of: weighing the raw materials, uniformly mixing, and filling to obtain the product.
In another aspect, the invention provides the application of the health food for inhibiting tumor and improving immunity as a dietary therapy product for inhibiting tumor and improving immunity, wherein the oral dose is 5-10 g/day.
Preferably, the tumor is human lung cancer cell A549, human breast cancer cell MCF-7 or human chronic myelogenous leukemia cell K562.

The traditional Chinese medicine considers that the cordyceps sinensis enters the lung and kidney meridians, can tonify lung yin and kidney yang, is mainly used for treating kidney deficiency, impotence and spermatorrhea, soreness and pain of waist and knees, weakness after illness, chronic cough and weakness, cough and phlegmatic blood, spontaneous perspiration and night sweat and the like, and is the only traditional Chinese medicine capable of balancing and adjusting yin and yang. Record of Chinese medicine sea: cordyceps militaris, also known as Cordyceps militaris, is sweet and neutral in nature and flavor and enters lung and kidney meridians. The product has effects of invigorating kidney yang, nourishing lung yin, protecting lung, invigorating kidney, replenishing essence, and invigorating qi, and has good therapeutic effect on lung and kidney deficiency, chronic cough, asthma, and cough with excessive phlegm and blood. The Chinese herbal medicine compilation records: the fruiting body and polypide of Cordyceps militaris can also be used as a substitute for Cordyceps.
Pollen Pini is a kind of product which can be used as both medicine and food and is collected in ancient Chinese medical books. Li Shizhen is recorded in Ben Cao gang mu: pine pollen, sweet, warm and nontoxic; moisten heart and lung, tonify qi, dispel wind to stop bleeding. Pollen pini contains more than 200 kinds of nutrients such as saccharides, lipids, proteins, amino acids, nucleic acids, vitamins, minerals, etc., including 8 kinds of amino acids, 14 kinds of vitamins and 24 kinds of trace elements essential for human body, and is known as "micro-nutrient bank" and has multiple functions of regulating immunity, resisting fatigue, improving gastrointestinal function, regulating blood fat and protecting liver.
Selenium is a trace element necessary for human body, cannot be synthesized in human body, is an element easy to excrete, and gradually decreases with age. Researches find that selenium is closely related to the occurrence and development of tumors. Selenium can help to remove free radicals in vivo, repair membrane molecule damage, and prevent genetic variation or canceration of organism cells; selenium has an activating effect on a DNA repair system and can resist the damage of a carcinogen to DNA; selenium can induce tumor cell apoptosis.
The Euglena contains more than 20% of protein, essential vitamins, minerals, amino acids, unsaturated fatty acids, etc., is natural and comprehensive nutrient substance, is rich in beta (1, 3) glucan, and can stimulate non-specific immunity of human body.
The health food of the invention compounds each active component according to specific proportion, wherein: cordycepin, Cordyceps polysaccharide and adenosine in Cordyceps militaris have anti-tumor effect; selenium is a strong antioxidant and can assist in clearing free radicals in vivo and prevent cytogenetic variation; the compound of the euglena and the pine pollen can supplement natural and comprehensive nutrient substances and can stimulate the autoimmune system of a human body.
The invention has the beneficial effects that: the invention selects high-quality raw materials to form the health food for inhibiting tumor and improving immunity, the components are mutually matched and synergically, and the inhibition effect on cancer cells is obviously higher than that of a single component. Experimental results show that the health food has the effect of inhibiting human lung cancer cells A549, human breast cancer cells MCF-7 and human chronic myelogenous leukemia cells K562, and has reliable and safe effect and good taste.

Detailed Description
Sources of some of the raw materials in the following examples:
the cordyceps militaris powder is purchased from the development center of Shanghai national treasure enterprises, the content of the cordycepin is 1 percent, and the product is prepared by adopting the following method: the artificially cultured fresh cordyceps militaris sporocarp is prepared by drying, crushing and sieving processes.
The pine pollen is prepared by the following method: sieving manually collected pollen pini with a 120-mesh sieve, performing jet milling to break the walls, wherein the frequency of a wall breaking machine is 55 Hz-60 Hz, the pressure of an air source is 0.7-1.0 MPa, and the content of crude polysaccharide is 1.5%.
The selenium-rich malt flour is purchased from Tianjin Tianma Kangbi selenium technology limited company, the selenium content is 30mg/kg, and the product is prepared by the following method: the selenium-rich wheat sprout is prepared by cleaning, drying and pulverizing.
The gymnocyanin powder is purchased from the American Kemin company (KEMIN), the content of beta-glucan is 50%, and the product is prepared by the following method: the euglena seeds are prepared by culturing, concentrating, drying and sieving.
Example 1 health food for suppressing tumor and enhancing immunity
1. Formulation of
15 parts of cordyceps militaris powder, 13 parts of pine pollen, 13 parts of selenium-rich malt powder, 8 parts of naked algae powder, 12 parts of pumpkin powder, 12 parts of carrot powder, 12 parts of orange powder, 12 parts of malt powder and 1 part of stevioside.
2. Preparation method
The raw materials are uniformly mixed and filled to obtain a sample 1.
Example 2 health food for inhibiting tumor and enhancing immunity
1. Formulation of
20 parts of cordyceps militaris powder, 13 parts of pine pollen, 13 parts of selenium-rich malt powder, 8 parts of naked algae powder, 12 parts of pumpkin powder, 12 parts of carrot powder, 12 parts of orange powder, 12 parts of malt powder and 1 part of stevioside.
2. Preparation method
The raw materials are uniformly mixed and filled to obtain a sample 2.
Comparative example 1
1. Formulation of

Comparative example 4
1. Formulation of
49 parts of naked algae powder, 12 parts of pumpkin powder, 12 parts of carrot powder, 12 parts of orange powder, 12 parts of malt powder and 1 part of stevioside.
2. Preparation method
The raw materials are uniformly mixed and filled to obtain a comparative sample 4.
Experimental example 1
The samples 1 to 2 and the comparative samples 1 to 4 prepared in the examples and comparative examples were used to examine the inhibitory effects of these samples on human lung cancer cells A549, human breast cancer cells MCF-7 and human chronic myelogenous leukemia cells K562.
1. Principal materials and instruments
Human lung cancer cell A549, human breast cancer cell MCF-7 and human chronic myelogenous leukemia cell K562 were purchased from Bai Biotech Co., Ltd, Kyoto.
DMEM serum-free cell culture medium was purchased from Gibco; fetal Bovine Serum (FBS) and 100 Xpenicillin streptomycin mixed liquor are purchased from Hyclone company; trypsin was purchased from Roche; DMSO was purchased from Solarbio corporation; CCK-8 is available from Donjindo; PBS (phosphate buffered saline) was purchased from BBI life science; carbon dioxide incubators were purchased from Panasonic corporation; inverted optical microscope and fluorescence microscope were purchased from Nikon, Japan; desktop refrigerated centrifuges were purchased from HITACHI corporation; stat Fax-2100 enzyme linked immunosorbent assay (ELISA) instrument available from BIO-RAD; t25 cell culture flasks were purchased from Fisher; model HH-4 digital display thermostat water bath from Thermo corporation; YDS-20 type liquid nitrogen biological containers are purchased from Chengdu Jinfeng liquid nitrogen container GmbH; clean bench (SW-CJ-2D type) was purchased from NUAIRE corporation.
2. Experimental methods
2.1 tumor cell inhibition assay
A549, MCF-7 and K562 cells in logarithmic growth phase were selected as 1 × 105Each/mL was inoculated in a 96-well plate at 100. mu.L of DMEM medium per well. After the cells are cultured for 12h, adding 100 mu L (200 mu g/mL) of samples, and arranging 6 samples in parallel; the negative control group is that the cells are added with culture solution with the same volume; the blank group had only culture medium; standing at 37 deg.C for 5% CO2And incubating for 12h, 24h and 48h in an incubator with saturated humidity. After the culture is finished, carefully sucking out the original culture solution, adding 10 mu L of CCK-8 solution into each well, incubating for 1-4h, and then measuring the light absorption value under the wavelength of 450nm by using an enzyme-labeling instrument. The inhibition rate of the sample on the cells is calculated according to the following formula:
inhibition (%) (experimental OD value-negative OD value)/(negative OD value-blank OD value) × 100%
2.2 morphological Observation of tumor cells
A549, MCF-7 and K562 cells in logarithmic growth phase were selected as 1 × 105one/mL was inoculated in 12-well plates, 1mL DMEM medium per well. After 12h of cell culture, sample 1 was added, 1mL per well (200. mu.g/mL); the control group is cells added with culture solution with the same volume; standing at 37 deg.C for 5% CO2And incubating for 48h in an incubator with saturated humidity. After the culture was completed, the original culture solution was carefully aspirated, the cells were gently washed with PBS 2 times, and then stained with a certain volume of trypan blue, followed by observation with a fluorescent microscope. For K562 suspension cells, a certain volume of culture solution is directly sucked, centrifuged at 1000rpm for 5 minutes, washed once with PBS, centrifuged, added with a certain volume of culture medium, gently blown and uniformly mixed, added with trypan blue with the same volume for dyeing, and observed by a fluorescence microscope.
2.3 data analysis
Data were statistically analyzed using prism 8.0.
3. Results of the experiment
3.1 Effect of samples on the growth of human Lung cancer cells A549
A549 cells show different tolerance after the samples are acted for 12, 24 and 48 hours. The results are shown in table 1:
wherein, the sample 1 treats the A549 cells for 12h, and the sample promotes the cell proliferation of the A549 cells to reach 19.4%. After the A549 cells are treated for 24 hours, the sample 1 inhibits the proliferation of the A549 cells, and the inhibition rate reaches 22.3%. After 48 hours of treatment, the inhibition rate of the sample 1 on the proliferation of the A549 cells reaches 48.6 percent.
The results show that the inhibition effect of the sample 1 and the sample 2 on the A549 cells shows different effects along with different time, and the cordyceps sinensis polysaccharide has certain effect of promoting cell proliferation after being treated for 12 hours, and possibly has certain protection effect on the cells; with the increase of the treatment time, the cordycepin begins to exert the inhibition effect on the cell growth.

3.2 Effect of samples on the growth of human Breast cancer cells MCF-7
MCF-7 cells show different tolerance and inhibition effects after sample action for 12, 24 and 48 hours. The results are shown in table 2:
among them, samples 1 and 2 showed promotion-then-inhibition of MCF-7 cell proliferation, probably for the same reason as a549 cells. The highest inhibition rate of sample 1 at 48h reaches 13.4%.

3.3 Effect of samples on the growth of Chronic myelogenous leukemia cells K562
The K562 cell strain shows different tolerance and shows time-dependent effect after samples are treated for 12, 24 and 48 hours. The results are shown in Table 3:
among them, sample 1 had the best inhibitory effect, and after 12h of treatment of K562 cells, the inhibitory rate of sample 1 was 26.8%. After K562 cells are treated for 24 hours, the inhibition rate reaches 66.0%. After K562 cells were treated for 48h, the cell inhibition rate of sample 1 treatment continued to increase to 80.5%.

3.4 morphological Observation of samples after treatment of different cells
FIG. 1 is a photograph showing the morphology of A549 cells treated for 48 hours in sample 1 and control group, wherein A549 cells treated for 48 hours in sample are observed by a fluorescence microscope after staining. As can be seen, some debris appeared in the group of sample 1, i.e., the cell debris after cell disruption, most of the cells treated with sample 1 were disrupted, and the cells in the control group were intact.
FIG. 2 is a photograph showing the morphology of MCF-7 cells treated for 48 hours in sample 1 and control group, and the results of fluorescence microscopy show that most of the cells in the control group are intact, and the cell membrane of the sample 1 treated group is ruptured, and the content flows out.
FIG. 3 is a photograph showing the morphology of K562 cells after 48 hours of treatment in sample 1 and control group, and most of the cells in the treatment group are ruptured and the morphology of cell membranes cannot be distinguished after K562 cells are treated in sample 1 for 48 hours.
4. Conclusion
From the results of in vitro experiments, the optimal inhibition rates of the sample on A549, MCF-7 and K562 tumor cells are 48.6%, 13.4% and 80.5%, respectively. In a certain concentration range, the growth inhibition of tumor cells by samples 1 and 2 showed time-dependent effect. The specificity of the sample on the growth inhibition of tumor cells may be caused by different cell morphological structures on one hand and different cell signaling pathway regulation of three kinds of cancer cells on the other hand.
Claims (10)
1. A health food for inhibiting tumor and improving immunity is characterized by being prepared from the following raw materials in parts by weight: 13-25 parts of cordyceps militaris powder, 6-15 parts of pine pollen, 5-15 parts of selenium-rich malt powder, 3-12 parts of naked algae powder, 20-70 parts of fruit and vegetable coarse cereal powder and 0-5 parts of stevioside.
2. The health food for inhibiting tumor and improving immunity according to claim 1, which is prepared from the following raw materials in parts by weight: 15-20 parts of cordyceps militaris powder, 10-13 parts of pine pollen, 9-13 parts of selenium-rich malt powder, 6-8 parts of euglena powder, 48-60 parts of fruit and vegetable coarse cereal powder and 0-2 parts of stevioside.
3. The health food for suppressing tumor and enhancing immunity according to claim 1 or 2, wherein the cordyceps militaris powder is prepared by the following method: the artificially cultured fresh cordyceps militaris sporocarp is prepared by drying, crushing and sieving processes, wherein the content of cordycepin is 1 wt%.

The health food for inhibiting tumor and improving immunity according to claim 1 or 2, wherein the pine pollen is prepared by the following method: manually collected pollen pini is sieved by a 120-mesh sieve, and the pollen pini is subjected to jet milling to break the wall, wherein the content of crude polysaccharide is 1.5 wt%.
5. The health food for suppressing tumor and enhancing immunity according to claim 1 or 2, wherein the selenium-rich malt flour is prepared from selenium-rich malt sprouts by cleaning, drying and pulverizing, and the selenium content is 30-50 mg/kg.
6. The health food for suppressing tumor and enhancing immunity according to claim 1 or 2, wherein the gymnocyanine powder is prepared by culturing, concentrating, drying and sieving seeds of gymnocyanine, and has a β -glucan content of 50 wt%.
7. The health food for inhibiting tumor and improving immunity according to claim 1 or 2, wherein the coarse cereal fruit and vegetable powder is one or more selected from orange powder, pumpkin powder, carrot powder and malt powder.
8. The method for preparing a health food for suppressing tumor and enhancing immunity according to any one of claims 1 to 7, comprising the steps of: weighing the raw materials, uniformly mixing, and filling to obtain the product.
9. The use of the health food for suppressing tumor and enhancing immunity according to any one of claims 1 to 7 as a diet therapy product for suppressing tumor and enhancing immunity, wherein the oral dose is 5 to 10 g/day.
10. The use of claim 9, wherein the tumor is human lung cancer cell a549, human breast cancer cell MCF-7, or human chronic myelogenous leukemia cell K562.
