Abstract
本发明公开了一种与CD123蛋白特异性结合的多肽、多肽复合物、共递送系统及其制备方法与应用。该多肽中氨基酸排列的通式为X1‑DYDTR‑X2‑QGTD‑X3‑G‑X4‑DFRRISD‑X5;能够与CD123阳性细胞结合,抑制白血病的发展;多肽复合物能够提高该多肽与CD123阳性细胞的结合数量,单药即可抑制白血病的发展,延长白血病小鼠的生存期;该共递送系统可负载化疗药物用于肿瘤治疗,降低化疗药物对正常细胞的毒性。
Description
技术领域
本发明涉及生物医药技术领域,特别是涉及一种与CD123蛋白特异性结合的多肽、由该多肽与高分子材料形成的多肽复合物、含有该多肽复合物的共递送系统,该多肽、多肽复合物和共递送系统的制备方法与应用。
背景技术
急性髓系白血病(Acute myeloid leukemia,AML)是一种常见的恶性血液肿瘤,以异常增殖分化的不成熟髓系细胞在骨髓中浸润并抑制骨髓正常造血功能为特征,占急性白血病的70%左右。由于该病涉及大量的突变基因,靶向药物的治疗效果尚极为有限,因此,化疗仍然是该病的主要治疗手段。
现行AML化疗多采用以阿糖胞苷为基础的联合化疗方案,完全缓解率(治疗后检测患者的骨髓样品,其中白血病细胞比例低于5%的即为完全缓解)在70%左右。但是,化疗药物会使AML细胞上调某些表面蛋白的表达,降低细胞对化疗药物的敏感性,同时还引导细胞向骨髓、脾脏等基质细胞丰富的组织器官趋化归巢,在这些组织中形成微小残留病变,导致耐药性的产生和容易复发(患者在完全缓解后三年内的复发率高达50%-70%),使得AML预后不良(指患者获得治愈的可能性很低)。
此外,化疗药物普遍缺乏靶向性,会带来严重的毒副作用,老年患者对于化疗的耐受尤其差。目前国际上AML的五年总生存率也仅为25%左右(老年患者的生存率更是低于10%)。因此,AML的治疗一直面临重大挑战,迫切需要研发更为有效的新技术和新治疗手段。
有研究发现,CD123在正常的原始造血细胞和红系祖细胞中低表达或者不表达;虽然在骨髓细胞和B淋巴祖细胞中CD123表达量相对较高,但随着骨髓细胞和B淋巴祖细胞逐渐成熟,这些细胞表面CD123的表达量逐渐降低;当骨髓细胞和B淋巴祖细胞分别完全成熟成粒细胞和B淋巴细胞时,在粒细胞和B淋巴细胞中均检测不到CD123。但是,CD123在白血病干细胞及大部分白血病细胞表面高度表达,而且与AML预后不良密切相关。CD123不仅在AML中表达,在急性B淋巴细胞白血病、慢性粒细胞白血病、浆细胞样树突细胞肿瘤和毛细胞白血病等其他血液肿瘤中也有一定的表达。可见,CD123是白血病治疗的重要靶标蛋白。
CD123是白细胞介素3(Interleukin 3,IL-3)的受体,当与IL-3相结合时,AML细胞可以被激活而增殖。因此,阻遏CD123与IL-3的相互作用,是抑制白血病细胞增殖和诱导其凋亡的关键。目前已有抗CD123的抗体和抗体药物的报道,如7G3,属于鼠源性抗CD123单克隆抗体(Monoclonal antibody,MAb),可以与细胞表面CD123结合,从而阻止了CD123与IL-3相互作用,抑制了IL-3介导的白血病细胞增殖,使白血病细胞失去活性而死亡。又如CSL362,是鼠单克隆抗体(MAb 7G3)的人源化单克隆抗体,也具有特异性识别CD123并阻断AML细胞对IL-3响应的能力,使白血病细胞失去活性而死亡。如SL-401(Tagraxofusp),是一种靶向CD123的细胞毒素,由人IL-3与截短的白喉毒素(DT)重组融合而成,该药物可有效抑制白血病细胞的增殖。如SL-101,是将抗CD123单链Fv(scFv)与假单胞菌外毒素A融合制备得到的抗体偶联物,能够阻断由IL-3介导的信号传导途径,诱导白血病细胞的凋亡。上述化合物中,只有SL-401被美国FDA批准成为首个CD123靶向治疗药物,主要用于浆细胞样树突细胞肿瘤的治疗,其余药物均处于临床试验阶段(I期/II期),且在临床中尚未观察到突出疗效。
因此,研发新的CD123拮抗药物十分必要。
发明内容
本发明的目的是针对现有技术中存在的技术缺陷,第一方面,提供一种化学合成的多肽,其氨基酸排列的通式为X1-DYDTR-X2-QGTD-X3-G-X4-DFRRISD-X5;其中:
X1为空、或氨基酸D、或氨基酸序列为DD或GGDD的肽片段;
X2为A或R;
X3为I或D;
X4为C或N;
X5为空、或氨基酸D、或氨基酸序列为DD或DRR的肽片段。
为含有以下任一种氨基酸排列的多肽片段:
DYDTRAQGTDIGCDFRRISD、
DYDTRAQGTDDGCDFRRISD、
DYDTRRQGTDDGCDFRRISDD、
DDYDTRAQGTDIGCDFRRISDD、

制备上述多肽的方法,即使用标准固相多肽合成方法合成得到。
第二方面,本发明提供一种多肽复合物,由上述多肽与高分子材料复合得到,高分子材料能够负载更多的多肽。该多肽复合物,由包括1重量份的上述多肽和1-50重量份的高分子材料在内的原料制备得到;
优选的,所述高分子材料为高分子聚合物,所述多肽优选DDDYDTRAQGTDIGCDFRRISDDD或GGDDDYDTRAQGTDIGNDFRRISDDD;
更优选的,所述多肽复合物的平均水合直径在50-150nm。
所述高分子材料选自以下物质中的一种或几种:甲氧基聚乙二醇(mPEG)-聚己内酯(PCL)(mPEG-PCL)、二硬脂酰基磷脂酰乙醇胺(DSPE)-聚乙二醇2000(PEG2000)(DSPE-PEG2000)、聚乙烯己内酰胺-聚醋酸乙烯酯-聚乙二醇(Soluplus)、聚氧乙烯聚氧丙烯醚嵌段共聚物(聚氧乙烯和聚氧丙烯的分子量比为80:20)和聚乳酸-羟基乙酸共聚物(PLGA,聚乳酸和羟基乙酸的分子量比为50:50),优选甲氧基聚乙二醇(mPEG)-聚己内酯(PCL)(mPEG2000-PCL2000)、Soluplus(聚乙烯己内酰胺、聚醋酸乙烯酯和聚乙二醇的分子量比为57:30:13)、二硬脂酰基磷脂酰乙醇胺(DSPE)-聚乙二醇2000(PEG2000)(DSPE-PEG2000)。
由包括多肽DDDYDTRAQGTDIGCDFRRISDDD或GGDDDYDTRAQGTDIGNDFRRISDDD和甲氧基聚乙二醇(mPEG)-聚己内酯(PCL)(mPEG2000-PCL2000)或Soluplus(聚乙烯己内酰胺、聚醋酸乙烯酯和聚乙二醇的分子量比为57:30:13)的原料制备得到。
本发明提供上述多肽复合物的制备方法,将高分子材料和所述多肽溶于PBS溶液中,在25-75℃下反应20-120min,使高分子材料与多肽形成复合物,于室温静置至澄清透明,过滤除去游离多肽(例如用聚醚砜水系滤膜),即得到所述多肽复合物。
优选的,多肽复合物中多肽的负载率为60-100%。
第三方面,本发明提供一种共递送系统,由包括上述多肽复合物和化疗药在内的组分混合制得;所述化疗药选自长春新碱、阿霉素盐酸盐、喜树碱和环磷酰胺等中的一种或几种,优选阿霉素盐酸盐。
所述化疗药与所述多肽复合物的质量比为1:(10-80)。
本发明提供上述共递送系统的制备方法,将所述化疗药用水溶解,得到化疗药母液(浓度优选1-10mg/mL);将上述多肽复合物与所述化疗药母液在25-75℃下混合20-120min,于室温静置,过滤(例如用聚醚砜水系滤膜)去除游离的化疗药,得到所述共递送系统。
第四方面,本发明提供了上述多肽、多肽复合物、共递送系统在制备治疗白血病药物中的应用,白血病可以是急性髓系白血病、急性淋巴系白血病、慢性淋巴系白血病。
与抗体相比,本发明通过化学合成得到的多肽具有序列设计灵活性高、易于合成和修饰、成本相对较低、免疫原性低等优越性。实验证明,本发明提供的多肽能够与CD123阳性细胞特异性结合,且具有浓度依赖性;而在相同多肽浓度下,与CD123阴性细胞的结合很弱,说明该多肽具有靶向CD123阳性细胞的功能且亲和力较高;与此同时,该多肽还可以通过对CD123的拮抗,有效阻遏CD123与IL-3的相互作用,抑制IL-3介导的白血病细胞的活化和增殖。本发明提供的多肽复合物,是由上述多肽与高分子材料相结合形成的一种胶束形式的多肽复合物。实验证明,该多肽复合物能够负载多肽,且与CD123阳性细胞的结合量增加(与单用多肽相比),从而更好地抑制白血病细胞的活性。在对白血病动物的治疗实验中,本发明的多肽复合物可显著延长急性髓系白血病小鼠的生存期,具有治疗作用。本发明还在多肽复合物上再负载化疗药物,形成了一种拮抗CD123的多肽和化疗药物的共递送系统,实验证明,该共递送系统能够显著增强化疗药对CD123阳性细胞的杀伤力,同时降低化疗药对正常细胞的毒性(见实验9),说明本发明提供的共递送系统在肿瘤的靶向治疗应用中会有更显著优势。
本发明提供的靶向CD123的多肽、多肽复合物和共递送系统可为白血病的治疗提供可行的治疗策略,具有广阔前景。

具体实施方式
鉴于开发靶向CD123的多肽药物,对于白血病的有效治疗具有非常重要的意义,且截止目前尚未有文献报道靶向和拮抗CD123的多肽。本发明提供了一类靶向CD123的多肽,其氨基酸排列的通式为X1-DYDTR-X2-QGTD-X3-G-X4-DFRRISD-X5,其中,X1为空或氨基酸D或氨基酸序列为DD或GGDD的肽片段;X2为A或R;X3为I或D;X4为C或N;X5为空或氨基酸D或氨基酸序列为DD或DRR的肽片段;通式中D代表Asp(天冬氨酸)、Y代表Tyr(酪氨酸)、T代表Thr(苏氨酸)、R代表Arg(精氨酸)、Q代表Gln(谷氨酰胺)、G代表Gly(甘氨酸)、F代表Phe(苯丙氨酸)、I代表Ile(异亮氨酸)、S代表Ser(丝氨酸)、C代表Cys(半胱氨酸)、N代表Asn(天冬酰胺)。
在上述多肽(以下若无特殊说明,提到的多肽均为本发明提供的靶向CD123的多肽)的基础上,本发明还提供了一种多肽复合物。在上述多肽复合物的基础上,本发明还提供了一种共递送系统。
以下结合具体实施例,更具体地说明本发明的内容,并对本发明作进一步阐述,但这些实施例绝非对本发明进行限制。
下面将结合实施例对本发明的实施方案进行详细描述。实施例中未注明具体技术或条件者,按照本领域内的文献所描述的技术或条件,或者按照产品说明书进行。所用试剂或仪器未注明生产厂商者,均为可通过正规渠道商购获得的常规产品。
除非特别指明,以下实施例中所用的细胞株均购自国家实验细胞资源共享平台;所用MOLM-13、KG-1a和KG-1细胞均来自中国医学科学院血液病医院。
除非特别指明,以下实施例中所用水溶液的溶剂均为无菌超纯水溶液。
除非特别指明,以下实施例中所用的试剂均为分析纯试剂。
除非特别指明,以下实施例中所用的磷酸盐缓冲液均为1×PBS溶液(pH=7.4)。
实施例1:多肽及其制备
通过标准固相多肽合成方法制备得到本发明靶向CD123的多肽,其氨基酸序列如表1所示,均为白色粉末,纯度均≥98%,优选表1中编号为5#和6#所示氨基酸序列的多肽,代号为PO-6和PO-6N。为了验证本发明多肽的作用,在所有实验前用PBS缓冲溶液将表1所示的多肽配制成浓度为1mM的多肽母液。

实施例2:多肽复合物及其制备
多肽复合物的制备方法包括以下步骤:
(1)将1-50mg高分子聚合物和1mg多肽溶于PBS溶液中,经超声涡旋溶解得到溶液A;高分子聚合物选自甲氧基聚乙二醇(mPEG)-聚己内酯(PCL)(mPEG2000-PCL2000)、二硬脂酰基磷脂酰乙醇胺(DSPE)-聚乙二醇2000(PEG2000)(DSPE-PEG2000)、聚乙烯己内酰胺-聚醋酸乙烯酯-聚乙二醇(Soluplus)、聚氧乙烯聚氧丙烯醚嵌段共聚物(聚氧乙烯和聚氧丙烯的分子量比为80:20)、聚乳酸-羟基乙酸共聚物(PLGA,聚乳酸和羟基乙酸的分子量比为50:50)中的一种或几种,优选甲氧基聚乙二醇(mPEG)-聚己内酯(PCL)(mPEG2000-PCL2000,可商购)、Soluplus(可商购,聚乙烯己内酰胺、聚醋酸乙烯酯和聚乙二醇的分子量比为57:30:13)、二硬脂酰基磷脂酰乙醇胺(DSPE)-聚乙二醇2000(PEG2000)(DSPE-PEG2000)。
(2)将溶液A在25-75℃恒温水浴中反应20-120min,获得溶液B。
(3)将溶液B放于室温静置约1-2小时,至其澄清透明,用0.22μm聚醚砜水系滤膜过滤,除去游离的多肽,收集到的滤液中含有多肽复合物。
用该方法制备得到的多肽复合物中多肽和高分子材料结合形成胶束形式,通常平均水合粒径可在50-150nm范围内变化;以多肽复合物中负载的多肽量占步骤(1)加入多肽量的百分比为多肽复合物中多肽的负载率,收集到的多肽复合物多肽负载率可在50-100%,优选60%-100%。
按照以上方法,制备得到一系列多肽复合物C,仅是调整了使用的原料组成和制备参数,具体见表2。

实施例3:共递送系统及其制备
共递送系统的制备方法包括以下步骤:
(1)将化疗药(长春新碱、阿霉素盐酸盐、喜树碱、或环磷酰胺等)用无菌水溶解,配制成浓度为1-10mg/mL的化疗药母液,化疗药优选阿霉素盐酸盐。
(2)将步骤(1)得到的化疗药母液与以上多肽复合物按化疗药与多肽复合物的质量比1:(10-80)混合,在25-75℃恒温水浴中混合20-120min,以将化疗药包封在多肽复合物中,获得溶液D。
(3)将溶液D于室温静置,用0.22μm聚醚砜水系滤膜过滤,除去游离的药物,即得到多肽复合物与化疗药形成的共递送系统,其平均水合粒径为50-200nm。
以共递送系统中包封的化疗药量占步骤(2)加入化疗药量的百分比为共递送系统中化疗药的包封率,本实施例共递送系统中化疗药的包封率为30-99%,优选40%-99%。
本实施例共递送系统的使用浓度遵照各化疗药的使用要求。
按照以上方法,制备得到一系列多肽复合物与化疗药组成的共递送系统,仅是调整了所加化疗药的参数,具体见表3。

实验1:流式细胞术检测CD123在MOLM-13、KG-1a、KG-1、K562及HUVEC细胞中的表达
分别收集MOLM-13、KG-1a、KG-1、K562及HUVEC细胞至1.5mL Ep管中,PBS润洗一遍,经1500rpm,5min离心后,取沉淀并重悬于100μL含0.5wt%胎牛血清白蛋白的PBS溶液中,之后加入异硫氰酸荧光素酯(Fluorescein Isothiocyanate,FITC)标记的抗CD123抗体(FITCanti-human CD123,Biolegend),每100μL细胞悬液中加入2μL FITC标记的抗CD123抗体,室温下避光孵育30min。孵育后,PBS润洗一遍,在1500rpm下离心5min,取沉淀并重悬于100μLPBS溶液中,利用流式细胞仪检测细胞阳性率,结果如图1所示。
图1表明,MOLM-13细胞、KG-1细胞、KG-1a细胞的CD123表达阳性率均高于50%,属于表达CD123的阳性细胞;HUVEC、K562细胞的CD123表达阳性率均低于20%,属于CD123阴性细胞。
因此,在后续实验中使用MOLM-13细胞、KG-1细胞、KG-1a细胞作为表达CD123的阳性细胞。
实验2:流式细胞术检测本发明多肽与MOLM-13、K562细胞的结合情况
按每孔2×105个细胞将MOLM-13、K562细胞分别种于48孔板中,加入FITC标记的表1中所列多肽,使多肽终浓度分别为0.5μM、1μM、5μM、10μM。在37℃、5%CO2培养箱中孵育0.5h。孵育后,PBS润洗一遍,在1500rpm下离心5min,弃掉上清,每孔加入100μL PBS溶液重悬细胞,300目筛网过滤,用流式细胞仪检测MOLM-13、K562细胞的平均荧光强度(MeanFluorescence Intensity,MFI),以MFI表示多肽在细胞上的结合量。图2和图3显示了多肽与细胞的结合量。
图2以多肽PO-6为例,显示PO-6能够与CD123阳性的MOLM-13细胞结合,且随着PO-6浓度的增加,细胞的平均荧光强度增强,说明多肽在细胞上的结合量增加,该结合具有浓度依赖性。在相同的PO-6浓度下,CD123阴性的K562细胞上的平均荧光强度非常低,且显著低于MOLM-13细胞的平均荧光强度,说明PO-6可与CD123阳性细胞结合,而与CD123阴性细胞的结合很弱。可见,PO-6具有靶向CD123阳性细胞的功能。
图3以多肽PO-6N为例,显示PO-6N能够与CD123阳性的MOLM-13细胞结合,且随着浓度的增加,细胞的平均荧光强度增强,说明结合的多肽数量增加,该结合具有浓度依赖性。在相同的PO-6N浓度下,CD123阴性的K562细胞的平均荧光强度非常低,且显著低于MOLM-13细胞的平均荧光强度,说明PO-6N与CD123阴性细胞的结合很弱。
表1中其它多肽与此实验的结果相同,即:与CD123阳性的MOLM-13细胞结合,与CD123阴性的K562细胞的结合很弱,不再一一列示。
实验3:多肽复合物水合粒径分布
按照实施例2和实施例3中多肽复合物及共递送系统的制备方法制备多肽复合物C-1及其与阿霉素盐酸盐组成的共递送系统。吸取1mL上述共递送系统的溶液置于比色皿中,使用Nano ZS90纳米粒度分析仪检测水合粒径尺寸分布。
得到的结果如图4、图5所示。
由图4可知,多肽复合物C-1的平均水合粒径约为77nm。由图5可知,多肽复合物C-1与化疗药阿霉素盐酸盐组成的药物共递送系统的平均水合粒径约为72nm。
图4和图5中,多肽复合物和药物共递送系统的粒径尺寸均呈正态分布,且峰宽较窄,说明多肽复合物C-1、多肽复合物C-1与化疗药阿霉素盐酸盐组成的药物共递送系统均形成了稳定的纳米颗粒,且尺寸均匀。此外,颗粒的粒径在100nm以下,有利于其发挥纳米尺寸效应。
对表2和表3中的其它多肽复合物和共递送系统也进行了此实验,得到类似结果,因此不再一一赘述。
实验4:流式细胞术检测本发明多肽复合物对MOLM-13细胞的亲和力
按每孔2×105个细胞将MOLM-13细胞种于48孔板中,分别加入FITC标记的表2中所列多肽复合物,使多肽复合物的终浓度分别为0.1μM、0.3μM、0.5μM、1μM。在37℃、5%CO2培养箱中孵育0.5h,然后用PBS润洗一遍,在1500rpm下离心5min,弃掉上清,每孔加入100μLPBS溶液重悬细胞,300目筛网过滤;多肽复合物具有绿色荧光,细胞若获得绿色荧光,说明多肽复合物结合到了细胞上,用流式细胞仪检测细胞平均荧光强度(MFI),以MFI表示多肽在细胞上的结合量。
以多肽复合物C-1为例,结果如图6所示。
图6结果表明,多肽复合物C-1能够与MOLM-13细胞结合,且随着多肽浓度的增加,细胞的平均荧光强度增强,具有浓度依赖性;说明多肽复合物C-1具有靶向CD123阳性细胞MOLM-13的功能。
表2中其它的多肽复合物与此实验的结果相同,即:能够与MOLM-13细胞结合,且随着多肽浓度的增加,细胞的平均荧光强度增强,具有浓度依赖性,不再一一列示。
实验5:流式细胞术检测本发明多肽复合物对KG-1a细胞的亲和力
同实验4,仅是将实验3中的MOLM-13细胞换成KG-1a细胞。以多肽复合物C-3为例,结果如图7所示。
图7结果表明,多肽复合物C-3能够与KG-1a细胞有效结合,且随着多肽浓度的增加,细胞的平均荧光强度增强,具有浓度依赖性;说明多肽复合物C-3具有靶向CD123阳性细胞KG-1a的能力。
表2中其它的多肽复合物与此实验的结果相同,即:能够与KG-1a细胞结合,且随着多肽浓度的增加,细胞的平均荧光强度增强,具有浓度依赖性,不再一一列示。
实验6:流式细胞术检测本发明多肽复合物对KG-1细胞的亲和力
同实验4,仅是将实验3中的MOLM-13细胞换成KG-1细胞。以多肽复合物C-1’为例,结果如图8所示。
图8结果表明,多肽复合物C-1’能够与KG-1细胞有效结合,且随着多肽浓度的增加,细胞的平均荧光强度增强,具有浓度依赖性;说明多肽复合物C-1’具有靶向CD123阳性细胞KG-1的能力。
表2中其它的多肽复合物与此实验的结果相同,即:能够与KG-1细胞结合,且随着多肽浓度的增加,细胞的平均荧光强度增强,具有浓度依赖性,不再一一列示。
实验7:激光共聚焦显微镜观察本发明多肽、多肽复合物与MOLM-13的结合情况
MOLM-13细胞(每孔2×105个)与FITC标记的本发明多肽(以PO-6、PO-6N为例)、多肽复合物(见表2)在37℃条件下孵育30min,多肽的终浓度均为0.5μM(多肽复合物的培育体系中多肽的终浓度也为0.5μM)。孵育后,收集细胞并用PBS溶液洗两遍,在1500rpm下离心5min,弃掉上清,每孔加入400μL多聚甲醛的PBS水溶液(含4wt%多聚甲醛),在室温下避光放置30min,使细胞被多聚甲醛固定。用PBS溶液润洗一遍,在2000rpm下离心5min,将细胞滴于玻片上自然晾干,然后使用含DAPI(4',6-二脒基-2-苯基吲哚,4',6-diamidino-2-phenylindole)的封片剂封片。用激光共聚焦显微镜观察并拍照(Ex=488nm(FITC);Ex=405nm(DAPI))。以不做任何处理的MOLM-13细胞作为对照组,即为Con,多肽PO-6和PO-6N及相应的多肽复合物(以C-1、C-1’为例)的照片如图9、图10所示。
图9和图10中,FITC标示的是多肽,DAPI标示的是细胞核,Merge标示的是FITC和DAPI叠加后的效果照片。
图9表明,与Con相比,FITC标记的PO-6、多肽复合物C-1与MOLM-13细胞孵育0.5h后,在细胞表面能够观察到明显的绿色荧光,且C-1孵育后细胞表面的荧光明显强于PO-6孵育后的荧光,说明PO-6和多肽复合物C-1均具有特异性靶向MOLM-13细胞并与之结合的功能,且C-1的结合功能更强。
图10表明,与Con相比,FITC标记的PO-6N、多肽复合物C-1’与MOLM-13细胞孵育0.5h后,在细胞表面能够观察到明显的绿色荧光,且C-1’孵育后细胞表面的荧光明显强于PO-6N孵育后的荧光,说明PO-6N和多肽复合物C-1’均具有特异性靶向MOLM-13细胞并与之结合的功能,且C-1’的结合功能更强。
表2中其它的多肽复合物(C-2~C-6,C-2’~C-6’)与此实验的结果相同,即:与MOLM-13细胞孵育0.5h后,在细胞表面能够观察到明显的绿色荧光,且多肽复合物孵育后细胞表面的荧光明显强于对应多肽孵育后的荧光,不再一一列示。
实验8:本发明共递送系统对MOLM-13细胞活性的影响
设置以下实验组并配置实验用母液,包括:
1)多肽PO-6组(PO-6):实施例1得到的多肽母液(多肽浓度为1mM);
2)阿霉素组(Dox):阿霉素盐酸盐用无菌水溶解得到1mg/mL(1.7mM)的Dox母液;
3)高分子聚合物组(mPEG2000-PCL2000):称取15mg mPEG2000-PCL2000粉末,用1mL PBS溶液溶解,得到15mg/mL(3.3mM)的mPEG2000-PCL2000母液。
4)多肽PO-6和Dox联合组(PO-6+Dox):实验组1)的多肽母液和实验组2)的Dox母液分别用PBS溶液稀释后混合得到混合液,混合液中多肽的浓度为36.2μM,Dox的浓度为17.24μM,多肽PO-6与Dox的重量比为10:1;
5)多肽复合物组(C-1):将表2所列的多肽复合物C-1用PBS溶液稀释至PO-6的浓度为36.2μM;
6)阿霉素胶束组(M-Dox):将实验组2)的Dox母液和实验组3)的mPEG2000-PCL2000母液用PBS溶液稀释至mPEG2000-PCL2000的浓度为3.3μM,Dox的浓度为17.24μM,然后按mPEG2000-PCL2000与Dox的重量比为150:1混合。
7)本发明共递送系统组(C-1-Dox):将表3所列的共递送系统用PBS溶液稀释至多肽的浓度不高于36.2μM,Dox的浓度不高于17.24μM。
实验过程:按每孔8×104个细胞将MOLM-13细胞种于96孔U型板中。各实验组中每孔加入相应的药物母液,使各孔Dox终浓度为0.1μM,对应的多肽终浓度为0.21μM。在37℃,5%CO2培养箱中孵育24h,然后用PBS清洗细胞,再向每孔加入110μL CCK8稀释液(含100μLRPMI 1640培养基+10μL CCK-8),孵育2h,用酶标仪测定450和630nm波长下的吸光度值(OD值),按照公式1计算细胞存活率;对照组为不加药物处理的细胞,记为Con;
公式1:细胞存活率=OD450-630(实验组)/OD450-630(对照组)×100%,结果如图11所示。
图11表明,当共递送系统中Dox浓度为0.1μM、PO-6为0.21μM时,PO-6组、Dox组、C-1组、M-dox组、PO-6+Dox组、C-1-Dox组的细胞存活率分别为98%、57%、93%、38%、55%、34%。
该结果说明,与PO-6组、Dox组、C-1组、M-Dox组、PO-6+Dox相比,在相同Dox浓度下,共递送系统赋予了Dox靶向性,因此C-1-Dox针对MOLM-13细胞的毒性(细胞存活率为34%)比PO-6+Dox组(细胞存活率为55%)、M-Dox组(细胞存活率为38%)的细胞毒性更强,具有显著性差异。
本实验中的共递送系统组(第7)组)用表3中其它共递送系统替换,结果表明,各组分浓度相同的前提下,本发明共递送系统组对MOLM-13细胞的细胞存活率在28-37%范围内,均低于PO-6+Dox组和M-Dox组。
实验9:本发明共递送系统对HUVEC细胞活性的影响
按每孔1×104个细胞将HUVEC细胞种植于96孔板中。分组同实验8。各实验组中每孔加入相应的药物母液,使每孔Dox终浓度为1.0μM,对应的多肽终浓度为2.1μM。在37℃,5%CO2培养箱中孵育24h,然后用PBS清洗细胞,再向每孔加入110μL CCK8稀释液(含100μLRPMI 1640培养基+10μL CCK-8),孵育2h,用酶标仪测定450和630nm波长下的吸光度值(OD值),按照公式1计算细胞存活率;对照组为不加药物处理的细胞,记为Con。以多肽PO-6、多肽复合物C-1为例,结果如图12所示。
图12表明,在所测试浓度范围内,mPEG2000-PCL2000组、PO-6组、C-1组的细胞存活率均在90%以上;Dox、M-Dox、PO-6+Dox各组存活率在50%左右,而C-1-Dox组的细胞存活率大约在60%左右,说明C-1-Dox对CD123的靶向性没有增加Dox对HUVEC细胞的毒性,反而有所减弱,间接说明了PO-6与低表达CD123的HUVEC细胞之间的结合较低,有利于减少Dox对正常细胞的杀伤。
本实验中的共递送系统组(第7)组)用表3中其它共递送系统替换,结果表明,各组分浓度相同的前提下,本发明共递送系统组对HUVEC细胞的细胞存活率在56-68%范围内,均高于Dox组、PO-6+Dox组和M-Dox组。
实验10:本发明多肽复合物对AML1-ETO小鼠生存期的影响
本实验所采用的小鼠为6-8周龄的雌性C57鼠(来自中国医学科学院基础医学研究所的动物实验中心),在SPF级环境中饲养。用200μL乙二胺四乙酸钠盐(EDTA)的PBS溶液悬浮1×106个AML1-ETO小鼠原代脾脏细胞(GFP标记),通过尾静脉注射到经亚致死剂量辐照(450cGy)的C57小鼠体内。注射后第11天从小鼠尾静脉取血,用流式细胞仪检测外周血白血病细胞阳性率已高于10%,可确认小鼠已发病。确认发病的小鼠用于本实验,从移植后的第12天开始,尾静脉注射以下各组药物:
(1)表2中的多肽复合物C-1(多肽PO-6与mPEG2000-PCL2000的重量比为1:15),按多肽重量计10mg/kg(对应mPEG2000-PCL2000的给药剂量为150mg/kg),记作C-1组;
(2)无菌PBS溶液,作为对照组,记作Con组;
(3)经无菌PBS溶液溶解的mPEG2000-PCL2000,150mg/kg,记作mPEG2000-PCL2000组。每隔两天给一次药,结果见图13。
图13结果表明,Con组小鼠中位生存期为19天,mPEG2000-PCL2000组小鼠中位生存期为20天,C-1组小鼠中位生存期为25天,即C-1可显著延长AML1-ETO小鼠的生存期(p<0.05),说明本发明提供的多肽复合物对白血病有治疗作用。
本发明通过上述实施例来说明本发明的工艺方法,但本发明并不局限于上述工艺步骤,即不意味着本发明必须依赖上述工艺步骤才能实施。所属技术领域的技术人员应该明了,对本发明的任何改进,对本发明所选用原材料的等效替换及辅助成分的添加、具体方式的选择等,均落在本发明的保护范围和公开范围之内。
以上所述仅是本发明的优选实施方式,应当指出的是,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的内容。

1.一种与CD123蛋白特异性结合的多肽,其特征在于,其氨基酸排列的通式为X1-DYDTR-X2-QGTD-X3-G-X4-DFRRISD-X5;其中:
X1为空、或氨基酸D、或氨基酸序列为DD或GGDD的肽片段;
X2为A或R;
X3为I或D;
X4为C或N;
X5为空、或氨基酸D、或氨基酸序列为DD或DRR的肽片段。
2.根据权利要求1所述多肽,其特征在于,为含有以下任一种氨基酸排列的多肽片段:

优选的,所述高分子材料为高分子聚合物,所述多肽优选DDDYDTRAQGTDIGCDFRRISDDD或GGDDDYDTRAQGTDIGNDFRRISDDD;
更优选的,所述多肽复合物的平均水合直径在50-150nm。
5.根据权利要求4所述多肽复合物,其特征在于,所述高分子材料选自以下物质中的一种或几种:甲氧基聚乙二醇(mPEG)-聚己内酯(PCL)(mPEG-PCL)、二硬脂酰基磷脂酰乙醇胺(DSPE)-聚乙二醇2000(PEG2000)(DSPE-PEG2000)、聚乙烯己内酰胺-聚醋酸乙烯酯-聚乙二醇(Soluplus)、聚氧乙烯聚氧丙烯醚嵌段共聚物(聚氧乙烯和聚氧丙烯的分子量比为80:20)和聚乳酸-羟基乙酸共聚物(PLGA,聚乳酸和羟基乙酸的分子量比为50:50),优选甲氧基聚乙二醇(mPEG)-聚己内酯(PCL)(mPEG2000-PCL2000)、Soluplus(聚乙烯己内酰胺、聚醋酸乙烯酯和聚乙二醇的分子量比为57:30:13)、二硬脂酰基磷脂酰乙醇胺(DSPE)-聚乙二醇2000(PEG2000)(DSPE-PEG2000)。
6.根据权利要求5所述多肽复合物,其特征在于,由包括多肽DDDYDTRAQGTDIGCDFRRISDDD或GGDDDYDTRAQGTDIGNDFRRISDDD和甲氧基聚乙二醇(mPEG)-聚己内酯(PCL)(mPEG2000-PCL2000)或Soluplus(聚乙烯己内酰胺、聚醋酸乙烯酯和聚乙二醇的分子量比为57:30:13)的原料制备得到。
7.一种制备权利要求4或5或6所述多肽复合物的方法,其特征在于,将高分子材料和所述多肽溶于PBS溶液中,在25-75℃下反应20-120min,使高分子材料与多肽形成复合物,于室温静置至澄清透明,过滤除去游离多肽(例如用聚醚砜水系滤膜),即得到所述多肽复合物;
优选的,多肽复合物中多肽的负载率为60-100%。
8.一种与CD123蛋白特异性结合的共递送系统,其特征在于,由包括权利要求4或5或6所述多肽复合物和化疗药在内的组分混合制得;所述化疗药选自长春新碱、阿霉素盐酸盐、喜树碱和环磷酰胺等中的一种或几种,优选阿霉素盐酸盐。
9.根据权利要求8所述共递送系统,其特征在于,所述化疗药与所述多肽复合物的质量比为1:(10-80)。
10.一种制备权利要求8或9所述共递送系统的方法,其特征在于,将所述化疗药用水溶解,得到化疗药母液;将权利要求4或5所述多肽复合物与所述化疗药母液在25-75℃下混合20-120min,于室温静置,过滤(例如用聚醚砜水系滤膜)去除游离的化疗药,得到所述共递送系统。
11.权利要求1或2所述多肽、权利要求4或5或6所述多肽复合物、权利要求8或9所述共递送系统在制备治疗白血病药物中的应用,白血病包括但不限于急性髓系白血病、急性淋巴系白血病、慢性淋巴系白血病。
