Background
Mosquitoes are important vectors of many infectious diseases, and over 70 million people worldwide are killed by mosquito bites each year. The mosquito not only disturbs peace and affects sleeping life, but also carries more than 300 pathogenic microorganisms, is very easy to cause diseases such as dengue fever, filariasis, malaria and the like, and seriously harms the safety and health of human beings. The mosquito repellent is an effective means for solving the problem of mosquito-transmitted diseases. At present, mosquito repellent products in the market are roughly divided into two types. The insecticidal spray, the traditional mosquito-repellent incense, the electric heating mosquito-repellent incense tablet, the liquid and the like belong to pyrethroid chemical insecticides, the chemical mosquito-repellent incense has high toxicity and pungent taste, and the long-term frequent use of the chemical mosquito-repellent incense not only harms the human health and reduces the human immunity, but also causes the environment pollution by the generated smoke and has adverse effect on the respiratory system. In addition, mosquito repellent and itching relieving products such as toilet water, mosquito repellent spray, mosquito repellent bracelet, mosquito repellent patch and the like mostly take chemosynthetic repellent DEET (DEET) or mosquito repellent ester as a main component, and ethanol is mostly used as a solvent. The long-term contact may induce nervous system diseases, and the skin has certain permeability and irritation, and children use the medicine can also cause systemic urticaria and the like. In addition, such mosquito repellent products can only provide an actual effective protection time of 3 to 4 hours, and cannot effectively protect the safety and health of the consumer for a long time. Along with the continuous improvement of the safety and health consciousness of people, the natural, safe, nontoxic and long-acting mosquito repellent product can meet the requirements.
The citronella oil is a pale yellow to pale brown yellow clear liquid essential oil, and the main components of the citronellol and citronellal have good mosquito repellent effect. The herba Nepetae Catariae is perennial herb of Nepeta of Labiatae, has fragrance of whole plant, can be used for medicine, and has good bioactivity on mosquito. If the two are mixed for use, the defects of single effect and low mosquito repelling efficiency of single essential oil can be overcome.
The nano emulsion is a thermodynamic stable system which is clear and transparent or semitransparent in appearance and low in viscosity and is formed spontaneously by water, oil, a surfactant, a cosurfactant and the like in a proper proportion, and the particle size is 1-100 nm. Nanoemulsions have many advantages not comparable to other formulations: (1) the liquid is isotropic transparent liquid, belongs to a thermodynamic stable system, can not be layered after hot-pressing sterilization or centrifugation, and can be stored for a long time; (2) the process is simple, no special equipment is needed in the preparation process, and spontaneous formation can be realized; (3) the viscosity is low, so that the use discomfort can be reduced; (4) has slow release and targeting effects, and can improve solubility of the medicine. Therefore, when the mosquito repellent is used for the essential oil preparation, the long-acting mosquito repelling can be realized, and the effective mosquito repelling time can be prolonged. Therefore, the natural, safe, nontoxic, long-acting and stable mosquito repelling product is really necessary to be provided, and a new and better product choice can be provided for wide consumers.
Disclosure of Invention
The invention mainly aims to provide a long-acting mosquito-repellent nano spray with the combined action of citronella oil and catmint oil and a preparation method thereof. Aims to solve the problems that chemical components such as pyrethroid, deet, anophelifuge, solvent ethanol and the like in the prior mosquito repellent product are harmful to human bodies and the mosquito repellent action time is short. According to the invention, water is used as a water phase solvent, and natural plant essential oil such as citronella oil and catnip oil is embedded in a three-dimensional network structure formed by emulsion oil phase stearic acid, so that the purposes of safe mosquito repelling and long-acting release are realized, the mosquito repelling product does not irritate skin, and the effective mosquito repelling time is prolonged. Meanwhile, the emulsion also increases the solubility of the essential oil in water and improves the bioavailability of the essential oil.
The technical scheme of the invention is as follows: a mosquito-repellent nano-emulsion spray is composed of the following raw materials in parts by weight: 0.01-10% of citronella oil, 0.01-10% of catmint oil, 0-10% of mint oil, 0-5% of lavender oil, 1-50% of glycerol, 0-1% of hexanediol, 0-1% of octanediol and the balance of surfactant and water.
In some embodiments, the surfactant is one or more of tween, sorbitol, triethanolamine oleate, potassium oleate, medium chain triglycerides, polyglycerol 10-stearate, polyglycerol 10-oleate. In some embodiments, the surfactant is polyglycerol 10-stearate, polyglycerol 10-oleate, and medium chain triglycerides.
In a specific embodiment, the mosquito-repellent nanoemulsion spray is prepared from the following raw materials in parts by weight: 3.5% citronella oil, 2.5% catmint oil, 1% peppermint oil, 1% lavender oil, 3% polyglycerol 10-stearate, 5% polyglycerol 10-oleate, 2% medium chain triglyceride, 45% glycerol, 0.8% hexanediol, 0.2% octanediol, 36% deionized water.
The invention also provides a preparation method of the mosquito-repellent nanoemulsion spray, which comprises the following steps:
s1, dissolving the surfactant in glycerol, heating and dissolving to form a surfactant solution;
s2, adding the mixed solution of the plant essential oils such as citronella oil and catnip oil into the surfactant solution to be used as an oil phase;
s3, dissolving hexanediol and octanediol in deionized water to be used as a water phase;
s4, shearing the oil phase at a high speed for 10min, and adding the water phase while shearing to form a coarse emulsion;
and S5, homogenizing the sheared coarse emulsion under high pressure at 1100bar under the condition of circulating for 15 times to obtain the plant essential oil nanoemulsion spray.
Compared with the prior art, the invention has the beneficial effects that:
(1) the mosquito-repellent compound essential oil provided by the invention is prepared by compounding the mosquito-repellent, antibacterial and refreshing citronella essential oil, the catmint oil for repelling mosquitoes, clearing heat, diminishing inflammation and stopping bleeding, the mint oil for repelling mosquitoes, relieving itching, dispelling wind and heat, and the lavender oil for sterilizing, relieving itching, controlling oil and tendering skin, wherein the raw material components supplement each other, so that the synergistic effect is realized, and the defect of a single essential oil is overcome.
(2) The invention takes natural plant essential oil as a main raw material, takes deionized water to replace ethanol as a solvent, has the advantages of environmental protection, health, no pollution, low toxicity, no irritation, obvious mosquito repelling and itching relieving effects and the like, has no adverse reactions such as red swelling, pruritus, burning sensation and the like to skin, simultaneously has the effects of clearing heat and inducing resuscitation, can relieve discomfort in summer, and ensures the biological safety of the spray,
(3) the invention adopts the nano-emulsion preparation obtained by shearing and homogenizing, can realize slow release and greatly enhance the mosquito repelling time.
(4) The preparation method disclosed by the invention is simple in preparation process, easy to control reaction, good in stability, capable of realizing industrialization and wide in popularization prospect in the field of mosquito repellent products.

Detailed Description
In order to make the objects, technical solutions and advantages of the present invention more apparent, the present invention will be described in further detail below with reference to the accompanying drawings and embodiments. It should be understood that the specific embodiments described herein are merely illustrative of the invention and are not limiting.
Fig. 1 shows an implementation flow of a preparation method of a botanical long-acting mosquito-repellent nano spray provided by an embodiment of the invention.
The invention is described in detail below with reference to the figures of the specification. Reagents and equipment used in the present invention are conventional in the art unless otherwise specified.
Example 1
A plant long-acting mosquito-repellent nano spray comprises the following components in percentage by weight:
3.5% citronella oil, 2.5% catmint oil, 1% peppermint oil, 1% lavender oil, 3% polyglycerol 10-stearate, 5% polyglycerol 10-oleate, 2% medium chain triglyceride, 45% glycerol, 0.8% hexanediol, 0.2% octanediol, 36% deionized water.
The preparation method of the plant long-acting mosquito-repellent nano spray comprises the following specific steps:
s1, heating and dissolving 0.3g of polyglycerol 10-stearate, 0.5g of polyglycerol 10-oleate and 4.5g of glycerol to form a surfactant solution;
s2, accurately weighing 0.35g of citronella oil, 0.25g of catmint oil, 0.1g of mint oil, 0.1g of lavender oil and 0.2g of medium-chain triglyceride, uniformly mixing in a test tube, adding into a surfactant, placing in a water bath magnetic stirrer, setting the temperature at 75 ℃ and the rpm at 500, and heating for 1h to obtain an oil phase;
s3, accurately weighing 0.08g of hexanediol, 0.02g of octanediol and 3.6g of deionized water, placing the weighed materials into a beaker, placing the beaker on a magnetic stirrer to stir, setting the temperature at 25 ℃, adjusting the rotating speed to 500rpm, and stirring for 30min to obtain a clear aqueous solution which is a water phase;
s4, shearing the oil phase at a high speed of 12000rpm for 10min, and slowly adding the water phase while shearing to form a coarse emulsion;
and S5, homogenizing the sheared coarse emulsion under high pressure at 1100bar under the condition of circulating for 15 times to obtain the plant essential oil nanoemulsion spray 1.
Example 2
A plant long-acting mosquito-repellent nano spray comprises the following components in percentage by weight:
6% of citronella oil, 1% of mint oil, 1% of lavender oil, 3% of polyglycerol 10-stearate, 5% of polyglycerol 10-oleate, 2% of medium-chain triglyceride, 45% of glycerol, 0.8% of hexanediol, 0.2% of octanediol and 36% of deionized water.
A plant essential oil nanoemulsion spray 2 was prepared essentially according to the procedures and experimental conditions of example 1. Specifically, the method comprises the following steps: 0.3g of polyglycerol 10-stearate and 0.5g of polyglycerol 10-oleate were weighed out and dissolved in 4.5g of glycerol to form a surfactant solution.
0.6g of citronella oil, 0.1g of peppermint oil, 0.1g of lavender oil and 0.2g of medium chain triglyceride are mixed uniformly and added into the surfactant to serve as an oil phase, and a coarse emulsion is formed with the water phase under high-speed shearing, wherein the shearing speed is 12000rpm, and the time is 10 min. And (3) homogenizing the sheared coarse emulsion under high pressure at 1100bar under the condition of circulating for 15 times to obtain the plant essential oil nanoemulsion spray 2.
Example 3
A plant long-acting mosquito-repellent nano spray comprises the following components in percentage by weight:
6% of catmint oil, 1% of mint oil, 1% of lavender oil, 3% of polyglycerol 10-stearate, 5% of polyglycerol 10-oleate, 2% of medium-chain triglyceride, 45% of glycerol, 0.8% of hexanediol, 0.2% of octanediol and 36% of deionized water.
A plant essential oil nanoemulsion spray 3 was prepared essentially according to the procedure and experimental conditions of example 1. Specifically, the method comprises the following steps:
0.3g of polyglycerol 10-stearate and 0.5g of polyglycerol 10-oleate were weighed out and dissolved in 4.5g of glycerol to form a surfactant solution.
0.6g of catnip oil, 0.1g of mint oil, 0.1g of lavender oil and 0.2g of medium-chain triglyceride are uniformly mixed and added into a surfactant to be used as an oil phase, and the oil phase and a water phase form a coarse emulsion under high-speed shearing, and then the coarse emulsion is further homogenized under high pressure to obtain the plant essential oil nanoemulsion spray 3.
Mosquito repelling experiment
Experimental animals: mice, 20 g. + -. 2g, 120, male and female halves. Testing insects: causing several culex mosquitoes.
Experimental drugs: alcohol, essential oil nanoemulsion spray.
Grouping experiments: negative control group (alcohol), positive control group (example essential oil nanoemulsion spray).
The experimental method comprises the following steps: the experimental mice were divided into 12 groups of 10 mice each, and 3 parallel experiments were performed for each drug. Fixing the limbs of the mouse on the wood plate, subtracting villi with abdominal diameter of 2cm, and mixing with the medicine at a ratio of 1.5uL/cm 2 Uniformly spreading the mixture on the exposed abdomen, placing the mice into 60 × 60 × 60cm Culex fatigans cages every 1h, placing 50 female mosquitoes in each cage, standing for 3min, observing the condition of blood absorption before mosquitoes, recording the number of bites of the mosquitoes, and calculating the effective protection rate. The effective protection rate is above 80%, and the protection effect is lost when the effective protection rate is less than 80%.

The mosquito repelling test results are as follows: the effective protection time of the negative control group is 1 h; example 1 the effective protection time of the citronella oil and catnip oil compound essential oil emulsion is 8 h; the effective protection time of examples 2 and 3 is 5h and 3h respectively. The results prove that the citronella oil and the catmint oil respectively have good mosquito repelling effect, and the combined action effect of the citronella oil and the catmint oil is better.
