背景技术
由包括病原体(例如真菌、卵菌、细菌、线虫和有害昆虫)在内的微生物所引起的植物病害导致的植物产量损失,不仅在农业和园艺领域,而且在造林、花园中心、私人花园和观赏植物中都是全球关注的问题。许多有价值的作物和观赏植物非常容易受到病害,如果没有人类的干预,它们很难在自然界中生存。水(如湖泊养鱼场)中养殖产品的损失也令人关注。
栽培植物往往比它们的野生近缘植物更容易受到病害,因为大量相同的种类或品种(具有同一遗传背景)紧密地生长在一起,有时超过数千平方公里。在这些条件下,由病原生物所引起的病害可以迅速蔓延。例如,植物病原体疫霉(Phytophthora)会产生攻击一系列植物、蔬菜和水果的根和茎的孢子,种植者特别关注它,因为它能够污染水源,还能够留在植物残骸和土壤中多年而不被发现。据估计,单单是被称为“21世纪植物毁灭者”的疫霉每年在全球范围内就造成每种作物20-70亿美元的损失(Roy等人,2012Review of PlantPathology,Vol 6)。
检测植物病害有许多方法。例如,在检测植物病原体种类时,农民通常使顾问农艺师从土壤或植物材料(例如叶或根)中取样,并分析样本中是否存在植物病原体。使用实验室测试在远离取样的地方进行外部分析。这些实验室测试能够包括分子技术,例如ELISA、PCR(PCR和实时PCR)、免疫荧光(IF)、流式细胞术、荧光原位杂交(FISH)和DNA微阵列。土壤或植物材料样本的外部实验室测试存在一些问题,例如需要从样本中提取微生物,而且取样仅选择小样本,不一定反映土壤的真实状况。此外,这些技术中的一些还不能区分死的或活的微生物材料。
在某些情况下,所选土壤样本中的病原生物水平可能太低而无法检测,因此,在对分离的土壤区域取样时,尽管存在造成损害的量的病原体,但所检测到的植物病原体水平将是无效的。外部实验室分析要求将样本从取样点运输到实验室,在实验室需要纯化样本以备分析,然后进行分析。这会导致延迟提供分析结果。检测能够引起植物病害的生物体(如植物病原体)方面的任何延误,都可能导致病害的蔓延和更多的植物受到影响。
也能够使用原地横向流装置来测试样本是否存在病原生物。这些装置需要农民从植物上取样,例如从叶上取样。该方法从植物样本中提取核酸或蛋白质,能够检测到植物病原体的存在。然而,每个植物样本仅代表被测植物。因此,每个样本并不代表整个植物种植区域。此外,植物病原体检测呈阳性的植物样本表明植物已经受到病原体的影响,这可能太迟而无法防止由病原体造成的损害,也太迟而无法防止植物病原体传播到周围的植物。
早期检测对植物健康的威胁和病害(例如害虫和由微生物(如真菌、卵菌、细菌和线虫)所引起的植物病害),就能够通过适当的作物管理策略(如通过杀虫剂应用、杀真菌剂应用和病害特异性化学剂应用的病媒控制以及生物防治)促进对病害的防治。此外,在苗圃中进行早期检测将能够生产和供应无病害植物。而且,在水系统(如灌溉系统和含水层)中进行早期检测将有助于确定农业和园艺用水的安全性。因此,需要提供用于从土壤或水中采集和检测这些有害病原体种类的有效且简单的方法和装置,其能够在植物生长或潜在的植物生长场所或者供水场所使用,以提供基本上无需从土壤、水或植物材料中分离的微生物或病原体种类。然后,这样允许进行后续分析。
在我们的PCT公开WO 2016/097726和WO 2017/207756中,描述了用于检测土壤或水中植物病原微生物的装置,尤其是在病原体影响植物之前进行检测,该申请包含一系列病原体,并且该发明尤其适用于该申请中所列举的病原体。
美国专利公开2013/0334042描述了在田间原地检测空气传播的病原体孢子,但该方法需要相当长的分析时间来检测病原体。此外,该技术不能用于检测生长基质或水中的病原微生物。
美国专利4054491公开了微生物测试装置,其中将样本吸入吸管样腔室中,样本在那里与生长培养基接触。
WO 03/046136涉及用于检测微生物的单管滤网,其包含布置在腔室内的培养基。
一些病原体种类(如真菌、卵菌和细菌)可能长期存在于生长基质或水中并休眠,它们能够被环境变化(如雨、热或其他与天气有关的条件)激活。一旦被激活,它们就能够与植被接触,通常接触植被的根部,它们由此能够进入植被并对特定作物造成相当大的损害。植被可包括全部范围的农业和园艺作物,例如水果作物(如果园和葡萄园)、花卉生产、花园中心和观赏花园植物以及生长在自然环境(如森林)中的树木。本发明可用于在苗圃中早期采集和更快速分析引起植物病害的生物体(例如病原微生物(真菌、卵菌和细菌)),其中种植供应的植物能够使人们更确信幼苗植物是无病害的。病原生物也可能存在于水系统中,如灌溉用水、含水层、水花园、水库、水箱和提供养鱼场的湖泊。
PCT公开WO 94/08042将樟疫霉(Phytophthora cinnamomi)描述为在整个热带和温带地区所发现的最重要的植物病原体之一,该发明特别适用于疫霉型病原体,并描述了使用涂覆有化学引诱剂的固体试纸条来检测培养基中的病原体,其中的生物体是可移动的,需要形成样本溶液来将固体试纸条放入其中。
发明内容
在一个实施方案中,本发明涉及测量与生长基质(例如土壤)中微生物的存在相关的生长基质健康度,尤其是在种植作物之前或生长季节。微生物的存在(无论它们是致病的还是有益于特定作物的)对植物生长和作物产量有着重要的影响。对土壤和其中的微生物的了解能够影响适合在所讨论的土壤中生长的作物的性质和/或能够指示应当使用来消灭有害病原体的补救性化学品的性质和数量,从而改善植物生长和提高作物产量。
通常而言,土壤分析包括种植者向分析实验室提供土壤样本,通常约500克至1千克土壤,以及他们希望知道样本中是否存在的微生物的列表。然后,样本的分析包括纯化以获得足够数量的足够纯材料的微生物,以用于表征和寻找相关微生物,这同样是耗时和昂贵的。利用分子生物学技术能够从纯化的材料中确定所存在的微生物的身份。从土壤中获取能够用于微生物鉴别和后续分析的样本的这个过程可能既耗时又昂贵。通常而言,整个检测过程可能要花3或4周的时间。此外,用目前所使用的一些分子生物学技术,土壤中所存在的死微生物材料也会产生阳性结果,从而给出误导性信息。
本发明提供能够从生长基质或水系统原地采集微生物的技术,以避免需要提供生长基质或水的样本。本发明还指出,微生物应在其采集后进行培养以增加浓度以便于检测。本发明还提供了对样本进行实验室处理以获得用于微生物检测的材料。因此,本发明提供了用于鉴别生长基质或水系统中的微生物的更快、更便宜和更可靠的方法。
因此,本发明提供了将微生物直接从生长基质或水系统采集到容器中,该容器能够用来直接鉴别微生物;在优选实施方案中,容器包含用于微生物生长的生长培养基。
因此,本发明提供了取样装置,其包括能够插入生长基质或水系统中的中空探针,其中该探针包含微生物生长培养基。
探针可以用作微生物的容器,或者可以从远离插入到生长基质或水中那一端的末端从容器中延伸出来,并提供可释放的封盖,微生物能够由此迁移入容器并保存在容器中。
我们已发现,当这样的探针放置在生长基质或水系统中时,土壤中的微生物会迁移到探针中,它们能够保留在那里用于进行分析。
生长培养基可以对种植者希望知道生长基质或水系统中是否存在的微生物具有特异性。备选地,可以使用一种或多种生长培养基,以便能够采集多种微生物。
因此,本发明装置的使用使得能够直接从生长基质或水系统中获得和采集微生物并进行培养,这样含有一种或多种微生物的装置就能够被带到实验室检测所存在的微生物,而无需进一步纯化或处理样本。这对于在轮作决定在此种植哪种作物之前对田地进行预筛选特别有用。
在优选实施方案中,本发明还提供了装置的用途,该装置包括中空探针,其连接到具有可释放盖子的容器,探针和容器均含有微生物生长培养基,用于从生长基质和/或水或水系统中获取微生物样本随后进行微生物检测。
本发明还提供了用于检测生长基质中是否存在微生物的方法,包括将根据本发明的探针放置在生长基质或水系统中,以便采集微生物并将探针和任选包含微生物的容器运送到实验室进行分析以确定所存在的微生物的性质。然后,能够将分析结果反馈给种植者,提供处理生长基质或水或进行其他方式所需的信息,以改善他们作物的生长和产量。
术语生长基质包括任何可种植植物的天然和合成基质,包括土壤、椰壳、泥煤、沙子、水及其混合物。
如本文所使用的术语“生长培养基”包括促进待检测微生物生长的任何物质,微生物可以是植物病害微生物或致病微生物种类或损害植物的害虫以及有益微生物。根据感兴趣的微生物选择生长培养基或生长培养基成分。生长培养基能够包含氨基酸或醇、植物提取物或特定植物化合物(例如植物激素、植物蛋白质或植物信号传导化合物)、糖、有机酸、酚类或其他蛋白质(例如酪蛋白)、果胶,这些物质的任何衍生物也可用作引诱剂。生长培养基的选择将取决于待检测的微生物,例如病害微生物或致病微生物。
生长培养基将来自生长基质或水系统的微生物吸引到中空探针中,然后进入容器(如果使用)中。微生物用生长培养基以既促进微生物生长、又将微生物引导到探针中并任选进入容器中的方式提供。例如,中空探针可以是可被设计成放置在生长基质或水系统内部的探针、管或针,在探针、管或针的另一端可连接到提供容器的小室上,该容器能够储存从生长基质或水系统中提取到的微生物。中空探针、管或针的至少一部分内表面可涂覆生长培养基(例如固体培养基),并且涂覆模式可以使其促进微生物向探针上方移动并进入使用这种容器小室的容器中。生长培养基能够促进微生物沿着探针向上移动而进入容器小室中,并随着微生物沿着探针向上的移动而生长。因此微生物就在培养基上生长。
例如,当装置配置成使用时使容器小室位于中空探针、管或针的顶端,而中空探针、管或针的底端配置在生长基质或水系统内,如果生长培养基是作为垂直条带提供在中空探针、管或针的内表面上的,则它将促进微生物沿着条带向上移动并进入容器小室。
所采用的生长培养基将取决于要在生长基质或水系统中确定是否存在的微生物的性质。生长培养基还可含有能够杀灭或减少进入装置的非待检测微生物的微生物数量的组分(如杀真菌剂或抗菌剂)。
本发明中所使用的生长培养基将吸引特定的微生物,然后所述微生物将通过中空探针、管或针而进入容器中。生长培养基有助于微生物的生长,根据所采集的微生物,能够具有许多不同组成。Jeffers和Martin(Plant Disease,1986,Vol.80No.11)以及Guo和Ko(Applied Environmental Microbiology,July 1993,Vol.59,No.7,p2323-2325)中描述了合适的生长培养基的例子。
在一个实施方案中,生长培养基包含抗生素和/或养分。抗生素的存在能够防止不希望的细菌的生长。在另一实施方案中,生长培养基包含杀虫剂或杀真菌剂,抗生素、杀虫剂和/或杀真菌剂的存在允许感兴趣的微生物生长并防止不感兴趣的微生物的生长,所述不感兴趣的微生物例如可能已经进入中空探针、管或针中的其他真菌和细菌。在一个实施方案中,生长培养基包含硝基苯或异黄酮类。生长培养基能够含有琼脂、养分、醇、氨基酸、杀真菌剂、杀虫剂、抗生素、硝基苯、植物提取物和/或异黄酮类的任何组合。
如果使用容器小室,则中空管、探针或针中的生长培养基浓度可以低于容器中的生长培养基浓度,因为这能够促进微生物进入容器。生长培养基可另外包含保持中空管、探针或针中培养基的pH的缓冲液。容器中的生长培养基还可以包含浓度低于中空管、探针或针中的培养基缓冲液浓度的缓冲液。容器中的更低缓冲液浓度意味着容器中培养基的pH水平能够改变。在一个实施方案中,中空管、探针或针中的生长培养基包含缓冲液,且容器中的生长培养基不包含缓冲液。容器中没有缓冲液意味着培养基的pH水平能够改变。
在一个实施方案中,生长培养基为琼脂培养基。
如在使用容器小室的优选实施方案中所述的,容器还包括使生物体能够增殖的微生物用生长培养基,以在容器中提供足够高浓度的微生物进行后续分析。增殖所需的时间根据生物体的性质而不同,但我们已发现,使用本发明的技术就能够比现有技术更快地完成增殖,并且本发明提供了简单的原地采集装置,不需要进一步的样本操作。
本发明可应用于任何微生物,在其生命周期的任何阶段,例如,适用于由致病生物体(如真菌、卵菌和细菌)所产生的任何类型的孢子,包括细菌孢子和活动孢子(如游动孢子)。
在一个实施方案中,本发明可应用于包括至少一种疫霉种的任何植物病原体的采集。“疫霉”包括疫霉属的所有种。
本发明可以应用于的其他病原体包括稻瘟病菌(Magnaporthe)、葡萄孢属(Botrytis)、旋孢霉属(Cochiliobolus)、柄锈菌属(Puccinia)、胶锈菌属(Gymnosporangium)、驼孢锈菌属(Hemileia)和镰孢属(Fusarium)的所有种。

在采集植物病原体的地方,生长培养基对于植物致病物种将通常具有特异性。生长培养基可对一种类型的致病物种(例如特定的植物病原体)具有特异性,或者它可吸引一种以上类型的植物致病微生物。在一个实施方案中,使用对几种不同植物病原体具有特异性的植物病原体生长培养基。备选地,所使用的不同植物病原体生长培养基可对不同的植物病原体具有特异性。
中空管、探针或针应当坚固,并能抵抗生长基质中的腐蚀。塑料探针或塑料管是特别有用的。生长培养基应该以这样的量提供,以便(如优选地)如果一些生长培养基已从管或探针中滤沥出而进入生长基质中,则留在管或探针中的生长培养基处于浓度梯度中,其中更靠近容器的生长培养基存在的浓度高于生长基质中所放置的装置那一端的浓度。换言之,在使用时将与生长基质接触的递送装置一端的浓度比另一端的低。另一端可在使用时将微生物递送至容器中,该浓度梯度可开始就提供或者可在采集系统运行期间原位形成。尽管这种梯度不是必需的,但它能够有助于使微生物被带上中空管、探针或针并可能进入容器。

中空管、探针或针的尺寸和形状(包括横截面)能够根据待使用的装置的位置以及待检测微生物的性质进行选择。也可根据所采集微生物的性质,选择中空管、探针或针的内表面上的生长培养基涂层的厚度、位置和浓度。我们已发现,4至10厘米长、具有0.5至5平方厘米横截面积的中空管、探针或针是特别有用的,0.1至1厘米、特别是0.2至0.5厘米厚的生长培养基涂层是非常有效的。
当在生长基质或水系统中使用时,生长培养基不应被生长基质或水系统以及其中所包含的材料灭活。它还应在使用它的特定环境下所经受的温度范围内保持稳定。当在生长基质中使用时,生长培养基应当在生长基质所含的水分中具有一定的溶解度,并且当在水中使用时,应对它进行选择以在使用它时所处的条件下具有所需的水中溶解度。当生长培养基作为涂层提供在中空探针、管或针的内表面上,例如被保持在凝胶中或被吸附在载体(例如附着在中空探针、管或针的内表面上的膜或薄膜)上,它就可被释放到生长基质中吸引微生物,同时也保留在管或探针的内表面上以将微生物引导到容器中。这可以通过在中空管、探针或针的内表面上提供作为两层或更多层的生长培养基来实现。然而,在中空管、探针或针的内表面上保留足够的生长培养基是很重要的。
我们已发现,氨基酸和/或C1到C4一元醇及其混合物以及二价金属(特别是钙)是特别有用的生长培养基,并且它们也作为化学引诱剂起作用,特别是用于疫霉和腐霉孢子。我们还发现,可将它们方便地提供在膜(如尼龙或硝化纤维素膜)上或者凝胶(如琼脂)中。
在用致病微生物实施本发明时,一旦有活性,生长基质中的致病微生物就将被生长培养基吸引,并将向生长培养基移动或生长,生长培养基是以随后引导微生物进入中空管、探针或针的方式提供的,在优选实施方案中向上穿过管并进入容器。通过这种方式,就能够从生长基质或水系统中分离和采集致病生物体,并进入容器。然后能够将探针和/或容器送到实验室进行分析,在许多情况下能够足够快地进行分析,以便在生物体与特定植被发生重大相互作用之前获得物种特异性结果,从而能够在致病生物体对植被造成重大损害之前采取补救行动。
本发明的系统能够用于采集任何特定的微生物。例如,在一个实施方案中,它可包括设计来允许所讨论的微生物通过并排除可能包括其他微生物的其他材料的过滤系统。在本发明的一个实施方案中,致病生物体用生长培养基也包括在装置的容器中,以确保致病生物体不仅被吸引到中空管、探针或针中,而且被吸引向上流过中空管、探针或针并进入容器。在这种情况下,能够在中空管、探针或针内以及还在容器内以逐渐增加的量提供生长培养基,以确保生物体被引导到容器中。
尽管是任选的,但在一个实施方案中,本发明所采用的采集材料包括过滤器。当使用过滤器时,它选择性允许被采集的微生物到达容器,而防止不同形状和大小的物种到达容器中的培养基。通过将选择性过滤器与感兴趣微生物的特异性生长培养基结合使用,该装置能够用于采集感兴趣的微生物。备选地,在优选实施方案中,通过在管、探针或针的内表面上所提供的生长培养基中以及任选地在采集材料中提供抗生素和抗真菌剂来进行化学或生物过滤。
如所描述的,可以通过在与正种植于那里的材料邻近的生长基质中放置一个或多个本发明的装置来在农业和园艺学中实施本发明。在例如田间的装置之间的最佳距离将会取决于作物的性质、生长基质的类型、盛行的气候等。这些全都能够通过反复试验来确定,尽管初步结果建议装置组为例如每公顷10至20台装置将会是足够的。在另一个实施方案中,可以在新耕作的生长基质或可用的水系统中采用本发明,以获得样本进行后续分析来确定它们对于公共消费或使用以及在农业和园艺学中进行使用的适合性,以及确定添加材料(例如肥料、杀虫剂、杀昆虫剂、杀真菌剂等)的需要。因此,本发明进一步提供了横跨田地或水系统分布的本发明的装置的阵列。在另一实施方案中,可将本发明的一个或多个装置放置在水系统中,以便帮助评估水系统的纯净度。
一旦特定微生物已被采集到本发明的装置中,含有该微生物的装置就能被送到实验室进行分析。在使用容器的情况下,该容器优选配备可释放盖子,能够取下盖子并对容器内容物进行微生物分析(如LAMP、Taqman、PCR和DNA条形码分析)。本发明允许在生长基质中进行种植之前对其健康状况进行简单和快速的测试,并能用于在生长季节监测病害,以提供和指导喷洒实践。它还能用于监测水系统(如灌溉系统、含水层或水培作物)中的病害,另外还能用于监测化学处理的效率。

通过参考附图来说明本发明,所述附图显示了准备好进行使用的本发明的装置。图1显示了中空的探针或管(1),其在内部携带用于感兴趣微生物的生长培养基层(2)。所述探针或管向下通入到生长基质或水系统中,生长基质或水系统的表面显示在(3)处。一些生长培养基沥滤出而进入生长基质中以吸引微生物(5),如由箭头所指示的将微生物引导进入所述中空管中,如由箭头(4)所示沿着所述管向上。微生物在与生长培养基接触期间进行生长,然后通入到包含生长培养基(6)的容器(7)中。微生物样本(8)被储存在容器内,可将装置从生长基质或水系统中取出,并送至实验室,在实验室中可以进行微生物分析。一旦将微生物已从装置中清除,就可以对其进行清洗,并将其返回给种植者重新使用。
具体实施方式
参照以下实施例说明本发明。
实施例1:使用图中所示的取样装置对胡萝卜田土壤中的微生物进行取样和随后的分析。
设计了几种专门用于采集卵菌种的取样装置,将其放置在土壤中。中空探针和容器包含含有琼脂的生长培养基,所述生长培养基对卵菌生长具有特异性。在生长季节,将这些装置和探针一起放置在靠近胡萝卜的土壤中。2周后,收集装置并运送至实验室。所有的装置都在容器里含有可见的卵菌种纯培养物。通过DNA提取、特定基因组区域的PCR扩增和该区域的测序,鉴定了每个装置中存在的微生物。靠近一些装置的胡萝卜显示了空洞斑的可见症状,并鉴定出了许多种类的腐霉(堇菜腐霉(P.violae)、林栖腐霉(P.sylvaticum)、宽雄腐霉(P.dissotocum)、畸雌腐霉(P.irregulare)和间型腐霉(P.intermedium))为致病因子。
实施例2:使用图中所示的取样装置,对草莓地的椰壳和水中的微生物进行取样和后续分析。
设计了几种采样装置,用于真菌和真菌样微生物的广谱筛选和采集。将这些装置放置在用于种植草莓的椰壳袋中,以及在草莓多隧道内的每个排水沟末端所收集的水中。中空探针和容器中包含含有琼脂的生长培养基,其对真菌和卵菌的生长具有特异性。2周后,收集装置并运送至实验室。装置的容器中都含有至少一种微生物的可见培养物,在某些情况下含有几种微生物。通过DNA提取、特定基因组区域的PCR扩增和该区域的测序,鉴别了每个装置中存在的微生物。通过这种方式,对许多微生物进行了采样和鉴定(例如恶疫霉(Phytophthora cactorum)、卢氏腐霉(Pythium lutarium)、宽雄腐霉、灰葡萄孢菌(Botrytis cinerea)和Chlonostachys rose),为种植者提供了有关病害压力位置和生长系统中有益微生物分布的有用信息。
Claims (23)
1.取样装置,其包括能够插入生长基质或水系统中的中空探针,所述探针含有微生物用生长培养基,其中所述生长培养基含有化学引诱剂,特别是植物病原体化学引诱剂。
2.根据权利要求1所述的取样装置,其还包括从远离插入所述生长基质或水系统一端的所述探针末端延伸出的容器。
3.根据权利要求1或权利要求2所述的取样装置,其中所述生长培养基对种植者希望知道所述生长基质或水系统中是否存在的一种或多种微生物具有特异性。
4.根据前述任一权利要求所述的取样装置,其中所述生长培养基成分选自:氨基酸或醇,选自植物激素、植物蛋白质和植物信号传导化合物的植物提取物或植物化合物,糖,有机酸,酚类,或者选自酪蛋白的其他蛋白质,果胶以及它们的任何衍生物。
5.根据前述任一权利要求所述的取样装置,其中所述生长培养基是静电或离子生长培养基。
6.根据权利要求2至5中任一项所述的装置,其中所述生长培养基包括用于所述微生物的生长培养基,并以既促进所述微生物生长又引导所述微生物沿着所述探针和/或进入所述容器的方式提供。
7.根据权利要求2至6中任一项所述的装置,其中所述中空探针、管或针的至少一部分内表面涂覆有生长培养基,且涂层的模式使得所述微生物在所述生长培养基上生长时,所述涂层能够促进所述微生物向所述容器小室移动。
8.根据前述任一权利要求所述的装置,其中所述生长培养基包含抗生素和/或养分。
9.根据前述任一权利要求所述的装置,其中所述生长培养基包含杀虫剂或杀真菌剂。
10.根据权利要求2至9中任一项所述的装置,其中所述中空管、探针或针中的所述生长培养基成分的浓度低于所述容器中的所述生长培养基成分的浓度。
11.根据前述任一权利要求所述的装置,其中所述生长培养基包含缓冲液,其维持所述中空管、探针或针中所述培养基的pH。
12.根据权利要求11所述的装置,其中所述容器中的所述生长培养基包含缓冲液,其浓度低于所述中空管、探针或针中的所述培养基中的所述缓冲液的浓度。
13.根据前述任一权利要求所述的装置,其中所述生长培养基是提供在载体上的。
14.根据权利要求13所述的装置,其中所述生长培养基含有胶凝剂,由此将所述生长培养基粘附在所述装置的中空管、探针或针的内部。
15.根据权利要求13或权利要求14所述的装置,其中所述生长培养基的载体包括膜或薄膜。
16.根据前述任一权利要求所述的装置,其中所述中空管、探针或针为4至10厘米长,具有0.5至5平方厘米的横截面积。
17.根据权利要求16所述的装置,其中所述生长培养基以0.1至1cm的厚度涂覆在所述管、探针或针的内表面上。
18.根据前述任一权利要求所述的装置,其中所述生长培养基含有所选氨基酸和/或C1至C4一元醇及其混合物。
19.根据权利要求2至18中任一项所述的装置,其中所述容器中包含生长培养基,由此将所述致病生物体吸引至所述中空管、探针或针中,并沿着所述中空管、探针或针向上流入所述容器中。
20.根据权利要求19所述的装置,其中所述生长培养基在所述中空管、探针或针内以及所述容器中以逐渐增加的量提供。
21.取样装置,其包括能够插入生长基质或水系统中的中空探针、管或针,所述探针含有微生物用生长培养基,其中所述中空探针、管或针的至少部分内表面涂覆有生长培养基,且涂层模式使得所述微生物在所述生长培养基上生长时,所述涂层能够促进所述微生物向所述容器小室移动。
22.根据前述任一权利要求所述的装置用于从生长基质或水中获取微生物样本并随后进行所述微生物检测的用途。
23.检测生长基质或水中是否存在微生物的方法,所述方法包括将根据权利要求1至22中任一项所述的装置的探针置于所述生长基质或水中以将微生物采集到所述装置,并将含有所述微生物的装置运送至实验室进行分析以确定所述装置内存在的所述微生物的性质。
