Weighing the ingredients, and then processing the obtained ingredients through pretreatment, extraction, wine preparation, freezing and filtration; the pretreatment comprises crushing the obtained ingredients; the extraction comprises the steps of soaking the crushed ingredients in fen-flavor liquor, performing multi-stage countercurrent percolation extraction and obtaining an extracting solution; the wine blending comprises adding Chinese liquor and yellow wine into the extract. The nourishing nutritional wine provided by the invention adopts a special extraction process, extracts more abundant nutritional components, and remarkably improves the immunity of animals.

Technical Field
The invention belongs to the field of food wine and an extraction method, and particularly relates to nourishing three-penis wine and an extraction method thereof.
Background
The Chinese herbal medicine wine is a nourishing drink integrating traditional Chinese medicines for building up the body and wine, takes alcohol as a good solvent of the traditional Chinese medicine components, and the traditional Chinese medicines and the alcohol exert effects in a synergistic manner, so that the curative effect of the traditional Chinese medicines is improved. The preparation of Chinese herbal medicine wine is to soak Chinese herbal medicine in wine for some time to dissolve the effective components of the Chinese herbal medicine in the wine, and then to filter and eliminate dregs before drinking.
The existing extraction process of Chinese herbal medicine wine is that the Chinese herbal medicines are directly soaked in the koji wine or alcohol for a period of time, water is added to reduce the alcohol content, and a proper amount of granulated sugar and auxiliary materials are added to make the wine have moderate taste, but the extraction of the nutrient components in the wine solution is too little, and the nourishing performance of the wine is greatly reduced. Patent CN103060158A discloses a kidney-tonifying three-penis wine, which is prepared from penis cervi, testis Et penis bovis seu Bubali, penis canitis, fructus Lycii, Poria, cortex Cinnamomi, radix Puerariae, and herba Menthae by soaking in 500 g of 50 ° Chinese liquor for 720 hr, filtering, and blending with sweet yellow wine and water. According to the method, the Chinese herbal medicines are soaked in the white spirit to extract the active ingredients of the Chinese herbal medicines, however, the extracted ingredients of the obtained Chinese herbal medicines are very limited, a large amount of valuable active ingredients still remain in the filtered residues, the nutrition and nourishing performance is greatly reduced, and the obtained three-animal whip wine has the technical problems of being too turbid, uncomfortable in taste and the like.
In order to solve the above problems, research and development personnel for medicines are continuously developing various tonic wines with high nutrient contents.
Disclosure of Invention
In order to solve the problems in the prior art, the invention provides the nourishing three-animal penes wine and the extraction method thereof, the extracted useful components of the nourishing three-animal penes wine have high extraction rate and unique taste, and the clarity meets the sensory requirements, namely, the wine is clear and transparent and has no sediment or suspended matters.

According to the technical scheme, distilled liquor and fermented liquor are used as liquor bases, edible and medicinal animal and plant and food additives are added, blending and mixing are carried out, the original liquor base style of the alcoholic beverage is changed, and the requirements of national standard GB/T27588-2011 are met.
Preferably, the nourishing three-whip wine has the alcoholic strength of 34-36% vol and the total sugar of 54-66 g/L. By adopting the technical scheme, the alcohol content and the total sugar content of the nourishing three-animal-penis wine are adjusted through a large number of experiments and the requirements of nutrition and nourishing of human bodies, and the taste of the nourishing three-animal-penis wine is adjusted to meet the requirement of the good taste of the food wine.
Preferably, for the nourishing three-animal penes wine, the dosage of the fen-flavor liquor is 10000L multiplied by N% multiplied by 90% ÷ M, and the dosage of the grape distilled liquor is 10000L multiplied by N% multiplied by 10% ÷ G; wherein M is the alcohol content of the fen-flavor liquor, N is the numerical value of the alcohol content of the tonic three-whip liquor, and G is the alcohol content of the grape distilled liquor. The wine base with the proportion realizes the wine flavor of the nourishing three-animal penes wine, and the nourishing three-animal penes wine has unique taste.
The second aspect of the invention provides an extraction method of nourishing three-animal penes wine, weighing the above ingredients, and then processing the obtained ingredients through pretreatment, extraction, wine preparation, freezing and filtration; the pretreatment comprises crushing the obtained ingredients; the extraction comprises the steps of soaking the crushed ingredients in fen-flavor liquor, performing multi-stage countercurrent percolation extraction and obtaining an extracting solution; the wine blending comprises adding Chinese liquor, yellow wine and grape distilled liquor into the extracting solution. By adopting the technical scheme and the novel extraction process, the useful components in the animal and plant medicinal materials are fully extracted, the content of total solids in the extracting solution is improved, and the content of dry extract in the prepared wine is improved.
Preferably, for the extraction method, the weight of the fen-flavor liquor is 45-49 times of the weight of the crushed ingredients. By adopting the technical scheme, the full amount of the fen-flavor liquor in the liquor is nearly equal to the full amount of the fen-flavor liquor in the liquor prepared by extracting and adding the fen-flavor liquor, so that the full extraction of useful components is further ensured.
Preferably, for the extraction method as described above, the extraction comprises: the height of the ingredients of the fen-flavor liquor for soaking is not less than 15 cm. By adopting the technical scheme, the crushed ingredients can be completely soaked in the fen-flavor liquor in the soaking process.
Preferably, for the extraction method, the speed of the percolation is 140 +/-10L/hr, and the temperature of the percolation is 35-40 ℃. By adopting the technical scheme, the useful components in the ingredients are extracted to the maximum extent through percolation in the temperature range; the percolation speed (percolation speed of 5-stage counter-current, average about 28 + -2L/hr per tank) is used for percolation, so that the useful components are completely extracted before the solvent is used up.
Preferably, for the extraction method as described above, said freezing comprises: the freezing temperature is-6.5-7.5 deg.C, and the freezing time is not less than 24 hr. The adoption of the adjustment of the freezing temperature and the freezing time ensures that the clarity of the wine reaches the sensory requirement after the wine is filtered by the diatomite filter and the plate and frame filter, namely the wine is clear and transparent and has no sediment or suspended matters.
The third aspect of the invention provides application of the nourishing three-penis wine in the fields of medical treatment and beauty.
The fourth aspect of the invention provides application of the nourishing three-animal penes wine prepared by the extraction method in the fields of medical treatment and beauty.
The beneficial effects created by the invention are as follows:
the nourishing three-animal penes wine prepared by the invention adopts a new extraction process, so that the active ingredients in the medicinal materials are extracted more completely, the extraction rate of useful ingredients is high, the taste is unique, the clarity meets the sensory requirement, namely, the wine is clear and transparent, and has no precipitate or suspended matters.

Example 1 Tonic Sanbian wine and extraction method thereof
This example describes a tonic three-whip wine and its extraction method, and the formulation of the tonic three-whip wine is shown in table 1 below.

The calculation formula of the total volume of the added grape distilled wine is as follows: 10000L 35.0% × 10% ÷ alcohol content of the wine.
All the medicinal materials in the formula are respectively selected to remove impurities and foreign matters according to different requirements. The processing is carried out according to the pharmacopoeia of the people's republic of China and the processing specifications of the traditional Chinese medicines in Shandong province (2002 edition).
The method comprises the following steps: pretreatment of
(A) Ginseng radix, cortex Cinnamomi, fructus Lycii, Poria, rhizoma Dioscoreae, fructus Zanthoxyli, and fructus Foeniculi to remove impurities and non-medicinal parts (conventional method, specifically, see the pharmacopoeia of people's republic of China 2015 edition and the processing Specification of decoction pieces of Chinese medicinal materials in Shandong province 2012 edition).

The 60-65% of fen-flavor liquor refers to the volume percentage of the concentration of ethanol contained in the fen-flavor liquor.

As can be seen from Table 4, the tonic three-whip wine obtained by the method of examples 1 to 3 of the present invention meets the requirements of the national standard GB/T27588-2011 in alcohol content, total sugar content, total acid, dry extract and total ester.
In addition, the inventor takes distilled liquor and fermented liquor as liquor bases through experiments, adds edible and medicinal animals and plants and food additives, and carries out blending and mixing to obtain the alcoholic liquor with the changed original liquor base style, adjusts the alcoholic strength and the total sugar content of the tonic three-animal penis liquor, and adjusts the taste of the tonic three-animal penis liquor to meet the requirements of food liquor on the best quality. The wine base with unique mixture ratio realizes the wine flavor of the nourishing three-animal penes wine, and the nourishing three-animal penes wine has unique taste. By adopting a new extraction process, useful components in the animal and plant medicinal materials are fully extracted, the content of total solids in an extracting solution is improved, and the content of dry extract in prepared wine is improved. The full extraction of useful components is further ensured by adding the fen-flavor liquor close to the full amount of the fen-flavor liquor in the prepared liquor. And the ingredients crushed in the soaking process can be ensured to be completely soaked by the fen-flavor liquor by adjusting the soaking amount of the liquor. Parameters such as the temperature of percolation are set, and useful components in the ingredients are extracted to the maximum extent through the percolation in the temperature range; the percolation speed (percolation speed of 5-stage counter-current, average about 28 + -2L/hr per tank) is used for percolation, so that the useful components are completely extracted before the solvent is used up. By adjusting the freezing temperature and the freezing time, the clarity of the wine can meet the sensory requirements after being filtered by the diatomite filter and the plate and frame filter, namely the wine is clear and transparent and has no sediment or suspended matters.
Example 5 extraction Performance test of Tonic Sanbian wine extraction method
The tonic three-whip wines of examples 1 to 3 were weighed and extracted by the conventional method (cyclic extraction) and the extraction methods described in examples 1 to 3, respectively, to obtain the experimental results of comparative examples 1-1, 2-1 and 3-1 and the experimental results of examples 1-2, 2-2 and 3-2 of the present invention, as shown in table 5. As can be seen from Table 5, the extraction methods of examples 1 to 3 of the present invention showed an average increase in the total solid content volume of the extract solution of 10.1% and an average increase in the dry extract volume of 10.0% as compared with the cyclic extraction.

Further, the inventors conducted the same experiment with reference to the method of CN103060158A, and found that the same compounding ratio of CN103060158A, extracted by the method described in CN103060158A, did not reach 47% by volume of the total solid content of comparative examples 1-1, 2-1 and 3-1, nor did the concentration of the dry extract reach 41% by volume of the total solid content of comparative examples 1-1, 2-1 and 3-1.
The tonic three-whip wines obtained in examples 1 to 3 were tested for collagen, hemoglobin, mucin, ginsenoside, ginseng alkene, and β -sitosterol, and it was found that the presence of various nutritional components was detected in all of examples 1 to 3.
Example 6 testing of Tonic Sanbian wine for animal toxicity and immune Performance
1. Observation of acute toxic effects in animals
30 healthy Swiss mice were used, weighing 10. + -.1 g, male and female halves. The tonic three-whip wine obtained in example 1 was concentrated to 1: 10, by gavage of mice. The experiment was carried out once in the morning and once in the evening, each time 0.3ml/10g mouse weight, and then continuously observed for 10 days. During the experiment, the mice have good activity and appetite, the back hair is glossy, the expression and the urine are not abnormal, and none of 30 used mice die, which is also an abnormal phenomenon. Similar experimental results were obtained by performing toxicity experiments using examples 2 to 3, respectively. The dosage of the experiment is more than 70 times of the clinical dosage, which shows that the nourishing three-penis wine has no obvious acute toxicity.
2. Effects on body weight of Normal animals
30 healthy male Swiss mice were used, weighing about 20g, and were randomly divided into two groups according to weight balance. The mice obtained in example 1 were re-filled with the tonic three-animal penes wine and physiological saline 0.2ml/10g once a day for 10 consecutive days, and the body weights were recorded before and after administration. The body weight experiments were performed using examples 2 to 3, respectively. The experimental result shows that the nourishing three-animal penis wine has no influence on the weight of normal animals.
3. Influence on animal sports physical strength
30 healthy Swiss mice with weight of about 10g are divided into groups according to weight balance and randomness, and the physiological saline and the tonic three-penis wine obtained in the example 1 are respectively added with 0.2ml/10g of mice to be re-filled with stomach, and the mixture is taken once a day for 10 days. On the eleventh day of the experiment, the rotating rod is adjusted to rotate at a speed of 10 circles/minute, the animals are placed on the rod one by one, and the retention time of the animals on the rod is recorded, namely the time of physical consumption of the animals during movement and the bearing time of the physical strength of the animals. Physical force experiments were performed using examples 2 to 3, respectively. The result shows that the nourishing three-penis wine has the function of improving the physical strength and the movement tolerance of animals.
4. Influence on body weight, plasma cortisol content and adrenal weight of yang-deficient animals.
100 healthy male Swiss mice weighing about 15 g were injected subcutaneously with 0.5% hydrocortisone 0.1 ml/day for 9 consecutive days. During the period, the weight of the animals is obviously reduced, about 12 g on average, about 40 percent of the animals die, and the surviving animals all have loose hair, aversion to cold, inappetence and cachexia. On the 10 th day of the experiment, the animals were divided into two groups according to the weight random equilibrium, and the animals were re-filled with normal saline and the tonic three-whip wine obtained in examples 1 to 3 in an amount of 0.2ml/10g, once a day for 12 consecutive days, and the weight was recorded every 4 days during the experiment, and the results were recorded. On the 21 st day of the experiment, the eyeballs of the mice are removed, blood is taken from the orbital veins, heparin is used for anticoagulation, after centrifugation, plasma is quantitatively sucked, and the plasma cortisol content is measured by using an EJ-2008r immune counter. Differences between groups were compared using the T-test method. Weighing blood and adrenal gland, taking adrenal gland on both sides, fixing in Bownis solution for 24 hours, then sucking surface liquid with filter paper, removing fat and other tissues, weighing wet weight, converting to adrenal gland mg wet weight/10 mouse weight, and performing statistical treatment by an inspection method. The effects on plasma cortisol and adrenal weight suggest that the tonic triple penes obtained in examples 1 to 3 increase the body weight of experimental mice, and enhance the adrenal endocrine function of mice, and the weight of adrenal glands of mice increases.

4. A method for extracting tonic three-animal penes wine is characterized in that the ingredients of claim 1 or 3 are weighed, and then the obtained ingredients are processed by pretreatment, extraction, wine preparation, freezing and filtration;
the pretreatment comprises crushing the obtained ingredients;
the extraction comprises the steps of soaking the crushed ingredients in fen-flavor liquor, performing multi-stage countercurrent percolation extraction and obtaining an extracting solution;
the wine blending comprises adding Chinese liquor, yellow wine and grape distilled liquor into the extracting solution.
5. The extraction method according to claim 4, wherein the weight of the fen-flavor liquor is 45-49 times of the weight of the crushed ingredients.
6. The extraction method according to claim 4, wherein the extracting comprises: the height of the ingredients of the fen-flavor liquor for soaking is not less than 15 cm.
7. The extraction method according to claim 4, wherein the speed of the percolation is 140 ± 10L/hr, and the temperature of the percolation is 35-40 ℃.
8. The extraction method according to claim 4, wherein the freezing comprises: the freezing temperature is-6.5 to-7.5 ℃, and the freezing time is not less than 24 hours.
