Abstract
本发明公开了一种含魔芋多糖的痤疮微生态失衡改善制剂及其在化妆品中的应用。所述痤疮微生态失衡改善制剂包括魔芋提取物、乳酸杆菌发酵产物、酵母提取物、神经酰胺及马齿苋提取物。本发明所述具有调节痤疮微生态失衡的组合物能够被皮肤表面的益生菌所吸收利用,并且有选择性的促进皮肤表面益生菌的生长同时抑制病原菌,特别对于痤疮丙酸杆菌有很好的抑制作用。本发明所述具有调节痤疮微生态失衡的组合物应用在化妆品中具有良好的治疗、减少或者预防皮肤上出现的痤疮症状,改善失衡的痤疮皮肤菌群。
Description
技术领域
本发明涉及一种具有改善痤疮皮肤微生态失衡的组合物及其应用,属于化妆品技术领域。
背景技术
痤疮是一种非常普遍的皮肤炎症,包括皮脂腺炎。虽然痤疮是多种因素相互作用的结果,但痤疮的确切病因仍不清楚。目前研究发现,皮肤微生物与宿主免疫的相互作用在本病中起着重要作用,痤疮患者的微生物组成和活性受到干扰。皮肤有益菌就会降低,从而促使病原菌既痤疮丙酸杆菌大量增殖,导致皮肤菌群发生严重紊乱,皮肤屏障受损,皮肤自身防御力不足,加重痤疮发展进程,引发一系列皮肤疾病。
目前对于痤疮的治疗主要包括局部或者口服抗生素、维生素a类或者光子治疗,但是通过使用抗生素抑制和减少痤疮丙酸杆菌的增殖,短期对痤疮患者有很好的疗效,但是从长远角度来看,痤疮患者皮肤微生态失衡依然没有解决,使用抗生素在杀灭病原菌的前提下,皮肤正常菌群也会被破坏,皮肤屏障功能依然受损,导致痤疮患者病情容易反复或者加重。
发明内容
本发明所要解决的技术问题是:如果促进皮肤正常有益菌的增殖,通过占位、细菌素释放等来抑制和减少病原菌生长,就能维持皮肤pH值,调节皮肤微生态的失衡的状态,促进皮肤屏障恢复,加强皮肤自身抵抗力。
为了解决上述技术问题,本发明提供了一种含魔芋多糖的痤疮微生态失衡改善制剂,其特征在于,包括魔芋提取物、乳酸杆菌发酵产物、酵母提取物、神经酰胺及马齿苋提取物。
优选地,所述魔芋提取物的制备方法为:
步骤1):将魔芋精粉溶解于pH为5~6的缓冲液中;
步骤2):在步骤1)所得的溶液中加入纤维素酶在水浴酶解,得到魔芋多糖的水解液;
步骤3):将步骤2)所得的水解液采用旋转蒸发仪浓缩至原来体积的1/5,然后加入无水乙醇搅拌,过滤,过滤所得固体真空干燥,得到魔芋提取物,即含较多魔芋低聚糖的混合物。
更优选地,所述步骤1)所得溶液的浓度为5-20g/L。
更优选地,所述步骤2)中纤维素酶的加入量为50-100u/g;水浴酶解的温度为40℃。
更优选地,所述步骤3)中过滤所采用的滤网规格为100目。

本发明还提供了上述含魔芋多糖的痤疮微生态失衡改善制剂在化妆品中的应用。
优选地,所述含魔芋多糖的痤疮微生态失衡改善制剂的添加量为化妆品总质量的1-60%;所述化妆品为水剂、精华液、乳液、面霜、面膜或防晒产品。
本发明所述具有调节痤疮患者皮肤微生态失衡的组合物中,所述的魔芋提取物水解液是魔芋精粉(含大量高分子葡苷聚糖)通过酶水解成多种小分子片段的低聚糖水解液,该水解液中包含大量低聚糖对于皮肤微生态具有良好的调节作用,已知魔芋多糖作为潜在益生元对于双歧杆菌、乳酸菌具有很好的促增殖作用,其低聚糖葡萄糖和甘露糖的比例1:1.6对皮肤益生菌非常友好,并且对于抑制痤疮丙酸杆菌以及金黄色葡萄球菌增殖有一定功效。广谱上来说,在皮肤表面可以起到保湿、抗菌、防止紫外线辐射作用。作用于皮肤内层可以促进皮肤微循环,抗氧化延缓皮肤衰老,并可对损失皮肤修复。功效广泛、可以多方面高效能起到调节皮肤微生态平衡,达到护肤美肤的效果。
所述的乳酸杆菌发酵溶胞产物主要成分包含氨基酸、小肽、多糖、B族维生素等有益护肤的成分,可加强角质层代谢,去除细胞自由基,抑制脂质的过氧化,激活机体修复性能,抑制细胞酪氨酸酶活性,具有修复、美白、抗氧化的功能。其中含有乳酸菌素等具有抑制病原菌作用的多肽化合物,是天然的,对皮肤具有镇静作用,同时粘附于表皮细胞,对皮肤形成保护屏障,保护其免受外界环境的攻击。并且乳酸杆菌发酵溶胞产物富含乳酸和乳酸钠,是人体天然保湿因子(NMF)中主要的水溶性酸类,吸湿性强,有效增强角质层的水合作用,防止皮肤水分流失;乳酸和乳酸钠组成缓冲溶液,对皮肤具有良好的亲和性,可调节皮肤酸碱值。乳酸菌发酵溶胞产物使各种介质的比例恢复平衡,利于免疫系统的调控,缓解皮肤炎症。
所述的酵母发酵提取物主要成分为多肽、氨基酸、核苷酸、B族维生素和微量元素等。能抵抗皮肤衰老,对皮肤细胞活性和胶原蛋白合成都具有良好的促进作用。重建皮肤细胞,提升弹力蛋白质和更新真皮层的新陈代谢,有舒缓受损皮肤,有益皮肤整体健康。
所述的马齿苋提取物具有出色的抗菌消炎作用,提取物中的生物碱和黄酮对于人体内常见致病性皮肤真菌都有明显的抑制作用,可以降低毛细血管通透性,减轻炎症和水肿。0.5~5%即可发挥出色的抗炎,抗刺激,抗光毒性的功效。马齿苋提取物中的黄酮和皂甙可以去自由基,起到抗氧化和延缓皮肤衰老的作用。
所述的神经酰胺是存在于皮肤的一种脂肪类的鞘氨醇。在表皮角质层形成过程中发挥着重要作用。
本发明的组合物采用本行业通用方法制成,将各原料加入搅拌混合均匀即可制得该组合物。
本发明的关键点在于通过魔芋多糖具有益生元的潜在特性,从皮肤微生态失衡的角度来舒缓和改善痤疮患者的肌肤问题。魔芋多糖独特的促进皮肤益生菌生长,减少和抑制痤疮丙酸杆菌的增殖的作用,可以调节和改善失衡的皮肤微生物组成,间接促进皮肤屏障恢复。并且通过和假丝酵母发酵产物的促细胞自噬效应的协同,减轻炎症反应,加快痤疮患者恢复健康肌肤。
与现有技术相比,本发明的有益效果在于:本发明通过魔芋提取物、乳酸杆菌发酵溶胞产物、酵母发酵产物以及马齿苋提取物的协同增效,使得本发明所述具有调节痤疮皮肤微生态失衡的组合物具有良好的舒缓和抗痤疮功效,魔芋提取物的多糖成分能够被皮肤菌群利用加速增殖,同时可以选择性的保护益生菌抑制病原菌。含较多的乳酸杆菌发酵溶胞产物,包括乳酸等成分,可以调节皮肤PH,调节皮肤菌群的生存环境,使其更益于有益菌的生长,同时还具有软化角质、光滑肌肤、滋润保湿和提亮美白的功效。通过马齿苋提取物协同使用假丝酵母发酵产物所含有的β-葡聚糖细胞自体吞噬作用,加速痤疮炎症因子的代谢和排除,减轻痤疮肌肤的炎症反应,配合使用神经酰胺提供更好的屏障修复和舒缓功效。
本发明通过魔芋提取物、乳酸杆菌发酵产物、酵母提取物、神经酰胺以及马齿苋提取物的生物协调作用,能够有效的改善痤疮,调节皮肤微生态的菌群,增强肌肤屏障系统,改善痤疮患者肌肤状态,达到保湿、舒缓、祛痘等复合功效,可作为功效成分应用在化妆品当中。本发明具有调节痤疮皮肤微生态失衡的组合物能有效平衡痤疮患者肌肤微生物菌群,还能增强肌肤免疫系统,改善肌肤状态,达到祛痘效果。
具体实施方式
为使本发明更明显易懂,兹以优选实施例,作详细说明如下。
本发明所述具有调节痤疮皮肤微生态失衡的组合物中,各个成分含量均以组合物总质量为100%计。所述具有调节痤疮皮肤微生态失衡的组合物中,各个成分含量均以精华液总质量为100%计。其中,魔芋精粉购于中国BLG公司;乳酸杆菌发酵溶胞产物购于德国CLR公司(即Chemisches Laboratorium Dr.Kurt Richter GmbH);假丝酵母发酵产物购于法国silab公司;马齿苋提取物购于韩国百朗德公司(Bioland);神经酰胺购于北京华工科创生物技术有限公司。其中,乳酸杆菌发酵溶胞产物为发酵乳杆菌的溶胞产物,酵母发酵产物为假丝酵母的发酵产物滤液。
各实施例与对比例中所述魔芋提取物的制备方法为:
步骤1):将魔芋精粉溶解于pH为5~6的缓冲液中;
步骤2):在步骤1)所得的溶液中加入纤维素酶在水浴酶解,得到魔芋多糖的水解液;
步骤3):将步骤2)所得的水解液采用旋转蒸发仪浓缩至原来体积的1/5,然后加入无水乙醇搅拌,过滤,过滤所得固体真空干燥,得到魔芋提取物,即含较多魔芋低聚糖的混合物。
实施例1
本实施例提供的痤疮微生态失衡改善制剂包括魔芋提取物30%、乳酸杆菌发酵溶胞产物30%、酵母发酵产物15%、马齿苋提取物5%、神经酰胺5%和余量去离子水,将上述各成分混合即可。
实施例2
本实施例提供的痤疮微生态失衡改善制剂包括魔芋提取物25%、乳酸杆菌发酵溶胞产物25%、酵母发酵产物5%、马齿苋提取物5%、神经酰胺5%和余量去离子水,将上述各成分混合即可。
实施例3
本实施例提供的痤疮微生态失衡改善制剂包括魔芋提取物10%、乳酸杆菌发酵溶胞产物10%、酵母发酵产物25%、马齿苋提取物15%、神经酰胺5%和余量去离子水,将上述各成分混合即可。
对比例1
本对比例不含马齿苋提取物,仅包括魔芋提取物20%,乳酸杆菌发酵溶胞产物20%,酵母发酵产物20%,神经酰胺5%,其余为去离子水。
将上述各成分混合即可。
对比例2
本对比例不含马齿苋提取物,仅包括魔芋提取物20%,乳酸杆菌发酵溶胞产物20%,马齿苋提取物20%,神经酰胺5%,其余为去离子水。
将上述各成分混合即可。
对比例3
本对比例不含马齿苋提取物,仅包括魔芋提取物20%,酵母发酵产物20%,马齿苋提取物20%,神经酰胺5%,其余为去离子水。
将上述各成分混合即可。
对比例4
本对比例不含马齿苋提取物,仅包括魔芋提取物20%,酵母发酵产物20%,马齿苋提取物20%,神经酰胺5%,其余为去离子水。
将上述各成分混合即可。
对实施例和对比例进行功效测试,方法如下:
(1)竞争生长测试
测试实施例和对比例在益生菌与病原菌同时存在时对菌群生产情况的影响和作用。
分别配置浓度为1wt%的实施例1-3和对比例1-4的实验介质(胰蛋白胨),然后选取菌群在上述介质中培育,所有培养皿在37℃厌氧孵育,24小时计算细菌数目。菌群为戊糖乳杆菌和痤疮丙酸杆菌混合菌群;表皮葡萄球菌和痤疮丙酸杆菌混合菌群,菌株取自志愿者皮肤表面,筛分后进行培养分别移植到实验介质中去。
测试结果见表1。
从上述表1的实验结果可以看出,本发明的实施例1-3制备得到的组合物能够有效的促进皮肤正常菌群戊糖乳杆菌以及表皮葡萄球菌的增殖,对痤疮皮肤的病原菌有抑制作用,尤其是实施例1对增强皮肤微生态有积极的作用。
对比例1-4由于不包含其中一种组合物成分,相较于实施例1-3对于皮肤正常菌群戊糖乳杆菌和表皮葡萄球菌增殖效果并不优越,调节痤疮患者的皮肤微生态较差。这表明本发明物的相互配合,能够有效促进益生菌的增殖抑制病原菌。对比例3由于缺乏魔芋提取物,相较于对比例1、2、4其促进益生菌抑制病原菌效果较差,说明魔芋提取物作为潜在益生元作用,在本发明中起到了一定促进有益菌生长抑制病原菌的作用。
(2)调节痤疮皮肤微生态测试
调节痤疮皮肤微生态测试具体检测方法如下:
选取35名志愿者年龄在18-30岁之间患有相同程度痤疮症状的男性或者女性,将其分为7组,每组5人,分别使用所述具有调节痤疮皮肤微生态失衡的组合物实施例1-3和对比例1-4,使用样品28天,一天早晚二次。使用效果由VISIA-CR皮肤测试仪(美国Canfield公司)在使用前后分别对毛孔、平整度和卟啉进行定量评估。
具体检测结果如下表2-4所示。

由上表可知,实施例1-3和对比例1-4对痤疮患者皮肤毛孔、平整度以及卟啉都有较好的改善效果。其中卟啉作为痤疮杆菌的代谢产物,在紫外灯下可以一定程度的反应痤疮的改善程度以及病原菌的抑制程度。实施例1可以看出其对于卟啉值在使用后的降低是很明显的,具有显著改善痤疮患者皮肤菌群平衡作用。平整度的改善可以看出,该实施例1-3的组合物对修复受损细胞,促进新陈代谢,帮助受损皮肤愈合都有很好的疗效。
(3)皮肤屏障修复测试
保护皮肤屏障通过经皮失水率测试仪(Tewameter TM300;德国CK)检测、经皮水分流失TEWL值指标来测定,通过两组温度、湿度不同的传感器测定表皮由角度层水分分散失形成的不同点的水蒸气压梯度,测量经表皮蒸发的水分量,TEWL值用来评价皮肤屏障功能。
具体检测方法如下:选取20名志愿者,使用清洁产品清洁手臂内侧肌肤,在试验室静坐20min左右。进行皮肤第一次数据采集,然后涂抹样品,在涂抹样品1h、2h和4h采集数据,每个位置测三次取平均值。
具体测试结果如表5、6所示。

从实施例1-3和对比例1-4可以看出选择的组合物对人体含水量和经皮水流失都有所改善,说明本发明的组合物具有改善保湿和修复屏障的效果。并且实验结果表明实施例1的改善效果最佳,总的有效率在95%-100%左右,该组合物中魔芋提取物含量最高。而对比例3的实验结果相较于其他是较差的,对比例3组合物中不含魔芋提取物。可以得出魔芋提取物在人体保湿和皮肤屏障修复方面有显著效果。
Claims (10)
1.一种含魔芋多糖的痤疮微生态失衡改善制剂,其特征在于,包括魔芋提取物、乳酸杆菌发酵产物、酵母提取物、神经酰胺及马齿苋提取物。
2.如权利要求1所述的含魔芋多糖的痤疮微生态失衡改善制剂,其特征在于,所述魔芋提取物的制备方法为:
步骤1):将魔芋精粉溶解于pH为5~6的缓冲液中;
步骤2):在步骤1)所得的溶液中加入纤维素酶在水浴酶解,得到魔芋多糖的水解液;
步骤3):将步骤2)所得的水解液采用旋转蒸发仪浓缩至原来体积的1/5,然后加入无水乙醇搅拌,过滤,过滤所得固体真空干燥,得到魔芋提取物,即含较多魔芋低聚糖的混合物。
3.如权利要求2所述的含魔芋多糖的痤疮微生态失衡改善制剂,其特征在于,所述步骤1)所得溶液的浓度为5-20g/L。
4.如权利要求2所述的含魔芋多糖的痤疮微生态失衡改善制剂,其特征在于,所述步骤2)中纤维素酶的加入量为50-100u/g;水浴酶解的温度为40℃。
5.如权利要求2所述的含魔芋多糖的痤疮微生态失衡改善制剂,其特征在于,所述步骤3)中过滤所采用的滤网规格为100目。
9.权利要求1-8任意一项所述的含魔芋多糖的痤疮微生态失衡改善制剂在化妆品中的应用。
10.如权利要求9所述的应用,其特征在于,所述含魔芋多糖的痤疮微生态失衡改善制剂的添加量为化妆品总质量的1-60%;所述化妆品为水剂、精华液、乳液、面霜、面膜或防晒产品。
